Verfahren zum antimikrobiellen Ausrüsten bzw. zum Schützen von Textilfasern gegen Mikroorganismen Die vorliegende Erfindung betrifft ein Verfahren zum antimikrobiellen Ausrüsten bzw. zum Schützen von Textilfasern gegen Mikroorganismen.
Dieses Verfahren ist dadurch gekennzeichnet, dass man als Wirkstoff mindestens einen O,N-Diphenylcarb- aminsäureester der Formel I

in der R1 einen durch mindestens eines und höchstens 3 gleiche oder verschiedene Halogenatome substituierten Phenoxyrest, von den Symbolen R2, R3 und R4 mindestens eines Chlor oder Brom, die anderen unabhängig voneinander Wasserstoff, Chlor oder Brom, R5 und R7 unabhängig voneinander Wasserstoff, Halo gen, niederes Alkyl, niederes Alkoxy, niederes Halo genalkyl, Nitro und Hydroxyl, R6 Wasserstoff, Halogen, niederes Alkyl, niederes Alkoxy, Dialkylamino, Hydroxyl, und X Sauerstoff oder Schwefel bedeuten, verwendet.
In der Formel I stellt R1 insbesondere einen der folgenden halogenierten Phenoxy-Reste dar: 4-Chlorphenoxy, 4-Bromphenoxy, 2,4-Dichlorphenoxy, 2,4-Dibromphenoxy, , 2,4,5-Trichlorphenoxy, 2-Chlor-4-bromphenoxy, 2-Brom-4-chlorphenoxy. Niederes Alkyl und niederes Alkoxy als Reste R5 bis R7 weisen 1 bis 4 Kohlenstoffatome auf. Als Halo genalkyl ist Trifluormethyl bevorzugt. Die Alkylsubsti- tuenten in einer Dialkylamino-Gruppe sind vorzugsweise niedere Reste mit 1 bis 4 Kohlenstoffatomen. R5, Rfi und/oder R7 kann Fluor, Chlor, Brom und Jod, insbe sondere aber Fluor, Chlor und/oder Brom sein.
Die neuen 0,N-Diphenyl-carbaminsäureester werden erhalten, indem man ein Phenol der Formel<B>11</B> als solches oder in Form eines seiner Alkali- oder Erd- alkalimetallsalze entweder a) mit Phosgen oder Thiophosgen in ein Säure chlorid der Formel III

umsetzt. In den Formeln I bis V haben die Symbole R1 bis R7 und X die unter Formel I angegebenen Bedeu tungen.
Dieses Herstellungsverfahren wird vorzugsweise in Gegenwart eines Lösungs- oder Verdünnungsmittels sowie eines säurebindenden Mittels (Protonenacceptors) durchgeführt.
Als Lösungs- oder Verdünnungsmittel kommen bei spielsweise in Betracht: Kohlenwasserstoffe, halogenierte Kohlenwasserstoffe, Amide, Äther und ätherartige Ver bindungen, Ketone, usw. Säurebindende Mittel sind vor zugsweise organische Basen, z. B. tertiäre Amine, wie Pyridin, Triäthylamin usw., anorganische Basen, wie die Hydroxide und die Carbonate von Alkali- und Erd- alkalimetallen.
Nachstehend wird die Herstellung einiger Diphenyl- carbaminsäureester der Formel I beschrieben. Die Tem peraturen sind in Celsiusgraden angegeben. Daran an schliessend sind in Tabelle 1 Diphenyl-carbaminsäure- ester aufgeführt, die auf den in der vorangehenden Arbeitsweise beschriebenen Wegen unter Verwendung der entsprechenden 2-Hydroxy-diphenyläther bzw. 2- Phenoxyphenyl-kohlensäurechloriden und Phenyliso- cyanaten bzw. Anilinen erhalten wurden.
