"Nouveaux dérivés de tétracycline et leur préparation"
Les tétracyclines constituent actuellement le groupe d'antibiotiques le plus répandu en thérapeutique. Leur large spectre antimicrobien permet le traitement efficace d'une notable série de processus infectieux.
Ces produits s'absorbent avec facilité par voie orale,

mais leur emploi systématique par cette voie peut présenter de sérieux inconvénients et, entre autres, pour citer les plus importants: la manifestation de superinfections intestinales, d'avitaminoses K et de fréquentes intolérances gastro-intesti- nales (vomissements, diarrhées).
L'emploi de composés apparentés est donc d'un grand intérêt. Pourtant, tous sont mal tolérés par voie intramuscu- laire, leur absorption étant peu abondante et inconstante ; voies hypodermiques et intraveineuses sont encore plus défavora- bles.
Dans les dernières années, on a obtenu des composés hydrosolubles par l'addition à la tétracycline de divers groupes d'un caractère très hydrophile.
Ces composés sont mieux tolérés et parfaitement absorbés ; il reste pourtant un problème, à savoir qu'étant donné leur absorption et élimination facile, il faut les administrer d'une façon réitérée, toutes les 6 - 8 heures,
L'invention concerne l'obtention de dérivés de la tétracycline peu solubles dans l'eau, de caractère lipophile nar- qué, stables et d'une absorption lente, complète et constante, et parfaitement tolérés par voie intramusculaire.
Les produits faisant l'objet du présent brevet sont de nouveaux sels des tétracyclines, de la formule générale T.(H03S - 0 - R)n CI) dans laquelle T est une molécule de tétracycline suivant la for- mule figurant dans la USP XVII, page 109. ou d'un dérivé théra- peutiquement acceptable, contenant le noyau de tétracycline et présentant une activité antimicrobienne, R est un radical alkyle, saturé ou non, à chaîne linéaire ou ramifiée, de 6 à 18 atomes de carbone, et n peut être égal à 1 ou 2.
Le radical tétracyclinique T peut donc être fourni par la tétracycline elle-même ou par n'importe quel composé actif

de celle-ci qui contient dans sa structure le noyau tétracycline, que ce composé soit soluble ou insoluble.
Les nouveaux alkyl-sulfates de tétracyclines de formule I se caractérisent par une notable diminution de leur solubilité dans l'eau par rapport à la tétracycline initiale mais, par contre, par leur solubilité accrue dans des solvants peu po- laires comme le chloroforme et les solvants hydroxyliques, tels que les alcools aliphatiques et les glycols. Du fait de ces caractéris- tiques spéciales de solubilité, le coefficient de partage K (CHCI3/tampon pH 6,8) est, pour ces alkyl-sulfates, beaucoup plus élevé et certaines de leurs valeurs ne sont obtenues pour aucune tétracycline utilisée dans la thérapeutique actuelle.
La valeur du coefficient de partage indiqué, qui est une mesure du caractère lipophile du composé, est considérée par divers auteurs comme un indice de sa plus grande absorption par voie buccale, et de son aptitude à atteindre des concentrations hématiques mieux soutenues.
Pour obtenir ces sels, on part des tétracyclines bases que l'on traite avec la quantité stoechiométrique d'acide alkyl-sulfurique (obtenu, par exemple, en faisant passer à travers une résine cationique fortement acide, une solution hydro-alcoolique du sel alcalin ou ammonique de ce sulfate), ou bien on fait réagir directement un sel inorganique de la tétra- cycline appropriée avec un sel alcalin ou ammonique de l'alkyl- sulfate, dans chaque cas en solution aqueuse ou en solution hydro- alcoolique.
Dans le premier cas, le composé formé est isolé par évaporation du solvant, avec ou sans addition d'un autre solvant qui entratne l'eau par voie azéotropique, et dans le second cas, le dérivé obtenu par précipitation est séparé, après lavage, par filtration ou centrifugation, séchage direct sous vide élevé ou lyophilisation, ou encore, après dissolution dans un alcool approprié, évaporation du solvant jusqu'à siccité.

