La présente invention se rapporte à des produits de transformation de Céphalosporine C et à leurs dérivés, ainsi Que'-- des procédés pour la préparation de ces composés.
Le brevet anglais n 810.196 décrit un procédé de sépa- ration d'un nouvel antibiotique, la Céphalosporine C, à partir des produits d'un processus de fermentation utilisant une espace de Céphalosporiun. La structure de la Céphalosporine C était alors inconnue et de ce fait a rendu plus difficiles les travaux sur la préparation et l'identification des produits de transfor- mation et dérivés de la Céphalosporine C qui pourraient également présenter des activités biologiques utiles.
Certains travaux, toutefois, ont été effectués sur le traitement par les acides de la Céphalosporine C. C'est ainsi

L'hydrolyse de la Céphalosporine C, Cc ou CA ou de leurs dérivés à chaîne latérale protégée est effectuée de préfé- rence en utilisant un acide minéral dilué, et l'acide chlorhydrique dilué convient particulièrement. D'autres acides appropriés sont les acides sulfoniques et les résines de polystyrène sulfonées.
Pour l'exécution de ce procédé, la solution traitée par l'acide contenant la Céphalosporine C, Cc' CA ou un dérivé protégé par une chaîne latérale est généralement laissée à reposer jusqu'à ce qu'on ait obtenu un rendement relativement élevé de noyau, et la durée de ce séjour dépend de la sévérité des conditions nais est normalement de l'ordre de quelques jours. Les noyau:: pe@vent être alors séparés des produits de répction par des procédés con- nus en soi, par exemple l'électrophorse sur papier et/ou la chro- matographie sur papier.
En utilisant ces procédés, les noyaux sépa- rés peuvent généralement être identifiés sur le papier par le fait qu'ils ne présentent pas d'activité antibiotique par eux-mêmes aux concentrations utilisées.(L'essai consiste à placer le papier d'électrophorèse en contact avec ces plaques ensemencées de Star:-::,.- lococcus aureus /--Souche Oxford-7 mais possèdent une activité après un traitement par le chlorure de phénylacétyle provoquant la phénylacétylation. Ce traitement peut être avantageusement effec- tué en pulvérisant d'abord sur le papier de la M-pyridine dans de l'acétone à 50% puis du chlorure de phénylacétyle (1,25% volue/ volume) dans l'acétone.
Le dérivé phénylacétyle peut être séparé par des moyens chromatographiques et électrophorétiques.

Dans une autre forme de l'invention, des noyaux de compo- sés de Céphalosporine CA sont préparés par un procédé s ivant lequel on traite la Céphalosporine C ou un dérive protégé par une chaîne latérale de Céphalosporine C par un acide de fanon à former le noyau, puis on traite le'noyau ainsi formé par une base tertiaire faible pour foriner le noyau de Céphalosporine CA
Les conditions dE réaction pour la préparation du noyau. de Céphalosporine C à partir d'un dérivé protegé par une chaîne latérale de Céphalosporine C entraînent de prférerce l'emrloi d'acide chlorhydrique, particulièrement aux concentrations compri- ses entre 0,1 et 0,3 N. La réaction est de préférence exécutée dans un solvant organique miscible à l'eau, par exemple l'acéto- nitrile.
Avantageusement, la réaction est effectuée dans l'obseu- rité et en atmosphère inerte, par exemple en atmosphère d'azote.
Les conditions de réaction pour la préparation de nova@x de composés de Céphalosporine C, Cc et CA à partir de compos s de Céphalosporine C, cc et CA respectivement peuvent varier conside- rablement mais un procédé préféré comprend l'emploi d'acide chlon- hydrique à une concentration allant du pH 0 au pH 2,5 à une tempé- rature comprise entre -10 et + 50 C environ pendant une durée de 12 heures environ à 30 jours, les conditions étant choisies l'in- térieur de ces limites de façon à obtenir un rendement élevé de produits d'hydrolyse. Un procédé qui convient remarquablement bien comprend l'emploi d'acide chlorhydriuue 0,1 N à une température d'environ 20 C pendant 3 jours environ.
Lors du traitement de la Céphalosporine C par un acide, d'une concentration dans la gamme indiquée plus haut, au :noins