A. a) In 500 ml Toluol leitet man bei 0 etwa 100 g Phosgen ein. Zu dieser Lösung tropft man bei 0 bis 5 C eine Lösung von 289,5 g 4,2',4'-Trichlor-2-hy- droxy-diphenyläther in 700 ml Toluol und leitet noch mals 100 g Phosgen ein. Unter Rühren setzt man dem Reaktionsgemisch dann eine Lösung von 111,3g Tri- äthylamin in 200 ml Toluol zu. Nach mehrstündigem Stehen bei Raumtemperatur wird überschüssiges Phosgen entfernt, das Triäthylamin-Hydrochlorid abgetrennt und das Lösungsmittel des Filtrates abdestilliert. Der Rück stand wird fraktioniert, das O-[2-(2',4'-Dichlorphenoxy)- 5-chlorphenyll-kohlensäurechlorid siedet bei 180-184 und 0,5 Torr.
b) 35,2 g O-[2-(2',4'-Dichlorphenoxy)-5-chlorphe- nyl]-kohlensäurechlorid, gelöst in 300 ml Aceton, wer den bei 15 bis 20 tropfenweise mit einer Lösung von 32,4 g 3,5-Dichloranilin in 150 ml Aceton versetzt.
Man lässt das Reaktionsgemisch dann 3 Stunden bei Zimmertemperatur stehen, giesst es auf Wasser und filtriert den nach einigen Stunden ausfallenden Nieder schlag ab. Nach mehrmaligem Umkristallisieren aus Cyclohexan und Benzol/Petroläther erhält man den N-(3,5-Dichlorphenyl)-carbaminsäure-O- [2-2',4'-dichlorphenoxy)-5-chlorphenyl l-ester mit dem Fp.: 132-134 .
B. a) Zu einer Lösung von 289,5 g 2',4'-Dichlor- 2-hydroxy-4-chlor-diphenyläther in 400 ml Chloroform gibt man eine Lösung von 150 g Thiophosgen in 400 ml Chloroform und kühlt auf 5 C ab. Unter intensivem Rühren wird dann eine wässrige Natriumhydroxid- Lösung, 50 g Natriumhydroxid gelöst in 790 ml Wasser, so zugetropft, dass die Temperatur 15 nicht über schreitet. Das Reaktionsgemisch wird anschliessend 2 Stunden bei Zimmertemperatur gerührt, die orga nische Phase abgetrennt, mit Wasser gewaschen und über Natriumsulfat getrocknet. Nach dem Abdestillie- ren des Lösungsmittels wird der Rückstand im Vakuum fraktioniert.
Das 2-(2',4'-Dichlorphenoxy)-5-chlor- phenyl-thiokohlensäurechlorid siedet bei 175 bis l84 0,05 Torr. (nrD20 = 1,6275).
b) Zu einer Lösung von 36 g O-[2-(2',4'-Dichlor- phenoxy) - 5 - chlorphenyl] - thiokohlensäurechlorid in 150 ml Benzol wird eine Lösung von 24,2 g 3,5-Di- methylanilin in 150 ml Benzol bei 5 C innerhalb von 1,5 Stunden zugetropft. Das Gemisch wird dann 12 Stunden bei Zimmertemperatur gerührt, mit Wasser ausgeschüttelt und über Natriumsulfat getrocknet. Nach dem Abdestillieren des Lösungsmittels hat der Rück stand der O-[2-(2',4'-Dichlorphenoxy)-5-chlorphenyl]- thiocarbaminsäure-N-(3,5-dimethylphenyl)-ester, aus Benzol/Petroläther umkristallisiert, den Fp: 102-l04 .
C. Zu einer Suspension von 30 g 4-Brom-5-chlor- 2-hydroxy-diphenyläther in 100 ml Ligroin wird eine Lösung von 19,8 g 3-Brom-phenylisocyanat in 50 ml Ligroin zugetropft. Nach Zugabe von 2 ml Triäthyl- amin wird die Mischung eine Stunde am Rückfluss erhitzt und dann gekühlt. Der ausgefallene Niederschlag wird abgetrennt, und aus Benzol/Petroläther (1 :1) umkristallisiert.
Der N-(3-Bromphenyl)-carbaminsäure-O-[2-(4-brom- phenoxy)-5-chlorphenyl]-ester hat den Fp. 116 .

feste Körper dar, welche durch Umkristallisie- ren gereinigt werden können. Sie zeichnen sich durch eine geringe Toxizität für Warmblüter aus und sind für die Haut in den, in Betracht kommenden Konzen trationen reizlos. Ein weiterer Vorteil der neuen Carb- aminsäurecster ist ihre Farblosigkeit oder geringe Eigen farbe. Diese Eigenschaft erschliesst ihnen viele Verwen dungsgebiete, welche stark farbigen Verbindungen ver schlossen sind.
