Verfahren zur Darstellung von 1-Phenyl-2 - 3-dimethyl-4-dimethylamino-ö-pyr azolon. Bei den bisher bekannten Verfahren zur Darstellung von 1-Phenyl-2 # 3-dimethyl-4-di- methylamino-5-pyrazolon musste aus 1-Phenyl- 2 - 3-dimethyl-4-nitroso-5-pyrazolon zunächst die entsprechende 4-Aminoverbindung dar gestellt werden.
Die freie 4-Aminoverbindung, deren Reinigung umständlich ist, wurde dann in die 4-Dimethylaminoverbindungübergeführt.
Den Gegenstand der vorliegenden Erfin dung bildet ein Verfahren, bei dem das 1-Phenyl-2 # 3-dimethyl-4-dimethylamino-5- pyrazolon aus der entsprechenden 4-Nitroso- v erbindung in einem einzigen Arbeitsgang in guter Ausbeute gewonnen wird. Es be steht darin, dass man das 1-Phenyl-2 # 3-di- methyl-4-nitroso-5-pyrazolon, Formaldehyd oder dessen Polynieren und reduzierende Mittel aufeinander zur Einwirkung bringt.
Als solche kommen zum Beispiel geeignete Metalle, wie Zink oder Eisen, in mineral- oder organischsaurer Lösung in Betracht. Man kann einen geeigneten Katalysator zu setzen, wie zum Beispiel Kupfersulfat.
Man kann das Verfahren auch so aus- führen, dass man zunächst teilweise reduziert und dann Formaldehyd und reduzierende Mittel einwirken lässt.
Man kann das Verfahren noch weiter ver einfachen und in einem einzigen Arbeits gange aus 1-Pbenyl-2 # 3-dimethyl-5-pyrazolon zu dem gewünschten Endprodukt gelangen, indem man 1-Phenyl - 2 . 3 - dimethyl - 5 -pyr- azolon durch Einwirkung von Nitrit in saurer Lösung in 1-Phenyl-2 # 3-dimethyl-4-nitroso- 5-pyrazolon überführt und dieses, ohne es von dem Reaktionsgemisch zu trennen, in Gegenwart von Formaldehyd oder dessen Polymeren mit reduzierenden Mitteln be handelt.
<I>Beispiele</I> 1. 22 Teile 1-Phenyl-2. 3-dimethyl-4- nitroso-5-pyrazolon werden in 100 Teilen 50 o/oiger Essigsäure. aufgeschwemmt und unter allmählicher Zugabe von 30 Teilen Zinkstaub bei gleichzeitigem Zutropfen von 30 Teilen 10 o/oiger Formaldehydlösung bei etwa 40<B>0</B> 15 Minuten lang lebhaft gerührt. Dann werden 25 Teile Eisessig und weitere 60 Teile 10 %iger Formaldehydlösung all mählich zugegeben und unter langsamer Steigerung der Temperatur weitere 40 Teile Zinkstaub nach und nach eingetragen.
Das Rühren wird noch etwa 1'/4 Stunden lang fortgesetzt. Dann wird alkalisch gemacht, die entstandene 4-Dimethylaminoverbindung mit Benzol ausgeschüttelt und aus Ligroin oder dergleichen umkristallisiert.
2. 22 Teile 1-Phenyl-2.3-dimethyl-4- nitroso-5-pyrazolonwerden unter gutem Rühren in 500 Teilen Wasser suspendiert, sodann 8 Teile Paraformaldehyd zugefügt und unter Kühlung 350 Teile 25 %ige Salzsäure hin- zugegossen. Unter fortgesetztem Rühren werden dann allmählich 80 Teile Zinkstaub einge tragen, dem etwas Kupfersulfat beigemischt ist, indem gleichzeitig die Temperatur lang sam gesteigert wird.
Nach dem Aufhören der Wasserstoffentwicklung wird filtriert und das gebildete 1-Phenyl - 2 # 3-dimethyl - 4 - di- methylamino-5-pyrazolon in der üblichen Weise isoliert und gereinigt.
3. 3,8 Teile 1-Phenyl-2'. 3-dimethyl-5- pyrazolon werden in 10 Teilen 40 %iger Schwefelsäure unter Zusatz von 50 Teilen Wasser gelöst und unter Kühlung und Tur- binieren allmählich mit einer Lösung von 1.55 Teilen Natriumnitrit in 15 Teilen Wasser versetzt.
Dabei scheidet sich grünfarbiges 1-Phenyl-2 . 3-dimethyl-4-n itroso- 5 - pyrazol o n ab. Es werden dann 40 Teile 40 0%iger Schwefelsäure zugegeben und unter Kühlung allmählich 6 Teile Zinkstaub mit etwas Kupfersulfat eingetragen, während man gleich zeitig 6 Teile 10 %iger Formaldehydlösung zutropfen lässt. Das Rühren wird fortgesetzt, bis die Flüssigkeit annähernd entfärbt ist.
