Synthèse d'aspartame.
L'invention concerne une nouvelle synthèse pour l'aspartame, c'est-à-dire l'ester méthylique d'a-L-aspartyl-L-phénylalanine. Tout comme le sucre de canne et de betterave, l'aspartame possède des propriétés édulcorantes et il est utilisé comme agent édulcorant pour les produits alimentaires et les boissons comme décrit dans le brevet belge 717. 373.
L'aspartame est un dipeptide et, comme tel, il est formé avec une liaison amido entre un groupe carboxy activé d'un acide aminé et le groupe amino d'un autre acide aminé. L'activation est nécessaire pour accroître la vitesse et le rendement de la condensation. Le peptide pur désiré nécessite la protection de tous les autres groupes fonctionnels n'intervenant pas dans la formation de la liaison peptide.
Enfin, les groupes protecteurs sont éliminés.
On peut préparer l'aspartame en faisant réagir l'anhydride L-aspartique N-protégé avec l'ester méthylique de L-phénylalanine. De la sorte, on obtient un mélange de l'ester méthylique de la L-aaspartyl-et de la L-ss-aspartyl-L-phénylalanine, ce qui nécessite la séparation des isomères. Les groupes N-protecteurs utilisés sont un des groupes N-protecteurs habituellement utilisés dans la chimie des peptides tels que les groupes benzyloxycarbonyle (Z) (brevet français 2.040. 473) et t-butyloxycarbonyle (brevet britannique 1. 339. 101) ou, afin d'éviter l'élimination finale du groupe N-protecteur, une simple protonation (brevet des Etats-Unis d'Amérique 3.901. 871).
Dans DE-2. 064. 482, on décrit la séparation des isomères a et ss (l'isomère ss ne possède pas des propriétés édulcorantes), ce qui conduit cependant à une réduction du rendement et à un accroissement des frais. Dans DE-2.608. 174 et EP-27. 319, on

décrit et revendique l'utilisation du palladium sur du charbon comme catalyseur en vue d'éliminer les groupes protecteurs par hydrogénation catalytique.
Au cours de l'élimination du groupe Z, il se forme de l'anhydride carbonique qui doit être éliminé afin d'éviter un arrêt de la réaction. Cette opération est habituellement effectuée sous un courant d'hydrogène, ce qui donne lieu à une perte de quantités importantes et dangereuses de ce gaz explosif.
L'invention concerne un procédé de synthèse d'aspartame, ce procédé consistant à condenser un dérivé d'acide L-aspartique complètement protégé avec l'ester méthylique de L-phénylalanine en présence d'un agent de condensation obtenu par la réaction entre un composé hétérocyclique azoté et un chlorure d'acide organique ou inorganique, puis éliminer les groupes protecteurs par hydrogénation avec un donneur d'hydrogène autre que l'hydrogène moléculaire, en présence d'un acide.
L'utilisation d'un dérivé d'acide L-aspartique complètement protégé permet d'éviter la formation de l'isomère ss. Des agents de condensation du type indiqué ci-dessus peuvent être obtenus aisément, tandis qu'ils sont peu coûteux et efficaces dans des conditions modérées. L'élimination des groupes protecteurs avec un donneur d'hydrogène autre que l'hydrogène moléculaire peut être effectuée dans un appareil simple sans devoir recourir à des réactifs gazeux dangereux. La présence d'un acide dans la dernière étape de déprotection permet d'éviter la formation du sous-produit inopportun que constitue la dicétopipérazine.

