Procédé de préparation de dérivés de la céphalosporine C La présente invention a pour objet un procédé de préparation de dérivés de la céphalosporine C.
Dans le brevet britannique No 810196, on décrit un antibiotique résistant à la pénicillinase, la c6pha- losporine C, que l'on isole des produits de fermenta tion d'une espèce de céphalosporium.
La céphalosporine C est relativement stable en présence des acides, ne subissant pas de perte d'ac tivité décelable par séjour dans de l'acide chlorhy drique 0,1 N à température ordinaire pendant 4 h. L'acide chlorhydrique concentré chaud agissant pen dant une durée de l'ordre de 18 h dégrade la molé cule de la céphalosporine C et détruit en même temps son activité.
Dans le Symposium de la Fondation Ciba sur les aminoacides et les peptides à l'activité antimétaboli- que, 1958, pp. 205-223, on indique que le traite ment de la céphalosporine C par l'acide chlorhydri que 0,1 N à 20,) C fournit au moins deux composés acides et trois composés neutres, qui sont tous posi tifs à la ninhydrine. Les composés neutres se sépa rent facilement sur des chromatogrammes sur papier traités avec une solution butanol-acide acétique-eau (4 : 1 : 4).
L'un d'eux (Rr,, 0,15) présente une activité antibactérienne et a été dénommé céphalosporine C, La préparation et les propriétés de la céphalosporine C,. sont décrites plus en détail dans l'exposé du bre vet britannique No 912360.
On a maintenant découvert qu'en soumettant la céphalosporine C à une hydrolyse acide ménagée, on peut former un produit ayant un caractère totale- ment distinct des produits de dégradation qui résul tent du traitement acide décrit ci-dessus. Ainsi, en traitant la céphalosporine C conformément à l'inven tion, on scinde la molécule de la céphalosporine C en deux fragments, à savoir un noyau cyclique et une chaîne latérale qui est l'acide .a-amino adipique.

On a également découvert qu'un type de struc ture noyau-chaîne latérale similaire se retrouve dans des produits de transformation de la céphalosporine C qui étaient connus jusqu'ici sous les noms de cé- phalosporine C. et céphalosporines CA .

dans laquelle R représente le radical d'une base hété- rocyclique tertiaire faible reliée au groupe CH., par son atome d'azote tertiaire, par exemple de la pyri- dine, de la collidine ou de la quinoléine.
Les céphalo sporines C #l, représentées ci-dessus sous la forme de zwitterion,peuvent être préparées comme décrit dans l'exposé du brevet britannique Nû, 912541. Comme dans la céphalosporine C, la chaîne latérale de la céphalosporine C, et des céphalosporines C_' est le radical acyle de l'acide a-amino-adipique, HOOC - CH(NH.,) - (CH,),, - CO -. Le noyau de la céphalosporine C est également appelé acide 7-amino-céphalosporanique.
Il a été constaté que la propriété de résistance à la pénicillinase dont jouissent la céphalosporine C, la céphalosporine C,. et les céphalosporines CA dépend principalement de la structure du noyau et que la préparation de ces noyaux par mise en aeuvre de l'invention ouvre la voie à la préparation de compo sés analogues à ces céphalosporines, notamment par adjonction d'autres chaînes latérales à ces noyaux par acylation sur le groupe 7-amino. Les nouvelles céphalosporines ainsi obtenues présentent une activité similaire, mais modifiée et parfois augmentée,
en comparaison avec l'activité de la céphalosporine C elle-même.
Conformément à l'invention, on obtient ces noyaux en soumettant de la céphalosporine C, de la céphalosporine C, ou une céphalosporine CA, ou un dérivé de ces céphalosporines dans lequel le groupe amino de la chaîne latérale a-amino-adipyle est pro tégé, à une hydrolyse en présence d'un agent hydroly- tique acide pendant une durée qui, en fonction de la température et des autres conditions de la réaction, est telle que la chaîne latérale a-amino-adipyle, pro tégée ou non protégée, soit enlevée et que la struc ture soit conservée.
Les noyaux peuvent ensuite être isolés sous forme libre ou sous forme de leurs sels d'addition d'acides, par un ajustage approprié du pH de la solution. Par exemple, si l'on ajuste le pH de la solution à environ 4,5, on peut isoler les noyaux libres. Il est entendu que le terme acide comprend les matières échan- geuses d'ions acides.
Le groupe 2,4-dinitro-phényle est un groupe pro tecteur particulièrement approprié. Un autre groupe utilisable est le groupe 2-nitro-4-carbométhoxy-phé- nyle. Le dérivé 2,4-dinitro-phénylé de la céphalospo rine C peut être préparé par réaction de la céphalo sporine C avec du 1-fluoro-2,4-dinitro-benzène, et le dérivé 2-nitro-4-carbométhoxy-phénylé peut être pré paré par réaction de la céphalosporine C avec du 1- fluoro-2-nitro-4-carbométhoxy-benzène.