deux réactions s'amorcent.Ces réactions sont (1) l'hydrolyse de la Céphalosporine C qui ouvre les liaisons joignant le noyau à la chaîne latérale et (2) la réaction de l'acide avec la molécule de 'Céphalosporine C pour fournir la Céphalosporine Cc. En outre, aussitôt qu'une certaine quantité de Céphalosporine Cc est obte- nue dans le mélange de réaction, une. troisième réaction s'amorce, l'hydrolyse de la Céphalosporine Cc ouvrant la liaison joignant le noyau de la Céphalosporine Cc à la chaîne latérale. En plus, une certaine quantité de noyau de Céphalosporine Cc se forme par réaction de l'acide avec le noyau de Céphalosporine C.
C'est ainsi, que si.-le traitement de la Céphalosporine C par l'acide est effec- tué dans le but d'obtenir le noyau de Céphalosporine C, les con- ditions de réaction sont choisies de façon à obtenir une propor- tion relativement élevée de noyau de Céphalosporine C et une pro- portion relativement faible de noyau de Céphalosporine Cc De même, si la réaction est effectuée dans le but d'obtenir le noyau de Céphalosporine Cc, les conditions de réaction sont choisies de façon à obtenir une proportion relativement élevé de noyau de Céphalosporine Cc.
Les conditions les plus avantageuses pour la réaction peuvent être établies par des expériences simples et on a trouvé qu'au départ de Céphalosporine C, l'acide chlorhydrique 0,ln à 20 C pendant 3 jours environ procure un bon rendement de noyau de Céphalosporine C et l'acide chlorhydr que 0,5 N à 1 N à 20 C pendant 3 jours environ procure un bon rendement de noyau de Céphalosporince Cc Ces conditions peuvent être comparées au traitement par l'acide chlorhydrique 0,1 N à 20 C pendant 5 jours environ au bout desquels on obtient d'après le tableau 3 de la demande de brevet anglais n 8544/57 les meilleurs rendements de Céphalosporine Cc susceptible d'être obtenus à l'aide d'acide chlorhydrique 0,
1 N et au bout desquels le noyau de Céphalosporine C qui s'est formé à un stade antérieur a pratiquement disparu.
On notera que le noyau de Céphalosporine C n'est pas un des composes acides relevés .:lais non caractérisés dans le Symposium Ciba cité plus haut pour les raisons suivantes.Tout d'abord, aprèstraitement de la Céphalosporine C par de l'acide chlorhy-

drique 0,1 N à 20 C pendant plus de 5 jours on peut supposer qu'il ne reste pratiquement plus de noyau de Céphalosporine C. Les condi- tions optima,. pour obtenir les noyaux de Céphalosporine C sont de l'acide chlorhydrique 0,1 N pendant 3 jours et l'éventail des temps pour lesquels on obtient des rendements significatifs de noyau est étroit. C'est ainsi qu'après plus de 4 jours, il ne reste pra- tiquement pas de noyau de Céphalosporine C dans la solution.
En second lien' les expériences subséquentes ont montré que le noyau de Céphalospor@@@@@@ serait pas prés@@@@quantités suffisa pour donner une réaction positive à la ninhydrine, même si le trai- teillent n'avait duré que penda t la durée optimum de 3 jours. Dans ces conditions optima, le noyau est décelé à un moment ultérieur par phénylacétylation et par un essai d'activité comme indiqué plus haut. En tout cas, même lorsqu'une solution contenant le noyen de Céphalosporine C est concentrée au point qu'une réaction positive de la ninhydrine se produise à un degré décelable, on n'obtient pas la couleur bleue ou pourpre typique.
On obtient plu- tôt une couleur jaune brunâtre. Le noyau de Céphalosporine C n'est donc pas positif à la ninhydrine dans le sens généralement accerté,

Après électrophorèse dans de l'acide acétique à 10%, élution du papier et hydrolyse dans HCL-N à 105 pendant 16 heures, le composé fournit un peu de glycine (54 micro grammes donnant une tache d'intensité semblable à la tache de glycine obtenue par hydro- lyse de 50 microgrammes de Céphalosporine C) mais pas d'acide a-aminoadipique. Il ne contient donc pas la chaîne latérale de la molécule de Céphalosporine C.

