La présente invention est relative à un nouveau dérivé de l'acide l-éthyl-6-fluoro-7-(l-pipérazinyl)4-oxo-l,4-dihydroquinoléine-3-carboxylique, à savoir son ester benzoyloxyméthylique de la formule I:

L'invention se réfère, en outre, aux compositions pharmaceutiques contenant le composé de la formule

préparation. L'acide l-éthyl-6-fluoro-7-(l-pipérazinyl)4-oxo-l,4-dihydroquinoléine-3-carboxylique, également connu sous la dénomination de norfloxacine, est un agent antibactérien utilisé en particulier en médecine humaine dans le traitement des infections des voies urinaires.
Les propriétés pharmacocinétiques de la norfloxacine sont toutefois caractérisées par une absorption gastro-intestinale variant avec l'ingestion des aliments et par des concentrations sériques relativement bas-ses, du fait d'une distribution rapide vers les compartiments périphériques et d'une élimination rénale élevée, avec une demi-vie sérique d'environ 3 heures chez des sujets sains.
La norfloxacine, en tant que telle, n'est pas douée, par conséquent, d'une bonne biodisponibilité.
On a constaté de façon surprenante que le composé objet de l'invention, appelé ci-après pour une question de brièveté par l'abréviation PU 195, montre, comparativement à la norfloxacine, des avantages pharmacocinétiques considérables, tels qu'un pic plus élevé des niveaux sériques moyens de médicament et une demi-vie sérique fortement accrue.
On prépare le PU 195 au départ d'acide 6fluoro-7-chloro-l-éthyl-4-oxo-l,4-dihydro-quinoléine-3carboxylique (II), par réaction avec de la N-monotritylpipérazine (III), suivie d'une estérification du sel de potassium de l'acide 6-fluoro-1-éthyl-4-oxo-7-(4-tritylpipérazino)-1,4-dihydro-quinoléine-3-carboxylique (IV) avec du benzoate de chlorométhyle et d'une détritylation finale dans un milieu acide, d'après le schéma suivant:

L'exemple suivant illustre davantage le procédé de l'invention, sans pour autant en constituer une limitation.
Par conséquent, on peut utiliser, par exemple, d'autres solvants, réactifs, temps et températures de réaction appropriés sans sortir du cadre du présent brevet.
Exemple a) Préparation du dérivé de monotriphénylméthyl pipérazine (III)
On ajoute progressivement 12,71 g (45,6 millimoles) de chlorure de triphénylméthyle à 150 ml de chloroforme anhydre, contenant 8,2 g de pipérazine anhydre (95,6 millimoles). Ces additions sont réalisées toutes les 20 minutes, sous agitation, à la température ambiante. Au fur et à mesure que la réaction se développe, on observe la formation d'un précipité blanc, formé par le chlorhydrate de pipérazine. Lorsque les additions sont terminées, on maintient la solution sous agitation pendant une 1/2 heure, et on sépare ensuite le chlorhydrate de pipérazine par filtration. On lave la solution avec H20 plus de la triéthylamine (2 fois 100 ml) et ensuite uniquement avec de l'H20 (2 fois 100 ml). Après séchage sur Na2S04, on filtre le mélange et on évapore la phase chloroformique sous pression réduite. Production de
14,3 g (rendement de 95%).
Le produit est un solide blanc cristallin
(point de fusion de 168[deg.]-172[deg.]C). La structure du produit est confirmée par spectroscopie IR. Il est soluble dans les solvants chlorés, dans le benzène et dans le to-

dioxane, le formamide, l'éther, le n-heptane.
Analyse élémentaire:

% trouvé: C = 83,90; H = 7,45; N = 8,37.
b) Préparation du 6-fluoro-1-éthyl-7-(4trityl-pipérazino)-4-oxo-l,4-dihydro-quinolé-

On chauffe au reflux pendant 12 heures 30 g
(0,1 mole) d'acide 6-fluoro-7-chloro-1-éthyl-4-oxo-1,4dihydro-quinoléine-3-carboxylique (II), 36 g (0,11 mole) de monotritylpipérazine (III) et 300 ml de pyridine anhydre. On sépare par distillation la pyridine à pression réduite et on traite le résidu avec 500 ml de méthanol <EMI ID=6.1>
Le précipité est filtré à la pompe, mis en suspension dans du méthanol anhydre, à nouveau filtré et lavé avec de l'isopropanol.
On obtient ainsi 53 g (85%) de sel de potassium, que l'on sèche sous vide à 120[deg.]C pendant 4-5 heures.
Le produit, pur suivant la chromatographie sur couche mince, est directement utilisé dans l'étape suivante.
c) Préparation du 6-fluoro-1-éthyl-7-(4-trityl-pipérazino-4-oxo-l,4-dihydro-quinoléine3-carboxylate de benzoyloxyméthyle(V)
On met en suspension 6,6 g de sel de potassium (IV) dans 50 ml d'acétone anhydre et on les traite par 25,5 g de benzoate de chlorométhyle. On agite le mélange de réaction, on le chauffe au reflux pendant 2 heures, on le refroidit, on filtre le précipité, on le lave avec de l'eau distillée et on le cristallise avec de l'isopropanol. On obtient une poudre microcristalline crémeuse.
Rendement: 65%.Le produit,pur à la chromatographie sur couche mince, est directement utilisé dans l'étape suivante.
d) Préparation de l'ester de benzoyloxyméthyl-l-éthyl-6-fluoro-7-(l-pipérazinyl)4-oxo-1,4-dihydro-quinoléine-3-carboxylate