L'hydrolyse de la céphalosporine C, de la cépha losporine C, ou de leurs dérivés à chaîne latérale protégée, ou d'une céphalosporine C.@, peut être réa lisée au moyen d'un acide minéral dilué, par exem ple d'acide chlorhydrique 0,1 N à N. Comme autres acides appropriés, on peut mentionner les acides sul- foniques et les résines de polystyrène sulfonées.
Pour mettre en oeuvre le procédé, on peut laisser reposer la solution traitée par un acide, contenant de la cé phalosporine C, de la céphalosporine C,. ou une cé phalosporine CA ou le dérivé à chaîne latérale pro tégée, pendant un temps tel que le noyau soit séparé de la chaîne latérale, ce temps dépendant de la sévé rité des conditions mais étant normalement de l'ordre de quelques jours. En général, aux températures com prises entre - 10#, C et -I- 50,, C, les durées employées sont de 30 jours à 12 heures respectivement, par exemple de 3 jours avec de l'acide chlorhydrique 0,1 N à 20o C.
Ensuite, on peut séparer le noyau des produits de réaction, par exemple par électro phorèse, par chromatographie ou échange d'ions. Dans une méthode, on traite le produit de réaction par une résine échangeuse d'anions fortement basi que, on élue la résine au moyen d'un solvant et on recueille les fractions contenant le noyau. La résine est de préférence sous la forme d'acétate et le sol vant peut être de l'acide acétique.
Si l'on utilise ces techniques, on peut généralement identifier le noyau séparé sur un chromatogramme sur papier par la ca ractéristique d'absence d'activité antibiotique propre ment dite aux concentrations utilisées (par exemple lorsqu'on place le papier d'électrophorése au contact de plaques ensemencées de staphylocoque doré (sou che d'Oxford), mais d'activité après phénylacétyla- tion au moyen de chlorure de phénylacétyle. Pour cette phénylacétylation, on peut tout d'abord sou mettre le papier à une pulvérisation de pyridine M dans de l'acétone à 50 0/0,
puis à une pulvérisation de chlorure de phénylacétyle (1,25 % v/v) dans de l'acétone. Le dérivé phénylacétylé peut être séparé par chromatographie et électrophorèse.
Pour produire le noyau de la céphalosporine C,, , on peut préparer la céphalosporine C,. in situ à par tir de céphalosporine C. Ainsi, en traitant de la cé phalosporine C par un acide, on la transforme en céphalosporine C,. et en continuant le traitement acide, on obtient le noyau de la céphalosporine C, Il s'est avéré que l'acide chlorhydrique 0,5 N à N pendant moins de 1 jour sont les conditions préférées pour la production de céphalosporine C,. et que les conditions préférées pour la production de son noyau consistent en le traitement de la céphalosporine C par de l'acide chlorhydrique 0,5 N à N pendant 2 à 4 jours.
Le noyau de la céphalosporine C, obtenu con formément à l'invention par hydrolyse acide de la céphalosporine C ou d'un dérivé à chaîne latérale protégée de la céphalosporine C, peut être utilisé pour la production de noyaux de céphalosporines Ç@ , par traitement du noyau de la céphalosporine C par une base hétérocyclique tertiaire faible.
Lorsqu'on met en couvre l'invention en partant d'un dérivé à chaîne latérale protégée, il convient d'utiliser de l'acide chlorhydrique dilué, spéciale ment en concentration comprise entre 0,1 et 0,3N, en présence d'un solvant organique miscible à l'eau tel que l'acétonitrile. Avantageusement, on effectue la réaction à l'obscurité et sous une atmosphère inerte, par exemple sous azote.
Au cours du traitement de la céphalosporine C par un acide, de concentration comprise dans les li mites indiquées, au moins deux réactions entrent en jeu. Ces réactions sont (1) l'hydrolyse de la céphalosporine C qui scinde la liaison reliant le noyau à la chaîne latérale et (2) la réaction de l'acide avec la molécule de la cé phalosporine C produisant de la céphalosporine C'.
De plus, dès que de la céphalosporine C, est pré sente dans le mélange réactionnel, une troisième réac tion commence, à savoir l'hydrolyse de la céphalo sporine C. qui scinde la liaison reliant le noyau de la céphalosporine C. à la chaîne latérale. En outre, il se forme une certaine quantité de noyau de la céphalo sporine C,, par réaction de l'acide avec le noyau de la céphalosporine C.
