EMI0001.0002
In <SEP> der <SEP> Hauptpatentschrift <SEP> wurde <SEP> ein <SEP> Ver fahren <SEP> zur <SEP> Darstellung <SEP> eines <SEP> Pentaens <SEP> dar gelegt, <SEP> das <SEP> dadurch <SEP> gekennzeichnet.
<SEP> ist, <SEP> dass
<tb> man <SEP> 1-Oxy-3,7-clinietliyl-6-oxy-9-[2',6',6-trinie thyl-ey <SEP> clohexen- <SEP> (1') <SEP> -y1 <SEP> ] <SEP> - <SEP> nonadien- <SEP> (2,7) <SEP> -in- <SEP> (4)
Method of representing a pentaene.
EMI0001.0002
In <SEP> of the <SEP> main patent specification <SEP> <SEP> a <SEP> method <SEP> for the <SEP> representation <SEP> of a <SEP> pentaene <SEP> was presented, <SEP> the <SEP > marked with <SEP>.
<SEP> is, <SEP> that
<tb> man <SEP> 1-Oxy-3,7-clinietliyl-6-oxy-9- [2 ', 6', 6-trinie thyl-ey <SEP> clohexen- <SEP> (1 ') <SEP > -y1 <SEP>] <SEP> - <SEP> nonadiene- <SEP> (2,7) <SEP> -in- <SEP> (4)

<tb> the <SEP> triple bond <SEP> partially <SEP> hydrogenated, <SEP> that
<tb> formed <SEP> 1-oxy-3,7-diniethyl-6-oxy-9-trinie thylcycloliexenyl-nonatriene- <SEP> (2,4,7) <SEP> on <SEP> der
<tb> terminal <SEP> hydroxyl group <SEP> esterified, <SEP> one
<tb> Ally <SEP> reallocation <SEP>, <SEP> from <SEP> the <SEP> received <SEP> connection <SEP> the <SEP> 8 permanent <SEP> substituents <SEP> together <SEP> with <SEP> a <SEP> adjacent <SEP> hydrogen atom <SEP> splits off <SEP> and <SEP> then <SEP> saponifies the <SEP> vitamin <SEP> A ester <SEP> obtained .
It has now been found that the same pentaene can also be obtained by adding 1-Ox. #> - 3,7-dimethyl-6-oxy-9- [2 ', 6', 6'-t.rimethy 1 - ey elohexen- (1 ') -y1] -nonadiene- (2.7) - in- (4) partially hydrogenated on the triple bond by the action of 1 1VIol hydrogen, then the 1-oxy-3 formed,
7-dimethyl - 6-oxy-9-t.rimet.hyicyclohexenyl-nonatriene- (2,4, 7) treated with an allyl rearrangement-effecting agent and from the compound obtained the 8-position substituent together with a splitting off neighboring hydrogen atom.
This synthesis proceeds according to the following equation
EMI0001.0024
The starting compound for the present process is 1-oxy-3,7-dimethyl-6-oxy-9- [2 ', 6', 61-trimethyl-cyclohexen- (1 ') - yl] - nonadiene- (2, 7) -in- (4). This compound can be prepared by condensing 1-oxy-3-methyl-pentene- (2) -in- (4) by means of a Grignard reaction with 4- [2 ', 6', 6'-trimethyl-cyclohexene- ( 1')
-y1] -2-methyl-buten- (2) -al- (1).
The 1-oxy-3,7-dimethyl-6-oxy-9-trimethyl-cyclohexenyl-nonadiene- (2,7) -yne- (4) is subjected to the partial hydrogenation of the triple bond. Palladium-calcium carbonate and palladium-barium-sulfate can be used as catalysts, for example; Palladium carbon, onto which quinoline has been adsorbed before use, is particularly suitable. The hydrogen supply is interrupted after the calculated amount of hydrogen has been taken up.
The hydrogenated product does not need to be purified in isolation. It is a viscous, yellowish oil which does not absorb ultraviolet rays of a wavelength greater than 260 mu and which dissolves in arsenic trichloride or trichloroacetic acid-chloroform solution with a blue color.