Hierauf wird auf 70 0 erwärmt, und all- mählich werden 12 Teile 10 %igei# Formal- dehy dlösung und 12 Teile Zinkstaub mit etwas Kupfersulfat zugegeben. Das Rühren wird fortgesetzt, bis nur noch schwache Wasserstoffentwicklung stattfindet. Dann wird filtriert und das Filtrat alkalisch gemacht.
Das 1-Plienyl-2 . 3-diinethyl-4-dimethylainino- 5-pyrazolon wird mit Benzol ausgeschüttelt und durch Unikristallisieren gereinigt.
Anstatt den Formaldehyd als 10 0',!oige wässerige Lösung zutropfen zu lassen, kann man auch die entsprechende Menge als poly meren Formaldehyd (z. B. Paraformaldehy d) mit Zinkstaub gut gemischt bei sonst gleicher Arbeitsweise eintragen.
Man kann auch so verfahren, dass man die Nitrosoverbindung zunächst teilweise reduziert und dann weiter mit Formaldehyd oder dessen Polymeren und reduzierenden Mitteln behandelt.
4. 22 Teile 1-Phenyl-2 . 3-dimethyl-4- niti#oso-5-pyi-azoloij werden in 100 Teilen 50 %igei, Essigsäure aufgeschwemmt und unter allmählicher Zugabe von 30 Teilen Zinkstaub, zweckmässig in Gegenwart von etwas Kupfersulfat, bei etwa 40 0 15 Minuten lang lebhaft gerührt,
bezw. turbiniert. Dann werden 25 Teile Eisessig und 80 Teile 10 %ige Fornialdeliy dlösung allmählich zu- gegeben und unter langsamer Steigerung der Temperatur weitere 40 Teile Zinkstaub nach und nach eingetragen. Das Rühren wird noch etwa 1 '/4 Stunden lang fortgesetzt.
Dann wird alkalisch gemacht, die entstandene 4-Dimethylaininoverbindung mit Benzol aus geschüttelt und aus Ligroin oder dergleichen umkristallisiert.
5. 3,8 Teile 1-Phenyl-2 # 3-dimethyl-5- pyrazolon werden in 1.0 Teilen 40 0;'oiger Schwefelsäure unter Zusatz von 50 Teilen Wasser gelöst und unter Kühlung und Tur- biuieren allmählich mit einer Lösung von L,55 Teilen Natriumnitrit in 15 Teilen Wasser versetzt. Dabei scheidet sich grünfarbiges 1-Pheny 1-2 . 3-diinetliyl-4-iiitroso-5-pyrazolon ab.
Es werden dann 40 Teile 40 0,'oigei# Schwefelsäure zugegeben und unter Kühlung allmählich 6 Teile Zinkstaub und etwas Kupfersulfat eingetragen. Das Rühren wird fortgesetzt, bis die Flüssigkeit annähernd entfärbt ist. Es wird dann auf etwa 70 0 erwärmt, und allmählich werden 16 Teile 10 %iger Formaldehydlösung und 12 Teile Zinkstaub mit etwas Kupfersulfat zugegeben. Das Rühren wird fortgesetzt, bis nur noch schwache Wasserstoffentwicklung stattfindet. Dann wird filtriert und das Filtrat alkalisch gemacht.
Das 1-Phenyl-2 . 3-dimethyl-4-di- methylamino-5-pyrazolon wird mit Benzol ausgeschüttelt und durch Umkristallisieren gereinigt.
Process for the preparation of 1-phenyl-2 - 3-dimethyl-4-dimethylamino-ö-pyrazolone. In the previously known processes for the preparation of 1-phenyl-2 # 3-dimethyl-4-dimethylamino-5-pyrazolone, the corresponding 4-pyrazolone first had to be obtained from 1-phenyl-2 - 3-dimethyl-4-nitroso-5-pyrazolone -Amino connection can be provided.
The free 4-amino compound, which is cumbersome to purify, was then converted to the 4-dimethylamino compound.
The subject of the present invention is a process in which the 1-phenyl-2 # 3-dimethyl-4-dimethylamino-5-pyrazolone is obtained from the corresponding 4-nitroso compound in a single operation in good yield. It consists in bringing the 1-phenyl-2 # 3-dimethyl-4-nitroso-5-pyrazolone, formaldehyde or its polynating and reducing agents to act on one another.
Suitable metals, such as zinc or iron, in mineral or organically acidic solution, for example, come into consideration as such. One can put a suitable catalyst, such as copper sulfate.
The process can also be carried out in such a way that one first partially reduces and then allows formaldehyde and reducing agents to act.
