Le présent certificat d'addition a pour objet un vaccin combiné pour immuniser des chiens ou des renards simultanément contre la maladie de Carré, l'hépatite contagieuse du chien et les leptospiroses (maladie de Stuttgart et maladie de Weil), ainsi qu'un procédé de préparation de ce vaccin selon lequel on inactive de manière connue un produit contenant le virus de l'hépatite infectieuse du chien, obtenu par *:

tion de foies ou de rates de chiens ou de cultures de tissus* au moyen du formaldéhyde, puis on lie le formaldéhyde libre par addition d'une solution contenant 1,5 à 2 fois la quantité stoechiométrique de sels acides de l'acide sulfureux, en par* ticulier de bisulfite de sodium, on mélange le produit avec une suspension obtenue à partir d'une culture de tissu$ selon le brevet allemand N 1.138.888. et contenant un virus de la maladie de Carré médité par au moins 50 passages en série, avec une suspension de culture de Leptospira canicola tuée par refroidissement à très basse température et avec une suspension de Culture de Leptospira icterohaemorrhagiae, et on lyophilise ce mélange. Il n'est pas absolument nécessaire d'effectuer la lyophilisation.
On peut utiliser la suspension obtenue telle quelle pour la vaccination.
Mais la lyophilisation offre l'avantage que le vaccin obtenu est stable et conservable.
Il y a intérét à maintenir sensiblement égalée dans le mélange la portion agissant coûtée le virus de la maladie de Carré, qui est constituée par environ 25 % d'un produit obtenu à partir de culture de tissu contenant le Virus modifia de la maladie de Carré, 60 % de bouillon de boeuf ayant un pH de 7,6 et 15 % d'une solution de glucose à 50 %, et la portion agissant contre l'hépatite contagieuse du chien, qui comprend environ 90 % d'un extrait contenant le virus inactivé, de l'hépatite contagieuse du chien et 10 % d'une solution d'hydre xyde d'aluminim m à 2 %.
A 20 parties en volumes de ce mélangé on ajoute 1 partie en volume d'une suspension de culture de Leptospira canicola, réglée à une densité de 100.106 germes par ml à 1'ultra-microscope et tuée par refroidissement à très basse température, et 1 partie en volume d'une suspention de culture de Leptospira ioterohaemorrhagiae réglée à une densité de 100.106 germes par ml à l'ultra-microscope et

Avant la vaccination, aucun des chiens ne possède d'anticorps contre la maladie de Carré et l'hépatite contagieux se du chien, ni d'aglutinines contre Leptospira canioola et Leptospira icterohaemorrhagiae.
Quatre semaines après la vaccination, tous le chiens possèdent Une quantité considéra** ble et égale d'anticorps neutralisant les virus de la maladie de Carré et de l'hépatite contagieuse du chien, ainsi que d'ag- glutinines centre Leptoapira canicola et Leptospira ictérohaemorrhagiae.
On détermine les doses neutralisant le virus selon la méthode de Recd et Münch mentionnée ci-dessus. Infection d'épreuve avec le virus pathogène de la maladie de Carré et de l'hépatite contagieuse du chien
6 semaines après vaccination, on inocule aux chiens vaccinés,ainsi qu'aux 2 chiens témoins, la maladie de Carré par voie sous-cutanée avec une souche de virus de la maladie de
Carré pathogène pour le chien* Chez des chiens sensibles à la maladie de Carré, la souche de virus utilisée conduit à ce qu'on appelle le stade viral de la maladie de Carré, stade qui est caractérisé par courbe fébrile à deux commets* Au moment de la deuxième poussée fébrile, on peut déceler le virus de la maladie de Carré, dans les organes des chiens malades, par la réaction de déviation du complément.
Après l'infection d'épreuve, tous les chiens vaccinés restent en bon état général et apyrétiques, tandis que les deux chiens témoins non traitée présentent les symp- tomes typiques d'une infection par la maladie de Carré.

On met en évidence la présence du virus par la réaction de déviation du complément.
Deux semaines après l'infection par le virus de la maladie de Carré,on infecta tous les chiens vaccinés ainsi que les deux chiens témoins atteints d'hépatite contagieuse par une souche de virus de l'hépatite contagieuse du chien virulent pour le chien. Tous les chiens vaccinés restent bien portants et ne montrent aucun symptôme d'hépatite contagieuse, tandis que les animaux témoins non traités sont atteints de la fièvre de l'hépatite contagieuse du chien et une partie y succombe. On met en évidence la présence du virus de l'hépatite contagieuse du chien par l'autopsie des animaux et par la réaction de déviation du complément.
B) Examen des animaux de laboratoire.
En plus de l'essai sur le chien, on examine le vaccin préparé selon le procédé de la présente invention sur la souris blanche, le cobaye et le hamster doré, au point de vue tolérance et innocuité. Pour étudier l'activité du vaccin contre les leptospiroses (maladie de Stuttgart et maladie de Weil) on utilise aussi le cobaye et le hamster doré. Pour l'étude de la tolé- rance et de l'innocuité, on vaccine les souris blanches par voie intracérébrale et les cobaye* et les hamsters par voix souscutanée avec le vaccin redissous. On observe les animaux pendant 10 jours, il ne doivent as tomber malades.
Pour l'examen d'efficacité du vaccin contre Leptoapira canicola et Leptospira ictéorhaemorrhagiae, on vaccine à un intervalle d'une semaine des cobayes avec chacun 0,5 ml et des hamsters dorées avec 0,3 ml chacun du vaccin redissous. Deux semaines après vaccination, on fait une prise de sang aux animaux et on examine le sérum obtenu au point de vue de sa teneur en agglutinines (anticorps) contre Leptospira canicola et Leptospira ictérohaemorrhagiae.

Au lieu des examens sérologiques pour déterminer le anticorps contre les leptospires, l'efficacité du vaccin peut être étudiée par une infection d'épave avec des cultures de leptospires pathogènes. Le test représenté dans le tableau III est effectué de la même manière que celui du tableau II.
Au lieu de la sainée, on infecte des hamstern dorés, par voie intra. péritonéale, avec des cultures de Leptospira canicula pathogène.
Les animaux vaccinés restnt en bon état tandis que les animaux témoins non traités tombent malades ou meurent de leptospiroses.
Le diagnostic est confirma par le résultat de l'autopsie.

B) Examination of laboratory animals.