Diese neuen Diphenyl-carbaminsäureester der For mel 1 weisen sehr gute bakteriostatische und bakterizide, wie auch fungistatische und fungizide Eigenschaften auf. Dank ihrer chemischen Struktur zeigen diese Verbin dungen eine gute Substantivität zum Substrat von Textil fasern, dies selbst in grosser Verdünnung in wässrigen Lösungen. Sie werden daher vorzugsweise zum anti mikrobiellen Ausrüsten beziehungsweise zum Schützen von Textilfasern gegen Mikroorganismen verwendet.
Von den schweizerischen Patentschriften Nrn.454 829 und 459 977 sind in der Phenoxygruppe unsubstituierte O-Phenoxyphenyl-N-trifluormethylphenyl- carbaminsäureester als Anthelmintica umfasst, jedoch nicht beschrieben. Diese Verbindungen besitzen aber keine oder nur sehr schwache mikrobizide Eigenschaften.
Ganz ähnliche Struktur weisen die substituierten Phenylester der Carbanilsäure auf, aus dem belgischen Patent Nr. 562 631, dem französischen Patent Nummer 1 397 317 oder dem USA-Patent Nr. 3 142 646. Diese Verbindungen zeigen zwar biologische Aktivität im in vitro Versuch, sie haben aber nicht die den vorliegen den Verbindungen eigene Substantivität zum Substrat der Textilfaser, so dass in den Waschtests ihre Wirkung weitgehend verlorengeht.
Dasselbe kann von der aus dem deutschen Patent Nr.<B>881</B>458 als Desinfektionsmittel bekannten Verbin dung O-(2-Chlor-4-phenoxyphenyl)-carbamat gesagt werden.
Ferner ist in der französischen Patentschrift Num mer<B>881</B>458 ein Ester der Diäthyl-thiocarbaminsäure, das Natriumsalz der 3,5-Dichloro-2-(diäthylcarbamoyl- oxy)-benzosulfonsäure als Mottenschutzmittel beschrie ben.
Ein bevorzugtes Anwendungsgebiet der Diphenyl- carbaminsäureester der Formel I besteht im Entkeimen von Waschgut und zum Schützen von Waschgut gegen Befall durch Mikroorganismen. Man verwendet hierzu Spülflotten, die die genannten Carbaminsäureester mit Vorteil in Konzentrationen von etwa 5 bis 200 Teilen pro Million, bezogen auf die Flotte, enthalten.
Als waschaktive Substanzen enthalten die Wasch flotten beispielsweise anionaktive Verbindungen, wie durch lipophile Gruppen substituierte aromatische Sul- fonsäuren bzw. deren wasserlösliche Salze, etwa das Natriumsalz der Dodecylbenzolsulfonsäure, oder wasser lösliche Salze von Schwefelsäuremonoestern höhermole kularer Alkohole oder ihrer Polyglykoläther, beispiels weise lösliche Salze von Dodecylalkoholsulfat oder von Dodecylalkoholpolyglykoläthersulfat, oder Alkalisalze höherer Fettsäuren (Seifen), ferner nichtionogene wasch aktive Substanzen, wie Polyglykoläther höherer Fett alkohole,
ferner Polyglykoläther höhermolekular-alky lierter Phenole sowie auch sogenannte amphoteres waschaktive Substanzen, etwa Umsetzungsprodukte der Alkalisalze niederer Halogenfettsäuren mit lipophile Reste enthaltenden Polyalkylenpolyaminen, z. B. mit Lauryldiäthylentriamin.
Daneben kann die Flotte auch noch übliche Hilfs stoffe, wie optische Aufheller, Weichmacher, sauer reagierende Salze, wie Ammonium- oder Zinksilico- fluorid oder gewisse organische Säuren, wie Oxalsäure, ferner Appreturmittel, z. B. solche auf Kunstharzbasis oder Stärke, enthalten.
Als Waschgut, welches mit diese Wirkstoffe der Formel 1 enthaltenden Spülflotten entkeimt werden kann, kommt vor allem organisches Fasermaterial in Betracht, nämlich solches natürlicher Herkunft, wie cellulosehaltiges, beispielsweise Baumwolle, oder poly- peptidhaltiges, z. B. Wolle oder Seide, oder Fasermate rial synthetischer Herkunft, wie solches auf Polyamid-, Polyacrylnitril- oder Polyesterbasis, oder Mischungen obengenannter Fasern.