Les conditions qui conviennent le mieux pour l'obtention du produit désiré peuvent être déterminées expérimentalement, et on a cons taté que, lorsqu'on part de céphalosporine C, le trai tement par de l'acide chlorhydrique 0,1 N à 20o C pendant environ 3 jours convient pour la produc tion du noyau de la céphalosporine C et que le trai tement par de l'acide chlorhydrique 0,5 à 1 N à 20 C pendant environ 3 jours convient pour la pro duction du noyau de la céphalosporine C, Le noyau de la céphalosporine C forme des sels, par exemple des sels de sodium, de potassium et d'ammonium, par traitement avec un équivalent d'une base.
Les noyaux de la céphalosporine C, de la céphalosporine C. et des céphalosporines C.1 forment également des sels d'addition d'acide sur le groupe amino primaire, par exemple d'acides minéraux et d'autres acides forts, par exemple des chlorhydrates.
Le noyau de la céphalosporine C, que l'on a appelé acide 7-aminocéphalosporanique, possède une struc ture stérique semblable à celle de la partie corres pondante de la céphalosporine C obtenue par fer mentation du céphalosporium. Il peut également être identifié par les propriétés suivantes, après phényl- acétylation 1) La fraction de la préparation formée du noyau de la céphalosporine C migre légèrement plus vite que la céphalosporine C vers l'anode, lors de l'électrophorèse au pH 7 (avec de l'acétate de collidine aqueux comme solvant).
2) La fraction formée du noyau de la céphalospo rine C migre à une vitesse égale à environ la moitié de celle de la céphalosporine C vers l'anode au pH 4,5 (avec de l'acétate de pyridine aqueux comme solvant).
3) La fraction formée du noyau de la céphalospo rine C reste au voisinage du point d'origine dans l'électrophorèse au pH 4,0 (dans de l'acétate de pyridine aqueux), ne manifestant qu'une migra tion relativement faible vers l'anode. Dans ces conditions, la céphalosporine C migre encore vers l'anode presque aussi rapidement qu'au pH 7.
4) Après électrophorèse dans l'acide acétique à 10 %, élution du papier et hydrolyse dans l'acide chlorhydrique N à 1050 pendant 16 h, elle four nit un peu de glycine (50 #tg donnant une tache d'intensité semblable à celle de la tache de gly cine obtenue par hydrolyse de 50 [g de cépha- Iosporine C) mais pas d'acide a-amino-adipique. Elle ne contient donc pas la chaîne latérale de la molécule de la céphalosporine C.
5) La fraction formée du noyau de la céphalospo rine C présente une valeur Ru semblable à celle de la N-phénylacétylcéphalosporine C (0,08-0,15) dans le butanol-éthanol-eau (4 : 1 : 5 en volume).
6) Après chromatographie dans le butanol-éthanol- eau, ou après électrophorèse, et après avoir pul vérisé sur le papier une solution de gélatine à 0,1 %, contenant de la pénicillinase (concentra tion d'enzyme égale à 10 fois celle nécessaire pour inactiver une tache de 50 [.g de benzylpéni- cilline), elle donne une tache d'activité apparem ment non diminuée lorsqu'on pulvérise sur le pa pier du chlorure de phénylacétyle et de la py- ridine.
7) Lorsqu'on en dépose une goutte sur du papier, on pulvérise de l'acétate de pyridine au pH 7 (concentration 2 M de la pyridine) sur la tache, on suspend à l'état humide sur la vapeur d'une solution d'acétate de pyridine 2 M à 37o pendant 16 h, on sèche, on soumet à l'électrophorèse au pH 7 et on phénylacétyle, une tache active (due au dérivé phénylacétylé du noyau de la céphalo sporine<B>C.1</B> (pyridine) apparaît dans la position neutre, ainsi qu'une tache dans une position légè rement plus proche de l'anode que celle occupée par la céphalosporine C.
8) Après électrophorèse sur papier, le noyau donne une couleur brun jaunâtre avec la ninhydrine, qui vire par la suite au gris bleu (il a été publié que l'acide 6-aminopénicillanique ne donne pas une couleur normale avec la ninhydrine). Après élec trophorèse ou chromatographie sur papier, il peut être facilement décelé (comme la céphalosporine C), sous forme d'une tache absorbante sombre, en plaçant le papier devant une source de lu mière ultraviolette.
Le noyau de la céphalosporine C,., qui est la lac- tone de l'acide désacétyl-7-aminocéphalosporanique, a également une structure stérique semblable à celle de la partie correspondante de la céphalosporine C obtenue par fermentation de céphalosporium. Elle peut être encore identifiée par les propriétés sui vantes, après phénylacétylation.