The compound II obtained by partial hydrogenation is treated with an agent causing an allyl rearrangement which, if appropriate, simultaneously causes a substitution of the hydroxyl group, the hydroxyl group or
the substituent migrates from the 6-position to the 8-position and the neighboring double bond migrates to the 6-position. To introduce a new double bond in the compound III obtained in this way, water or z. B. split off an acid, for which the usual methods can be applied, provided that the ge sought pentaene (vitamin A) is stable under the relevant conditions.
An advantageous method, which achieves the alyl rearrangement and the elimination of water in a single reaction stage, consists in heating with a small amount of iodine in the presence of an inert solvent with a boiling point of 80 to 1500 C,
such as toluene or petroleum ether. Instead of iodine, the allyl rearrangement and dehydration can also be carried out with a small amount of a compound that easily splits off iodine, such as, for example, with hydrogen iodide, phosphorodiodide or haloiodides. The product corresponds to formula IV.
According to the process explained above, vitamin A is obtained, which has the same biological effectiveness as vitamin A obtained from natural materials. The process product is characterized by the absorption maximum occurring in the ultraviolet spectrum at 328 μm and by the color reactions characteristic of vitamin A. with antimony trichloride, arsenic trichloride, aluminum minilusilicate containing hydrogen chloride and trichloroacetic acid.
The product can be cleaned using the same methods as high concentrations of vitamin A obtained from natural materials (separation between solvents, chromatographic adsorption, gentle distillation, etc.). Just like natural vitamin A, the products must be protected from the damaging effects of light, air and heat. It is advisable to add antioxidants, which can also be present during the entire course of the synthesis; Tocopherols are particularly suitable as antioxidants.
The vitamin A obtained in this way is intended to be used as a medicinal product. <I> Example: </I> 10 parts by weight of 1-oxy-3,7-dimethyl-6-oxy-9 - [2 ', 6', 6'-trimethyl-cyclohexen- (1 ') -y1] -nona - diene- (2,7) -in- (4) are dissolved in 100 parts of methyl alcohol and letter addition of 0.3 parts by weight of 4% palladium-calcium carbonate catalyst with 0,
2 parts by weight of pyridine hydrogenated in a hydrogen atmosphere. After 1 mol of hydrogen has been absorbed, the hydrogenation is interrupted, whereupon the reaction mixture is filtered off from the catalyst. Evaporation of the solvent gives 1-oxy-3,7-dimethyl-6-oxy-9- [2 ', 6', 6'-trimethyl-cyelohexen- (1 ') -yl] -nona- in quantitative yield triene- (2,4,7). Light yellow, very viscous oil of n10 = 1.524.
The compound contains two active hydrogen atoms and shows no absorption above 260 niu in the ultraviolet absorption spectrum.
1 part by weight of 1-oxy-3,7-dimethy l-6-oxv-9-trimethyleyclohexenyl-nonatriene- (2,4,7) is dissolved in 20 parts by volume of petroleum ether with a boiling point of 80 to 1200 ° C. and while introducing nitrogen and vigorous stirring 0.1 part by weight of 50% aqueous hydroiodic acid is added at the boiling point.
The mixture is heated under reflux for a further 1/1 hour, cooled, washed with sodium bicarbonate solution and water and the petroleum ether solution is dried with sodium sulfate. After the solvent has been distilled off, an orange-yellow oil with a content of about <B> 10% </B> vitamin A is obtained.
The raw product obtained in this way can be purified, for example, in a continuous chromatogram through columns with little activated aluminum oxide using petroleum ether with a boiling point of 60 to 80 C. The elffiate of the yellow-greenish fluorescent main zone provides a yellow vitamin A preparation, its ultraviolet absorption spectrum a pronounced maximum at 325 m, u possesses.
The partially hydrogenated condensation product can also be heated with a small amount of iodine instead of hydrogen iodide to achieve the allyl rearrangement and dehydration. The presence of an inert solvent is advantageous, but not essential.
