Description
Verfahren zur Herstellung von Vaccinen. Es sind Verfahren bekannt, um aerobe und anaerobe Bakterien durch Oxydations mittel abzutöten. Diese Verfahren haben je doch den Nachteil, dass darnach hergestellte Vaecine nicht voll wirksam sind, da man bis jetzt stets soviel Oxydationsmittel zusetzte, dass zwangsläufig auch ein Teil der Anti gene, .die sehr empfindlich sind, zerstört wurde.
Ferner ist bekannt, zur Immunisierung Kulturfiltrate zu verwenden, die zwar frei sind von lebenden Bakterien und Bakterien- leibern, die aber gerade deshalb nicht alle antigenen Stoffe enthalten, so dass denn auch mit den Produkten dieser Verfahren keine 100 ö ixe Immunität erzielt werden kann.
Vorliegende Erfindung betrifft ein Ver fahren zur Herstellung von Vaccinen, z. B. aus aeroben oder anaeroben Bakterien, das dadurch gekennzeichnet ist, dass man in Bak terienkulturen den Bakteriengehalt anrei chert, und alsdann die so erhaltenen Konzen trate mit Mitteln behandelt, welche die Bazil- len unter Schonung der Antigene, durch Ein wirkung auf den Atmungsmechanismus der Zellen abtöten.
Zur Durchführung des erfindungsgemä ssen Verfahrens werden die Bakterien zum Beispiel auf optimalen Nährboden gezüchtet und alsdann auf irgendeine Weise, z. B. durch Absetzen, konzentriert.
Da zu den lebenswichtigsten und beim Proliferationstoffwechsel am stärksten betei ligten Apparaten der Bakterienzelle das Atmungssystem gehört, wurde nachSub stanzen gesucht, welche insbesondere den Atmungschemismus der Zelle im weitesten Sinne stören müssen. Dabei ist der Begriff der Atmung sehr weit zu fassen, im Sinne von Wieland.
Wenn es gelingt, durch Ein führung eines kräftigen Oxydations- oder Re- duktionsmittels die Oxydoreduktionsprozesse zu stören, muss die Proliferation der Zellen in Frage gestellt sein. Wenn die hierzu ver wendeten Substanzen - Oxydations- und Re duktionsmittel - zugleich auch andere Systeme in der Bakterienzelle schädigen, wird gleichzeitig bei geeigneter niedriger Konzen tration auch eine langsame Abtötung der Zelle vor sich gehen.
Aus der Gruppe der hierfür in Frage kommenden Oxydationsmittel seien genannt Superoxyde, wie H:0, und Chlorpräparate, aus der Gruppe der in Frage kommenden Reduktionsmittel, Aldehyde und Nitrite.
Die Verwendung von Formalin zur Ent giftung von Toxinen und zur Abtötung von Bakterien ist bekannt. Hingegen ist es neu, sehr niedrige Konzentrationen und eine lange Einwirkungsdauer anzuwenden, wodurcheine sehr schonende Umwandlung der Bakterien kolloide eintritt, derzufolge die Zellen ster ben, das antigene Vermögen jedoch in hohem Masse erhalten bleibt. Während frühere Un tersucher verhältnismässig hohe Konzentra tion von Formaldehyd oder von Oxydations mitteln und nur kurze Zeit, z.
B. 12 bis 24 Stunden, einwirken liessen, und die Beurtei- lung der Verwendbarkeit eines derartigen Materials, von dem nach dieser Zeit erreich ten Abschwächungs-, Abtötungs- und Um änderungsgrad abhängig machten, wird man bei Ausführung des erfindungsgemässen Ver fahrens mittels Formaldehyd oder Oxyda tionsmitteln diese in möglichst geringen Kon zentrationen verwenden,
dabei aber die Stoffe so lange auf die Bakterien einwirken lassen, bis letztere im Kultur- und Tierversuch als abgetötet gelten müssen. Die Einwirkungs- zeit kann dabei 1 bis 2 Wochen betragen.
