Verfahren zur Herstellung von Folsäure. Die Patentinhaberin hat eine Methode zur Herstellung von N-[(2-Amino-2-oxy-6-pteri- clyl) -methyl] -N- (R-S02) -p-amino-benzoe- säure-Verbindungen der folgenden Formel entwickelt:

worin R-S02 einen organischen Sulfonyl- rest, R' Wasserstoff oder Alkylreste und n die Zahl 0 oder eine ganze Zahl von 1-7 bedeu tet.
Unter diese Formel fallen somit zum Bei spiel. N-Substitutionsprodukte von p-Amino- benzoesäure, p-Amino-benzoylglutaminsäure, p - Amino - benzoyl - glutamyl- ghltaminsäuren, die bis zu 7 Ghitaminsäurereste im Molekül enthalten, sowie deren Ester.
Bei der Bezeich nung der Verbindungen der obigen allgemei nen Formel, die sowohl einen Glutaminsäure- rest als auch einen p-Amino-benzoesäurerest enthalten, wird das Stickstoffatom des Glut- aminsäurerestes mit dem Symbol N' und das Stickstoffatom des p-Amino-benzoesäure- restes mit dem Symbol N bezeichnet.
Die Verbindungen der obigen Formel sind von besonderem Wert, weil sie mit den Ver bindungen der Polsäuregruppen verwandt, sind bzw. sich leicht in solche überführen las sen. So lässt sich der Diäthylester der N'- [N- (2-Amino-4- oxy - 6 -ptelidyl-methyl)- N- (p-to- luol-sulfonyl) -p -amino -benzoyl] -1-ghttamin- säure leicht durch Hydrolyse mittels Alkali in die entsprechende Dicarboxyverbindiuig, überführen,
und letztere kann dann leicht. durch Abspalten des p-Toluolsulfonylrestes mittels Bromwasserstoff und eines Bromäceep- tors in Gegenwart einer aliphatisehen Car- bonsäure in eine Pteroyl-glutaminsäüre über-, geführt werden, welche identisch zu sein scheint mit dein L. Gasei factor bzw. Vit amin B, (Folrsäure) aus Leber.
Die Hydro lyse und die Abspaltung des Arylsulfonyl- restes kann gewünschtenfalls auch in umge kehrter Reihenfolge erfolgen.
Verbindungen der obigen Formel, in denen die Glutaminsäurereste die l(+)-Konfigura- tion besitzen, bieten ein besonderes Interesse, weil dies die Konfiguration des in den natür lichen Polsäureverbindungen enthaltenen Glutaminsäurerestes ist.

Wie aus dein Formelschema ersichtlich ist, erhält man durch Kondensation von N-(2- Formy l - 2 - keto - äthyl) - N - (ary 1 sulfonyl) - p- amino-benzoyl-Verbind-tingen (III) mit 2,4,5 Triamino-6-oxy-pyrimidin (VII) die N-[(2- Amino -4-oxy-6-pteridyl) -methyl] -N- (arylsul- fonyl)

Man kann zum Beispiel die :leetalgruppe von N-(3,3-Dialkoxy-2-keto-pro- py 1)-N -(arvlsulfonyl)- p - amino- benzoyl-Ver- bindungen (IV) mit einer wässerigen Mineral säure hy drolysieren. Ferner können die Ver bindungen (III) auch durch Oxydation von N- (3-Oxy-2 -keto -propyl) -N- (arylsulfonyl) -p- amino-benzoyl-Verbindungen (V)
beispiels weise mittels Chromi- oder Cupriacetat in einem. wässerigen Alkanol erhalten werden., Eine weitere Methode zur Herstellung der Verbindungen (III) besteht darin, dass N-(3 3Iethoxy-2-keto-propyl)-N-(arylsulfonyl)-p- amino-benzoyl-Verbindiuigen (VI) zum Bei spiel mittels Chromiacetat in wässerigem Me dium oxydiert werden.
Schliesslich kann man auch N-(2-Formyl-2-oxy-äthyl)-N-(arylsulfo- nyl)-p-amino-benzoyl-Verbindungen (II) bei spielsweise mit Chromi- oder Cupriacetat in alkoholischem oder wässerig-alkoholischem Medium oxydieren.
