Abstract
a presente invenção refere-se a um método para detectar uma proximidade espacial de um primeiro e um segundo epítopos (11, 21) de uma proteína ou de uma primeira e uma segunda proteínas (10, 20) de um complexo de proteína (1) em uma amostra de um indivíduo. o método compreende ligar um primeiro membro de ligação (30), que tem um primeiro oligonucleotídeo (31) conjugado ao mesmo, ao primeiro epítopo (11), ligar um segundo membro de ligação (40), que tem um segundo oligonucleotídeo (41) conjugado ao mesmo, ao segundo epítopo (21), e determinar se um efeito de transferência de energia de ressonância por fluorescência (fret) está presente ou não entre um fluoróforo doador (32) e um fluoróforo receptor (42), os quais estão associados ao primeiro oligonucleotídeo (31) e ao segundo oligonucleotídeo (41), sendo que a presença do efeito de fret indica uma proximidade espacial do primeiro e do segundo oligonucleotídeos (31, 41) e, dessa forma, a proximidade espacial do primeiro e do segundo epítopos (11, 21).