1) Par électrophorèse sur papier dans un tampon d'acétate de pyridine (pH 4,5), le noyau de la céphalosporine C,. se déplace vers la cathode ap proximativement sur la même distance que le noyau de la céphalosporine C se déplace vers l'anode.
2) Par électrophorèse sur papier avec un tampon d'acide acétique à 10<B>()/a,</B> le noyau de la céphalo sporine C,. migre vers l'anode deux fois et demie plus vite que le noyau de la céphalosporine C.
3) Par chromatographie dans le butanol-éthanol-eau (4: 1 : 5 en volume), la valeur R,,. du noyau de la céphalosporine C,. est de 0,33, alors que celle de la céphalosporine C est de 0,08-0,15 et celle du noyau de la céphalosporine CA est de 0,08.
Les noyaux des céphalosporines C.,, ont égale ment une structure stérique semblable à celle de la partie correspondante de la céphalosporine C obtenue par fermentation du céphalosporium. La céphalospo rine C., (pyridine) peut être encore identifiée par les propriétés suivantes: 1) Par électrophorèse sur papier au pH 4,0 dans un tampon d'acétate de pyridine, le noyau de cépha losporine C1 se déplace vers la cathode sur une distance semblable à la distance sur laquelle la céphalosporine C se déplace vers l'anode.
Lors que l'électrophorèse est effectuée dans de l'acide acétique à 10<B>%</B>, le noyau de la céphalosporine C_L (pyridine) se déplace environ deux fois plus vite vers la cathode que la céphalosporine C,t (pyridine).
2) Le noyau de la céphalosporine C_@ (pyridine) n'est pas détruit par l'action de la pénicillinase dans des conditions dans lesquelles 50 g de pénicilline G sont complètement détruits.
3) Le noyau de la céphalosporine C., (pyridine) se distingue du noyau de la céphalosporine C, sur les chromatogrammes sur papier traités au n-bu- tanol-éthanol-eau (4: 1 :5 en volume).
Les composés définis plus haut, c'est-à-dire les noyaux des céphalosporines C, C, et CA sont spécia lement importants, car ils sont des produits intermé diaires à partir desquels d'autres dérivés de la cépha losporine C doués d'activité biologique peuvent être préparés.
Les exemples suivants illustrent l'invention. Exemple 1 <I>Préparation de l'acide</I> 7-aminocéphalosporanique <I>(a) Préparation dit dérivé du</I> 1-flitoro-2,4-dinitro- benzène (FDNB) <I>et de la céphalosporine C.</I>
On a dissous 31,7 g du sel de sodium de la cé phalosporine C (pureté 85-90<B>%</B>) dans 635 ml d'eau contenant 25,5 g de bicarbonate de sodium anhydre. En agitant la solution, on a ajouté 25,5 ml de FDNB dissous dans 635 ml d'alcool éthylique. On a agité ce mélange à l'obscurité à température ordinaire pen dant encore 21/= h. On a ensuite ajusté le pH de la solution à 5 avec de l'acide chlorhydrique concentré et on a chassé l'alcool par distillation sous vide. Du FDNB en excès a été chassé en même temps.
Après avoir porté ce mélange au pH 7 avec du bicarbonate de sodium, on l'a extrait avec 3 X 1/z vol. d'éther pour éliminer l'excès de FDNB et laisser une solution aqueuse limpide. On a alors amené le pH à 2,5 avec de l'acide chlorhydrique concentré. II s'est formé un précipité collant, qui s'est dissous par extraction avec 3 X 1/:a vol. d'acétate d'éthyle. On a réuni les extraits dans l'acétate d'éthyle, on les a lavés à l'eau, séchés sur du sulfate de sodium anhy dre et évaporés à sec, ce qui a laissé 40,3 g d'un solide jaune.
Un dosage biologique par comparaison avec un échantillon pur du dérivé dinitrophénylé de la cé phalosporine C a montré que ce produit est d'une pureté d'environ 88 %.
<I>(b) Hydrolyse acide du dérivé</I> 2,4-dinitrophényle <I>(le</I> <I>la céphalosporine C.</I>
Le dérivé 2,4-dinitrophényle brut de la céphalo sporine C provenant de la préparation ci-dessus con tient une faible proportion d'une impureté basique colorée, que l'on a éliminée en dissolvant le produit solide dans de l'acétate d'éthyle et en lavant du HCl 0,2 N. On a ensuite séché l'acétate d'éthyle et on l'a chassé sous vide. 10,1 g de solide ont été réduits à 9,7 g par cette opération.