Die N- (3,3-Dialkoxy-2-keto-propyl) -N- (arylsulfonyl) -p-amino-benzoyl-Verbindungen (IV) können durch Erhitzen von 2,3-Oxido- propanal-dialkyl-acetalen der Formel:

Gegenstand der vorliegenden Erfindung ist nun ein Verfahren zur Herstellung von Folsäure. Dieses Verfahren ist dadurch ge kennzeichnet, dass man einen Ester einer sub stituierten 1-Gliitaminsäure der Formel:

in welcher R-SO2- einen durch Behand lung mit Halogenwasserstoff abspaltbaren or ganischen Sulfonylrest bedeutet, mit 2,4,
5- Triamino-6-oxy-pyrimidin kondensiert und dass man alsdann das Kondensationsprodukt einerseits durch Abspaltung - des Restes R-80.- mittels Halogenwasserstoff und an- derseits durch Hydrolyse der veresterten Carboxylgruppe bzw. Carboxylgruppen in Folsäure überführt.
Als Ausgangsstoffe kommen vor allem die Alkylester der genannten Säure in Betracht, beispielsweise die Methyl-, Äthyl-, n-Propyl-, Iso-propy 1-, Butyl-, Amy1-, Hexyl-, Nonyl- ester. Vorzugsweise werden Alkylester ver wendet, welche weniger als etwa 8 Kahlen stoffatome in den Alkylesterresten enthalten, obzwar man auch ändere Alkylester verwen den kann.
Die Gruppe R-S02-- ist vorzugsweise ein Ary lsulfonylrest, am besten der p-Toluolsul- fonylrest, weil die p-Toluolsulfonylhalogenide leicht zugänglich sind und die p-Toluolsulfo- nylderiv ate der in Frage stehenden Verbin dungen im allgemeinen in kristalliner Form anfallen. Ferner lässt sieh der p-Toluolsulfo- nylrest mit. Bromwasserstoff leichter und un ter Bildung kleinerer Mengen unerwünschter Nebenprodukte abspalten, als dies der Fall ist.
bei manchen andern Ary lsulfonylresten. Im merhin werden auch mit dem o-Toluolsulfo- nyl-, den Naphthalinsulfonyl-, Methylnaph- thalinsrdfonylresten usw., und ferner mit Al- kyjsulfony-1-, Aralkylsulfonyl- oder Cyclo- alkylsiilfonylresten, z.
B. Methansulfonyl-, ,L@tliansulfonyl-, Cyelohexylsulfonyl- und Phe- nylmethansulfonylresten, befriedigende Resul tate erzielt.
Die Arylsulfonylreste können allenfalls auch Substituenten enthalten, welche unter den Reaktionsbedingungen nicht reagieren, z. B. Chlor, Brom, ferner Methoxy-, Phenoxy-, Nitro- und ähnliche Reste.
Die als Ausgangsstoffe in Betracht fallen den Ketoaldehyde können durch Oxydation der entsprechenden Oxyaldehyde erhalten werden, welche ihrerseits durch Hydrolyse aus den entsprechenden Dialkylacetalen her gestellt werden können. Diese Hydrolyse wird zweckmässig mit einer wässerigen Mineral säure, vorzugsweise bei erhöhter Temperatur, durchgeführt. Es kommen verschiedene Di- alkylacetale in Betracht.
Aus praktischen Er wägungen heraus verwendet man jedoch vor zugsweise solche Verbindungen, deren Alkyl- reste der Dialkylacetalgruppe je weniger als 8 Kohlenstoffatome enthalten. Die Umsetzung erfolgt rasch bei oder unterhalb der Rück- flusstemper atur des Gemisches und ist im all gemeinen innerhalb von wenigen Minuten bis zu einer Stunde oder mehr -beendet. Zufrie denstellende Resultate werden dadurch er- zielt, dass das Gemisch während etwa. 15 Mi nuten auf ungefähr 60 C erhitzt wird.
Man kann gewünsehtenfalls auch wässerige, alko holische Mineralsäure verwenden und durch die grössere Löslichkeit der organischen Ver bindung in alkoholischer Mineralsäure die Umsetzung beschleunigen. Nach dem Erwär men kann das Wasser und die überschüssige Säure zusammen mit etwa, im Gemisch vor handenem Alkohol im Vakuum abdestilliert werden.
Der Rückstand, welcher aus der N- (2-For myl-2-oxy-- äthy 1) - N-a rylsulfonyl) -p- amino-benzoy 1-Verbindung besteht, ist für die meisten Zwecke hinreichend rein. Durch vor sichtige Verseifung und Ansäuern können sol ehe Produkte zu den entsprechenden Säuren hydrolysiert werden.
