Abstract
CONTROLES DE PICOS SINTÉTICOS PARA SEQUENCIAMENTO MEDIP LIVRE DE CÉLULAS E MÉTODOS DE USO DOS MESMOS. É aqui descrito um método de captura e análise de DNA metilado livre de células em uma amostra. O método envolve submeter a amostra à preparação da biblioteca para permitir o sequenciamento subsequente do DNA metilado livre de células. Uma quantidade predeterminada de fragmentos de DNA sintético de controle é adicionada à amostra. Os fragmentos de DNA sintético de controle têm, cada um, uma sequência de ácido nucleico conhecida que não se alinha a uma sequência de genoma alvo e pelo menos alguns dos fragmentos de DNA sintético de controle são metilados. A amostra é desnaturada e o DNA metilado livre de células e os fragmentos de DNA sintético de controle são capturados usando um aglutinante seletivo para polinucleotídeos metila-dos. O DNA capturado é amplificado e sequenciado.
