Process for the preparation of pyridine-3-aldehyde. Up to now, pyridine-3-aldehyde was only accessible in awkward ways and in poor yields. The method of Panizzon (Helvetiea Chimiea Acta, vol. 24, 11 [1941], 24 E), which has been considered the best up to now,
supplies the aldehyde in the last stage of the process in only 36% yield.
It has now been found that the pyridiri- 3-aldehyde can be obtained in a simple manner if, according to the invention, pyridyl-3-carbinol is brought into contact with an oxidation catalyst together with a hydrogen acceptor at a temperature of 200 to 400.degree.
The reaction is conveniently carried out as follows: Vapors of pyridyl-3-earbinol are passed with a stream of air through a tube filled with silver or copper and heated to about 250 to 400 ° C. The condensation of the reaction gases and subsequent rectification gives the Aldehyde in good yield and in pure form. It is best to use a copper pipe which can be heated by an electric furnace over a substantial part of its length.
Acts as a catalyst. on the one hand the inner wall of the copper pipe, on the other hand a filling of finely divided copper or silver, such as copper wire mesh, silver wire mesh, silver shavings or wool. The air dried with concentrated sulfuric acid and filtered through cotton wool. can either be blown or sucked through the pipe, whereby the flow velocity is measured with a rotameter. Before entering. The air is allowed to flow into the tube through the pyridyl-3-carbinol, which is heated to about 190 to 200 ° C., and it is loaded with the pyridyl-3-carbinol vapors.
After the air / vapor mixture has passed the pipe, the overheated reaction gases are condensed in a wide-lumen cooling coil that is cooled to -10 ° C. The condensate, which mainly consists of pyridine-3-aldehyde, pyridyl-3-carbinol and water, is collected in a collecting vessel that is also cooled to -10 ° C.
Pyridine-3-aldehyde can be used as an intermediate in the manufacture of medicinal products.
Example 1: With. With an air flow of 2.3 liters per minute, 100 g of pyridyl-3-earbinol are blown through a 1.6 m long copper pipe charged with rolls of copper wire and heated to 320 to 340 ° C. The concentration of pyridyl-3-carbinol in the air is on average 0.1 g / liter. After about 6 hours, the 100 g are blown through and air is allowed to flow through the apparatus for another 30 minutes.
The temperature of the cooling device is allowed to rise from -10 C to about + 10 C so that the reaction mixture, which is viscous at a low temperature, can drain off better. If necessary, it is rinsed with a little methanol. In this way, 95 to 105 g of a mixture are obtained which consists mainly of pyridine-3-aldehyde, pyridyl-3-earbinol and water.
By fractionating once at 12 mm, after a small forerun, 43 g of pyridine-3-aldehyde from 85 to 90 ° C. and 38 g of pyridyl 1-3-carbinol from 143 to 145 ° C. are obtained. The yield of Al- dehyde, based on converted pyridyl-3-carbinol, is thus 70% of theory. Example 2:
With an air flow of 3.8 liters per minute, 80 g of pyridyl-3-earbinol are blown through a 1.6 m long copper pipe that is filled with silver chips and heated to 280 to 300 ° C. The concentration of pyridyl-3-earbinol is on average 0.1 g / liter. After about 4 hours the 80 g are blown through. Then proceed as described in example 1.
In addition to 31.3 g of pyridyl-3-carbinol, 36.5 g of pyridine-3-aldehyde are obtained, that is to say 771 / o of theory, based on converted pyridyl 1-3-earbinol.
Example 3: 200 g of Pyridi-l-3-carbinoI are blown with an air flow of 3.2 liters per minute. through a 1.6 m long copper pipe covered with silver shavings and heated to 300 to 310 C. The concentration of pyridyl-3-caxbinol is on average 0.1 g / liter. After about 9 hours, the 200 g are blown through. The procedure is then as described in Example 1.
In addition to 90 g of pyridyl-3-earbinol, 91.5 g of pyridine-3-aldehyde are obtained, that is to say 84% of theory, based on converted pyridyl-3-carbinol.