On a dissous 9,7 g du dérivé du DNP et de la céphalosporine C dans un mélange de<B>100</B> ml d'acé- tonitrile, 65 ml d'eau et 34 ml d'acide chlorhydrique 1 N. On a ensuite laissé reposer cette solution à l'obs curité, à température ordinaire et sous azote pendant 64 h. On a ensuite ajouté 200 ml d'eau à la solution et on a extrait le tout avec 5 X 100 ml d'acétate d'éthyle. On a amené le résidu aqueux (270 ml) d'acide 7-aminocéphalosporanique au pH 7,2 avec une solution saturée de bicarbonate de sodium et on a refroidi à 0o C.
<I>(c) Isolement (le l'acide</I> 7-arriiriocéphalospor-anique <I>11I</I> (7-ACA).
Après l'hydrolyse et l'extraction à l'acétate d'éthyle du stade (b) ci-dessus, il reste une solution acide contenant principalement du 7-ACA et de la lactone de 7-ACA. On a ajusté cette solution au pH 6,7 et on l'a fait passer dans une colonne de Dowex-1 <B>(CI-).</B> La lactone de 7-ACA traverse la colonne et apparaît dans le filtrat sortant de la résine. Après lavage à l'eau, le 7-ACA peut être élué avec de l'acide chlorhydrique 0,05 N.
Les frac tions contenant le 7-ACA donnent un précipité cris tallin qui parait être la forme zwitterion de cette ma tière, alors que la majeure partie du 7-ACA reste en solution comme chlorhydrate. Le spectre infra rouge du solide est représenté à la fig. 1 du dessin. Les solutions du chlorhydrate peuvent être phényl- acétylées pour fournir la benzylcéphalosporine.
Exemple 2 <I>Préparation du noyau de la céphalosporine C</I> n <I>partir (le</I> céphalosporine <I>C</I> On a dissous 2 g de sel de sodium de céphalo sporine C dans 30 ml d'eau, on a amené le pH à 2,5 par addition de Dowex 50 X 8 (H--), on a séparé la résine par filtration, on l'a lavée avec 10 ml d'eau et on a ajouté 10,2 ml d'HCl 1 N au filtrat et aux eaux de lavage réunis. On a maintenu la so lution à 20 C pendant 3 jours et on l'a fait passer dans une colonne de Dowex-1 (forme acétate) de 2,1 cm de diamètre X 7 cm.
On a recueilli le percolat en fractions. de 5 ml (1 à 12) et on a élué la colonne avec de l'eau jusqu'à ce qu'un total de 34 fractions ait été recueilli. On a ensuite com mencé une élution avec de l'acide acétique 0,5 N et on a recueilli encore 66 fractions. La densité optique à 260 mI, a été mesurée pour chaque fraction.
Les fractions 2-16 ont été réunies et concentrées sous vide et 312 mg de céphalosporine C, se sont séparés sous forme cristalline. Les fractions 36-45 contenaient la majeure partie du noyau de céphalo sporine C, qui a fourni un dérivé phénylacétylé ac tif. Ces fractions ont été réunies et séchées sous congélation (40 mg). Par phénylacétylation, ce pro duit a donné 250 unités d'activité sur Staph. aureus, par mg du produit original.
Le nouveau composé a encore été purifié par électrophorèse de la fraction appropriée provenant de la colonne de Dowex-1 (acétate). L'électro phorèse a été effectuée dans une cellule d'électropho- irèse sur papier à écoulement continu Beckman/ spinco Modèle CP. Le tampon utilisé a été préparé en ajoutant de la pyridine à de l'acide acétique 0,05 N jusqu'à ce que le pH monte à 4,0. On a fait fonctionner la cellule à courant constant (40 mA), la tension étant de 880 V.
L'échantillon, dans un tampon de 20 ml, a été fourni au rideau pendant 24 h et 32 fractions ont été recueillies, les fractions étant numérotées de la cathode (1) à l'anode (32). Le volume de chaque fraction était d'environ 12 ml. A la fin de l'expérience, on a pulvérisé de la ninhy- drine sur le rideau de papier. Ceci a révélé une forte bande de matière de très faible mobilité qui a été entraînée hors du rideau dans les fractions 13-15, une bande de matière fortement acide qui avait coulé dans la mèche d'anode, et une faible bande d'un produit (correspondant à l'acide a-aminodipi- que) qui avait migré vers l'anode et a été entraîné hors du rideau dans la fraction 24.
Pour déterminer la position du nouveau composé, on a appliqué 10 touches de 10 [l de chaque frac tion sur du papier et on a pulvérisé sur le papier de la pyridine 1 M dans. de l'acétone à 50 %, puis du chlorure de phénylacétyle à 2 % dans de l'acétone. Après contact du papier avec des plaques ensemen cées de Staph. aureus et incubation des plaques,
on a trouvé une grande zone d'inhibition dans une po sition correspondant à la fraction 22 et une plus petite dans une position correspondant à la frac tion 23.