tion du composé obtenu à partir de ses solutions alcooliques grâce à l'addition d'un grand volume d'un solvant très peu polai- re, par exemple de l'éther éthylique ou de l'hexane, avec ensui- te une dessiccation sous vide du précipité formé,
Finalement, un procédé à grand rendement consiste à soumettre les solutions alcooliques mentionnées, préalablement concentrées à un volume convenable, à un traitement d'atomisation dans un courant d'air chaud.
Les produits ainsi obtenus, dont le degré de pureté est généralement excellent, peuvent être soumis à une purifica- tion complémentaire, normalement par reprécipitation ou par des lavages aqueux d'une solution de ces produits dans un solvant peu polaire.
Les exemples non limitatifs suivants illustrent la préparation des composés nouveaux suivant l'invention.
Exemple 1
Dans 800 ml d'eau distillée soumise à une agitation énergique, on ajoute simultanément et à des vitesses similaires les solutions suivantes: 48,09 g (0,1 mole) de chlorhydrate de tétracycline dissous dans 40Q ml d'eau et 28,8 g (0,1 mole) de lauryl-sulfate sodique dissous dans 400 ml d'eau.
A la fin de l'addition, on prolonge l'agitation pendant environ 30 minutes et le précipité obtenu, sous la forme de flocons légers, est laissé au repos, on décante le liquide surnageant, on lave deux fois par remise en suspension du préci- pité dans environ 800 ml d'eau, en éliminant chaque fois l'eau par décantation, on filtre sur toile fine, on lave sur le filtre et finalement on presse.
On dissout la masse pâteuse qui retient l'eau par in- terposition, dans 400 ml d'alcool éthylique à 96 , on filtre à travers une plaque de verre poreux et on élimine le solvant sous vide jusqu'à siccité, en triturant jusqu'à obtention d'une

poudre fine et en dessé 'tant finalement sous vide élevé à
50-60 C pendant plusieurs heures.
On obtient ainsi 67,5 g (rendement de 95%) de lauryl-sulfate de tétracycline sous forme de poudre jaune claire, soluble dans les alcools aliphatiques, les glycols, l'acétone et le dioxane, légèrement soluble dans l'éther et le benzène et insoluble dans l'eau. Point de fusion 128-130 C.

De la même manière, on peut, au lieu du chlorhydrate de tétracycline, utiliser les chlorhydrates de 5-oxytétracycline, de 6-méthylène-oxytétracycline ou de déméthylchlorotétracycline à titre d'exemples de composés contenant le noyau tétracyclinique, ce qui donne naissance aux alkyl-sulfates correspondants.
Exemple 2
On ajoute lentement et avec agitation une solution de 48,09 g (0,1 mole) de chlorhydrate de tétracycline dans envi- ron 400 ml d'eau distillée, à une autre solution de 28,8 g (0,1 mole) de lauryl-sulfate de sodium dans environ 1000 ml d'eau distillée. On poursuit l'agitation pendant environ 30 minutes et le précipité formé est lavé par décantation et remise en sus- pension dans de l'eau, on dissout avec agitation dans 250 ml d'alcool éthylique à environ 40-50 , on concentre la soluti.on for-

mée à la moitié de son volume et on l'ajoute à 1 litre d'éther éthylique.
On filtre le précipité, on triture avec de l'éther de pétrole et on dessèche sous vide pour obtenir, avec un rende- ment de 90%, un produit ayant les mêmes caractéristiques que celui décrit dans l'exemple précédent, Exemple 3
On ajoute à des vitesses identiques et à jets fins, à environ 40 1 d'eau soumis à une agitation intense, une solu- tion de 2,404 kg (5 moles) de chlorhydrate de tétracycline dans 20 1 d'eau distillée et une solution de 1,44 kg(5 moles) de lauryl-sulfate de sodium dans 20 1 d'eau distillée.
On lave le précipité formé, une fois par remise en suspension dns envi- ron 30 1 d'eau, et l'ensemble est centrifugé jusqu'à obtention d'une masse pâteuse qui conserve un degré d'humidité variable suivant la vitesse et la durée de centrifugation; finalement, cette pâte est émiettée et dissoute dans environ 15 1 d'alcool isopropylique, préalablement chauffé à 45-50 C. Là solution alcoolique concentrée jusqu'à un volume approprié sous vide est soumise à une atomisation dans un appareil approprié, pour obte- nir ainsi un produit de caractéristiques physiques et chimiques identiques à celles du produit de l'Exemple 1, sauf en ce qui concerne la microstructure qui est beaucoup plus poreuse, ce qui se traduit par une densité apparente inférieure.
ExempLe 4
2,642 g (Smnoles) de chlorhydrate de déméthylchloro- tétracycline dissous dans envron 200-250 ml d'eau distillée sont traitée en agitant avec une solution de 1,442 g (5mmoles) de lauryl-sulfate de sodium dans 15 ml d'alcool à 10% ; leprécipité, filtré et lavé avec de l'eau, est dissous dans de l'éthanol à
96 et le solvant est évaporé jusqu'à siccité sous pression ré- duite et à basse température. On obtient avec un rendement de
98% le lauryl-sulfate de déméthylchlorotétracycline poudre jaune

