Verfahren zur Darstellung von f-Alanin, fl-Alanin kann auf verschiedenen Wegen synthetisch gewonnen werden. Ein solcher Weg ist die Behandlung von Bernsteinsäure imid mit Hypobromiden. Neuerdings wurde Cyanessigester katalytisch zu ss-Alaninester hydriert (Berichte der deutschen chemischen Gesellschaft 74 [1941] S. 256).
Nach einer kurzen Angabe (Berichte der deutschen che mischen Gesellschaft 8 [1875] S. 1597) soll fl-Alanin auch .durch Reduktion der Cyan- essigsäure mit Zink und Schwefelsäure ent stehen. Alle diese Verfahren sind für -die technische Darstellung von fl-Alanin unge eignet.
Es wurde nun gefunden, dass ss-Alanin mit vorzüglicher Ausbeute und in technisch leicht ausführbarer Weise dadurch gewon nen werden kann, dass man cyanessigsaure Salze in ammoniakalischer Lösung bei Ge genwart eines Nickelkatalysators hydriert. Man kann sowohl in wässeriger als auch in alkoholischer Lösung arbeiten. Vorteilhaft ist ,die Verwendung der cyanessigsauren Alkali- salze, besonders des Kaliumsalzes, da in die sem Falle .die nachträgliche Abtrennung des Alkalis in sehr einfacher Weise gelingt.
Die Vorteile des neuen Verfahrens gegen über den bekannten Darstellungen von ss-A,la- nin liegen auf der Hand. Einerseits sind die Cyanessigsäure und ihre Salze sehr leicht zugänglich.
Gegenüber der Darstellung aus Cyanessiges.ter liegt der wesentliche Fort schritt weniger in der Verwendung von Sal zen der Cyanessigsäure als in der Möglich keit, Nickelkatalysatoren an .Stelle des kost spieligen Platinoxyds, das bei der Hydrie rung des Esters in saurer Lösung notwendig ist, zu benützen. Ausserdem wird nach dem neuen Verfahren in alkalischer Lösung ge arbeitet, wodurch sich die Gefässfrage wesentlich vereinfacht.
Die Hydrierung der Cyanessigsäure bezw. ihrer Salze in ammoniakalischer Lösung mit Nickelkatalysator verläuft sehr rasch bei Anwendung von hohem Druck. Sie gelingt aber auch unter gewöhnlichem bezw. wenig erhöhtem Druck, nur geht sie in diesem Falle wesentlich langsamer vor sich. Auch die Temperatur kann innerhalb weiter Grenzen schwanken, ohne dass durch ihre Erhöhung wesentlich mehr als eine Beschleunigung er zielt wird. .
<I>Beispiel 1:</I> 61,5 !Gewichtsteile cyanessigsaures Ka lium werden im Rühra.utoklaven mit 500 Raumteilen<B>18%</B> ammoniakhaltigem 'Methyl alkohol, 5 Gewichtsteilen Raneynickel und Wasserstoff hydriert. Bei 100 Atmosphären und 90 wird die berechnete Menge Wasser stoff in etwa<B>15</B> 'Minuten aufgenommen. Nach Abfiltrieren des Katalysators wird das Fil trat im Vakuum auf 'j,, seines Volumens ein gedampft, dann mit 35 Gewichtsteilen Eis essig und 100 Teilen Wasser versetzt und bei nahe zur Trockne eingedampft.
Versetzt man nun mit Alkohol, so beginnt alsbald die Aus scheidung des ss-Alanins, das nach einmali gem Umkristallisieren vollkommen rein ist. Die Ausbeute beträgt über 75ö der Theorie. Beispiel <I>2:</I> <B>21,25</B> Gewichtsteile Cyanessigsäure wer- den, in 250 Gewichtsteilen 28 öigem wässe rigem Ammoniak gelöst, mit 5 Gewichtsteilen Nickelkatalysator versetzt und mit Wasser- Stoff hydriert.
Nach dem Abfiltrieren des Nickels und dem Entfernen des gelösten Nickels mit Schwefelwasserstoff erhält man beim Eindampfen der filtrierten wässerigen Lösung das ss-Alanin.
<I>Beispiel 3:</I> 42,6 Gewichtsteile Cyanessigsäure wer den in 50 Teilen Wasser gelöst und mit Kali lauge von 50ö neutralisiert. Unter Zusatz von. 450 CTe-,vichtsteilen 30 % igem wässerigem -zlmmoniak und 20 Gewichtsteilen Nickel katalysator wird im Schüttelautoklaven bei 30 Atmosphären und 20 mit Wasserstoff hydriert. Nach beendeter Reduktion (4 bis 5 Stunden)
und Entfernen des Katalysators wird teilweise eingedampft und mit 45 Ge- wiehtsteilen Eisessig versetzt. Die Kristalli sation<I>des ss-</I> Ahinins aus der weiter konzen trierten Lösung erfolgt auf Zusatz von Alko hol.