Par séchage sous congélation de la fraction 22, on a obtenu un résidu qui était trop petit pour pou voir être pesé avec précision. En solution aqueuse, le spectre d'absorption ultraviolet du produit a mon tré un plateau à 260 m#t, et son poids total (43 I,g) a été estimé d'après l'extinction à cette longueur d'onde, en admettant que le poids moléculaire de la substance est égal à celui de l'acide 7-aminocéphalo- sporanique, et que son extinction moléculaire est égale à celle de la céphalosporine C. On a phényl- acétylé le produit dans de l'acétone aqueuse conte nant un tampon au phosphate de pH 6,5.
L'activité du dérivé phénylacétylé contre Staph. aureus a cor respondu à 1450 unités par mg de produit original. La céphalosporine C présente une activité de 8-10 unités par mg contre le même organisme.
Le groupe amino de l'acide 6-aminopénicillanique (noyau de la pénicilline) a une basicité relativement faible, le pH isoélectrique publié pour ce composé étant de 4,3 (Batchelor, Doyle, Naylor et Rolinson, 1959).
Ceci est facile à comprendre, étant donné que le groupe amino est en (3 à la fois par rapport à un groupe CON et à un groupe CH - S -. Pour des raisons analogues, le groupe amino de l'acide 7 aminocéphalosporanique devrait être faiblement ba sique et la substance devrait migrer vers l'anode au pH 7.
Les propriétés du nouveau dérivé de la cépha losporine C (décrites aux pages 5 et 6) dénotent qu'il est en fait l'acide 7-aminocéphalosporanique. Exemple 3 <I>Préparation de</I> <I>l'acide</I> 7-phénylacétamidocéphalosporanique On a traité le sel de sodium de la céphalosporine C avec de l'acide chlorhydrique dilué à température ordinaire, en procédant comme décrit précédemment.
On a neutralisé la solution résultante, contenant l'acide 7-aminocéphalosporanique et d'autres pro duits de dégradation faiblement acide de la céphalo sporine C, et on l'a tamponnée au pH 7,0 avec une solution de bicarbonate de sodium sous atmosphère de C02. On a ajouté lentement du chlorure d'acé tyle (1,2 équiv., basé sur la quantité de céphalospo rine C utilisée)
dans de l'acétone à la solution neutre préalablement refroidie à 0o et mélangée avec suffi samment d'acétone pour que la concentration finale de l'acétone soit de 50 % après l'addition de la solution de chlorure de phénylacétyle. Au bout d'une heure, on a amené le pH du mélange à 5,0 (électrode de verre) et on a chassé l'acétone sous vide.
On a alors abaissé le pH à 2,0 et on a extrait à l'acétate de butyle l'acide 7-phénylacétamidocéphalosporani- que, ensemble avec la N-phénacétyl-céphalosporine C. et la N-phénylacétyl-céphalosporine C. On a sé paré l'acétate de butyle et on l'a extrait avec de l'eau amenée au pH 5,0 à l'équilibre par addition d'alcali. On a séché l'extrait aqueux sous congéla tion.
Par chromatographie sur papier de 150 ug du produit dans du n-butanol-éthanol-eau (4: 1 : 5 en vol.) on a constaté qu'il contenait trois composants actifs contre Staph. aureus (voir fig. 1).
Le compo sant actif principal est l'acide 7-phénylacétamidocé- phalosporanique. Les autres composants actifs sont la N-phénylacétyl-céphalosporine C,, et la N-phényl- acétyl-céphalosporine C qui toutes deux n'ont été extraites que partiellement par l'acétate de butyle.
On a séparé l'acide 7-phénylacétamidocéphalo- sporanique des autres composants actifs du mélange brut (17 unités/mg contre Staph. aureus) par chro matographie sur une colonne de rouleau de papier Grycksbo (IKB-Produkter, Suède) et par distribution à contre-courant.
L'élution de la colonne de papier a été effectuée avec du n-butanol saturé d'eau. Dans une expérience préliminaire, le sel de sodium de l'acide 7-phényl- acétamidocéphalosporanique a été obtenu de la co lonne de papier sous forme d'une poudre présentant une activité de 167 unités/mg. Ce produit est claire ment impur.
Les distributions à contre-courant préliminaires ont été effectuées dans deux systèmes solvants. Le premier système consiste en volumes égaux d'acé- tate de butyle et d'acide acétique à 1 % (solvant I), et le second en n-butanol et tampon de phosphate de sodium 0,05 M au pH 6,0 (solvant II).
