Verfahren zur Darstellung von Vitamin Bi. Die Herstellung von Vitamin Bi, aus gehend aus 2-Methyl-4-amino-5-(thioform- amido-methyl)-pyrimidin und aus 2-Methyl- 2-alkoxy-3-chlor-tetrahydrofuran, ist bereits bekannt.
Wir haben nun gefunden, dass man Vit amin B, mit ausgezeichneter Ausbeute auch so herstellen kann, dass man 2-Methyl-2-formyl- oxy-3-chlor-tetrahydrofuran der Einwirkung von 2-11letl-lyl-4-amino-5-(thioformamido-me- thyl)-pyrimidin unterwirft. Man führt die Umsetzung zweckmässig in Ameisensäure als Reaktionsmedium aus, zweckmässig mit etwa 90-100%iger Ameisensäure. Das Aufein- anderwirken der beiden Reaktionskomponen ten führt man zweckmässig bei 50-70 , so zum Beispiel vorteilhaft durch Erhitzen während 40 Stunden auf 50 , aus.
Die allfällige Aufarbeitung des Reak tionsgemisches wird zum Beispiel so .ausge führt, dass man dazu abs. Alkohol und Salz säuregas enthaltenden äbs. Alkohol zugibt und nach Stehen das ausgeschiedene Vitamin- B,-Chlorhydrat durch Filtrieren abtrennt. Das so gewonnene Vitamin B, ist, wie sich zeigte, schon in diesem Zustand weiss und auch die Ausbeute ist vorzüglich. Eine Rei nigung durch das Pikrat oder Pikrolonat ist überflüssig.
Die Bereitung des für die erfindungs gemässe Herstellung von Vitamin B1 verwen deten 2-Methyl-2-formyloxy-3-chlor-tetrahy- drofurans, welche bis jetzt unbekannt war, kann so geschehen, dass man Aceto-chlor- butyrolacton oder Aceto-chlor-propylalkohol oder 2-Methyl-2-oxy-3-chlor-tetrahydrofuran oder dessen Äther, zum Beispiel den mit <I>y</I> - A.ceto <I>- y</I> - chlor - propylalkohol gebildeten Äther, der Einwirkung von Ameisensäure unterwirft,
öder 2-Methyl-2,3-dichlor-tetra- hydrofuran der Einwirkung von ameisensau ren Salzen und/oder Ameisensäure unter wirft. So zum Beispiel kann man das 2-Me- thyl - 2 - formyloxy-3-chlor -tetrahydrofuran durch Einwirkenlassen der berechneten Men gen wasserfreien galiumformiats auf 2-Me- thyl-2,3-dichlor-tetrahydrofuran herstellen.
Man kann aber auch das Produkt auf die Weise herstellen, dass man die Verbindung, welche bis jetzt für Aceto-chlor-propylalko- hol gehalten wurde, deren wahre Konstitu tion aber durch Stevens und Stein (siehe Journal of the American Chemical Society, volume 62, Seite 1045) ermittelt wurde, mit 100%iger Ameisensäure erhitzt.
Man kann feiner aber auch noch so verfahren, dass man Acetochlor-butyrolacton mit 1 142a1 Wasser und mit einer überschüssigen Menge unge fähr 100%iger Ameisensäure während eini gen Stunden kocht und dann das Produkt durch fraktionierte Destillation gewinnt. Beispiele: 1. Man gibt zu 15,5 g 2-Methyl-2,3-di- chlor-tetrahydrofuran (über dessen Darstel lung siehe schweiz. Patent Nr. 225777) 20 cm' 100%ige Ameisensäure und 9 g was serfreies pulverisiertes Kaliumformiat. Die Reaktion tritt ein; man entfernt die Reak tionswärme durch Kühlen. Inzwischen er wärmt sich das Reaktionsgemisch auf etwa 60 .
