Verfahren zur Herstellung eines neuen antibiotisch wirksamen Stoffes. Es wurde gefunden, dass ein neuer anti biotisch wirksamer Stoff dadurch hergestellt werden kann, dass man mittels einer Nähr lösung einen Vertreter einer Gruppe von Actinomyceten züchtet, welche einen Farb stoff bilden, der die Nährlösung in der natür lichen Konzentration gelb färbt und der als ziegelrotes Produkt vom Schmelzpunkt 254 isoliert werden kann.
Der aus den Nährlösungen isolierte Farb stoff zeigt starke antibiotische Wirksamkeit und stellt ein wirksames Chemotherapeutieum gegen eine Reihe von Mieroorganismen, z. B. Staphylococcus aureus, Bacillus subtilis und Baeterium coli, dar. Ausserdem hat. er sich über raschenderweise als tumorhemmend erwiesen. Diese Hemmung tritt auch bei den reinen Produkten in Erscheinung.
Der antibiotisch wirkame Farbstoff ist im folgenden als Aeti- noehyrsin bezeichnet.
Es sind in den letzten Jahren eine Anzahl von Antibiotica aus Actinomyees-Stämmen iso liert worden. Unter anderem haben Waksman und Mitarbeiter (USA-Patent. Nr. 2378449, Waksman und Tishler, :
l. Biol. Chem. 142, 519 (1942) und Scienee 98 (1943) Suppl. Seiten 8 und 10) bei der Züchtung des Aetinomyces antibioticus aus den Nährlösungen einen Stoff isoliert den sie als Aetinomycin bezeichneten, und der ebenfalls bacteriost.atische Eigen schaften hat. Dieser bekannte, antibiotisch wirksame Stoff hat. mit Actinoehrysin zahl reiche Berührungspunkte, das Aetinochrysin unterscheidet sich jedoch von ihm auch in charakteristischer Weise.
Insbesondere unter scheidet es sich hinsichtlich des Stickstoffge haltes (12,27 % N beim Actinochrysin, 13,35 % N beim Actinomycin), seiner Löslichkeit in Aceton, in dem es erheblich löslicher ist als Aetinomycin, und durch die Tatsache, das es im Gegensatz zum Actinomycin kein Diacetyl- deriva.t bildet.
Auch hat das Leukoacetyl- derivat des Aetinochrysins einen niedrigeren Stickstoffgehalt als das Leukoacetylderivat des Aetinomycins. Besonders charakteristisch und zur leichten Unterscheidung der beiden Antibiotica geeignet ist ihr verschiedenes Ver halten gegenüber Claisenlauge. Während beim Actinomyein in Claisenlaage eine Rotfärbung auftritt, die rasch wieder verschwindet,
ist die Rotfärbung beim Actinochrysin bestän diger.
Aetinochrysin kristallisiert aus Essigester in alizarinroten hexagonalen Bipyramiden. Es ist eine sehr schwache, zweisäurige Base mit einer wahrscheinlichen Summenformel C4oH57011N7 und einem errechneten Moleku- largewicht von 811,5.
Die Züchtung der actinoehrysinbildenden Aetinomyceten kann in an sich bekannter Weise im Oberflächenverfahren oder submers erfol gen. Das submerse Verfahren gibt. bessere Aus beuten an Antibiotieutn und die Zeiten bis zur optimalen Konzentration des Actino- chrysins in der Nährlösung werden erheblich abgekürzt. Infolgedessen wird das submerse Verfahren bevorzugt.
Die Isolierung des Aetinochrysins aus der Kulturflüssigkeit kann durch Extraktions- und bzw. oder Adsorp- tionsverfahren geschehen. Als Extraktionsmit tel eignen sich organische Lösungsmittel, ins besondere Butylacetat, als Adsorptionsmittel kommt in erster Linie Aluminiumoxyd in Frage. Zur Abtrennung des Aetinochrysins von seinen Verunreinigungen wird das Roh produkt z.
B. in einem organischen Lösungs mittel gelöst und die Lösung chromatogra._ phisch mit Aluminiumoxyd behandelt. Das Produkt lässt sich aus einem Gemisch aus gleichen Teilen Essigester und Benzol um- kristallisieren. Der Schmelzpunkt liegt dann bei 254 . Aus <I>f</I> ührungsb eispieZ: 1.
Stammkultur Der Stamm Actinomyees Bo 103 ist im Northern Regional Research Laboratory of the USA-Department of Agriculture in Peoria, Illinois unter der Nummer NRRL-2250 de poniert.