The process can be further simplified and the desired end product can be obtained in a single operation from 1-pbenyl-2 # 3-dimethyl-5-pyrazolone by 1-phenyl - 2. 3-dimethyl-5-pyrazolone is converted into 1-phenyl-2 # 3-dimethyl-4-nitroso-5-pyrazolone by the action of nitrite in acidic solution and this, without separating it from the reaction mixture, in the presence of formaldehyde or its polymers are treated with reducing agents.
<I> Examples </I> 1. 22 parts of 1-phenyl-2. 3-dimethyl-4-nitroso-5-pyrazolone are dissolved in 100 parts of 50% acetic acid. Suspended and with the gradual addition of 30 parts of zinc dust with simultaneous dropwise addition of 30 parts of 10% formaldehyde solution at about 40 <B> 0 </B> vigorously stirred for 15 minutes. Then 25 parts of glacial acetic acid and a further 60 parts of 10% strength formaldehyde solution are gradually added and a further 40 parts of zinc dust are gradually added while the temperature is slowly increased.
Stirring is continued for about 1/4 hour. It is then made alkaline, the 4-dimethylamino compound formed is extracted by shaking with benzene and recrystallized from ligroin or the like.
2. 22 parts of 1-phenyl-2,3-dimethyl-4-nitroso-5-pyrazolone are suspended in 500 parts of water with thorough stirring, then 8 parts of paraformaldehyde are added and 350 parts of 25% hydrochloric acid are poured in with cooling. With continued stirring, 80 parts of zinc dust are gradually added to which a little copper sulfate is added by slowly increasing the temperature at the same time.
After the evolution of hydrogen has ceased, it is filtered and the 1-phenyl-2 # 3-dimethyl-4-dimethylamino-5-pyrazolone formed is isolated and purified in the usual manner.
3. 3.8 parts of 1-phenyl-2 '. 3-dimethyl-5-pyrazolone is dissolved in 10 parts of 40% strength sulfuric acid with the addition of 50 parts of water, and a solution of 1.55 parts of sodium nitrite in 15 parts of water is gradually added while cooling and turbocharging.
Here, green-colored 1-phenyl-2 separates. 3-dimethyl-4-n itroso- 5 - pyrazole on from. 40 parts of 40% strength sulfuric acid are then added and 6 parts of zinc dust with a little copper sulfate are gradually introduced with cooling, while 6 parts of 10% strength formaldehyde solution are added dropwise at the same time. Stirring is continued until the liquid is nearly discolored.
This is followed by heating to 70 °, and gradually 12 parts of 10% strength formaldehyde solution and 12 parts of zinc dust with a little copper sulfate are added. Stirring is continued until only a weak evolution of hydrogen takes place. It is then filtered and the filtrate made alkaline.
The 1-plienyl-2. 3-diinethyl-4-dimethylainino-5-pyrazolone is extracted by shaking with benzene and purified by unicrystallization.
Instead of adding the formaldehyde dropwise as a 10% aqueous solution, you can also enter the corresponding amount as polymeric formaldehyde (e.g. paraformaldehyde) mixed well with zinc dust with otherwise the same procedure.
One can also proceed in such a way that the nitroso compound is first partially reduced and then treated further with formaldehyde or its polymers and reducing agents.
4. 22 parts of 1-phenyl-2. 3-dimethyl-4- niti # oso-5-pyi-azoloij are suspended in 100 parts of 50% strength acetic acid and stirred vigorously for 15 minutes with the gradual addition of 30 parts of zinc dust, advantageously in the presence of a little copper sulfate ,
respectively turbinated. Then 25 parts of glacial acetic acid and 80 parts of 10% strength formaldehyde solution are gradually added and a further 40 parts of zinc dust are gradually added with a slow increase in temperature. Stirring is continued for about 1/4 hour.
It is then made alkaline, the resulting 4-dimethylainino compound is shaken out with benzene and recrystallized from ligroin or the like.
5. 3.8 parts of 1-phenyl-2 # 3-dimethyl-5-pyrazolone are dissolved in 1.0 part of 40% sulfuric acid with the addition of 50 parts of water and, with cooling and turbulence, gradually with a solution of L, 55 parts of sodium nitrite in 15 parts of water are added. Here, green-colored 1-Pheny 1-2 separates. 3-diinetliyl-4-iitroso-5-pyrazolone.
40 parts of 40% sulfuric acid are then added and 6 parts of zinc dust and a little copper sulphate are gradually introduced with cooling. Stirring is continued until the liquid is nearly discolored. It is then heated to about 70 °, and 16 parts of 10% formaldehyde solution and 12 parts of zinc dust with a little copper sulphate are gradually added. Stirring is continued until only a weak evolution of hydrogen takes place. It is then filtered and the filtrate made alkaline.