2. Le noyau de Céphalosporine CA (pyridine) n'est pas dé- truit par l'action de pénicillinase dans des conditions détruisent complètement 50 microgrammes de pénicilline G.
3. Noyau de Céphalosporine CA (pyridine) se distingue du noyau de Céphalospotine Cc sur des chromatogrammes sur papier obtenus à l'aide d'un mélange n-butanol-eau (4:1:5 en volume).
Les composés de l'invention définis plus haut, c'est-à- dire les noyaux de Céphalosporine C, Cc et CA sont particuli re- ment importants parce qu'ils sont des produits intermédiaires à partir desquels d'autres dérivés de Céphalosporine C peuvent être préparés, ces autres dérivés présentent une activité piolgique.
Les exemples suivants illustrent l'invention : EXEMPLE 1. - Préparation de l'acide 7-amino-céphalosporanique (a) Préparation du dérivé l-fluoro-2:4-dinitrobenzine (FDNB) de Céphalosporine C
On dissout 31,7 g du sel de sodium de Céphalosp- rine C (pureté 85-90%) dans 635 cm3 d'eau contenant 25,5 g de bicarbonate de sodium anhydre. On ajoute à cette solution agitée 25,5 cm3 de FDNB dissous dans 635 cm3 d'alcool éthylique. On agite ce mélange dans l'obscurité à la température ordinaire pendant 2 1/2 meures encore. Le pH de la solution est alors réglé à 5 au moyen d'acide chlorhydrique concentré et l'alcool est chassé par distillation dans le vide. L'excès de FDNB se sépare à ce stade.
Le mélange, après avoir été porté au pH 7 à l'aide de bicarbonate de sodium est extrait par 3 x 1/2 cm3 d'éther pour éliminer l'excès de FDNB et laisser une solution aqueuse limpide.
Le pH est alors réglé à 2,5 à l'aide d'acide chlorhydrique concen- tré. Il se forme un précipité collant qui se dissout lorsque le mélange est extrait par 3 x 1/2 volume d'acétate d'éthyle. Les extraits d'acétate d'éthyle rassemblés sont alors lavés à l'eau, séchés sur du sulfate de sodium anhydre et sèches, ce qui laisse 40,3 g d'un solide jaune.
L'essai biologique par comparaison avec un échantillon pur du dérivé dinitrophéynle de Céphalosporine C assigne un

degré de pureté d'environ 88% à cette matière.

Le dérivé 2:4-dinitrophényle brut de la Céphalosporine C obtenu ci-dessus contient une petite- d'une impureté basique colorée qui est éliminée en dissolvant la matière solide dans l'acé- tate d'éthyle et en lavant par fi.Cl 0,2 N. L'acétate d'éthyle est ensuite séché et éliminé dans le vide. 10,1 g de matière solide sort réduits.à 9,7 g par ce procédé.
On dissout 9,7 g du dérivé DNP de Céphalosporine C dans un mélange d'acétonitrile (100 cm3) eau (65 cm3) et acide chlorhy- drique N (34 cm3). Cette solution est ensuite conservée dans l'ob- scurité à la température ordinaire sous azote pendant 64 heures. On ajoute ensuite 200 cm3 d'eau à la solution et on extrait le tout par 5 x 100 cm3 d'acétate d'éthyle. Le résidu aqueux de 170 cm3 d'acide 7-amino-céphalosporanique est ensuite réglé au pH 7,2 par une solution saturée de bicarbonate de sodium et refroidi à 0 C.
(c) Isolation de l'acide 7-amino-céphalosporanique III (7-ACA)
Après hydrolyse et extraction par l'acétate d'éthyle sous (B) ci-dessus, il reste une solution acide contenant principale- ment le 7-ACA et le 7-ACA -(lactone) ; cette solution est réglée au pH 6,7 et passée dans une colonne de Dowex 1 (Cl-). Le 7-ACA lac- tone passe par la colonne et se retrouve dans le filtrat de ré- sine. Après lavage à l'eau, le 7-ACA peut être lavé à l'acide chlorhydrique 0,05 N . Des fractions contenant le 7-ACA fournissent un précipité cristallin qui semble être la forme zwitterion de la matière tandis que la plus grande partie du 7-ACA reste en solution sous forme de chlorhydrate. Le spectre dans l'infrarouge de la matière solide est indiqué à la Fig.l.
Les solutions de chlorhydra- te peuvent être phénylacétylées pour fournir la benzyl-Céphalospo- rine.

