ohlenstoffseitenkette mit einer Carboxylgr uppe und im Polyhydrophen- anthrenrest Acyloxy- und/oder Oxogruppen enthalten, in die entsprechenden Säurechloride überführt, Diazomethan zur Einwirkung bringt und die entstandenen Diazoketone mit Säuren zu den entsprechenden Estern umsetzt. Die Überführung der freien Säure in das Säure chlorid kann nach den üblichen Methoden erfolgen, z.
B. mit Hilfe von Thionylchlorid, Phosphortrichlorid oder Phosphorpentachlorid. Als Veresterungsmittel für die Diazoketone lässt sich jede beliebige organische oder an organische Säure verwenden. Es kommt vor wiegend Essigsäure in Betracht, jedoch sind z. B. auch Schwefelsäure, Propionsäure, Öl säure, Bernsteinsäure oder Benzoesäure ver wendbar. Das Verfahren wird zweckmässigerweise wie folgt ausgeführt Das Säurechlorid, welches aus der als Ausgangsstoff dienenden Carbonsäure, z. B.
durch Behandlung mit Thionylchlorid erhalten wird, wird in überschüssige ätherische Di- azomethanlösung eingetragen und so lange stehen gelassen, bis die Stickstoffentwicklung beendet ist. Meistens scheidet sich das Di- azoketon dann kristallin ab. Das Diazoketon wird sodann z. B. durch Kochen mit Eisessig in das entsprechende Acetoxymethylketon über geführt.
Durch Verseifung kann daraus die freie Oxyverbindung gewonnen werden.
Die so erhaltenen neuen Verbindungen sollen als solche in der Therapie Verwendung finden.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines Triketonor- cholanyl-acetoxymethyl-ketons, welches da durch gekennzeichnet ist, dass man Dehydro- cholsäurehalogenid mit Diazomethan zu dem entsprechenden Diazoketon umsetzt und dieses Produkt mit Eisessig erhitzt.
<I>Beispiel:</I> 10g Dehydrocholsäure werden mit 30 g 5 Thionylchlorid und 300 cm'' Benzol zum Sieden erhitzt. Es wird lebhaft Salzsäure entwickelt. Die Substanz geht bald in Lösung. Nach zwei Stunden wird die dunkel gefärbte Reaktions- lösung im Vakuum abdestilliert, wobei Kri stallisation des Rückstandes eintritt. plan ver reibt die Kristalle des Dehydrocholsäure- chlorids mit trockenem Aceton und saugt ab. Das so gereinigte weisse Säurechlorid schmilzt bei '08-209 unter Zersetzung.
Hau löst das Delivdrocliolsäureclilorid in trockenem Aceton und giesst in überschüssige ätherische Diazomethanlösung. Es wird leb haft Stickstoff entwickelt. Nach einiger Zeit scheiden sich feine Nadeln ab, die dann nach Stehen über Nacht abgesaugt und im Exsik- kator getrocknet werden.
Das Diazoketon wird in trockenem Eis essig gelöst und '/# Stunde unter Rückfluss zum Sieden erhitzt. Dabei findet Abspaltung von Stickstoff statt, die volumetrisch verfolgt werden kann. Man lässt erkalten und spritzt mit Wasser an. Das Triketonorcholanyl- acetoxymethyl-keton scheidet sich in langen Nadeln aus, die aus Aceton, Methanol und verdünntem Eisessig, eventuell unter Ver wendung von Tierkohle umgelöst werden. Schmelzpunkt 222 . Ausbeute 3,5 g.