On dissout 3 g du dérivé tritylé (V) préparé suivant le procédé décrit dans l'étape précédente, dans
15 ml de dioxane; on y ajoute 5 ml d'HCl 2N sous agita-tion et on agite le mélange à la température ambiante pendant 2 heures. Après addition d'eau, on filtre le triphénylcarbinol précipité et on amène le filtrat à pH 8 avec du bicarbonate de sodium.
On obtient un précipité blanc crémeux, que l'on filtre et cristallise dans un mélange d'éthanol
et d'eau 50/50. Rendement de 69%. Point de fusion de
172[deg.]-173[deg.]C.

La présente invention se réfère également à tous les aspects industriellement applicables concernant l'utilisation du composé de l'invention comme agent thérapeutique. Un aspect essentiel de l'invention consiste, par conséquent, en les compositions pharmaceutiques convenant à l'administration orale contenant, comme principe actif, des quantités prédéterminées et thérapeutiquement efficaces de PU 195 en plus des excipients usuels, sous la forme de comprimés, de comprimés enrobés de sucre, de capsules, de sirops, de granules, etc.
Ces formulations contiendront de 50 à 600 mg de principe actif et seront administrées deux à quatre fois par jour.
Il doit être entendu que la présente invention n'est en aucune façon limitée aux formes de réalisation ci-dessus et que bien des modifications peuvent y être apportées sans sortir du cadre du présent brevet.

preparation. 1-Ethyl-6-fluoro-7- (1-piperazinyl) 4-oxo-1,4-dihydroquinoline-3-carboxylic acid, also known under the name of norfloxacin, is an antibacterial agent used in particular in human medicine in the treatment of urinary tract infections.
The pharmacokinetic properties of norfloxacin are however characterized by gastrointestinal absorption varying with the ingestion of food and by relatively low serum concentrations, due to rapid distribution to the peripheral compartments and high renal elimination , with a serum half-life of approximately 3 hours in healthy subjects.
Norfloxacin, as such, is therefore not endowed with good bioavailability.
It has been found, surprisingly, that the compound which is the subject of the invention, called hereinafter for the sake of brevity by the abbreviation PU 195, shows, compared to norfloxacin, considerable pharmacokinetic advantages, such as a higher peak. mean serum drug levels and a greatly increased serum half-life.
PU 195 is prepared from 6fluoro-7-chloro-1-ethyl-4-oxo-1,4-dihydro-quinoline-3carboxylic acid (II), by reaction with N-monotritylpiperazine (III), followed of an esterification of the potassium salt of 6-fluoro-1-ethyl-4-oxo-7- (4-tritylpiperazino) -1,4-dihydro-quinoline-3-carboxylic acid (IV) with benzoate chloromethyl and a final detritylation in an acid medium, according to the following scheme:

Example a) Preparation of the monotriphenylmethyl piperazine derivative (III)
12.71 g (45.6 millimoles) of triphenylmethyl chloride are gradually added to 150 ml of anhydrous chloroform, containing 8.2 g of anhydrous piperazine (95.6 millimoles). 6.6 g of potassium (IV) salt are suspended in 50 ml of anhydrous acetone and treated with 25.5 g of chloromethyl benzoate. The reaction mixture is stirred, heated to reflux for 2 hours, cooled, the precipitate is filtered, washed with distilled water and crystallized from isopropanol. A creamy microcrystalline powder is obtained.
Yield: 65%. The product, pure on thin layer chromatography, is used directly in the next step.
d) Preparation of the benzoyloxymethyl-1-ethyl-6-fluoro-7- (1-piperazinyl) 4-oxo-1,4-dihydro-quinoline-3-carboxylate ester

The present invention also refers to all the industrially applicable aspects concerning the use of the compound of the invention as a therapeutic agent. An essential aspect of the invention therefore consists of pharmaceutical compositions suitable for oral administration containing, as active ingredient, predetermined and therapeutically effective amounts of PU 195 in addition to the usual excipients, in the form of tablets, sugar-coated tablets, capsules, syrups, granules, etc.
These formulations will contain from 50 to 600 mg of active principle and will be administered two to four times a day.
It should be understood that the present invention is in no way limited to the above embodiments and that many modifications can be made thereto without departing from the scope of this patent.