Préparation du noyau de Céphalosporine C à partir de
Céphalosporine C

On. dissout 2 g du sel de sodium de Céphalosporine C dans 30 cm3 d'eau, on règle le pH à 2,5 par addition de Dowex 50 x 8 (H+), on filtre la résine et on lave avec 10 cm3 d'eau, et on ajoute 10,2 cm3 de N-HC1 au filtrat combiné aux eaux de lavage. La solution est maintenue à 20 C pendant 3 jours et ajoutée à une colonne de Dowex-1 (torse acétate) de 2,1 cm de diamètre x 7 cm.
Le liquide qui s'écoule est recueilli par fractions de 5 cm3 (1 à 12) et la colonne est lavée à l'eau jusqu'à ce qu'on ait re- cueilli un total de 34 fractions. Le lavage peut alors être entre- pris à l'aide d'acide acétique 0,5 N et on obtient encore 66 frac- tions. La densité optique à 260 m u est mesurée pour chaque frac- tion.
Les fractions 2 à 16 sont rassemblées et concentrées dans le vide, ce qui provoque la séparation sous forme cristalline de Céphalosporine Cc (312 mg). Les fractions 36 à 45 contiennent la plus grande partie du noyau de Céphalosporine C qui fournit un dérivé phénylacétyle actif. Ces fractions sont rassemblées et séchées par congélation (40 mg). Par phénylacétylatio, cette matière fournit 250 unités d'activité contre Staph. aureus par mg de produit original.
Le nouveau composé est encore purifié par électrophorèse de la fraction appropriée provenant de la colonne de Dewex-1 (acétatel L'électrophorèse est effectuée dans une cellule d'élec- trophorèse sur papier à passage continu Beckman/spinco Mod le CP.
Le tampon utilisé est préparé en ajoutant de la pyridine à de l'acide acétique 0,05 N jusqu'à ce que le pH atteigne 4,0. La cellule travaille à courant constant (40 mA), le potentiel ?tant 880v. L'échantillon, dans un tampon de 20 cm3 est amené sur le rideau en 24 heures et on recueille 32 fractions, les fractions étant numérotées de la cathode (1) à l'anode (32).

The compounds of the invention defined above, i.e. the nuclei of Cephalosporin C, Cc and CA are particularly important because they are intermediates from which other derivatives of Cephalosporin C can be prepared, these other derivatives exhibit piolgique activity.
The following examples illustrate the invention: EXAMPLE 1. Preparation of 7-amino-cephalosporanic acid (a) Preparation of the 1-fluoro-2: 4-dinitrobenzine (FDNB) derivative of Cephalosporin C
31.7 g of the sodium salt of Cephalosprin C (85-90% purity) are dissolved in 635 cm3 of water containing 25.5 g of anhydrous sodium bicarbonate. 25.5 cm3 of FDNB dissolved in 635 cm3 of ethyl alcohol are added to this stirred solution. This mixture is stirred in the dark at room temperature for a further 2 1/2 hours. The pH of the solution is then adjusted to 5 using concentrated hydrochloric acid and the alcohol is distilled off in a vacuum. The excess FDNB separates at this point.
The mixture, after being brought to pH 7 using sodium bicarbonate, is extracted with 3 × 1/2 cm3 of ether to remove the excess of FDNB and leave a clear aqueous solution.
The pH is then adjusted to 2.5 using concentrated hydrochloric acid. A sticky precipitate forms which dissolves when the mixture is extracted with 3 x 1/2 volume of ethyl acetate. The combined ethyl acetate extracts are then washed with water, dried over anhydrous sodium sulfate and dried, which leaves 40.3 g of a yellow solid.
The bioassay by comparison with a pure sample of the dinitrophyll derivative of Cephalosporin C assigns a

