g) la réaction est économique, car seules des matières inorganiques peu coûteuses sont consommées (c'est-à-dire le sulfate ferreux, le trioxyde de chrome, l'acide sulfurique).-
Bien que l'acétone ait été indiquée comme solvant, certaines substitutions limitées peuvent lui être faites.
Ainsi, par exemple, le tétrahydrofurane peut être employé, mais il est sujet à une plus lente vitesse d'oxydation dans la position 3-hydroxy. La butanone se oomporte d'une façon analogue à l'acétone dans la réaction; toutefois, elle est sujette à certaine inconvénients car 1'isolement du produit et la récupération du solvant ne sont pas aussi faciles qu'avoo l'acétone, en grande partie à cause du point d'ébullition plus élevé de la butanone.
Il est possible dans une certaine mesure d'employer un solvant supplémentaire en même temps que l'acétone aux fins de l'augmentation do la solubilité des stéroïdes dans le m6lan- ge réactionnel. Toutefois, les mélanges do solvants employés, par exemple le 1,1-diohlorom6thane et l'acétone, se sont montrés susceptibles d'une réaction incomplète ot ont réduit la sélectivité de l'oxydation. En outre, on doit employor un

La réaction proprement dite est effectuée à des températures dans la gamme de 0 à 25 C, l'extrémité iatériaura de la gamme de température étant meilleure pour les #5-atéroides qui n'ont pas de substituants 6-méthyle (ou alcoyle). Cet aspect de l'invention observé expérimentalement tourne apparemment autour de la plus grande stabilité de la double liaison C-5,6 lorsqu'un substituant 6-méthyle est présent sur la molécule.de stéroide. L'isomérisation en #4 se produit bien plus lentement dans le cas des 6-méthylstéroïdes.
Inversement, la plus grande stabilité de la double liaison en C-5,6 dans les 6-méthylstéroides permet de réduire la durée de la réaction d'oxydation, d'utiliser moins d'acétone pour maintenir le stéroïde en solution et d'employer l'extrémité supérieure de la gamme de température de réaction de 0 à 25 C.
La réaction est très sélective, pourvu quo le temps de contact de réaction soit maintenu dans les limites pres- orites de moine d'environ 6 minutes, une gamme de 3 à 6 minutes étant préférée. Les doubles liaisons isolées disposées ailleurs dans la molécule do stéroïde ne sont pas affectées en général par l'acide chromique utilisé comme réactif.
Généralement, des stéroïdes ayant une double liaison en C-7, C-9 (11), C-6, C-14 (15), C-16 peuvent être oxydés sans donner lieu à des concentrations gênantes de produits secondaires indésira- bles,
Le choix de la quantité d'eau appropriée pour arrêter la réaction par le sulfate ferreux et l'isomérisation subsé- quente in situ ne présenta pas de problème particulier car en général, les #5-3-cétones sont plus solubles dans l'acétone aqueuse aux températures de reflux que les #4-3-cétones cor- respondantes. En gros, la quantité d'eau présente aveo le

sulfate ferreux est environ égale au volume de solution acéto- nique initiale.

3-cétone saturée (dont on sait qu'elle se oétalise facilement).

Les composés 0 5-3-c6to-6-alooylés n'ont pas besoin d'être isolée lorsqu'ils doivent être utilisés pour obtenir les 3-4thyléne-oétale. Ainsi, le produit de la réaotion d'oxy- dation peut être extrait aveo le dichlorométhano, l'extrait étant lavé deux fois avec de l'eau et concentré à sec. En réalité, les composés de l'invention ont un spectre étendu,

