Verfahren zur Abtrennung der Begleitstoffe mit Lipoideharakter und ges Adrenalins von dem Nebennierenrindenhormon. ' Extrakte, die das Nebennierenrindenhor- mon enthalten, müssen von den darin beson ders reichlich vorhandenen Begleitstoffen von Lipoidcharakter einerseits und dem Adre- naIin anderseits befreit werden, da diese durch ihren störenden Geschmack und ihre Giftwirkung die orale oder parenterale Ver abreichung des Nebennierenrindenhormons erschweren.
Die bekannten Verfahren zur Reinigung von Extrakten, die das Nebennierenrinden- hormon enthalten, bestehen in selektiver Ex traktion bezw. fraktionierter Fällung der Extrakte mit organischen Lösungsmitteln. Diese Verfahren sind langwierig und erfor dern grosse Mengen organischer Lösungs- mittel.
Die vorliegende Erfindung betrifft ein Verfahren zur Abtrennung der Begleitstoffe mit Lipoidcharakter und des Adrenalins von dem Nebennierenrindenhormon, welches da- durch gekennzeichnet ist, dass man in wäss- rigen Extrakten, welche die genannten Stoffe enthalten, einen Niederschlag einer Ferrover- bindung, wie beispielsweise Ferrohydroxyd, Ferrophosphat,
Ferrokarbonat oder Ferro- sulfid erzeugt, und die an diesem Nieder schlag adsorbierten Begleitstoffe abtrennt. Der Niederschlag adsorbiert, wie Versuche ergaben, die Begleitstoffe lipoidartiger Natur und das Adrenalin, so dass in der Lösung das reine Nebennierenrindenhormon verbleibt. Nach Abtrennung des erwähnten Nieder schlages wird gegebenenfalls die Lösung konzentriert.
Auf diese Weise erhält man ein stark wirksames und gut verträgliches Nebennierenrindenhormon-Präparat.
Für die Entfernung von Eiweiss aus Hor monlösungen ist die Erzeugung von Nieder schlägen von Ferroverbindungen schon be kannt. Dass aber auch lipoidartige Begleit- stoffe und Adrenalin unter Verwendung von Niederschlägen von Ferroverbindungen von dem Nebennierenrindenhormon abgetrennt werden können, war nicht vorauszusehen.
Für die Gewinnung von Ausgangslösun gen für das erfindungsgemässe Verfahren können, wie üblich, die Nebennieren mit ver dünntem Alkohol extrahiert. werden. Es lassen sieh hierzu sowohl zerkleinerte frische Nebennierenrinden als auch Nebennierenrin- dentrockenpulver verwenden; aber auch die ganzen Nebennieren, frisch oder getrocknet. können als Ausgangsmaterial benutzt werden.
Die verdünnt allzoholischen Lösungen werden zweckmässig durch Eindampfen im Vakuum von denn Alkohol befreit, und dann in den zuriickbleibenden wässrigen Lösungen die er findungsgemässe Reinigung durch Erzeugung eines Niederschlages einer Ferroverbindung durchgeführt. Da in sauren Lösungen die Niederschläge von Ferroverbindungen löslich sind, ist es zweckmässig, für das erfindungs gemässe Verfahren neutrale bezw. schwach alkalische Lösungen zu verwenden.
Der be vorzugte p11-Bereich für das vorliegende Ver fahren ist pH 7-9; gegebenenfalls kommt auch noch eine etwas stärkere alkalische Re aktion, wie pH 10, in Betracht; doch ver grössert sich mit zunehmendem Alkali-ehalt die Gefahr der Schädigung des Hormons.
Als günstigste Arbeitstemperatur kommt für das erfindungsgemässeVerfahren Zimmer temperatur in Betracht. Aber auch bei schwach erhöhter Temperatur bis etwa<B>50'</B> C ist das erfindungsgemässe Verfahren ausführ- bar. Bei noch höheren Temperaturen besteht die Gefahr der Schädigung des Hormons.
Wenn die erfindungsgemässe Reinigung durch Erzeugung eines Niederschlages voll Ferrokarbonat oder Ferrophosphat bewirkt wird, so entsteht je nach dem pH der Lösung entweder ein neutrales oder ein basisches Karbonat bezw. Phosphat.
Will man die erfindungsgemäss gewonnenen gereinigten Präparate noch von anorganischen Salzen be freien, so kann man die wässrigen Lösungen des Hormons, gegebenenfalls nach Norheri- gem Einengen, in an sieh bekannter Weise mit organischen Lösungsmitteln, die mit Wasser nicht mischbar sind, wie beispiels weise Benzol, Äther, Chloroform. Diehlor- < ithylen oder Essigester extrahieren, wobei das Hormon in die organischen Lösungsmittel übergeht, die anorganischen Salze dagegen in der wässrigen Lösung zurückbleiben.