Die Oxydation des Oxyaldehyds zum Keto- aldehyd kann durch Erhitzen in alkoholischem oder wässerig-alkoholisehem Medium mittels Cupriacetat oder Chromiacet.at erfolgen. Die Oxydation ist im allgemeinen praktisch nach etwa einer halben Stunde oder weniger be endet, sofern das Gemisch auf etwa 50 bis 80 C erhitzt wird. Wird die Oxydation mit tels Cupriacetat durchgeführt, so bildet. sich ein Niederschlag von Kupferoxyd, welcher leicht. durch Filtrieren entfernt. werden kann.
Der Alkohol bzw. der wässerige Alkohol. kann dann im Vakuum. abdestilliert werden, wobei die N-(2-Formyl-2-keto-äthyl)-N-(arylsulfo- nyl)-p-aminobenzoyl-Verbindung im allgemei nen in Form eines viskosen, gelblichen Sirups anfällt.
Wird Chromiacetat als Oxydationsmittel verwendet, so kann der Ketoaldehyd aus dein Gemisch gewonnen werden, indem das Lö sungsmittel abgedampft und der Rückstand so lange mit Wasser gewaschen wird, bis in der Waschlösung die grüne Farbe der Chrom salze verschwunden ist. Gewünschtenfalls kann der Rückstand vor dem Waschen zur Erleichterung des Beseitigens der Chromsalze in einem organischem Lösungsmittel gelöst werden.
Die N- (2-Formiyl-2-keto-ätKyl)-N- (R-S02) - p - amino - benzoy 1- Verbindungen können auch durch Oxydation aus N-(3-Oxy- 9-keto-propy 1) - N - (arylsulfonyl) -p-amino-ben- zoyl-Verbindungen hergestellt werden, und zwar ebenfalls in alkoholischem oder wässerig alkoholischem Medium unter Verwendung von Cupriacetat oder Chromacetat praktisch in gleicher Weise,
wie dies oben für die Oxyda- 1 ion der Hy droxy aldehydverbindungen be schrieben wurde. Das Gemisch wird im all gemeinen während 5 bis 30 Minuten auf eine Temperatur von etwa 40 bis 80 C erhitzt und der Ketoaldehyd hierauf aus dem Gemisch in der oben beschriebenen Weise gewonnen.
Schliesslich kann man die Ketoaldehyde in analoger Weise auch durch Oxydation von N - (3 - Methoxy - 2 - keto - propyl) -N- (arylsulfo- iil)-p-amino-benzoyl-Verbindungen gewin nen. Immerhin ist es in diesem Fall im all gemeinen nötig, das wässerig-alkoholische Re aktionsgemisch während einer bis mehreren Munden unter Rückfluss zu erhitzen, um prak- t iscli vollständige Oxydation zu bewirken.
N -(3,3-Dialkoxy-2-keto-propyl)-N-(aryl- sulfonyl)-p-amino-benzoyl-Verbindungen las sen sich leicht zu den N-(2-Formyl-2-keto- äthyl )-N- (arylsulfonyl)- p-amino-benzoyl-Ver-. Bindungen hydrolysieren, indem das Ketoace- tal mit wässeriger oder wässerig-alkoholischer Mineralsäure, vorzugsweise während etwa 5 bis 30 Minuten, bei einer Temperatur von un- gefähr 30 bis 8Ö C, erwärmt wird.
Das Keto- aldehyd kann aus demReaktionsgemischdurch Wegdampfen des Wassers und des Alkohols und durch Waschen des öligen Rückstandes mittels Wassers, bis er säurefrei ist, gewon nen werden.
Die Kondensation der N'- [N- (2-Formyl-2- keto-äthy 1)-N-(arylsulfonyl)-p-ainino-benzoyl] - 1-gIutaminsäureester mit dem 2,4,5-Triamino- 6-oxy-pyrimidin unter Bildung der entspre chenden N-[(2-Amino-4-oxy-6-pteridyl)-me- thyl 1 -N - (arylsulfonyl) -p - amino-benzoyl-Ver- bindungen lässt sich leicht durch Vermischen der Substanzen, vorzugsweise zusammen mit wasserfreiem Natriumacetat,
in Eisessiglösung und durch Stehenlassen des Gemisches wäh rend einer bis mehreren Stunden bei Zimmer temperatur durchführen. Das 2,4,5-Triamino- 6-oxy-pyrimidin wird gewöhnlich in Form seines Dihydrochlorids, welches sich leichter handhaben lässt, verwendet. Das Gemisch wird vorzugsweise unter Stickstoffatmosphäre oder in Gegenwart eines andern inerten Gases gerührt. Die Reaktion verläuft in befriedi gender Weise unter Bildung der gewünschten Produkte in verhältnismässig reichlicher Aus beute.
