Verfahren zur Herstellung von 3-(Bis-diogypropyl)-amino-4-oxy-3'-amino-4'- ogyar senobenzol-for uialdehydnatr ium-snlfogylat. Es ist bekannt, stickstoffhaltige Arseno- benzole herzustellen, welche ein monooxyal- kyliertes Stickstoffatom,
sowie einen For- maldehydrest oder den .Sulfoxylatrest an einem andern Stickstoffatom enthalten. Alle diese Verbindungen haben das Ge meinsame, dass sie Monooxyalkylverbindun- gen sind.
Es wurde nun gefunden, dass man Arseno- benzolmonoformaldehydnatriumsulf oxylate der allgemeinen Formel

worin R und R' Oxyalkylreste bedeuten, da durch herstellen kann, dass man Bisoxyalkyl- aminobenzolarsinsäuren, welche im Benzol kern noch weitere Substituenten enthalten können, mit aminosubstitnierten Benzolarsin- säuren,
die ebenfalls noch weitere Substitu- enten im Benzolkern enthalten können, in molekularen Mengen mischt und durch Re duktionsmittel in Arsenobenzole überführt und die erhaltenen Arsenobenzole mit For- maldehydnatriumsulfoxylat zur Umsetzung bringt. Zu denselben Verbindungen gelangt man, wenn man auf die entsprechenden oxyalkyl- freien Aminoarsenobenzolsulfoxylate zwei Holeküle Alkylenoxyd einwirken lässt.
Es gelingt hierdurch, die an und für sich intramuskulär- nicht verwendbaren Sulfoxyl- verbindungen durch Einführung zweier Oxyalkylgruppen an dasselbe Stickstoff atom so grundlegend zu verbessern, dass sie, intramuskulär eingespritzt, reizlos vertra gen werden, ohne dass die intravenöse Wir kung dadurch aufgehoben wird. Diese Ver- bindungen können also sowohl intravenös als intramuskulär gebraucht werden, was für den Arzt eine erhebliche Vereinfachung be deutet.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung des 3-(Bis- dioxypropyl) -amino-4 -.'oxy-3'-amino-4'-oxy- arsenobenzolformaldehydnatriumsulfoxylats, welches dadurch gekennzeichnet ist, dass man 3 - (Bis - dioxypropyl) - amino- 4 - oxybenzol -1- arsinsäure mit 3-Amino-4-oxybenzol-l-arsin- säure in molekularen Mengen mischt,
auf die Mischung ein Reduktionsmittel einwirken lässt und das so erhaltene Arsenobenzol mit Formaldehydnatriumsulfoxylat zur Umset zung bringt.' Die neue Verbindung stellt ein in Wasser leicht lösliches, in Äther und Äthylalkohol unlösliches gelbes Pulver dar. Sie soll als Heilmittel Verwendung finden.
<I>Beispiel;</I> ,35 g 3-(Bis-dioxypropyl)-amino-4-oxyben- zol-1-arsinsäure werden mit 22,9 g 3-Amino- 4-oxybenzol-l-arsinsäure und 28 g Jodkali gemischt, in 580 cm' zehnprozentiger Salz säure gelöst und unter Rühren mit 77 cm' 50%iger unterphosphoriger Säure versetzt. Die Temperatur steigt unter gleichzeitiger Gelbfärbung der Flüssigkeit auf etwa 35 .
Sobald die Temperatur wieder fällt, wird die klare gelbe Lösung in Äthylalkohol ein gerührt, das ausgeschiedene salzsaure '3-(Bis- dioxypropyl)- amino- 44- oxy- 3'-amino -4'- oxy- arsenobenzol abgesaugt und mit Äther ge waschen. Das gelbe Präparat löst sich leicht in Wasser und Methylalkohol und hat einen Arsengehalt von 22,59%.
An Stelle von unterphosphoriger Säure können auch andere Reduktionsmittel, wie zum Beispiel Hydrosulfit, verwendet werden.
Man löst j55 g salzsaures 3-(Bis-dioxy- propyl) - amino -4--oxy-3'-amino-4'- oxyarseno- benzol in 275 cm' Methylalkohol und 275 cm' Wasser und lässt eine Lösung von 35 g F'ormaldehydnatriumsulfoxylat in 56 cm3 Wasser bei etwa 27 einwirken. Es scheidet sich ein gelbes Produkt aus, das auf Zugabe von Natriumcarbonatlösung bis zur schwach alkalischen Reaktion vollkommen in Lösung :geht.
Wird diese klare, gelbe Flüs sigkeit in ein Gemisch von Äthylalkohol und Wasser eingerührt, so fällt ,3-(Bis-dioxypro- pyl) - amino-4-oxy-3'- amino -4'- oxyarsenoben- zolf ormaldehydnatriumsulf oxylat

als gelber Niederschlag aus, der abgesaugt und mit Äther gewaschen wird.
Die Verbindung löst sich leicht in Was ser mit schwach alkalischer Reaktion und hat einen Arsengehalt von 19,44%. Ihre wässrige Lösung lässt sich sowohl intravenös wie auch intramuskulär ohne jede Reizwir kung auf das Gewebe injizieren, während die entsprechende Mono-;glycid-Verbindung, 3 Dioxypropyl-amino - 4- oxy - 3'- amino-4'- oxy - arsenobenzolsulfoxylat, bei der intramusku lären Injektion starke Nekrosen hervorruft.
Die als Ausgangsstoff dienende 3-(Bis- dioxypropyl)-amino-4-oxybenzol-l-arsinsäure wird durch Erhitzen einer wässrig methyl- alkoholischen Lösung von 3-amino-4-oxyben- zol-l-arsinsaurem Natrium mit überschüssi gem Glycid erhalten.