Man hält es ungefähr eine halbe Stunde lang auf dem siedenden Wasserbad, während dessen sich das Reaktionsgemisch schwach braun färbt. Man kühlt das Reaktions gemisch, gibt 50 cm' trockenen Äther hinzu, filtriert das abgeschiedene Mineralsalz ab, dampft die ätherische Lösung ein und destil liert dann die Ameisensäure bei ungefähr 20 mm Hg ab. Das zurückgebliebene 01 wird in einem Vakuum von 1,5 mm Hg destilliert. Es destillieren etwa 12 g 2-Methyl-2-formyl- oxy-3-chlor-tetrahydrofuran über, welches in diesem Vakuum ungefähr bei 86 siedet.
Das Produkt bildet eine farblose Flüssigkeit, deren spez. Gewicht bei 15 ungefähr um <B>1,208</B> liegt. Chlorgehalt: 21,8%.
Man hält 5-6 cm' vom obigen Produkt, 5 cm 3 ungefähr 99%ige Ameisensäure und 5 g 2-Methyl-4-amino-5-(thioformamido-me- thyl)-pyrimidin während 24 Stunden bei 50 im Thermostat. Zur blass braunen Lösung gibt man ungefähr 45 cm3 abs. Alkohol und 5 cm' ungefähr 20 % Salzsäuregas enthalten den abs. Alkohol hinzu, kocht die Lösung auf und kühlt dann. Nach einigen Stunden Ste hen filtriert man die Vitamin B-Kristalle und wäscht mit abs. Alkohol aus.
Man ge winnt 6 g Vitamin Bi, welches schon in die sem Zustand weiss ist und unter Zersetzung um 246 schmilzt. Chlorgehalt: um 19,8r.
2. Man kocht 13,6 g Acetochlor-propyl- alkohol bezw. dessen von Stevens und Stein beschriebenen Äther (siehe Journal of the American Chemical Society, Volume 62, Seite 1046, rechte Kolonne, Absatz 1 des Ver suchsteils) mit 16 cm' wasserfreier Ameisen säure während 3 Stunden unter Rückfluss; dann destilliert man die Ameisensäure ab. Aus dem Rückstand kann man durch Frak tionieren das 2-Methyl-2-formyloxy-3-chlor- tetrahydrofuran gewinnen.
Es ist aber vor teilhafter, den Rückstand vorerst noch einmal mit wasserfreier Ameisensäure zu erwärmen, dann die Ameisensäure zu entfernen und den Rückstand im Vakuum zu fraktionieren. Man gewinnt etwa 10-12 g 2-Methyl-2-formyl- oxy-3-chlor-tetrahydrofuran.
Man hält 12 cm' vom obigen Produkt, 7.0 cm' ungefähr 90 % ige Ameisensäure und 10 g 2-Methyl-4-a.mino-5-(thioformamido-me- thyl)-pyrimidin während 40 Stunden bei 50 im Thermostat. Man gibt zum Reaktions gemisch 60 cm' abs. Alkohol und 10 cm' un gefähr 20 % Salzsäuregas enthaltenden abs. Alkohol hinzu. Man erhält einen dicken Kri stallbrei, aus feinen Kristallen bestehend. Man erhitzt diesen, kühlt mit Eiswasser und putscht nach Stehen.
Man gewinnt 9 g wei sses Vitamin BI, welches unter Zersetzung um 236 schmilzt.
3. Man gibt zu 16,2 g Acetochlor-buty- rolacton 1,8 cm' Wasser und 16-17 cm' un gefähr 100%ige Ameisensäure und kocht unter Rückfluss während 5 Stunden. Dann destilliert man die Ameisensäure bei vermin dertem Druck ab und destilliert den dun kelfarbigen Rückstand im Vakuum bei 1,5 mm Hg. Man erhält 11-12 g 2-Methyl- 2-formylogy-3-chlor-tetrahydrofuran, wel ches 2270 Chlor enthält.
Man hält 10-12 cm' vom obigen Pro dukt, 10 cm' ungefähr 90 % ige Ameisensäure und 10 g 2-Methyl-4-amino-5-(thioformamido- methyl)-pyrimidin während 40 Stunden bei 50 im Thermostat. Die Aufarbeitung des Reaktionsgemisches kann nach den Methoden der Beispiele 1 und 2 geschehen. Man erhält <B>10,7</B> g Vitamin B1, welches schon in diesem Zustand weiss ist und um 236 schmilzt.
