<B>Verfahren</B> zur <B>Herstellung eines</B> Octahydroisochinolins. Es ist bekannt. (0. Schnider und J. Heller bach, Helv. Chim. Acta, 33 [1950], 1437), dass man 1- Benzyl - 2 - alkyloctahydroisochinoline der allgemeinen Formel VI herstellen kann, indem man ss-Cyelohexen-(1)-yl-äthylamin (I) mit substituierten oder unsubstituierten Phe- nyiessigsäuren kondensiert,
die entstehenden Säureamide mit wasserentziehenden Mitteln zu den entsprechenden 1-Benzyl-3,4,5,6,7,8- hexahydroisochinolinen cyelisiert und diese zu den Octahydroverbindungen hydriert. Die an schliessende N-Alkylierung ergibt dann die im aromatischen Ring substituierten oder un- substituierten 1-Benzyl-2-alkyl-1,2,3,4,5,6,7,8- octahydroisochinoline.
Es wurde nun gefunden, dass man zu den gleichen 1-Benzyl-2-alkyl-1,2,3,4,5,6,7,8-octa- hydroisochinolinen der allgemeinen Formel VI und deren Salzen dadurch gelangen kann, dass man ein Phenylessigsäurehalogenid der Formel

mit 1-Alky lamino-2- [cyclohexen- (1') -y1 ] -äthan (11I) umsetzt, das dabei entstehende Amid IV zum entsprechenden 1-Benzyliden-2-alkyl- 1,2,3,4,5,6,7,
8 - octahydroisochinolin der allge- meinen Formel V cyclisiert und dieses in Gegenwart von Katalysatoren hydriert.

