Verfahren zur Herstellung von 5-Amino-uraeil-sulfosäure-(4) und deren N-substituierten Derivaten Die vorliegende Erfindung betrifft ein Ver- fahren zur Herstellung von 5-Aniino-uracil- sulfosäure-(4) und deren N-substituierten Derivaten.
Es wurde gefunden, dass bei der Reduktion von 5-Nitro-uracil mit Dithionit (pIypodisnlfit) in stark alkalischer Lösung eine neue Verbin dung, die 5-Amino-uracil-sulfosäure-(4), in fast quantitativer Ausbeute gebildet wird. Die Um setzung erfolgt nach erfolgtem Schema:

Die Durchführung des erfindungsgemässen Verfahrens erfolgt in der Weise, dass die Re aktionslösung ständig ätzalkalische Reaktion aufweist. Das Dithionit wird zweckmässig por- tionenweise zugegeben.
Die Umsetzung erfolgt vorteilhaft. bei einer Temperatur von etwa 10-90 , vorzugsweise bei 30-35 . Die Umset- zung kann mit allen wasserlöslichen Dithio- niten, wie Natriumdithionit, Kalitundithionit, C'alciumdithionit usw., durchgeführt werden.
Als Ausgangsmaterial für die Reduktion kann man auch das rohe Kaliiunsalz des 5- Nitro-uracils verwenden, wie es nach der De- eai-bo .plici-ting des nitro-uracil-carbonsauren Kaliums anfällt. Die an den beiden Stickstoff- atomen des Nitro-uracilringes- befindlichen Wasserstoffatome können auch substituiert sein, z. B. durch Alkyl-, Aryl- oder Aralkyl- reste.
Die erfindungsgemäss hergestellte 5-Amino- uracil-siilfosäure-(4) ist ein wichtiges Zwi schenprodukt für weitere Synthesen.
Das folgende Beispiel soll die Erfindung näher erläutern, ohne sie zu beschränken. <I>Beispiel</I> 5-A mino-urucil-sul <I>f</I> osäure-(4) 0,2 Mol (31,4 g) 5-Nitro-uracil werden mit 200 cm3 3n-Natronlauge übergossen und ver rührt.
Zu dem entstandenen Brei des Di- natriiunsalzes werden unter kräftigem Uüh- ren abwechselnd Natriumdithionit in kleinen Mengen und 3n-Natronlauge in der Weise zu gegeben, dass die Reaktion der sich alsbald bil denden Lösung stets ätzalkaliseh bleibt und das Dithionit vor Zugabe weiterer Mengen verbraucht ist. Dies kann z. B. durch Tupfen auf Methylenblaupapier geprüft werden.
Die Temperatur der Reaktionsmischung 'wird durch Aussenkiihlumg bei etwa 30 gehalten. Es werden insgesamt etwa 95-100g tech nisches, wasserfreies Natriumdithionit und noch weitere 200 cm3 3n-Natronlauge ver braucht. Bei Beendigung der Umsetzung soll die Lösung noch ätzalkalisch reagieren und etwas unverbrauchtes Dithionit enthalten. Die Reaktionsmischung wird dann auf 50 er wärmt und durch ein Filter gesaugt.
Dies geschieht zur Entfernung von evtl. Sandkör nern aus dem Dithionit und von etwaigen dunkelbraunen Flocken, welche sich bei Ver wendung von unreinem 5-Nitro-uracil bilden. Nachher wird mit Eisessig neutralisiert, wobei sich das primäre Natritunsalz der 5-Amino- uracil-sulfosäure-(4) sofort in derben, pris matischen, flachen Nadeln abscheidet. Nach dem Abkühlen ist die Abscheidung nahezu vollständig.
Das gebildete Salz wird nach lutrzem Stehenlassen von der Mutterlauge scharf abgesaugt und ohne weitere Reinigung durch 5n-Salzsäure in die freie Sulfosäure übergeführt.
Diese fällt in groben, fast farblosen Kri stallkörnern aus. Die Ausbeute beträgt 36 bis 38 g, i. e. 85-90% der theoretischen Ausbeute.
Man kann für die Umsetzung auch das rohe Kalittmsalz des 5-Nitro-uracils verwen den, wie es nach der Decarboxylierumg des nitro-tuaeil-earbonsaturen Kaliums (2 Stunden bei 160 ) anfällt. In diesem Fall wird 1 Äqui valent Lauge weniger gebraucht. Die Sutlfo- säure wird in gleicher Ausbeute erhalten, je doch hellbraun verfärbt.
Die Sehwerlösliehkeit des oben erhaltenen primären Natrittmsalzes der 5-Amino-urae.il- stüfosäure-(4) wird durch die Anwesenheit der andern Natriumsalze in nahezu gesättigter Lösung verursacht.
Wird das Salz in reinem MTasser gelöst, so ist es in der Hitze leicht und auch in der Kälte noch zieiulich löslich. Die Lösung rea giert neutral. Das Salz kristallisiert daraus in feinen, meist sternförmig verwachsenen Nadeln oder gröberen Parallelogrammen. Diese enthalten 2 Mol Kristallwasser, das im Va- ktum schon bei Raumtemperatur unter Ver witterung abgegeben wird.
Analyse: C4II105N3SNa - 21120 (229,2) berechnet: 1I20 13,58% gefunden: 1r20 13,14% Die 5-Amino-uracil-sulfosätire-(4) kann leicht durch Umkristallisieren aus Wasser unter Zuhilfenahme von Tierkohle oder durch Umfällung mit Ammoniak /Salzsäure gereinigt. werden. Sie scheidet sich in kristallwasser- freien rhombischen Blättchen ab. Die Löslich keit in Wasser beträgt bei 20 0,355 g / 100 em3 (Quarzgefäss, bidestilliertes Wasser, Thermo stat).
Die Säure schmilzt nicht, sondern zer setzt sich, rasch erhitzt, unter Aufblähung bei 280 (Kupferblock auf 270 vorerwärmt).

It was found that when 5-nitro-uracil is reduced with dithionite (pIypodisnlfit) in a strongly alkaline solution, a new compound, 5-amino-uracil-sulfonic acid- (4), is formed in almost quantitative yield. The implementation is based on the following scheme:

The method according to the invention is carried out in such a way that the reaction solution constantly exhibits an alkaline-caustic reaction. The dithionite is expediently added in portions.

Afterwards it is neutralized with glacial acetic acid, whereby the primary sodium salt of 5-aminouracil-sulfonic acid (4) is immediately deposited in coarse, prismatic, flat needles. After cooling, the deposition is almost complete.
After standing for a long time, the salt formed is sharply suctioned off by the mother liquor and converted into the free sulfonic acid with 5N hydrochloric acid without further purification.
This precipitates in coarse, almost colorless crystal grains. The yield is 36 to 38 g, i. e. 85-90% of the theoretical yield.
You can also use the crude potassium salt of 5-nitro-uracil for the reaction, as is obtained after decarboxylation of the nitro-tuaeil-carbonate potassium (2 hours at 160).
