Abstract
La présente invention concerne des procédés et trousses pour la préparation de bibliothèque de séquençage pour détecter une anomalie chromosomique au moyen d'ADN acellulaire (cfDNA) sans nécessiter un isolement préalable de l'ADN acellulaire depuis une fraction liquide d'un échantillon pour essai. Selon certains modes de réalisation, le procédé comprend la réduction de liaison entre l'ADN acellulaire et des protéines nucléosomiques sans déroulement de l'ADN acellulaire depuis les protéines nucléosomiques. Selon certains modes de réalisation, la réduction de liaison comprend également la congélation et la décongélation de l'échantillon pour essai avant la réduction de la liaison entre l'ADN acellulaire et les protéines nucléosomiques. Selon certains modes de réalisation, l'échantillon pour essai est un échantillon de sang périphérique prélevé d'une femme enceinte comportant l'ADN acellulaire de la maman et du ftus, les procédés pouvant être utilisés pour détecter une anomalie chromosomique ftale tel qu'une variation du nombre de copies. Selon d'autres modes de réalisation, l'échantillon pour essai est un échantillon de sang périphérique prélevé d'un patient connu ou suspecté d'être atteint d'un cancer, les procédés pouvant être utilisés pour détecter des anomalies chromosomiques dans l'ADN acellulaire du patient. L'invention concerne également des trousses pour la détection de variation du nombre de copies du ftus au moyen des procédés selon la présente invention.