Zur näheren Veranschaulichung der Er findung dienen die folgenden Beispiele: Beispiel <I>1:</I> 40 kg frische Nebennieren werden mehr fach mit 70%igem Alkohol ausgezogen. Die vereinigten Extrakte "v-erden im Vakuum bei niedriger Temperatur zur Befreiung von Al- koliol auf etwa 10 Liter eingeengt.
In die konzentrierte wässrige Lösung rührt man eine konzentrierte Lösung von 2 kg Ferrosulfat (Eisenvitriol) ein und bringt dann durch Zu gabe von Caleium bezw. Barium- oder Stron- tiumbydroxyd das pH auf 8,5-9,0. Dabei entsteht eine voluminöse Fällung von Ferro- hydroxyd, die alle störenden Begleitstoffe von Lipoideharakter und das Adrenalin mit sieh niederreisst.
Man entfernt den Nieder schlag durch Filtrieren oder Zentrifugieren und bringt das klare Filtrat mit Phosphor säure auf ein pH von 6,5. Dabei tritt eine leichte Fällung von Calciumphosphat bezw. Barrom- oder Strontiumphosphat ein,
die <B>.</B> a bfiltriert und verworfen wird. Der auf diese reise erhaltene Extrakt wird nun auf das gewünschte Volumen eingeengt und ist in dieser Form für orale oder parenterale Ver abreichung geeignet.
Beispiel <I>2:</I> 10 kg Nebennierenpulver, gewonnen durch Vakuumtrocknung. werden mehrfach mit 70%igem Alkohol ausgezogen. Die -vereinig ten Ausziige werden zur Entfernung des Al kohols im Vakuum auf etwa 9 Liter einge engt. Das wässrige Konzentrat wird mit einer konzentrierten wässrigen Lösung von 1400 g Ferrochlorid versetzt. und mit Natronlauge bezw. Kalilauge oder Ammoniak auf pH 8,5 bis 9 gebracht.
Die dabei entstehende Fällung von Ferrohy droxyd wird durch Abfiltrieren oder Abschleudern entfernt und verworfen. Das Filtrat wird mit Phosphorsäure auf pH 6,5 gebracht, die dabei eintretende Fäl lung aus der Lösung abfiltriert und ver worfen. Das Filtrat. enthält das Nebennieren rindenhormon frei von störenden Lipoiden und Adrenalin. Zur Entfernung der gelösten Elektrolyte wird es in bekannter Weise wiederholt mit Benzol ausgezogen, wobei das Hormon in die Benzolphase geht. Die verei nigten Benzolauszüge werden im Vakuum durch Abdampfen vom Benzol befreit.
Der Abdampfrück,stand kann in einer beliebigen Menge von 0,9 % iger Kochsalzlösung oder in einer analogen für Injektionszwecke ver wendbaren isotonischenLösung aufgenommen werden. Er ist gut verträglich und hoch wirksam.
Beispiel <I>3:</I> 40 kg frische Nebennierenrinden werden, wie in Beispiel 2 angegeben, mit Alkohol ausgezogen. Nach Entfernen des Alkohols wird das ivä.ssrige Konzentrat mit einer kon zentrierten Lösung von 1000 g Ferrochlorid versetzt und mit Natrium- bezw. Kalium karbonat auf ein pH von 8,5-9 gebracht. Die auf diese Weise entstehende Fällung von Ferrokarbonat schlägt Adrenalin und stö rende Lipoide aus der Lösung nieder.
Sie wird durch Abfiltrieren oder Abschleudern abgetrennt und verworfen. Das Filtrat wird mit Phosphorsäure auf pH 6,5 gebracht, der dabei ausfallende Niederschlag abfiltriert. Das Filtrat enthält das Nebennierenrinden- hormon frei von Lipoiden und Adrenalin und kann in dieser Form oral oder parenteral ver abreicht werden.
<I>Beispiel</I> 10 kg Nebennierenrindenpulver werden, wie in Beispiel 1 angegeben, extrahiert. Nach Abdampfen des Alkohols wird das wässrige Konzentrat mit 1350 g Ferrosulfat versetzt, worauf man mit Natriumphosphatlösung das pH auf 8,5-9 bringt. Bei diesem pH erfolgt eine kräftige Ausscheidung von Ferrophos- phat, die in gleicher Weise wie in den Bei spielen 1-3 angegeben, Adrenalin und alle andern störenden Begleitstoffe niederschlägt.