intense, soluble dans les alcools aliphatiques et l'acétone, un peu soluble dans l'éther, insoluble dans l'éther de pétrole et très insoluble dans l'eau, p. f. 136-138 .
Le spectre dans l'infrarouge présente les bandes prévues par la théorie.
Par ce mode opératoire, on peut de même obtenir les produits mentionnés dans l'exemple 1..
Exemple 5
A 3,04 g (6,25 mmoles) de (2-hydroxyéthyl)-pipéra- zinométhylène-tétracycline (mépicycline) dissous dans 25 ml de méthanol/eau (1/1) et additionnés de 6,25 ml de HCl 1 N on ajoute une solution de 1,801 g (6,25 mmoles) de lauryl-sulfate de sodium dans 10 ml de méthanol/eau (1/1). Le précipité formé est filtré et lavé à l'eau; on dissout dans de l'alcool, on élimine celui-ci sous pression réduite jusqu'à siccité ou on dessèche directement le précipité humide sous vide élevé en présence d'anhydride phos- phorique.
On obtient avec un rendement de 94% le lauryl-sulfate de mépicycline (relation molaire 1/1 des deux constituants) qui est une poudre jaune, insoluble dans 1.'eau, très soluble dans le méthanol, soluble dans l'alcool éthylique, les alcools aliphati- ques et l'acétone, assez soluble dans les glycols et peu soluble dans l'éther, p.f. 140-142 C. Le spectre dans l'infrarouge pré- sente les bandes prévues par la théorie.
Si l'on utilise, au lieu de la tétracycline soluble mentionnée, la pyrrolidino-méthyltétracycline ou la tétracycline- L-méthylènelysine, comme exemples de tétracyclines hydrosolubles, on obtient les alkyl-sulfates correspondants avec des propriétés physico-chimiques analogues. Comme exemple, on citera: le lauryl- sulfate de pyrrolidino-méthyltétracyclinc p.f. 132 C, ayant les mêmes solubilisés que son homologue décrit dans l'exemple, sauf une certaine augmentation de la solubilité dans l'eau; le lauryl- sulfate de tétracycline-L-méthylènelysine, p.f, 156-158 C, un peu soluble dans l'eau, peu soluble dans l'alcool, le méthanol et le

propylène glycol, et insoluble dans l'acétone et léther.
Exemple 6
0,971 g (2 mmoles) de (2-hydroxyéthyl)-pipérazino- méthylènetétracycline dissous dans la quantité la plus petite possible de méthanol, et additionnés au préalable de 4 ml de HCl 1 N, sont traités en agitant avec une solution de 1,152 g (4 mmoles) de lauryl-sulfate de sodium dans 10 ml d'eau/méthanol (1/1). Le précipité grumeleux est lavé avec du méthanol à 10% et finalement 2 fois avec de l'eau distillée; il est séché sous pression réduite pendant plusieurs heures.
On obtient, avec un rendement de 90-92%, le bis- (lauryl-sulfate) de mépicycline, sous la forme d'une poudre d'un jaune intense, un peu soluble dans l'eau, soluble dans les alcools aliphatiques, les glycols et l'acétone et insoluble dans l'éther; point de fusion 124-125 . On obtient un spectre dans l'infra- rouge conforme à la théorie.
On peut remplacer la mépicycline par n'importe quelle tétracycline hydrosoluble telle que celles antérieurement men- tionnées à la fin de l'exemple 5.
Exemple 7
On ajoute lentement et avec agitation une quantité de tétracycline base équivalant à 4,444 g (0,01 mole) du composé anhydre, à une solution de 2,664 g(0,01 mole) d'acide lauryl-sulfu- rique dans environ 50 ml d'alcool éthylique à 80% jusqu'à disso- lution complète.
On filtre la solution pour éliminer les impuretés possibles et on évapore jusqu'à siccité sous pression réduite, On triture le résidu et on sèche sous vide élevé. On obtient avec un rendement de 98 à 99% le lauryl-sulfate de tétracycline, ayant des caractéristiques identiques à celles du produit décrit antérieurment dans l'exemple 1.