Après huit transferts dans le solvant I (10 ml de la couche su périeure et 20 ml de la couche inférieure), la con centration de l'acide 7-phénylacétamidocéphalospo- ranique a atteint un maximum entre les tubes 6 et 7, alors que les autres composants actifs ont été trouvés dans les tubes 0 et 1. La matière recueillie dans les tubes 5, 6 et 7 a titré approximativement 50 unités par mg. La chromatographie sur papier de cette matière dans du n-butanol-eau-éthanol (4: 5 : 1 en vol.) a montré que l'acide 7-phénylacétamidocéphalo- sporanique était la seule substance active présente.
Après huit transferts dans le solvant II (20 ml de chaque couche), le phénylacétamidocéphalospora- nate de sodium a montré un sommet entre les tubes 3 et 4, mais la majeure partie du restant de la ma tière active est restée dans le tube 0.
L'acide 7-phénylacétamidocéphalosporanique s'est déplacé vers l'anode à presque la même vitesse que la pénicilline G, en électrophorèse sur papier dans un tampon d'acétate de collidine au pH 7. Par incu bation avec de la pyridine aqueuse à 37 , l'acide 7- phénylacétamidocéphalosporanique a été partielle ment converti en un dérivé actif du type de la cé phalosporine C A (pyridine). Ce dérivé s'est comporté comme une substance neutre en électrophorèse au pH 7,0.
Une quantité d'environ 10 ug d'acide 7-phényl- acétamidocéphalosporanique n'a pas été appréciable- ment inactivée par une solution de pénicillinase con tenant 10 fois la quantité d'enzyme inactivant com plètement 50 #tg de pénicilline G dans des conditions semblables.
Exemple 4 <I>Préparation du noyau de la</I> céphalosporine C=1 (pyridine) <I>à partir de céphalosporine</I> C.1 (pyridine) On maintient de la céphalosporine C_1 (pyridine) dans de l'acide chlorhydrique 0,5 N ou 1 N à tempé rature ordinaire pendant 3 jours ou plus.
Lorsque les produits de cette réaction sont analysés par élec trophorèse sur papier dans un tampon d'acétate de pyridine (pH 4,0) suivie de mise en contact du papier avec des plaques d'agar ensemencées de Staph. au- reus, une unique zone active due à la céphalosporine C A (pyridine) non transformée est révélée, à la con centration utilisée. La céphalosporine CA (pyridine) se comporte comme si elle n'avait pas de charge nette au pH 4,0.
Cependant, des bandes de papier dupli quées sur lesquelles on a pulvérisé de la pyridine M dans de l'acétone aqueuse à 50 % et du chlorure de phénylacétyle à 0,125 % dans de l'acétone sèche,
révèlent une zone active additionnelle sur les plaques ensemencées, due aux produits de la phénylacétyla- tion du noyau de la céphalosporine C, (pyridine). Le noyau de céphalosporine CA se comporte comme une base au pH 4,0. Il migre vers la cathode sur une distance un peu plus grande que la distance sur la quelle la céphalosporine C elle-même migre vers l'anode dans la même expérience. Il reste en posï- tion neutre dans l'électrophorèse au pH 7.
Ce com portement est en accord avec les propriétés prévues du noyau de la céphalosporine CA (pyridine), (voir la position 9 de la définition du noyau de la céphalo sporine C). On a formé un composé avec les mêmes propriétés par incubation du noyau de la céphalo sporine C avec de l'acétate de pyridine 2 M au pH 7 pendant 16 h. Par phénylacétylation, ce composé a fourni un dérivé phénylacétylé actif. Les propriétés du noyau de la céphalosporine CA ont été étudiées et elles se sont montrées coïncider avec les proprié tés décrites plus haut pour ce noyau.
Exemple 5 Préparation <I>du</I> noyait <I>de la céphalosporine C,.</I> <I>à partir de</I> céphalospotine C, On a maintenu 50 mg de céphalosporine C dans 1,33 ml de HCl 0,5 N ou 1 N à 20o C pendant 2 ou 3 jours. Dans ces conditions, l'acide 7-aminocéphalo- sporanique, qui était présent dans la solution chlor hydrique normale après 24 h, a disparu après 3 jours.
Après 4 jours, la zone active révélée par phénylacé- tylation du noyau de la céphalosporine C, était encore proche de la valeur maximum, mais il ne res tait que des traces de céphalosporine C, Le noyau de la céphalosporine C,, paraît donc être le produit final relativement stable en milieu acide d'une série de dérivés actifs ou potentiellement actifs, formés par traitement acide ménagé de la céphalosporine C.