Die Carbaminsäureester verleihen in den vorstehend genannten Konzentrationen sowohl der Flotte als auch dem damit behandelten Waschgut eine weitgehende und remanente Keimfreiheit gegen Staphylococcus- und Coli- formen, die selbst nach Belichten des Wirkstoffes bzw. der damit behandelten Ware bestehen bleibt. Sie zeich nen sich durch ihre Lichtbeständigkeit auf dem damit behandelten Waschgut sowie durch ihre hohe Aktivität und Wirkungsbreite gegen grampositive und gram negative Organismen aus.
Die neuen Carbaminsäureester sind auch gegen die Schweissgeruch erzeugende Bakterienflora sehr wirksam und darum und wegen ihrer geringen Toxizität als desodorierende Mittel für Wäsche oder als Zusätze für kosmetische Mittel, wie Salben oder Cremen, geeignet.
Die bakteristatische und fungistatische Wirkung der Carbaminsäurcester der Formel I wird durch folgende vergleichende Versuche bestimmt.
Methode: Zur Bestimmung der das Wachstum der verschie denen Mikroorganismen hemmenden Grenzkonzentra tion wird die Wirkstoff-Lösung mit dem noch heissen Agar vermischt. Der Agar wird dann in Platten aus gegossen, und nach dem Erstarren werden die Testkeime ausgestrichen. Man kann so die Grenzkonzentrationen bestimmen, bei denen sich die Mikroorganismen nicht mehr entwickeln.
Als Mikroorganismen wurden verwendet: <I>Versuch 1</I> A. Bakterien: Escherichia coli, Bacillus pumilus, Sarcina ureae, Staphylococcus aureus, Proteus vulgaris HXL. B. Fungi: 1. wenn die Carbamin- säureester der Formel I in Waschflotten auf mit Bakte rien infizierte Wäsche einwirken.
Dieser Versuch wird ausgewertet durch A. Bestimmung der Keimzahl in der Spülflotte, B. Bestimmung der Desinfektionswirkung auf dem Waschgut, C. Bestimmung der remanenten Wirkung auf dem Waschgut.
Diese Wäscheversuche wurden mit den Bakterien Staphylococcus atueus SG 511 und Escherichia coli ausgeführt.
Beispiel Die Wäscheversuche werden in Glasbehältern aus geführt, welche dermassen montiert sind, dass sie die Bewegung einer Waschmaschine nachahmen können. Diese Glasbehälter haben einen geschliffenen Deckel und etwa 100 ml Inhalt. Das gesamte verwendete Mate rial wird vor jedem Versuch sterilisiert entweder durch 20minutiges Erhitzen auf 120 C in einem Autoklav oder, wo dieses nicht möglich ist, durch Waschen mit einer Lösung, welche pro Liter Wasser 1 ml 80%3, a- (p -Tolyl) - dodecyl - trimethylammonium - methosulfat (Desogen R) enthält.
Das verwendete Waschmittel besteht aus 40 % Na triumseife, 31 % Natriumpolyphosphat (Waschalkali), 9 % Natriumpyrophosphat, 6 % Natriumsilikat, 3 Magnesiumsilikat, 0,5% Natriumäthylendiamintetra acetat, 5 % gebrannte Soda und 5,5 % Natriumsulfat.
1,25 g dieses Waschmittels werden unter Rühren und Sieden in ungefähr 250 ml sterilem Wasser auf gelöst, welches vorher weichgemacht wurde, indem man es durch eine Permutit-Ionenaustauschkolonne durch fliessen liess. Solches Wasser wird in der Folge als Per- mutit-Wasser bezeichnet. Die Waschmittellösung wird mit kaltem Permutit-Wasser auf 1000 ml verdünnt. Von den Versuchssubstanzen werden je 100 mg in 50 ml destillierter Methylcellosolve aufgelöst. Je 40 ml Waschmittellösung werden in jeden der sterilisierten Glasbehälter gegeben. Dann bereitet man mit Hilfe der Wirkstofflösungen in Methylcellosolve Waschflotten her, welche 6,25 und 25 ppm Versuchssubstanz im Bad enthalten.