Die als Ausgangsprodukte benötigten 1-Alkylamino-2-cyclohexen-(1')-yl-äthane III können z. B. durch Acylierung des ,B-Cyelo- hexen-(1)-yl-äthylamins (1) und nachfolgende Reduktion gewonnen werden.
Die sekundären Amine III führt man durch Umsetzung mit unsubstituierten oder substituierten Phenyl- essigsäurehalogeniden in die entsprechenden Phenylessigsäure- [cyclohexen- (1) -yl-äthyl] -N alkyl-amide IV über.
Durch Behandlung mit wasserentziehenden Mitteln, wie P0C13 oder P205, lassen sie sich zu den unsubstituierten oder substituierten 1- Benzyliden - 2 - alkyl - 1,2,3,4,5,6,7,8-octahydroisochinolinen cyclisie- ren, welche bei der Hydrierung die entspre chenden Benzylverbindungen liefern.
Die neue Synthese unterscheidet sich von der bekannten prinzipiell dadurch, dass die Isoehinolinverbindung V unter Bildung einer semicyclischen Doppelbindung entsteht. Die Verbindungen der allgemeinen Formel VI bilden sich deshalb durch Hydrierung einer C-C-Doppelbindung.
Die Erzeugnisse des vorliegenden Verfah rens können als Zwischenprodukte für die Herstellung von N-Alkylmorphinanen Ver wendung finden. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung des bereits be kannten 1-(3',4'-Dimethoxy-benzyl)-2-methyl- 1,2,3,4,5,6,7,8 - oetahydroisochinolins, das da durch gekennzeichnet ist, dass man ein Homo veratrumsäurehalogenid (z.
B. -chlorid oder -bromid) mit 1-Methylamino-2-cyelohexen-(1')- yl-äthan umsetzt, das dabei entstehende Amid zum 1-(3',4'-Dimethoxy-benzyliden)-2-methyl- 1,2,3,4,5,6,7,8 - octahydroisochinolin cyelisiert und dieses in Gegenwart von Katalysatoren hydriert.
<I>Beispiel:</I> Eine Suspension von 196,2 Gewichtsteilen Homoveratrumsäure in 2000 Gewiehtsteilen absolutem Benzol versetzt man langsam mit 230 Gewichtsteilen Phosphorpentachlorid. Nachdem die anfangs lebhafte Reaktion zum Stillstand kommt, wird 15 Minuten auf 60 C erwärmt und anschliessend das Benzol im Wasserstrahlvakuum bei etwa 30 C abge dampft. Um das bei der Säureehloridbildung entstehende Phosphoroxychlorid vollständig zu entfernen, wird der Rückstand wiederholt in Benzol gelöst und erneut im Vakuum ein geengt.
Man löst darauf das Homoveratrum- säurechlorid in 7.000 Gewichtsteilen absolutem Benzol und tropft langsam zu einer eisgekühl- ten Lösung von 278 Gewichtsteilen 1-11letliyl- amino-2-cyclohexen-(1')-yl-äthan (Herstellung beschrieben in Patentschrift Nr.292079) in 500 Gewiehtsteilen absolutem Benzol. Das Reaktionsprodukt wird darauf 3 Stunden bei Zimmertemperatur stehen gelassen und dann der Reihe naeh mit 3n Salzsäure, Wasser, 3n Soda und Wasser gewaschen.
Nach dein Abdestillieren des Lösungsmittels bleibt das Homoveratrumsäure- (cyclohexen-(1)-y 1-äthy 1)- N-methylamid als hellgelbes, viskoses Öl, das ohne weitere Reinigung weiterverarbeitet wird, zurück.
317,4 Gewiehtsteile Honioveratrumsäure- (eyelohexen-(1)-z-1-iithyl)-N-metliyl-amid wer den in 1000 Gewiehtsteilen absolutem Benzol gelöst und mit 306 Gewichtsteilen POCl3 zwei Stunden auf 95 C erwärmt und dann in Was ser gegossen. Nach der Zugabe von Äther wird die wässerige Phase, die die phosphor sauren Salze des Reaktionsproduktes enthält, abgetrennt und wiederholt mit Äther gewa schen. Die eiskalte, wässerige Lösung versetzt man darauf vorsichtig mit konzentrierter Natronlauge und nimmt die abgeschiedene Base in Äther auf, welcher nach dem Wasehen mit.
Wasser getrocknet und abdestilliert wird. Das zurückbleibende 1-(3',4'-Dimethoxy -ben- zyliden)-2-methyl-1,2,3,4,5,6,7,8-octahy droiso- chinolin wird in der 10faehen Menge Methanol gelöst und in Gegenwart von 60 Gewichts teilen Raney-Nickel. hydriert.
Nach der Auf nahme der für 1 liol berechneten Wasserstoff menge wird vom Katalysator abgetrennt und das Methanol abgedampft. Als Rückstand bleibt das 1-(3',4'-Dimethoxy-benzyl)-2-methyl- 1,2,3,4,5,6,7,8-oetahy droisoehinolin. Siedepunkt 135 C bei 0,001 mm; Oxalat, Schmelzpunkt 125-127 C (aus Methanol).
<B> Process </B> for <B> producing </B> an octahydroisoquinoline. It is known. (0. Schnider and J. Hellerbach, Helv. Chim. Acta, 33 [1950], 1437) that 1-benzyl - 2 - alkyloctahydroisoquinolines of the general formula VI can be prepared by ss-cyelohexen- (1) - yl-ethylamine (I) condensed with substituted or unsubstituted phenyiacetic acids,
the acid amides formed are cyelized with dehydrating agents to give the corresponding 1-benzyl-3,4,5,6,7,8-hexahydroisoquinolines and these are hydrogenated to give the octahydro compounds. The subsequent N-alkylation then gives the substituted or unsubstituted 1-benzyl-2-alkyl-1,2,3,4,5,6,7,8-octahydroisoquinolines in the aromatic ring.
It has now been found that the same 1-benzyl-2-alkyl-1,2,3,4,5,6,7,8-octa-hydroisoquinolines of the general formula VI and their salts can be obtained by using a phenylacetic acid halide of the formula

The 1-alkylamino-2-cyclohexen- (1 ') - yl-ethanes III required as starting materials can, for. B. by acylation of, B-Cyelohexen- (1) -yl-ethylamine (1) and subsequent reduction.
The secondary amines III are converted into the corresponding phenylacetic acid [cyclohexen (1) -ylethyl] -N alkyl amides IV by reaction with unsubstituted or substituted phenylacetic acid halides.
By treatment with dehydrating agents, such as POC13 or P205, they can be cyclized to the unsubstituted or substituted 1-benzylidene-2-alkyl-1,2,3,4,5,6,7,8-octahydroisoquinolines, which at the hydrogenation provide the corre sponding benzyl compounds.
The new synthesis differs from the known one in principle in that the isoquinoline compound V is formed with the formation of a semicyclic double bond. The compounds of the general formula VI are therefore formed by hydrogenation of a C-C double bond.
The products of the present process can be used as intermediates for the preparation of N-alkylmorphinans. The present patent is a process for the preparation of the already known 1- (3 ', 4'-dimethoxy-benzyl) -2-methyl-1,2,3,4,5,6,7,8 - oetahydroisoquinoline, the as is indicated by the fact that a homo veratrumsäurehalogenid (z.