La condensation du dérivé d'acide L-aspartique avec le chlorhydrate de l'ester méthylique de L-phénylanine peut être effectuée en utilisant des

quantités équimolaires de réactifs en présence d'un excès (jusqu'à 50%) de l'agent de condensation dans un solvant, par exemple, le dichlorométhane, le chloroforme, le tétrahydrofuranne, le toluène, le diméthylformamide, le diméthylacétamide, le diméthylsulfoxyde ou la N-méthylpyrrolidone. Le dichlorométhane, le chloroforme et le tétrahydrofuranne sont préférés.
La température réactionnelle peut se situer entre - 30 C et la température ambiante. Le temps de la réaction est généralement inférieur à 5 heures. On peut préparer les agents de condensation comme décrit par H. A. Staab (Angew. Chem., 1962, 74, 407).
La récupération du dipeptide protégé peut être effectuée en éliminant le solvant réactionnel, puis en dissolvant le résidu dans un solvant tel que le diméthylformamide, le diméthylacétamide, le diméthylsulfoxyde ou la N-méthylpyrrolidone, par dilution avec un alcool alkylique inférieur (de préférence, l'alcool méthylique ou l'alcool éthylique) et par précipitation moyennant une addition d'eau. La recristallisation peut être effectuée dans un mélange de diméthylformamide et d'un alcool alkylique inférieur (de préférence, l'alcool méthylique ou l'alcool éthylique). Les groupes protecteurs sont éliminés quantitativement du dipeptide protégé par hydrogénation de transfert catalytique en utilisant une quantité appropriée d'un catalyseur.
Le catalyseur peut être l'un ou l'autre des métaux nobles (de préférence, le palladium ou le platine) que l'on sait être appropriés, seuls, sous forme d'un complexe ou déposés sur un support. Le formiate d'ammonium est un donneur d'hydrogène préféré. L'hydrogénation peut être effectuée dans un solvant tel que le diméthylformamide, le diméthylacétamide, le diméthylsulfoxyde ou la N-méthylpyrrolidone, contenant un acide organique (de préfé-

rence, l'acide formique ou l'acide acétique) ou un acide inorganique (de préférence, l'acide chlorhydrique) et un alcool alkylique inférieur (de préférence, l'alcool méthylique ou l'alcool éthylique).
La température réactionnelle peut se situer entre 0 et 650C.
Pour la récupération finale de l'aspartame, on peut adopter n'importe quel procédé classique.
Les symboles et abréviations que l'on utilise dans la présente spécification ont les significations suivantes : AcOH = acide acétique glacial ; Bzl = benzyle ; Me méthyle. Les valeurs Rf ont été déterminées sur des plaques (préalablement enduites) de gel de silice 60 F254 (marque commerciale de "Merck") ; épaisseur de la couche : 0,25 mm ; longueur de la plaque : 20 cm ; on a utilisé les systèmes de développement suivants (toutes les proportions étant indiquées en parties en volume) :
Système A : benzène : benzine (60-80) : acétate d'éthyle (25 : 5 : 70) Système B : benzène : acétate d'éthyle : acide acétique : eau (10 : 10 : 2 : 1) (phase supérieure) Système C : n-butanol : acide acétique : eau (4 : 1 : 1) Système D : chloroforme : méthanol : hydroxyde d'ammonium à 32% (65 : 45 : 20).
Les analyses par chromatographie sur couche mince n'ont pas été effectuées dans des conditions classiques, si bien que les valeurs Rf peuvent changer, en particulier, à des températures différentes. Les points de fusion ont été déterminés dans des capillaires ouverts et ils ne sont pas corrigés. L'électrophorèse sur papier sous haute tension est effectuée avec un appareil"Pherograph-Original Frankfurt Type 64" sur un papier de Schleicher et Schüll n 2317 à un pH de 1, 2 (acide formique : acide acétique : eau = 123 : 100 : 777

en volume) à 1. 600 V (40 V/cm). La mobilité électrophorétique (E., 2) est donnée par rapport à celle de 1 l'acide glutamique.
Exemple 1 Etape 1 : Préparation de Z-Asp (OBzl)-Phe-OMe (V) par condensation.
A une solution de 0,2 g de thionyldiimidazole dans 300 ml de dichlorométhane, on a ajouté successivement, à OOC, 35,7 g de Z-Asp (OBzl)-OH, 21,6 g de HCl. H-PheOme et 11,2 ml de N-méthylmorpholine. Après agitation à cette température pendant une heure et pendant 3 heures à la température ambiante, on a éliminé le solvant par évaporation sous vide.
On a dissous le résidu dans 400 ml de diméthylformamide, on l'a concentré à un faible volume et on l'a dilué avec de l'alcool éthylique. On a précipité le produit par addition goutte à goutte d'eau et on a séparé le précipité par filtration. On a recristallisé le composé brut dans un mélange de diméthylformamide et d'alcool éthylique pour obtenir 46, 1 g de (I) (rendement : 89%).

