On entend ici par ac@-le des radicaux acyle d'acides carboxy liques tels qu'un radical acyle aliphatique inférieur, acyle aliphatique halogéné inTé- rieur, acyle aliphatique inférieur non saturé, acyle aliphatique inférieur substitué au moyen d'un groupe éther, benzoyle, benzoy le halogéné, benzoyle nitré, benzoy le alcoylé, acyle araliphat.ique, etc.
Les composés dont la préparation est. envisagée par l'invention sont des substances connues ayant des pro priétés antibiotiques ou servant de produits intermédiaires pour la préparation d'antibio tiques.
Les matières premières et les produits finals obtenus selon l'invention peuvent exis- ler sous forme d'isomères de structure (diastéréoisomères) et sous forme d'isomères optiques. La présente invention se rapporte à la préparation de composés ayant la forme diastéréoisomérique dite trans , pseudo (<B>!U</B>) ou thréo , en opposition à la forme diastéréoisomérique dite eus , rég.
ou érythro . Les groupes fixés sur les deux atomes de carbone asymétriques de ces diastéréoisomères trans possèdent la même configuration ou même arrangement spatial que les groupes fixés sur les deux atomes de carbone asymétriques dans la pseudo-éphé- drine et dans la thréose.
En raison de la difficulté à représenter graphiquement ces différences de structure, on utilisera les formules développées habi tuelles avec une annotation placée au-dessous ou à côté de la formule pour indiquer la structure particulière ou la configuration optique particulière du composé. Quand on indique forme trans , comme ci-dessus, on doit prendre la- formule dans son sens géné rique, c'est-à-dire comme représentant les iso mères (1)-trans et (d)-trans aussi bien: sous leur forme optiquement active individuelle que sous forme de leur mélange racémique.
Une formule de ce type ne représente donc; pas simplement. le mélange racémique optique.
Le présent brevet a pour objet un nou veau procédé de préparation de l'un de ces composés 1-nitrophényl2-amino-propanediol- 1,3, c'est-à-dire du trans-l-p-nitrophényl-2- dichloracétamido-propanediol-1,3, dont la forme optiquement active 1 est bien connue sous le nom de chloramphénicol . Toutes les formes optiques (d, 1 et dl) de ce composé sont connues et utilisées en thérapeutique; c'est surtout les formes 1 et dl qui présentent un fort pouvoir antibiotique (chloramphénicol et chloramphénicol racémique).

Ce procédé offre un avantage commercial par rapport aux procédés antérieurs qui con sistaient à protéger les groupes hydroxyle du diol de départ par acylation individuelle de ces groupes à l'aide d'un halogénure ou de l'anhydride d'un acide carboxylique tel que l'acide acétique, avant de soumettre le com posé O,O,N-triacylé à la. nitration.
Dans le procédé qui fait l'objet du présent brevet, on utilise comme halogénure d'acide soit un dihalogénure de l'acide carbonique, où X est un atome de carbone, soit un halogénure de thionyle (dihalogénure de l'acide sulfu reux), où X est. un -atome de soufre. Comme composé hétérocyclique intermédiaire, on obtient donc soit un carbonate cyclique, soit un sulfite cyclique du 1-phényl-?-diehloraeét- amido-propanediol-1,3, lequel est. ensuite sou mis à la nitration.

La réaction entre l'Halogénure d'acide et le trans-1-phényl-2-dichloracétamido-propane- diol-1,3 s'effectue d'habitude à une tempéra ture inférieure à 50 C. Lorsqu'on utilise un halogénure de thionyle, on obtient les meil leurs résultats à une température comprise entre 20 et 35 C environ, alors que la tempé rature optimum pour la réaction avec les halo- génures de carbonyle est inférieure à 20 C environ, et comprise, de préférence, entre -10 et. -f- 10 C.
Les quantités relatives des réactifs peuvent varier entre des limites assez larges. Toutefois, pour des raisons économi ques ainsi que pour la facilité de purification des produits, on utilise généralement un excès d'halogénure. Dans le cas des halo-énures de thionyle, une des manières les plus\ commodes d'effectuer la. réaction consiste à utiliser assez d'halogénure de thionyle pour qu'il fonctionne en tant que milieu réactionnel ou solvant.
A titre d'autres solvants pouvant. être utilisés pour la réaction entre l'halogénure d'acide et, le trans-1-phényl-2-dichloracét.amido-propane- diol-1,3, on peut citer des solvants organiques inertes tels que le chloroforme, le dichlorure de méthylène, etc.
