Verfahren zur Herstellung von Testosteron. Vorliegende Erfindung betrifft ein Ver fahren zur Überführung von Diketonen der Cyclopentanopolyhydrophenanthrenreihe in die entsprechenden Ogyketone, indem man in den als Ausgangsmaterial dienenden Diketonen eine Ketogruppe blockiert, die so erhaltene Verbindung der Einwirkung von Hydrie- rungsmitteln unterwirft und dann die Blok- kierung der Ketogruppe wieder aufhebt.
Als Ausgangsmaterialien kommen die ge sättigten und ungesättigten Diketone der Cyclopentanopolyhydrophenanthrenreihe in Betracht, wie z. B. die Androstandione, Preg- nandione, Allo-Pregnendione, Androstendione, Pregnendione usw.
Gegenstand des vorliegenden Patentes ist die Darstellung von Testosteron aus Andro- stendion. Dieses Verfahren ist besonders wertvoll, da es bisher nicht bekannt war, direkt vom Androstendion auf rein chemi schem Wege zum Testosteron zu gelangen.
Das erfindungsgemässe Verfahren ist da durch gekennzeichnet, dass man d4"-Andro- stendion-(3,17) in 3-Stellung mit einem Ke- tonreagens umsetzt, im so erhaltenen Pro dukt durch Behandlung mit einem Reduk tionsmittel die freie getogruppe in die OH- Gruppe überführt und dann aus dem so er haltenen Testosteronderivat durch hydroly- sierende Mittel das Testosteron freisetzt,
das als Arzneimittel oder als Zwischenprodukt zur Herstellung von Arzneimitteln Verwen dung finden soll.
Für die Überführung des Androstendions in sein Monoderivat sind als Ketonreageilzien die üblichen Stoffe, die Ketone in Derivate überzuführen vermögen, und aus denen durch Hydrolyse die freie Ketongruppe zurück gebildet werden kann, wie z. B. Semi- karbazid, Phenylhydrazin bezw. seine Sub- stitutionsprodukte, wie z. B. Nitro-phenyl- hydrazin, Hydrogylamin und dergleichen ge eignet.
Die erwähnten Ketonreagenzien haben die gemeinsame Eigenschaft, mit Ketonen- unter Wasserabspaltung Kondensationsprodukte zu bilden, die wieder aufgespalten werden kön nen, wobei die freien Ketone zuriickgebildet werden. Sie sind zum Beispiel beschrieben in Houben "Methoden der Organischen Che mie", 3. Aufl., z. Band, S. 572-573.
Um in dem Monoderivat die freie Keto- gruppe zur Hydroxylgruppe umzuwandeln, wird es einer Behandlung mit reduzierenden Mitteln unterworfen, die, wie sich zeigte, die blockierte Ketogruppe im wesentlichen um angegriffen lassen. Als besonders geeignet hat sich zum Beispiel die Reduktion mit Na trium in Alkohol erwiesen, jedoch können auch andere an sich bekannte Verfahren Ver wendung finden. So kann zum Beispiel die Reduktion nach dem Verfahren von Meer wein und Ponndorf mit Alkoholen in Gegen wart von Aluminium- oder Magnesium- Alkoholaten durchgeführt werden.
Um aus diesem Reduktionsprodukt das zugehörige Oxyketon (Testosteron) zu er halten, wird dasselbe, vorzugsweise ohne wei tere Reinigung, hydrolytisch aufgespalten, vorzugsweise mit Säure, z. B. mit Salz säure, Schwefelsäure, Oxalsäure usw. am besten in alkoholischer Lösung.
Falls erforderlich, kann das Hydrierungs- produkt in bekannter Weise gereinigt wer den, z. B. durch Umkristallisation, Destilla tion im Hochvakuum oder indem man es der Behandlung mit besonderen Ketonreagenzien unterwirft, besonders dem sogenannten Girard Reagenz (Trimethyl-ammonium-Essigsäure- hydra,zid-hydrochlorid) usw.
Das erfindungsgemässe Verfahren sei an Hand des nachstehenden Beispiels noch näher erläutert: Beispiel: Zu einer Lösung von 1.0 g A",-Andro- stendion-(3,17) vom Schmp. 169 in 50 cm' Äthanol wurde in der Kälte die 1 llol ent sprechende Menge Semikarbazidacetat (aus 386 mg Semikarbazidhydrochlorid und 285 mg wasserfreiem Natriumaceta.tes in wenig Wasser bereitet) gegeben.