peuvent être préparés préalablement en partant, par exemple, du chlorhydrate de tétracycline et du lauryl-sulfate de sodium de la façon suivante. On fait passer 2,884 g (0,01 mole) de lauryl-sulfate de sodium dissous dans 30 ml d'alcool à 80% à travers la ml de résine échangeuse de cations, telle que la résine Merck 1 ou IR 120, sous la forme activée à l'acide, à une vitesse de 1-1,5 m1/min. On lave la colonne avec un excès d'alcool à 80% jusqu'à ce que l'éluat ne donne pas de réaction acide. La solution hydro-alcoolique obtenue peut s'utiliser directement comme pour la préparation avec la tétracycline base.
Ceci peut s'obcenir, par exemple, par neutralisation d'une solution aqueu- se du chlorhydrate de tétracycline à 10% avec des alcalis (par exemple du NaOH à 2%) jusqu'au pH de 4-5. On filtre le précipi- té obtenu, on lave à l'eau et on dessèche sous vide.
Il sera entendu que l'invention n'est pas limitée aux exemples de mise en oeuvre décrits ci-dessus, l'invention englobant toutes les autres formes de réalisation et produits équivalents.

Tetracyclines are currently the most widely used group of antibiotics in therapy. Their broad antimicrobial spectrum allows the effective treatment of a remarkable series of infectious processes.
These products are easily absorbed by the oral route,

but their systematic use by this route can present serious drawbacks and, among others, to name the most important: the manifestation of intestinal superinfections, avitaminosis K and frequent gastrointestinal intolerance (vomiting, diarrhea).
The use of related compounds is therefore of great interest. However, all of them are poorly tolerated by the intramuscular route, their absorption being scarce and inconsistent; hypodermic and intravenous routes are even more unfavorable.
In recent years, water-soluble compounds have been obtained by the addition to tetracycline of various groups of a very hydrophilic character.
These compounds are better tolerated and perfectly absorbed; however, there remains a problem, namely that given their easy absorption and elimination, they must be administered repeatedly, every 6 - 8 hours,
The invention relates to obtaining tetracycline derivatives which are sparingly soluble in water, of narcotic lipophilic character, stable and of slow, complete and constant absorption, and perfectly tolerated by the intramuscular route.
The products which are the subject of the present patent are new salts of tetracyclines, of the general formula T. (H03S - 0 - R) n CI) in which T is a tetracycline molecule according to the formula appearing in USP XVII. , page 109. or a therapeutically acceptable derivative, containing the tetracycline ring and exhibiting antimicrobial activity, R is an alkyl radical, saturated or not, straight or branched chain, of 6 to 18 carbon atoms, and n can be equal to 1 or 2.
The tetracycline radical T can therefore be provided by the tetracycline itself or by any active compound

thereof which contains in its structure the tetracycline ring, whether this compound is soluble or insoluble.
The new tetracycline alkyl sulphates of formula I are characterized by a marked decrease in their solubility in water compared to the initial tetracycline but, on the other hand, by their increased solubility in less polar solvents such as chloroform and hydroxylic solvents, such as aliphatic alcohols and glycols. Because of these special solubility characteristics, the partition coefficient K (CHCl3 / buffer pH 6.8) is much higher for these alkyl sulfates and some of their values are not obtained for any tetracycline used in the formulation. current therapy.
The value of the partition coefficient indicated, which is a measure of the lipophilic character of the compound, is considered by various authors as an index of its greater absorption by the oral route, and of its ability to achieve better sustained blood concentrations.
To obtain these salts, one starts with tetracycline bases which are treated with the stoichiometric amount of alkyl-sulfuric acid (obtained, for example, by passing through a strongly acidic cationic resin, a hydro-alcoholic solution of the alkali salt. or ammonia thereof), or an inorganic salt of the appropriate tetracyclin is reacted directly with an alkali or ammonium salt of the alkyl sulfate, in each case in aqueous solution or in aqueous-alcoholic solution.
In the first case, the compound formed is isolated by evaporation of the solvent, with or without the addition of another solvent which entrains the water by azeotropic route, and in the second case, the derivative obtained by precipitation is separated, after washing, by filtration or centrifugation, direct drying under high vacuum or lyophilization, or alternatively, after dissolution in a suitable alcohol, evaporation of the solvent to dryness.

