Description

  

  Verfahren zur Darstellung der     4,4'-Diaminodiphenylsulfondiphosphordimetylamidsäure.       Das     4,4'-Diaminodiphenylsulfon,    dem eine  sehr starke bakterizide Wirkung zukommt,  konnte sich, seiner hohen Toxizität wegen,  nicht in die Therapie einbürgern.  



  Es wurde nun gefunden, dass die     Phos-          phamidsäuren    des     4,4'-Diaminodiphenylsul-          fons    bei ungefähr gleich starker bakterizider  Wirkung eine wesentlich geringere     Toxizität     besitzen, so dass sie sich in ausgezeichneter  Weise für die Bekämpfung bakterieller In  fektionen eignen. Ihre Salze lassen sich in  üblicher Weise gewinnen.  



  Gegenstand des vorliegenden Patentes ist  ein Verfahren zur     Darstellung    der 4,4'       Diaminodiphenylsulfondiphosphordimethyl-          amidsäure,    welches dadurch     gekennzeichnet     ist, dass auf     4,4'-Diaminodiphenylsulfon    oder  ein Salz desselben     Phosphoroxychlorid    ein  wirken gelassen und das Reaktionsprodukt  mit     wässrigem        Dimethylamin    umgesetzt wird.  



  Die neue     4,4'-        Diaminodiphenylsulfon-          diphosphordimethylamidsäure    bildet leicht  wasserlösliche, neutral reagierende Salze, die  ohne     Gewebsschädigungen        injiziert    werden    können. Sie soll als Arzneimittel verwendet  werden.  



  <I>Beispiel:</I>  10 Gewichtsteile     4,4'-Diaminodiplienyl-          sulfondihydrochlorid    werden mit 30 Raum  teilen     Phosphoroxychlorid    so lange am Rück  flusskühler gekocht, bis vollständige Lösung  eingetreten ist.

   Dann giesst man die hell  bräunliche Flüssigkeit     unter    Rühren in eine  30  warme Mischung von 80 Raumteilen Ben  zol und 80 Raumteilen     Petroläther,        worauf     das     Diphosphorsäuretetrachlorid    des     Diamino-          diphenylsulfons    bald fest     wird.    Man setzt  noch 80     Raumteile        Petroläther    hinzu und lässt  erkalten.

   Nach dem Absaugen trägt man das  feuchtigkeitsempfindliche Chlorid unter Küh  lung bei 10 bis 20  in 70 Raumteile einer  wässrig     konzentrierten        Lösung    von     Dime-          thylamin    ein, indem man 40 bis 50 Gewichts  teile Eis hinzufügt.

   Nach     Beendigung    der       Umsetzung        wird    die Lösung im     Vakuum    auf       ein    kleines     Volumen    eingedampft, worauf das       Dimethylaminsalz    der     4,4'-Diaminodiphenyl-          sulfondiphosphordimethylamidsäure              Cleg2406N,SP-,     auskristallisiert. Man saugt es ab, wäscht es  mit Alkohol und kristallisiert es aus 50 Raum  teilen     abs.    Alkohol um. Aus dem     Dimethyl-          a.minsalz    wird die Säure durch Ansäuern in  Freiheit gesetzt.

  It has now been found that the phosphamic acids of 4,4'-diaminodiphenylsulfons have a significantly lower toxicity with approximately the same bactericidal effect, so that they are outstandingly suitable for combating bacterial infections. Their salts can be obtained in the usual way.



  The subject of the present patent is a process for the preparation of 4,4'-diaminodiphenylsulfondiphosphorodimethylamide acid, which is characterized in that 4,4'-diaminodiphenylsulfone or a salt of the same phosphorus oxychloride is allowed to act and the reaction product is reacted with aqueous dimethylamine.



  The new 4,4'-diaminodiphenylsulfon-diphosphordimethylamic acid forms easily water-soluble, neutrally reacting salts that can be injected without damaging the tissue. It is intended to be used as a medicine.



  <I> Example: </I> 10 parts by weight of 4,4'-diaminodiplienyl sulfone dihydrochloride are refluxed with 30 parts by volume of phosphorus oxychloride until complete dissolution has occurred.

   The light brownish liquid is then poured into a warm mixture of 80 parts by volume of benzene and 80 parts by volume of petroleum ether, with stirring, whereupon the diphosphoric acid tetrachloride of the diaminodiphenyl sulfone soon solidifies. Add 80 parts of petroleum ether and let it cool down.

   After suctioning off, the moisture-sensitive chloride is introduced with cooling at 10 to 20 in 70 parts by volume of an aqueous concentrated solution of dimethylamine by adding 40 to 50 parts by weight of ice.

   After the reaction has ended, the solution is evaporated to a small volume in vacuo, whereupon the dimethylamine salt of 4,4'-diaminodiphenylsulfonediphosphorodimethylamic acid Cleg2406N, SP-, crystallizes out. It sucks it off, washed it with alcohol and crystallized it from 50 space divisions abs. Alcohol around. The acid is liberated from the dimethyl amine salt by acidification.

PATENTANSPRUCH: Verfahren zur Darstellung der 4,4' Diaminodiphenylsulfondiphosphordimethyl- amidsäure, dadurcb gekennzeichnet, dass auf 4,4'-Diaminodiphenylsulfon oder ein Salz desselben Phosphoroxychlorid einwirken ge lassen und das Reaktionsprodukt mit wäss- rigem Dimethylamin umgesetzt wird. Die neue 4,4'- Diaminodiphenylsulfondi- phosphordimethylamidsäure bildet leicht was serlösliche, neutral reagierende Salze, die ohne Gewebsschädigungen injiziert werden können.