Das Verfahren ,gemäss der Erfindung kann vorteilhafterweise auch bei der Rer- stellung von Mischvaocinen gegen mehrere Krankheiten Verwendung finden. Ein Bei spiel hierfür ist die Herstellung eines Misch vaccins gegen Rauschbrand und Malignes Oedem, bei der wie in nachstehendem Bei spiel angegeben, gearbeitet werden kann.
Beispiel: Auf optimalen Nährböden getrennt ge züchtete Kulturen von Rauschbrandbakterien und Bakterien des Malignen Oedeme werden durch Absetzen konzentriert, wonach man sie zur Schädigung des Atmungssystemes der Bakterien mit Formaldehyd behandelt.
Es hat sich dabei gezeigt, dass zur Ab tötung der Rauschbrandbaziilen die minimale nützliche Konzentration etwa 0,1 bis 0,6%ö für Maligne Oedembazillen aber etwa 0,5 bis 1,0,% Formaldehyd als 40%ige wässerige Formaldehydlösung ist.
Diese Konzentratio nen lässt man so lange auf die oben erwähn ten Kulturbodensätze einwirken, bis durch Tier- oder Kulturversuch die Abtötung der Mikroorganismen demonstriert werden .kann, was je nach der Temperatur und Konzentra tion in 8 bis,14 Tagen erreicht ist. Erst jetzt werden die beiden Kulturen zu gleichen Tei len gemischt und der Mischimpfstoff ist ge brauchsbereit. Infolge der hohen Konzentra tion des Antigens genügen zur Immunisie rung von Rindern kleine Mengen, z.
B. 2 bis 5 cm8.

The present invention relates to a method for the production of vaccines, e.g. B. from aerobic or anaerobic bacteria, which is characterized in that the bacterial content is enriched in bacterial cultures, and then the resulting concentrates are treated with agents that protect the antigens, the bacteria by acting on the respiratory mechanism kill the cells.
To carry out the method according to the invention, the bacteria are, for example, grown on an optimal nutrient medium and then in some way, e.g. B. by settling, concentrated.
Since the respiratory system is one of the most vital and most involved apparatuses of the bacterial cell in the proliferation metabolism, substances were sought which, in particular, must disrupt the cell's respiratory chemistry in the broadest sense. The concept of breathing is to be understood very broadly, in the sense of Wieland.
If it is possible to disrupt the oxidation reduction processes by introducing a powerful oxidizing or reducing agent, the proliferation of the cells must be called into question. If the substances used for this purpose - oxidizing and reducing agents - also damage other systems in the bacterial cell at the same time, the cell will also be slowly killed off at a suitable low concentration.
Superoxides, such as H: 0, and chlorine preparations, from the group of reducing agents, aldehydes and nitrites, may be mentioned from the group of suitable oxidizing agents.
The use of formalin to detoxify toxins and kill bacteria is known. On the other hand, it is new to use very low concentrations and a long exposure time, whereby a very gentle conversion of the bacteria to colloids occurs, as a result of which the cells die, but the antigenic capacity is retained to a large extent. The method according to the invention can advantageously also be used in the preparation of mixed vaocins against several diseases. An example of this is the production of a mixed vaccine against intoxicating burn and malignant edema, which can be used as indicated in the example below.
Example: Cultures of intoxicating bacteria and bacteria of malignant edema cultivated separately in optimal nutrient media are concentrated by settling, after which they are treated with formaldehyde to damage the respiratory system of the bacteria.
It has been shown that the minimum useful concentration for killing the intoxicating bacteria is about 0.1 to 0.6% for malignant edembacilli but about 0.5 to 1.0% formaldehyde as a 40% aqueous formaldehyde solution.
These concentrations are allowed to act on the abovementioned culture sediments until the killing of the microorganisms can be demonstrated by animal or culture experiments, which, depending on the temperature and concentration, is achieved in 8 to 14 days.