The products thus obtained, the degree of purity of which is generally excellent, can be subjected to a further purification, normally by reprecipitation or by aqueous washes of a solution of these products in a poorly polar solvent.
The following non-limiting examples illustrate the preparation of the new compounds according to the invention.
Example 1
In 800 ml of distilled water subjected to vigorous stirring, the following solutions are added simultaneously and at similar rates: 48.09 g (0.1 mol) of tetracycline hydrochloride dissolved in 40Q ml of water and 28.8 g (0.1 mole) of sodium lauryl sulfate dissolved in 400 ml of water.
At the end of the addition, stirring is continued for about 30 minutes and the precipitate obtained, in the form of light flakes, is left to stand, the supernatant liquid is decanted, washed twice by resuspending the preci - pité in about 800 ml of water, each time removing the water by decantation, filtered through a fine cloth, washed on the filter and finally pressed.

The precipitate is filtered off, triturated with petroleum ether and dried under vacuum to obtain, with a yield of 90%, a product having the same characteristics as that described in the preceding example, Example 3.

Example 5
To 3.04 g (6.25 mmol) of (2-hydroxyethyl) -piperazinomethylene-tetracycline (mepicycline) dissolved in 25 ml of methanol / water (1/1) and added with 6.25 ml of 1 N HCl a solution of 1.801 g (6.25 mmol) of sodium lauryl sulfate in 10 ml of methanol / water (1/1) is added. The precipitate formed is filtered off and washed with water; it is dissolved in alcohol, removed under reduced pressure to dryness or the wet precipitate is dried directly under high vacuum in the presence of phosphorus pentoxide.
Mepicycline lauryl sulfate is obtained with a yield of 94% (1/1 molar relationship of the two constituents) which is a yellow powder, insoluble in water, very soluble in methanol, soluble in ethyl alcohol, aliphatic alcohols and acetone, fairly soluble in glycols and sparingly soluble in ether, mp 140-142 C. The infrared spectrum shows the bands predicted by theory.
If, instead of the mentioned soluble tetracycline, pyrrolidino-methyltetracycline or tetracycline-L-methylenelysine are used as examples of water-soluble tetracyclines, the corresponding alkyl sulfates are obtained with similar physicochemical properties. As an example, mention will be made of: pyrrolidino-methyltetracyclinc lauryl sulfate m.p. 132 C, having the same solubilized as its counterpart described in the example, except a certain increase in the solubility in water; tetracycline-L-methylenelysine lauryl sulfate, mp, 156-158 C, slightly soluble in water, slightly soluble in alcohol, methanol and

propylene glycol, and insoluble in acetone and ether.
Example 6
0.971 g (2 mmol) of (2-hydroxyethyl) -piperazino-methylenetetracycline dissolved in the smallest possible quantity of methanol, and previously added with 4 ml of 1 N HCl, are treated while stirring with a solution of 1.152 g ( 4 mmol) of sodium lauryl sulfate in 10 ml of water / methanol (1/1). The lumpy precipitate is washed with 10% methanol and finally twice with distilled water; it is dried under reduced pressure for several hours.
Mepicycline bis- (lauryl-sulfate) is obtained with a yield of 90-92%, in the form of an intense yellow powder, slightly soluble in water, soluble in aliphatic alcohols, glycols and acetone and insoluble in ether; melting point 124-125. We obtain an infrared spectrum in accordance with the theory.
Mepicycline can be replaced by any water-soluble tetracycline such as those previously mentioned at the end of Example 5.
Example 7
A quantity of tetracycline base equivalent to 4.444 g (0.01 mole) of the anhydrous compound is added slowly and with stirring to a solution of 2.664 g (0.01 mole) of lauryl-sulfuric acid in about 50 ml of 80% ethyl alcohol until complete dissolution.
The solution is filtered to remove possible impurities and evaporated to dryness under reduced pressure. The residue is triturated and dried under high vacuum. Tetracycline lauryl sulfate is obtained with a yield of 98 to 99%, having characteristics identical to those of the product described previously in Example 1.

can be prepared beforehand starting, for example, from tetracycline hydrochloride and sodium lauryl sulfate as follows. 2.884 g (0.01 mole) of sodium lauryl sulfate dissolved in 30 ml of 80% alcohol is passed through the ml of cation exchange resin, such as Merck 1 or IR 120 resin, in the form acid activated, at a rate of 1-1.5 m1 / min. The column is washed with an excess of 80% alcohol until the eluate does not give an acid reaction. The hydro-alcoholic solution obtained can be used directly as for the preparation with tetracycline base.
This can be achieved, for example, by neutralizing a 10% aqueous solution of tetracycline hydrochloride with alkalis (eg 2% NaOH) to pH 4-5. The resulting precipitate is filtered off, washed with water and dried in vacuo.