Die in den vorangehenden Beispielen an gegebenen Reaktionsbedingungen können in verschiedener Hinsicht abgeändert werden: So zum Beispiel kann man die Herstellung von Vitamin B, auch bei höherer Temperatur aus- führen, zum Beispiel bei 70 , jedoch durch ein kürzere Zeit andauerndes Erhitzen.

The preparation of the 2-methyl-2-formyloxy-3-chloro-tetrahydrofuran used for the production of vitamin B1 according to the invention, which was previously unknown, can be done by using aceto-chlorobutyrolactone or aceto-chlorine propyl alcohol or 2-methyl-2-oxy-3-chloro-tetrahydrofuran or its ethers, for example those formed with <I> y </I> - A.ceto <I> - y </I> - chloro - propyl alcohol Ether subjected to the action of formic acid,
or 2-methyl-2,3-dichloro-tetrahydrofuran is subject to the action of formic acid salts and / or formic acid. For example, 2-methyl-2-formyloxy-3-chloro-tetrahydrofuran can be produced by allowing the calculated amounts of anhydrous galium formate to act on 2-methyl-2,3-dichloro-tetrahydrofuran.

The product forms a colorless liquid, the spec. Weight at 15 is around <B> 1.208 </B>. Chlorine content: 21.8%.
5-6 cm 'of the above product, 5 cm 3 of approximately 99% strength formic acid and 5 g of 2-methyl-4-amino-5- (thioformamido-methyl) -pyrimidine are kept in a thermostat for 24 hours at 50. About 45 cm3 abs are added to the pale brown solution. Alcohol and 5 cm 'approximately 20% hydrochloric acid gas contain the abs. Add alcohol, boil the solution and then cool. After a few hours of standing, the vitamin B crystals are filtered and washed with abs. Alcohol off.
You win 6 g of vitamin Bi, which is already white in this state and melts around 246 with decomposition. Chlorine content: around 19.8r.
2. 13.6 g of acetochloropropyl alcohol are boiled, respectively. its ether described by Stevens and Stein (see Journal of the American Chemical Society, Volume 62, page 1046, right column, paragraph 1 of the trial part) with 16 cm 'of anhydrous formic acid for 3 hours under reflux; then the formic acid is distilled off. The 2-methyl-2-formyloxy-3-chlorotetrahydrofuran can be obtained from the residue by fractionation.
However, it is more advantageous to first heat the residue again with anhydrous formic acid, then remove the formic acid and fractionate the residue in vacuo. About 10-12 g of 2-methyl-2-formyl-oxy-3-chloro-tetrahydrofuran are obtained.
One keeps 12 cm 'of the above product, 7.0 cm' of about 90% formic acid and 10 g of 2-methyl-4-a.mino-5- (thioformamido-methyl) -pyrimidine for 40 hours at 50 in the thermostat. 60 cm 'abs are added to the reaction mixture. Alcohol and 10 cm 'abs. Containing about 20% hydrochloric acid gas. Add alcohol. A thick crystal slurry is obtained, consisting of fine crystals. This is heated, cooled with ice water and put on after standing.
9 g of white vitamin BI are obtained, which melts by 236 with decomposition.
3. To 16.2 g of acetochloro-butyrolactone, 1.8 cm 'of water and 16-17 cm' of approximately 100% formic acid are added and the mixture is refluxed for 5 hours. The formic acid is then distilled off under reduced pressure and the dark-colored residue is distilled off in vacuo at 1.5 mm Hg. 11-12 g of 2-methyl-2-formylogy-3-chloro-tetrahydrofuran, which contains 2270 chlorine, are obtained .
Keep 10-12 cm 'of the above product, 10 cm' about 90% formic acid and 10 g of 2-methyl-4-amino-5- (thioformamido-methyl) pyrimidine for 40 hours at 50 in the thermostat. The reaction mixture can be worked up by the methods of Examples 1 and 2. <B> 10.7 </B> g of vitamin B1 are obtained, which is already white in this state and melts around 236.
The reaction conditions given in the preceding examples can be changed in various ways: For example, vitamin B can also be produced at a higher temperature, for example at 70, but by heating for a shorter time.