Für jede Versuchsserie wird ein Kontroll- test durchgeführt, bei dem der Waschflotte keine Wirk substanz beigegeben wird.
Dann wird 2 g rohes, nicht mit optischen Aufhellern behandeltes Baumwollgewebe, bestehend aus 13 Ron dellen zu 2,5 cm Durchmesser und drei Streifen der Dimension 2,5 ¿ 23 cm, jedem Bad zugegeben und 20 Minuten bei 40 gewaschen.
Die Waschflotte wird daraufhin verworfen und die Baumwollmuster mit 40 ml sterilem Permutit während 3 Minuten bei 40' C gespült. Diese Spülung wird noch zweimal wiederholt. Beim dritten Mal infiziert man das Spülwasser mit einer 24 Stunden auf Agar ge wachsenen Bakterienkultur.
Dieses Spülwasser wird zur Bestimmung der Keim zahl (A) verwendet.
Die Baumwollrondellen werden zur Bestimmung der Desinfektionswirkung (B) benützt, und die Baumwoll- streifen werden auf dem Trockenständer befestigt und an der Luft getrocknet. Am nächsten Tag werden daraus Rondellen von 2,5 cm Durchmesser gestanzt, welche der Bestimmung der remanenten Wirkung (C) dienen. A. Bestimmung der Keimzahl in der Spülflotte Das die Wirkstoffe enthaltende dritte Spülbad wurde mittels Keimen von Staphylococcus aureus SG 511 und Escherichia coli infiziert. 1 ml dieses Bades wurde 20 ml Nähragar [Mac Conkey, Difco Marmal, 9. Aus gabe (1953), Seite 131] bzw. Nähragar und Kalium tellurit zugegeben und das Ganze in Petrischalen ge gossen. Die Petrischalen wurden 24 Stunden bei 37 C bebrütet.
Die Anzahl lebensfähiger Bakterien pro ml Spülflotte wurde durch Auszählen der sich auf oder in den Agarflotten gebildeten Kolonien bestimmt.
Als Substrat wurde verwendet: für Staphylococcus aureus SG 511 Nähragar + - Kaliumtellurit und für Escherichia coli Nähragar.
Die Resultate sind in Tabelle 4 zusammengestellt:

Auf die entstandenen Agarplatten wurde je eine mit sterilem Permutit-Wasser gespülte Baumwollrondelle gelegt, die aus einem in einem mit Staphylococcus aureus SG 511 und Escherichia coli beimpften Spülbad gespülten Baumwollgewebe ausgestanzt worden war. Die so vor bereiteten Petrischalen wurden dann 24 Stunden lang bei 37' C bebrütet. Das Wachstum der Mikroorganismen auf der Agarplatte wurde dann visuell ermittelt. Die Resultate sind in Tabelle 5 angegeben, worin Wachstum unter dem Gewebe - _= kein Wachstum unter dem Gewebe + = Spuren von Wachstum unter dem Gewebe bedeuten.

solid bodies which can be cleaned by recrystallization. They are characterized by low toxicity for warm-blooded animals and are non-irritating to the skin in the concentrations in question. Another advantage of the new carbamic acid esters is that they are colorless or have little inherent color. This property opens up many areas of application for them, which are closed to strongly colored compounds.
These new diphenylcarbamic acid esters of the formula 1 have very good bacteriostatic and bactericidal, as well as fungistatic and fungicidal properties. Thanks to their chemical structure, these compounds show good substantivity to the substrate of textile fibers, even when they are highly diluted in aqueous solutions. They are therefore preferably used for anti-microbial treatment or to protect textile fibers against microorganisms.
Swiss Patent Nos. 454 829 and 459 977 include, but not describe, O-phenoxyphenyl-N-trifluoromethylphenyl-carbamic acid esters unsubstituted in the phenoxy group as anthelmintics. However, these compounds have no or only very weak microbicidal properties.
The substituted phenyl esters of carbanilic acid have a very similar structure, from Belgian patent No. 562 631, French patent No. 1,397,317 or US patent No. 3,142,646. Although these compounds show biological activity in an in vitro experiment, they However, they do not have the substantivity of the present compounds for the substrate of the textile fiber, so that their effect is largely lost in the washing tests.