Nach halb stündigem gelinden Sieden wurde bis zur be b nnenden Trübung mit heissem Wasser (etwa 100 cm') versetzt und über Nacht im Eis schrank aufbewahrt.. Es schieden sich 1,02 g Monoseinika.rbazon vom Zersetzungspunkt 234' ab; nach mehrmaligem Umkristalli- sieren aus verdünntem und reinem Alkohol bildete es entweder einzelne Drusen feiner Kristallnadeln oder schwere, stark licht brechende, von regelmässigen Flächen be grenzte kugelige Kristalle, die konstant bei -)45' unter Zersetzung schmolzen.
Das Mono semikarba,zon ist in geringem Grade löslich in Chloroform, Eisessig. Pyridin und Piperi- din; in der Ilitze auch in Äthyl-, Isopropyl- und Amylalkohol, sowie in Dioxan. Sehr schwer löslich oder unlöslich ist. es in Äther, Benzol, Essigester. Butanol, Cy clohexan, 3lalonester und Methylbenzoat.
300 mg Monosemikarbazon vom Schmelz punkt 245 wurden in 7,5 ein' a.bs. Isopro- panol gelöst und in der Siedehitze während 25 Minuten mit Natrium bis fast zur Sätti gung versetzt. Das heisse Gemisch wurde schnell ausbegossen, vom unverbrauchten Na: trium befreit, die, erstarrte Kristallmasse in Wasser gelöst und unter Kühlung mit v er- dünnter Schwefelsäure bis zur schiwach alka lischen Reaktion neutralisiert.
Das Reak tionsprodukt wurde sorgfältig mit Äther oder Chloroform ausgezogen und aus verdünntem Alkohol kristallisiert erhalten. Ausbeute 245 mg vom Schmelzpunkt l60-185 . In entsprechender Weise gelangt die Reduktion auch mit Natrium in Äthanol.
245 mg des bei 160-l85 schmelzenden Reduktionsproduktes wurden in einer Mi schung von 20 cm' Alkohol und 10 cm' 2-n wässeriger Schwefelsäure eine halbe Stunde gekocht. Nach üblicher Aufarbeitung durch Ausäthern wurde das Produkt in wenig Es sigester gelöst und mit niedrig siedendem Petroläther angespritzt. Es kristallisierten 24 mg in schönen, farblosen Drusen, die nach Umkristallisieren aus Essigester-, Petrol- äther, Essigester-Hexan und verdünntem Al kohol bei 151-156 schmolzen.
Die optische Drehung dieses 3lisehkristallisates betrug (a)n@ --f- 79,2 (i. Alkohol; unter der An nahme, dass es aus Testosteron ((ah = 104 ) und Androstendiol ja]\ ü -55,5o) bestand, berechnet, sich ein Testosterongehalt von ungefähr 85 %. Der Mischschmelzpunkt dieses Kristallisates mit reinem Testosteron zeigt keine Depression.
44 mg des Mischkristallisates in 1 cm' Alkohol wurden mit einer Lösung von etwa 50 mg des Chlorides vom Trimethyl-ammo- nium-essigsäure-hydrazid in 4 ein' Alkohol vereinigt und nach Zusatz von 0,5 cm' Eis essig eine halbe Stunde in gelingem Sieden ge halten. Die Reaktionslösung wurde in Was ser gegossen, in dem zur annähernden Neu tralisation der Essigsäure 420 mg Na2C03 gelöst waren.
Nach dem Entfernen des keton- freien Anteils durch Ausäthern wurde die wässrige Lösung mit verdünnter Schwefel säure angesäuert; sie trübte sich und wurde nach fünfstündigem Aufbewahren bei Zim mertemperatur ausgeäthert. Es wurde reines Testosteron erhalten, das nach Umkristalli- sieren aus verdünntem Alkohol bei 153 bis 154' schmolz.
Method of making testosterone. For the conversion of androstenedione into its monoderivative, the usual substances that are able to convert ketones into derivatives and from which the free ketone group can be formed back by hydrolysis are used as ketone reagents, such as. B. Semi- carbazide, phenylhydrazine or. its substitution products, such as B. nitro-phenyl hydrazine, hydrogylamine and the like ge suitable.