Dans le cas des halogénures de carbonyle, le solvant, préféré pour la réac tion est une amine organique tertiaire telle que la py ridine, la. N-éthy lpipéridine, la diméthylaniline, la N-méthy lniorpholine, la triéthylamine, etc.
La réaction avec les halog-éiiures cle carbo nyle, c'est-à-dire dans le cas où X est un atome de carbone, peut également être effectuée en présence d'eau. Le mode préféré consiste toute- fois à effectuer la réaction dans des condi tions anhydres, comme il est décrit ci-dessus.
Quand on opère en présence d'eau, on utilise un mélange réactionnel à deux phases, dontl'urne est, formée par un des solvants organiques inertes susmentionnés et. l'autre est. une phase aqueuse contenant une matière faiblement alcaline telle que le carbonate de calcium, l'hydroxyde de calcium, le carbonate de magnésium, le bicarbonate de sodium, etc., propre à neutraliser l'acide halohydrique formé ali cours de la réaction.
La nitration du noyau phénylique du com posé hétérocyclique peut être effectuée au moyen d'acide nitrique fumant, d'acide nitri- que à. 100% ou d'un mélange d'acide nitri- que concentré et d'acide sulfurique, le groupe nitro entrant.

Le composé cyclique nitré obtenu est transformé par hydrolyse modérée en le t,rans-1-p-nitrophényl-2.dicbloracétamido-pro- panediol-1,3. D'ordinaire, on effectue cette hydrolyse modérée ali sein d'un mélange formé d'eau et d'un solvant organique mis cible à. l'eau tel que le méthanol, l'éthanol, l'isopropanol, l'acétone, la métliy-létliyl-cétoiie, etc., en présence d'un agent hydrolysant acide ou alcalin.
On peut citer à titre d'agents hydrolysants, les hydroxydes, des métaux alcalins comme l'hydroxyde de sodium, l'hydroxyde de potassium, les hydroxydes d'ammonium quaternaires tels que l'hydroxyde de triméthylammonium et l'hydroxyde de triéthy lammonium, et les acides minéraux tels due l'acide chlorhydrique, l'acide bromhy dri- que et l'acide sulfurique.
On maintient la tem pérature au cours de 1'11y drolyse généralement au-dessous de 30 C environ, bien que, si on le -eut, on puisse chauffer le mélange réaction nel à l'ébullition pendant. quelques instants, har exemple 10 minutes ou moins. Un chauf fage prolongé provoquerait, la, transformation du t.ra.ns-1-p-nitrophényl-2-dichlora.c6tamido- propanediol-1,3 désiré en trans-1 -p-nitro- phényl-2-amino-propanediol-1,3, ce qui doit être évité.
D'une manière générale, l'hydro- h-se désirée se produit très rapidement et est habituellement terminée en quelques minutes à une demi-heure, à des températures com prises entre 20 et 50 C. Lors de l'hydrolyse., il. s'effectue une scission du noyau hétéro- cyclique entre l'atome X et les deux atomes d'oxygène voisins.
Les exemples suivants illustrent le pro cédé qui fait. l'objet de l'invention: Exemple .'1: On ajoute par petites portions 10 g de (dl) -trans-1 - phényl- 2-dichloracétamido - pro- panediol 1,3, en agitant, à 25 cm3 de chlorure de thionyle. On abandonne le mélange réac tionnel au repos à la température ambiante pendant une heure,
puis on ajoute de l'éther pour précipiter le sulfite cyclique intermé- ctiaire du (dl)-trans-1-phényl-2-dichloracét- aniido-propanediol-1,3; point de fusion: 118 à 119 C. Ce .composé ,a pour formule:

On ajoute par petites portions, en agi tant., 10 g de ce sulfite cyclique à 50 cm3 d'acide nitrique à.100 0/0, refroidi vers - 30 C par addition d'anhydride carbonique solide.
L'addition terminée, on verse le mélange réac tionnel sur de la glace pilée et on recueille le solide blanc qui se sépare. Ce composé est le sulfite cyclique du (dl)-trans-1-p-nitro- phényl - 2 - dichloracétamido - propanediol -1,3 qui, si on le veut., peut être purifié par recris tallisation au sein d'ét.hanol; point de fusion 175 à 176 C.
On obtient des résultats similaires en rem- plaçant l'acide nitrique à 100% utilisé comme décrit ci-dessus par -de l'acide nitrique fumant ou un mélange d'acides nitrique et sulfurique concentrés.