Nach der Umsetzung in Essigsäure kann das Produkt dadurch isoliert werden, dass die Essigsäure im Vakuum abdestilliert und der Rückstand mit Wasser gewaschen wird, wo durch Spuren von Essigsäure und anorgani schen Salzen entfernt werden. Auf diese Weise werden die N'-[N-(2-Amin.o-4-oxy-6-pteridyl- inethyl) -N- (arylsulfonyl) -p-amino -benzoyl] -1- glutaminsäureester in Form stark gefärbter Rückstände erhalten.
Beispiel: Man löst 330 mg rohen N'- [N- (2-Formyl-2 keto-äthyl)-N-(p-toluolsulfonyl)-p-amino-ben- zoyl]-1-glutaminsäure-diäthylester in 11 em3 Eisessig und vermischt die Lösung mit 107 mg 2,4, 5 -Triamino - 6 - oxy-pyrimidin-dihydrochlo- rid und 82 mg Natriumacetat. Das Gemisch wird bei Raumtemperatur unter einer Stick stoffatmosphäre während etwa 90 Minuten gerührt und dann im Vakuum konzentriert, um die Essigsäure grösstenteils zu entfernen.
Durch Waschen des Rückstandes mit Wasser erhält man ein Produkt, welches zur Haupt sache aus N'-[N-(2-Amino-4-oxy-6-pteridyl- methyl)-N- (p - toluolsulfonyl) -p- amino-ben- zoyl]-1-glutaminsäure-diäthylester besteht.
Das den N'-[N-(2-Amino-4-oxy-6-pteridyl- methyl)-N-(p-toluolsulfbnyl) -p-amino-ben- zoyl]-1-glutaminsäure-diäthylester enthaltende rohe Reaktionsprodukt wird mit 130 mg Phe nol und 2,5 em3 einer 26 % igen Lösung von Bromwasserstoff in Eisessig vermischt. Das Gemisch wird während 90 Minuten bei Raum temperatur gerührt und dann mit wasser freiem Äther verdünnt. Es scheidet sich Pte- royl-ghitaminsäure-äthylester-hydrobromid in Form eines braunen, hygroskopischen, festen Körpers aus dem Gemisch ab.
Der braune Körper wird durch Filtrieren isoliert und in etwa 2 ein?, Methanol gelöst, worauf die Lö sung durch Zugabe von 10 /o igem wässerigem Natriumhydroxyd stark alkalisch gestellt wird. Das alkalische Gemisch wird während 90 Minuten bei 40 C stehengelassen und dann mittels Mineralsäure auf ein pH von 3,0 angesäuert. Hierauf wird die Lösung mit 5 em3 Wasser verdünnt, während einer Stunde in einem Eisbad gekühlt und dann zentrifu giert.
Der feste Rückstand wird zuerst mit 3 em3 Wasser und dann dreimal mit 3 cm- Aceton gewaschen und hierauf im Vakuum über Schwefelsäure getrocknet. Man erhält auf diese Weise 89 mg Pteroyl-glutaminsäure, welche bei der mikrobiologischen Prüfung mit Lactobacillus easei eine Wirksamkeit von 48,8%, bezogen auf diejenige der reinen na- türlichen Säure, aufweist.
Der als Ausgangsmaterial verwendete N'- [N- (2-Formyl-2 - keto - äthyl) -N- (p-toluolsulfo ni-1)-p-amino-benzoyl]-1-glut.aminsäure-diäthyl- ester kann wie folgt erhalten werden:
Eine Lösung von 300 mg kristallisiertem N'- [N-(3-.#lethoxy-2 -keto-propyl) -N-(p-toluol- sulfonyl)-p-amino -benzoyl] -1-glutaminsäure- diäthylester in 25 em3 absolutem Äthanol, wel che eine katalytische Menge p-Toluolsulfon- säure und eine Menge Chromiacetat, die ge nügt, um der Lösung dauernd eine grüne Farbe zu verleihen, enthält, wird während drei Stun den unter Rückfluss sieden gelassen.
Hierauf dampft man die Lösung zur Entfernung des Äthanols ein und extrahiert den Rückstand mit Äthylessigester. Der Extrakt, wird mit, Wasser gewaschen und getrocknet, worauf der @lthylessigester im Vakuum abgedampft wird. Es bleibt ein Rückstand von 310 mg zurück.