Er wird auf einem Nährboden ge züchtet, der ausser der unten beschriebenen Nährlösung 16 3 % Aga.r enthält. 2.
Vorkultur Für die Vorkultur werden 300 cm- Erlen meyer mit je 50 cm3 Nährlösung 16 be schickt, sterilisiert und mit Impfmaterial, das von der Stammkultur gewonnen wurde, be- impft. Die Kölbchen werden etwa fünf Tage bei 30 auf einer Schüttelmaschine geschüttelt, wobei sich eine Submerskultur des Microorga- nismus bildet und die Nährlösung eine röt- lichgelbe Farbe annimmt.
Das Impfmaterial wird bakteriologisch auf seine Reinheit und Brauchbarkeit geprüft. Gleichzeitig wird die antibiotische Wirksamkeit getestet.. Hierbei zeigen sich gegenüber Staphylococcusaureus Hemmungswerte von 1:200 bis 1.:600 er reicht. 3.

Dann wird der Kessel auf 30 heruntergekühlt und unter sterilen Bedingungen mit etwa einem Liter der obigen Vorkultur beimpft. Dann wird der Hauptansatz unter Rühren und Belüften vier bis sechs Tage bei 29 bis 30 bebrütet. Der Luftdurchsatz beträgt im Anfang 500 Liter pro Minute, bei starkem Schäumen wird die Belüftung schwächer eingestellt.
Das p$ der Nährlösung ist im Anfang 6,5 bis 6,9; es steigt während der Bebrütung auf 8,8 bis 9,1 an. Die Lösung färbt sich rötlichgelb und zum Schluss des Wachstumsvorganges scheidet sich das Actinochry sin zum Teil in rötlichen Flocken ab, die sich beim Stehen der Flüssig keit oben abscheiden, während das Pilzmycel zu Boden sinkt.
4. Roh-Actinochrysin Die aus dem Submers-Kessel abgezogene Kulturflüssigkeit wird bei Zimmertemperatur in einem 1000-Liter-Rührkessel mit im Va kuum destilliertem Butylacetat, und zwar 200 auf 500 kg Kulturflüssigkeit, ausgerührt. Man lässt über Nacht stehen und scheidet am nächsten Morgen die tief gelb gefärbte Butyl- acetat-Lösung in einer Milchzentrifuge vom Kulturfiltrat ab.
Das Butylacetat wird unter vermindertem Druck bei 60 abdestilliert. Der Rückstand, eine rotbraune harzige Masse, wel che rohes Actinochrysin darstellt, wird im Vakuum getrocknet.. Ausbeute: 200 g.
5. Gereinigtes Actinochrysin Für die nun folgende ehromatographische Adsorption wird ein Gewichtsteil des obigen harzigen Rohproduktes in vier Gewichtsteilen Benzol gelöst, durch Faltenfilter filtriert, und über zehn Gewichtsteile, vorher mit Benzol angeschlämmten Aluminiumoxyd Merek (nach

Bei weite rem Zusatz von Essigester tritt das Aetino- ebrysin unten aus und wird aufgefangen. Man hört mit dem Essigesterzusatz auf, wenn der Durchlauf nur noch schwach gelb gefärbt ist..
Dann enthält -die Säule zwar noch einen, mit warmem Alkohol ablösbaren Farbstoff, im wesentlichen sind jedoch zwei Hauptfrak tionen erhalten worden, nämlich der Benzol- Vorlauf und die Essigester-Fraktion, -die die Hauptmenge Aetinoehrysin enthält.
Die Ausbeute an Actinochrysin - nach dem Abdestillieren des Essigesters - beträgt 40 bis 60 % die eingesetzten harzigen Roh- produktes. 6.
Aetinoehrysin-Kristallisat Man löst ein Gewichtsteil der Essigester- Fraktion in einem Gemisch von sieben Teilen Essigester und sieben Teilen Benzol, filtriert, dampft unter vermindertem Druck auf etwa ein Viertel Volumen ein und versetzt nach Abkühlung auf Zimmertemperatur mit dem selben Volumen über Quecksilber destillierten Schwefelkohlenstoff. Spontan oder nach Rei ben mit dem Glasstab fällt das kristallisierte Aetinoehrysin aus und wird nach 24stündigem Stehen bei 0 abgesaugt und nachgewaschen,
wobei zuerst ein Essigester-Schwefeikohlen- stoffgemisch, dann Ligroin verwendet wird. -Man trocknet auf dem Wasserbade; Ausbeute mindestens 7011/o des eingesetzten gereinigten Aetinochrysins, F. P. 254 .