about 88% purity to this material.
(B) Acid hydrolysis of the 2,4-dinitrophenyl derivative ¯¯¯ of Cephalosporin C
The crude 2: 4-dinitrophenyl derivative of Cephalosporin C obtained above contains a small of a colored basic impurity which is removed by dissolving the solid material in ethyl acetate and washing with fi.Cl 0.2 N. The ethyl acetate is then dried and removed in vacuo. 10.1 g of solid material comes out reduced to 9.7 g by this method.
9.7 g of the DNP derivative of Cephalosporin C are dissolved in a mixture of acetonitrile (100 cm3) water (65 cm3) and N hydrochloric acid (34 cm3). This solution is then stored in the dark at room temperature under nitrogen for 64 hours. Then 200 cm3 of water is added to the solution and the whole is extracted with 5 x 100 cm3 of ethyl acetate. The aqueous residue of 170 cm3 of 7-amino-cephalosporanic acid is then adjusted to pH 7.2 with a saturated solution of sodium bicarbonate and cooled to 0 C.
(c) Isolation of 7-amino-cephalosporanic acid III (7-ACA)
After hydrolysis and extraction with ethyl acetate under (B) above, an acidic solution remains, mainly containing 7-ACA and 7-ACA - (lactone); this solution is adjusted to pH 6.7 and passed through a Dowex 1 (Cl-) column. The 7-ACA lactone passes through the column and ends up in the resin filtrate. After washing with water, 7-ACA can be washed with 0.05N hydrochloric acid. Fractions containing 7-ACA provide a crystalline precipitate which appears to be the zwitterion form of the material while most of the 7-ACA remains in solution as the hydrochloride. The infrared spectrum of solid matter is shown in Fig. 1.
The hydrochloride solutions can be phenylacetylated to provide the benzyl-Cephalosporin.
EXAMPLE 2. -
Preparation of the Cephalosporin C nucleus from

We. dissolve 2 g of the sodium salt of Cephalosporin C in 30 cm3 of water, the pH is adjusted to 2.5 by adding Dowex 50 x 8 (H +), the resin is filtered and washed with 10 cm3 of water, and 10.2 cm3 of N-HCl is added to the filtrate combined with the washings. The solution is kept at 20 C for 3 days and added to a column of Dowex-1 (acetate torso) 2.1 cm in diameter x 7 cm.
The flowing liquid is collected in 5 cc fractions (1 to 12) and the column is washed with water until a total of 34 fractions have been collected. The washing can then be undertaken with 0.5N acetic acid and a further 66 fractions are obtained. The optical density at 260 m u is measured for each fraction.
Fractions 2 to 16 are combined and concentrated in vacuo, which causes separation in the crystalline form of Cephalosporin Cc (312 mg). Fractions 36-45 contain most of the Cephalosporin C nucleus which provides an active phenylacetyl derivative. These fractions are combined and dried by freezing (40 mg). Upon phenylacetylation, this material provides 250 units of activity against Staph. aureus per mg of original product.
The new compound is further purified by electrophoresis of the appropriate fraction from the Dewex-1 column (acetate) Electrophoresis is performed in a Beckman / Spinco Mod CP continuous pass paper electrophoresis cell.
The buffer used is prepared by adding pyridine to 0.05 N acetic acid until the pH reaches 4.0. The cell works at constant current (40 mA), the potential being 880v. The sample in a 20 cm3 buffer is brought to the curtain over 24 hours and 32 fractions are collected, the fractions being numbered from the cathode (1) to the anode (32).
The volume of each fraction is approximately 12 cc. At the end of the experiment, the paper curtain is sprayed with ninhydrin. This operation reveals a well marked band of material having only /

Cephalosporin CA (pyridine) is kept in 0.1 N'or N HCl at room temperature for 3 days or more.
When the reaction products are analyzed by electrophoresis on paper in pyridine acetate buffer (pH 4.0) and then the paper is contacted with agar plates seeded with Staph. aureus a single active area due to
Cephalosporin CA (pyridine) unchanged will reveal the concentration used. Cephalosporin CA (pyridine) behaves as if it did not show a net charge at pH 4.0.
However, other strips of paper which have been sprayed with M-pyridine in 50% aqueous acetone solution and 0.125 $ phenylacetyl chloride in anhydrous acetone reveal another active area on the sheets. This behavior is logical with respect to the properties that can be expected from the nucleus of Cephalosporin CA (pyridine) (see part 9 of the definition of the nucleus fraction of Cephalosporin C). A compound exhibiting the same properties is formed by incubation of a Cephalosporin C nucleus with 2M pyridine acetate at pH 7 for
16 hours and provides an active phenylacetyl derivative by phenylacetylation. The core properties of Cephalosporin CA are examined and coincide with the properties described above.