The ketone reagents mentioned have the common property of forming condensation products with ketones with elimination of water, which condensation products can be broken down again, the free ketones being reformed. They are described, for example, in Houben "Methods of Organic Chemistry", 3rd edition, z. Volume, pp. 572-573.
In order to convert the free keto group to the hydroxyl group in the mono derivative, it is subjected to a treatment with reducing agents which, as has been shown, can substantially attack the blocked keto group. Reduction with sodium in alcohol, for example, has proven to be particularly suitable, but other processes known per se can also be used. For example, the reduction can be carried out according to the Meerwein and Ponndorf process with alcohols in the presence of aluminum or magnesium alcoholates.
In order to keep the associated oxyketone (testosterone) from this reduction product, the same is hydrolytically split, preferably without further purification, preferably with acid, eg. B. with hydrochloric acid, sulfuric acid, oxalic acid, etc. is best in alcoholic solution.
If necessary, the hydrogenation product can be purified in a known manner, for. B. by recrystallization, distillation in a high vacuum or by subjecting it to treatment with special ketone reagents, especially the so-called Girard reagent (trimethyl ammonium acetic acid hydra, zid hydrochloride), etc.
The process according to the invention will be explained in more detail with reference to the following example: Example: To a solution of 1.0 g of A ", - androstenedione- (3.17) of melting point 169 in 50 cm 'of ethanol, the 1 llol appropriate amount of semicarbazide acetate (prepared from 386 mg of semicarbazide hydrochloride and 285 mg of anhydrous sodium acetates in a little water).
After half an hour of gentle boiling, hot water (approx. 100 cm ') was added until the cloudiness was quenching and the mixture was stored in the ice cabinet overnight. 1.02 g of monoseinika.rbazon separated from the decomposition point 234'; after repeated recrystallization from dilute and pure alcohol it either formed individual drusen of fine crystal needles or heavy, strongly refractive, spherical crystals delimited by regular surfaces, which constantly melted at -) 45 'with decomposition.
The mono semikarba, zon is slightly soluble in chloroform, glacial acetic acid. Pyridine and piperidine; in the Ilitze also in ethyl, isopropyl and amyl alcohol, as well as in dioxane. Is very sparingly soluble or insoluble. it in ether, benzene, ethyl acetate. Butanol, cyclohexane, 3lalonester and methyl benzoate.
300 mg of monosemicarbazone, melting point 245, were in 7.5 a.bs. Isopropanol dissolved and sodium was added to almost saturation at the boiling point for 25 minutes. The hot mixture was quickly poured out, freed from unused sodium, the solidified crystal mass was dissolved in water and, while cooling, was neutralized with dilute sulfuric acid until a weak alkaline reaction was achieved.
The reaction product was carefully extracted with ether or chloroform and obtained crystallized from dilute alcohol. Yield 245 mg, melting point 160-185. In a corresponding manner, the reduction also takes place with sodium in ethanol.
245 mg of the reduction product melting at 160-185 were boiled for half an hour in a mixture of 20 cm 'of alcohol and 10 cm' of 2N aqueous sulfuric acid. After the usual work-up by exothermic means, the product was dissolved in a little ethyl acetate and sprayed on with low-boiling petroleum ether. 24 mg crystallized in beautiful, colorless drusen which, after recrystallization from ethyl acetate, petroleum ether, ethyl acetate-hexane and dilute alcohol, melted at 151-156.
The optical rotation of this 3lisehkristallisates was (a) n @ --f- 79.2 (i. Alcohol; assuming that it consisted of testosterone ((ah = 104) and androstenediol yes] \ ü -55.5o) , calculated, a testosterone content of about 85%. The mixed melting point of this crystallizate with pure testosterone shows no depression.
44 mg of the mixed crystals in 1 cm 'of alcohol were combined with a solution of about 50 mg of the chloride of trimethylammonium-acetic acid hydrazide in 4 an' alcohol and after adding 0.5 cm 'of glacial acetic acid for half an hour successful boiling. The reaction solution was poured into water in which 420 mg of Na2CO3 had been dissolved to approximately neutralize the acetic acid.
After the ketone-free portion had been removed by ether, the aqueous solution was acidified with dilute sulfuric acid; it became cloudy and, after storing for five hours at room temperature, was etherified. Pure testosterone was obtained which, after recrystallization from dilute alcohol, melted at 153 to 154 '.