The same can be said of the compound O- (2-chloro-4-phenoxyphenyl) carbamate known from German Patent No. 881 458 as a disinfectant.
Furthermore, in French patent specification number <B> 881 </B> 458, an ester of diethyl thiocarbamic acid, the sodium salt of 3,5-dichloro-2- (diethylcarbamoyl oxy) -benzosulfonic acid, is described as a moth repellent.
A preferred field of application of the diphenylcarbamic acid esters of the formula I consists in the sterilization of laundry items and for protecting laundry items against attack by microorganisms. For this purpose, rinsing liquors are used which advantageously contain the carbamic acid esters mentioned in concentrations of about 5 to 200 parts per million, based on the liquor.
As detergent substances, the washing liquors contain, for example, anionic compounds such as aromatic sulfonic acids substituted by lipophilic groups or their water-soluble salts, such as the sodium salt of dodecylbenzenesulfonic acid, or water-soluble salts of sulfuric acid monoesters of higher molecular alcohols or their polyglycol ethers, for example soluble salts of Dodecyl alcohol sulfate or dodecyl alcohol polyglycol ether sulfate, or alkali salts of higher fatty acids (soaps), also non-ionic detergent substances such as polyglycol ethers of higher fatty alcohols,
also polyglycol ethers higher molecular alkylated phenols and so-called amphoteric detergent substances, such as reaction products of the alkali salts of lower halogen fatty acids with lipophilic radicals containing polyalkylene polyamines, eg. B. with lauryl diethylenetriamine.
In addition, the liquor can also contain customary auxiliaries such as optical brighteners, plasticizers, acidic salts such as ammonium or zinc silicofluoride or certain organic acids such as oxalic acid, and also finishing agents such. B. those based on synthetic resin or starch contain.
As laundry items which can be sterilized with rinsing liquors containing these active ingredients of formula 1, organic fiber material comes into consideration, namely those of natural origin such as cellulose-containing, for example cotton, or polypeptide-containing, e.g. B. wool or silk, or fiber material of synthetic origin, such as those based on polyamide, polyacrylonitrile or polyester, or mixtures of the above fibers.
In the concentrations mentioned above, the carbamic acid esters give both the liquor and the laundry treated with it an extensive and remanent sterility against Staphylococcus and Coli forms, which remains even after the active substance or the goods treated with it have been exposed to light. They are characterized by their light resistance on the laundry treated with them and by their high activity and range of effects against gram-positive and gram-negative organisms.
The new carbamic acid esters are also very effective against the bacterial flora which produce a sweat odor and, because of their low toxicity, are suitable as deodorants for laundry or as additives for cosmetic agents, such as ointments or creams.
The bacteristatic and fungistatic effect of the carbamic acid esters of the formula I is determined by the following comparative tests.
Method: To determine the limit concentration that inhibits the growth of the various microorganisms, the active ingredient solution is mixed with the still hot agar. The agar is then poured into plates and, after solidification, the test germs are streaked out. In this way one can determine the limit concentrations at which the microorganisms no longer develop.

This experiment is evaluated by A. determining the number of germs in the rinsing liquor, B. determining the disinfecting effect on the laundry, C. determining the remanent effect on the laundry.
These laundry tests were carried out with the bacteria Staphylococcus atueus SG 511 and Escherichia coli.
Example The laundry tests are carried out in glass containers that are mounted in such a way that they can imitate the movement of a washing machine. These glass containers have a polished lid and a capacity of around 100 ml. All material used is sterilized before each experiment either by heating it for 20 minutes to 120 C in an autoclave or, where this is not possible, by washing with a solution containing 1 ml of 80% 3, a- (p -Tolyl ) - dodecyl - trimethylammonium - methosulfate (Desogen R) contains.
The detergent used consists of 40% sodium soap, 31% sodium polyphosphate (washing alkali), 9% sodium pyrophosphate, 6% sodium silicate, 3 magnesium silicate, 0.5% sodium ethylenediamine tetra acetate, 5% burnt soda and 5.5% sodium sulfate.
1.25 g of this detergent are dissolved with stirring and boiling in approximately 250 ml of sterile water which has previously been softened by flowing it through a permutite ion exchange column. Such water is hereinafter referred to as permutite water.