Etape 2 : Préparation de H-Asp-PheOme (I) par hydrogé- nation.
On a dissous 41, 5 g de Z-Asp (OBzl)-PheOMe (V) dans 300 ml de diméthylformamide et on a dilué avec 150 ml d'acide acétique et 150 ml d'alcool méthylique. Tout en agitant, on a ajouté 8 g de charbon palladié à 10% en poids et 24 g de formiate d'ammonium. Au terme de la réaction, on a éliminé le catalyseur par filtration et on a évaporé le solvant jusqu'à siccité. On a repris le résidu avec de l'eau, le pH étant maintenu à 6 avec un appareil de titrage automatique, et on a ajouté de la triéthylamine,

provoquant ainsi la précipitation du produit. On a laissé reposer le mélange pendant une nuit dans un réfrigérateur. Après filtration, on a mis le produit en suspension dans de l'alcool éthylique et on a agité pendant 3 heures à la température ambiante.

Synthesis of aspartame.
The invention relates to a new synthesis for aspartame, that is to say the methyl ester of a-L-aspartyl-L-phenylalanine. Like cane and beet sugar, aspartame has sweetening properties and is used as a sweetening agent for food and beverages as described in Belgian patent 717. 373.
Aspartame is a dipeptide and, as such, it is formed with an amido bond between an activated carboxy group of one amino acid and the amino group of another amino acid. Activation is necessary to increase the speed and efficiency of condensation. The desired pure peptide requires the protection of all other functional groups not involved in the formation of the peptide bond.
Finally, the protecting groups are eliminated.
Aspartame can be prepared by reacting the N-protected L-aspartic anhydride with the methyl ester of L-phenylalanine. In this way, a mixture of the methyl ester of L-aaspartyl-and of L-ss-aspartyl-L-phenylalanine is obtained, which requires the separation of the isomers. The N-protecting groups used are one of the N-protecting groups usually used in the chemistry of peptides such as the benzyloxycarbonyl (Z) groups (French patent 2,040,473) and t-butyloxycarbonyl (British patent 1,339,101) or, in order to avoid the final elimination of the N-protecting group, a simple protonation (US Patent 3,901,871).

The invention relates to a process for the synthesis of aspartame, which process consists in condensing a fully protected L-aspartic acid derivative with the methyl ester of L-phenylalanine in the presence of a condensing agent obtained by the reaction between a heterocyclic nitrogen compound and an organic or inorganic acid chloride, then remove the protective groups by hydrogenation with a hydrogen donor other than molecular hydrogen, in the presence of an acid.
The use of a fully protected L-aspartic acid derivative avoids the formation of the ss isomer. Condensants of the type indicated above can be readily obtained, while they are inexpensive and effective under moderate conditions. The removal of the protecting groups with a hydrogen donor other than molecular hydrogen can be carried out in a simple apparatus without having to resort to dangerous gaseous reagents. The presence of an acid in the last deprotection step makes it possible to avoid the formation of the untimely by-product that is diketopiperazine.

The hydrogenation can be carried out in a solvent such as dimethylformamide, dimethylacetamide, dimethylsulfoxide or N-methylpyrrolidone, containing an organic acid (preferably

(formic acid or acetic acid) or an inorganic acid (preferably hydrochloric acid) and a lower alkyl alcohol (preferably methyl alcohol or ethyl alcohol).
The reaction temperature can be between 0 and 650C.
Any conventional process can be adopted for the final recovery of aspartame.