mais diffèrent de propriétés pharmacologiques, comparativement aux #4-3-cétones. Dans certains cas, l'action biologique est renversée. Par exemple, loraque dos rata subissent l'ova- riotomie le sixième jour après l'accouplement et reçoivent par voie sous-cutanée et voie orale la 6a-méthyl-17a-acétoxy- progestérone (0,1 mg) pendant 7 jours et les composés #3 de l'invention à la même dose, seuls les rats qui ont reçu la 6a-méthyl-17a-acétoxyprogestérone restent gravides.
Cette aotion prcgeatationnelle très sélective rond les composés de l'invention très utiles lorsque la prédominance des effets indésirables peut limiter l'application des substances progestationelles connues.
L'invention est illustrée par les exemples suivante, donnés à titre non limitatif
Exemple
1,0 g de #5-pregnèn-3ss-ol-20-one fut diosoun dans 175 ml d'acétone. La solution fut refroidie dans la glace pendant 10 minutes et on ajouta rapidement 2 ml do solution de Jonos, puis le mélange fut maintenu au bain pendant 3 minutes en agitant énergiquement. La couleur vira do l'orangé au brun verdâtre. Le mélange réactionnel fut ensuite versé dans un ballon à fond rond oontenant 100 ml d'une solution aqueuse qui, contenait 1 g de sulfate ferreux et 1 ml d'acide sulfurique concentré. Ce mélange fut chauffé au bain de vapeur sous agitation jusqu'à ce quo l'acétone no soit plus chass"e par distillation, Le mélange fut refroidi et filtré pour récu- p6ror le précipité granulaire.
Il tut lavé sur le filtre avec de grandes quantités d'eau, recueilli et séché. Le poids total de produit brut fut de 1,05 g et la couleur fut légère- mont vert grisâtre. Cette matière fut broyée dans un mortier aveo 250 mg de charbon activé, et transférée quantitativement dans une cartouche d'extraction (50 mg de oharbon supplémen-

tairea utilisée pour le transfert), qui fut placée dans un appareil d'extraction de Soxhlet et on effectua une extraction par épuisement à l'éther. L'extrait d'éther fut transféré dans un tube à essai et concentre encore à un volume final d'environ
2 ml. les oristaux furent filtrés, lavés sur le filtre avec un peu d'hexane, recueillis et séchés.
On obtint 945 mg de progestérone, fondant entre 128,5 et 1309C, (a)D200 dans le chloroforme, Les cristaux étaient blancs et furent trouvée identiques sous tous les rapporte avec un échantillon au- thentique (par détermination du point de fusion mixte et comparaison des spectres infrarouges).
Exemple 2
A une solution de 1,0 g de déhydroépiandrostérone dans 135 ml d'acétone, refroidie au bain d'eau à 0 C, on ajouta
2,1 ml de solution de Jones et le mélange fut agité (au bain de glace) pendant 3 minutes. Le mélange réactionnel fut versé dans un ballon oontenant 0,7 g de sulfate ferreux, 0,5 ml d'acide sulfurique et 85 ml d'eau, puis chauffé au bain de vapeur sous agitation junqu'à ce que de l'acétone ne soit plus chassée par distillation, Le précipité granulaire fut filtré, lavé à l'eau, séché et reoristallisé dans l'éther. On obtint
875 mg d'androstène-dione fondant entre 171,5 et 173QC, (a)D 189 (chloroforme). Le produit tut trouvé identique à un échantillon authontique.

On ajouta au résidu (après l'oxydation précédente) 10 ml d'acide acétique cristallisable et 5 ml furent chassés par distillation sous vide pour éliminer le dichlorométhane résiduel. On ajouta à cette solution pendant le refroidissement à +159C, 1 ml d'une solution de 32 % d'acide bromhyarique dans l'acide aoétique (préalablement refroidie à 159C). La mélange tut maintenu sous agitation un certain temps à la température ambiante (15 à 20 minutes) et pendant cette période, il se transforma progressivement en une boue cristalline. Cotte boue fut refroidie à 16 C et maintenue pendant 15 minutes à cette température, filtrée, et les cristaux, aveo un peu d'éther, turent recueillis et séch6a dans un dessiccateur préparé aveo de la chaux sodée.
On obtint 1,12 g de 17a-hydroxy-21-