On dissout 2 g de ce sulfite .cyclique du (dl) -trans-1-p-nitx ophényl-2-dichloracétamido- propanediol1,3 -dans 50 cm3 d'éthanol tiède. On ajoute 5 cm3 d'acide chlorhydrique con centré et on fait. bouillir le mélange pendant deux à trois minutes. On refroidit. le mélange réactionnel, on le neutralise au moyen d'une solution aqueuse à 10%, d'hydroxyde d e sodium et on l'évapore au bain-marie pour chasser l'éthanol.
On épuise le résidu aqueux au moyen d'acétate d'éthyle, on chasse l'acé- tat.e d'éthyle des extraits .et on reprend le résidu au moyen d'eau au sein de laquelle on le fait recristalliser. Le produit cristallisé blanc ainsi obtenu est le (dl)-trans-1-p-nitro- phényl-2-dichloracétamido-propanediol-1,3, de point de fusion 150 C.
On peut également effectuer de la manière suivante l'hydrolyse @du sulfite cyclique :du (dl) -trans-1-p-nitrophényl-2-dicbdoracétamido- propanediol-1,3 a) On dissout 3,7 g dudit sulfite cyclique dans 300 cm-3 d'éthanol, on .ajoute 225 crus d'une solution d'hydroxyde de sodium N/10 et on abandonne le mélange au repos à la tem pérature ambiante pendant une demi-heure. On neutralise le mélange réactionnel au moyen d'acide chlorhydrique étendu et on distille l'éthanol sous vide.
On épuise le résidu aqueux au moyen d'acétate d'éthyle, on mélange les extraits d'acétate d'éthyle et on distille l'acé tate d'éthyle. La recristallisation du résidu au sein de dichlorure d'éthylène donne le (dl) -trans-1-p-nitrophényl-2-dichlor acétamido- propanediol-1,3 à l'état pur. Point -de fusion 150 à 151 C.
b) On dissout 2 g du sulfite cyclique de (dl) -tr ans-1-p-nitrophényl-2-dichloracétamido- propanediol-1,3 dans 65 em3 d'éthanol.
On ajoute 5 cm3 d'acide chlorhydrique concentré et on abandonne le mélange au repos à la température ambiante pendant vingt minutes. On neutralise le mélange réactionnel au moyen d'une solution aqueuse à 10% d'hydroxyde de sodium et on chasse l'éthanol par distilla- tion sous vide. On épuise le résidu aqueux au moyen d'acétate d'éthyle, on mélange les extraits et on distille l'acétate d'éthyle.
La cristallisation du résidu donne le (dl)-trans- 1- p-nitrophényl- 2-dichloracétamido - propane- diol-1,3; point de fusion 150 à 151 C. <I>Exemple 2:</I> On ajoute lentement en l'espace de deux heures 7 g de phosgène à 9,5 g de (dl) -trans- 1-phényl..2-dichloracétamido-propanediol-1,3 dans 50 em3 de pyridine sèche, en maintenant la température .au voisinage de 0 à 5 C.
On verse le mélange réactionnel sur de la glace et on recueille le carbonate cyclique solide blanc du (dl)-tra.ns-1-phényl-2-dichloraeét- amido-propanediol-1,3 qui se sépare. Après lavage à Peau et .séchage, le produit fond à 191 à 196 C en se décomposant. Il a pour formule:

On verse par petites portions, en agitant, 9 g de ce carbonate cyclique dans 30 2m3 d'acide nitrique fumant maintenu à - 20 C par addition de petites portions d'anhydride carbonique solide.
L'addition terminée., on verse le mélange réactionnel sur de la. glace et on recueille le produit solide blanc. On fait. recristalliser le solide au sein d'éther, puis de méthanol, de manière à obtenir le carbo nate cyclique pur du (dl-t.rans-1-p-nitro- phényl-2-dichloracétamido-propanediol-1,3. Ce composé fond difficilement. à 180 à 185 C, mais peut cependant être analysé correcte ment.
Par hydrolyse modérée., il donne avec de bons 'rendements le (dl-trans-1-p-nitro- phényl-2-dichlora.cétamido-propauediol-1,3, en procédant comme suit: On dissout 2 g du carbonate cyclique du <B>(dl)</B> -trans-1-p-nitrophényl-2-dichloracét.amido- propanediol-1,3 dans 30 cms d'éthanol. On ajoute 1 cm3 d'acide chlorhydrique concentré et on abandonne le mélange au repos pendant dix minutes à la température ambiante. On neutralise le mélange réactionnel au moyen d'une solution d'hydroxyde de sodium, on filtre et on évapore le filtrat sous vide jus qu'à siccité.
