Abstract

La présente invention concerne des lymphocytes B génétiquement modifiés et leurs utilisations, par exemple, pour le traitement de diverses affections et maladies, notamment du cancer, de maladies cardiaques, de maladies inflammatoires, de maladies liées à l'atrophie musculaire, de maladies neurologiques et analogues. Dans certains modes de réalisation, l'invention concerne un lymphocyte B modifié isolé ("cellule CAR-B") pouvant exprimer un récepteur chimérique ("récepteur CAR-B"), ledit récepteur chimérique comprenant (a) un domaine extracellulaire; (b) un domaine transmembranaire; et (c) un domaine cytoplasmique qui comprend au moins un domaine de signalisation. Dans divers modes de réalisation, l'invention comprend un lymphocyte B modifié isolé, ledit lymphocyte B pouvant exprimer et sécréter une charge utile; ladite charge utile n'étant pas naturellement exprimée dans un lymphocyte B ou étant exprimée à des niveaux supérieurs à celui qui est naturellement exprimé dans un lymphocyte B. Dans divers modes de réalisation, la charge utile est un anticorps ou un fragment de ce dernier.