Die weitere Verarbeitung erfolgt wie in Bei- spiel 2 angegeben, jedoch mit dem Unter schied, dass zur Gewinnung des salzfreien Hormons die wässrige Lösung des Neben nierenrindenhormons nach dem Ansäuern mit Phosphorsäure mit Äther oder Chloroform ausgeschüttelt wird. Das Hormon geht in die Äther- bezw. Chlorofor-mphase und kann dar aus durch Abdampfen in hochgereinigter Form gewonnen werden.
<I>Beispiel 5:</I> 10 kg Nebennierenrindenpulver werden, wie in Beispiel 1 beschrieben, ausgezogen. Nach dem Entfernen des Alkohols durch Ab dampfen im Vakuum wird das wässrige Kon zentrat mit 1350g Ferrosulfat und mit einer Auflösung von 1280 g Natriumsulfid in Wasser versetzt. Gegebenenfalls wird nun mit Natronlauge alkalisch gemacht.
Der ent stehende Niederschlag von Ferrosulfid, der Adrenalin sowie andere störende Substanzen lipoidartiger Natur aus der Lösung nieder reisst, wird durch Filtrieren oder Zentrifu gieren abgetrennt. Anschliessend macht man mit Phosphorsäure lakmussauer, entfernt den überschüssigen Schwefelwasserstoff auf üb liche Waise durch Durchleiten von Stickstoff, v=orauf man die gereinigte Lösung des Hor mons, wie in den Beispielen 1 und 4 beschrie ben, weiterbehandelt.

Ferro carbonate or ferrous sulfide is produced, and the accompanying substances adsorbed on this precipitate are separated off. Experiments have shown that the precipitate adsorbs the accompanying substances of a lipoid-like nature and adrenaline, so that the pure adrenal cortex hormone remains in the solution. After the above-mentioned precipitate has been separated off, the solution is optionally concentrated.
In this way a highly effective and well tolerated adrenal cortex hormone preparation is obtained.
For the removal of protein from hormone solutions, the generation of precipitates from ferrous compounds is already known. However, it could not be foreseen that lipoid-like accompanying substances and adrenaline could also be separated from the adrenal cortex hormone using precipitates of ferrous compounds.
To obtain starting solutions for the process according to the invention, the adrenal glands can, as usual, be extracted with diluted alcohol. will. For this purpose, both crushed fresh adrenal cortex and dry adrenal cortex can be used; but also the whole adrenal glands, fresh or dried. can be used as starting material.
The diluted alcoholic solutions are expediently freed from the alcohol by evaporation in vacuo, and then the purification according to the invention is carried out in the remaining aqueous solutions by generating a precipitate of a ferrous compound. Since the precipitates of ferrous compounds are soluble in acidic solutions, it is appropriate for the process according to the invention to be neutral respectively. use weakly alkaline solutions.
The preferred p11 range for the present process is pH 7-9; if necessary, a somewhat stronger alkaline reaction, such as pH 10, can also be used; but the greater the alkali content, the greater the risk of damage to the hormone.
The most favorable working temperature for the process according to the invention is room temperature. The evaporation residue can be taken up in any amount of 0.9% saline solution or in an analogous isotonic solution that can be used for injection purposes. It is well tolerated and highly effective.
Example <I> 3: </I> 40 kg of fresh adrenal cortex are extracted with alcohol, as indicated in example 2. After removing the alcohol, the ivä.ssrige concentrate is mixed with a concentrated solution of 1000 g of ferrochloride and treated with sodium or. Potassium carbonate brought to a pH of 8.5-9. After the alcohol has evaporated, 1350 g of ferrous sulfate are added to the aqueous concentrate, whereupon the pH is brought to 8.5-9 with sodium phosphate solution. At this pH there is a strong excretion of ferrophosphate which, in the same way as in Examples 1-3, precipitates adrenaline and all other disturbing accompanying substances.
Further processing takes place as indicated in example 2, with the difference that the aqueous solution of the adrenal cortex hormone is extracted with ether or chloroform after acidification with phosphoric acid to obtain the salt-free hormone. The hormone goes into the ether resp. Chloroform phase and can be obtained from it in a highly purified form by evaporation.
<I> Example 5: </I> 10 kg of adrenal cortex powder are extracted as described in example 1. After the alcohol has been removed by evaporation in vacuo, the aqueous concentrate is treated with 1350 g of ferrous sulfate and a solution of 1280 g of sodium sulfide in water.