On dissout la gomme résiduelle clans l'acétate d'éthyle, on filtre la solution et on évapore l'acétate :d'éthyle. La cristalli sation de l'huile résiduelle au sein de l'eau donne le (dl)-trans-1-p-nitrophényl-2-dichlor- acétamido-propanediol-1,3; point de fusion de 1:i0 C après recristallisation au sein de dichlorure d'éthylène.
<I>Exemple 3:</I> On ajoute par petites portions 10 g de (1) -4 trans -.f1-tphéinyl- 2 -dichloracétamido-pro- panecliol-1,3, en agitant, à 30 cms de chlorure de thionyle, à la température ambiante. On abandonne le mélange réactionnel au repos à la température ambiante pendant. environ une heure, puis on ajoute de l'éther pour préci piter le sulfite cyclique du (1)-trans-1-phényl- 2-dichlora.cétamido-propanediol-1,3.
On ajoute par petites portions 9,5 g de ce sulfite cyclique, en agitant, à 50 em3 d'acide nitrique fumant, à - 30 C. On entretient la température au cours de l'addition vers - 20 C par addition d'anhydride carbonique solide. L'addition terminée, on verse le mélange réactionnel sur de la glace pilée et on recueille le sulfite cyclique solide blanc du (1) -trans -1- p-nitrophényl-2-diehloracétamidoç propanediol-1,3. Après recristallisation au sein d'éthanol, le produit pur fond à 171 à 172 C.
On dissout 4 g de ce sulfite cyclique nitré dans 125 em3 de méthanol. On ajoute 10 cm3 d'acide chlorhydrique concentré et on aban donne le mélange au repos à la. température ambiante pendant environ vingt minutes.
On neutralise le mélange réactionnel au moyen d'une solution d'hydroxyde de sodium à 101/o et on évapore au bain-marie pour chasser l'éthanol. On. épuise le résidu aqueux au moyen d'acétate d'éthyle, on chasse l'acétate d'éthyle du mélange des extraits et on re- prend le résidu au moyen d'acétate d'éthyle, au sein duquel on le fait recristalliser. Le produit cristallisé blanc ainsi obtenu est le (1) -trans- 1-p-nitrophényl-2-dichloracétamido- propanediol-1,3 (chloramphénicol), de point de fusion 150 à 151 C.
(a) D = -2505C dans l'acétate d'éthyle. Exemple <I>4:</I> On ajoute lentement ,en l'espace de trois heures 15 g de phosgène à 20 g de (l)-trans- 1- phényl@ 2c- dichloracétamido-propanediol-1,3 dissous dans 50 cm3 de pyridine sèche. On maintient la température au cours de l'addi tion au voisinage de 0 C.
L'addition terminée, on verse le mélange réactionnel scu un grand volume de glace, on recueille le carbonate cyclique blanc du (1)-trans-1-phényl-2-dichlor- acétamido-propanediol-1,3 et on le lave à l'eau.
On ajoute par petites portions 15 g de ce carbonate cyclique, en agitant., à 45 .em3 d'acide nitrique fumant, à - 20 C. On main tient la température au cours de l'addition à - 20 C par addition de petites portions d'anhydride carbonique solide au mélange réactionnel. Quand la totalité du carbonate cyclique a été ajoutée, on verse le mélange réactionnel sur un grand vol-Lime de glace. On recueille le carbonate cyclique blanc inso luble du (1)-trans-1-p-nitrophényl2-dichlor- aeétamido-propanediol-1,3, on le lave à l'eau et on le fait recristalliser au sein de méthanol; point de fusion 157 C.
On dissout 5 g de ce carbonate cyclique nitré dans 75 cms d'éthanol. On ajoute 3 em3 d'acide chlorhydrique concentré et on aban donne le mélange réactionnel .au repos à la température ambiantes pendant 10 minutes environ. On neutralise le mélange réactionnel au moyen d'une solution aqueuse d'hydroxyde de sodium, on filtre et on évapore le filtrat sous vide jusqu'à siccité. On reprend le résidu au moyen d'acétate d'éthyle, on filtre la solu tion et on chasse l'acétate d'éthyle par distil lation.
On fait cristalliser l'huile résiduelle au sein d'eau et. l'on obtient le (1)-trans-1-p- r-itrophényl- 2 - dichloracétamido- propanediol- 1-3 (chloramphénicol) ; point de fusion 150 à 151 C après recristallisation au sein d'acé tate d'éthyle. (a) D = - 25 5 C dans l'a.cé- tate d'éthyle.