Le noyau de la céphalosporine C,. a également été formé par mise en contact de céphalosporine C avec un poids égal de résine échangeuse d'ions Do- wex 50 X 8 (-H+) pendant 15-48 h à 20o C.
Des bandes dupliquées provenant d'ionogrammes effec tués dans un tampon d'acétate de pyridine de pH 4,5, sur lesquelles on avait pulvérisé du chlorure de phé- nylacétyle et que l'on avait mis en contact avec un organisme d'épreuve, ou sur lesquelles on avait pul vérisé de la ninhydrine, ont révélé que la position du noyau de la céphalosporine C,. correspondait à une zone jaune sur les bandes traitées à la ninhydrine.

British Patent No. 810196 describes a penicillinase resistant antibiotic, c6phalosporin C, which is isolated from the fermentation products of a species of cephalosporium.
Cephalosporin C is relatively stable in the presence of acids, undergoing no detectable loss of activity upon residence in 0.1N hydrochloric acid at room temperature for 4 h. Hot concentrated hydrochloric acid, acting for a period of around 18 hours, degrades the cephalosporin C molecule and at the same time destroys its activity.
In the Ciba Foundation Symposium on Amino Acids and Peptides with Antimetabolic Activity, 1958, pp. 205-223, it is reported that the treatment of cephalosporin C with 0.1N hydrochloric acid at 20 ° C provides at least two acidic compounds and three neutral compounds, all of which are ninhydrin positive. Neutral compounds readily separated on chromatograms on paper treated with a butanol-acetic acid-water solution (4: 1: 4).
One of them (Rr ,, 0.15) exhibits antibacterial activity and has been called cephalosporin C, The preparation and properties of cephalosporin C ,. are described in more detail in the disclosure of British Patent No 912360.
It has now been found that by subjecting cephalosporin C to controlled acid hydrolysis, a product can be formed which is completely distinct from the degradation products which result from the acid treatment described above. Thus, by processing cephalosporin C according to the invention, the cephalosporin C molecule is split into two fragments, namely a cyclic nucleus and a side chain which is α-amino adipic acid.

in which R represents the radical of a weak tertiary heterocyclic base linked to the CH 4 group, by its tertiary nitrogen atom, for example pyridine, collidine or quinoline.
Cephalosporins C # 1, shown above as the zwitterion, can be prepared as described in British Patent Disclosure No. 912541. As in cephalosporin C, the side chain of cephalosporin C, and cephalosporins C_ 'is the acyl radical of α-amino-adipic acid, HOOC - CH (NH 4) - (CH 4) ,, - CO -. The nucleus of cephalosporin C is also called 7-amino-cephalosporanic acid.
It was found that the property of resistance to penicillinase enjoyed by cephalosporin C, cephalosporin C ,. and CA cephalosporins depends mainly on the structure of the nucleus and that the preparation of these nuclei by implementing the invention opens the way for the preparation of compounds similar to these cephalosporins, in particular by adding other side chains to these rings by acylation on the 7-amino group. The new cephalosporins thus obtained exhibit a similar activity, but modified and sometimes increased,
in comparison with the activity of cephalosporin C itself.
According to the invention, these nuclei are obtained by subjecting cephalosporin C, cephalosporin C, or a cephalosporin CA, or a derivative of these cephalosporins in which the amino group of the α-amino-adipyl side chain is protected. to hydrolysis in the presence of an acidic hydrolytic agent for a period of time which, depending on the temperature and the other reaction conditions, is such that the α-amino-adipyl side chain, protected or unprotected, be removed and the structure retained.
The nuclei can then be isolated in free form or in the form of their acid addition salts, by appropriate adjustment of the pH of the solution. For example, if the pH of the solution is adjusted to about 4.5, the free nuclei can be isolated. It is understood that the term acid includes acidic ion exchange materials.
The 2,4-dinitro-phenyl group is a particularly suitable protecting group. Another useful group is the 2-nitro-4-carbomethoxy-phenyl group. The 2,4-dinitro-phenyl derivative of cephalosporin C can be prepared by reacting cephalosporin C with 1-fluoro-2,4-dinitro-benzene, and the 2-nitro-4-carbomethoxy-phenyl derivative can be prepared by reacting cephalosporin C with 1-fluoro-2-nitro-4-carbomethoxy-benzene.
The hydrolysis of cephalosporin C, of cephalosporin C, or their side chain protected derivatives, or of a cephalosporin C. @, can be carried out by means of a dilute mineral acid, for example of 0.1 N to N hydrochloric acid. As other suitable acids there may be mentioned sulphonic acids and sulphonated polystyrene resins.
In order to carry out the process, the solution treated with an acid, containing cephalosporin C, cephalosporin C, can be left to stand. or a ce phalosporin CA or the protected side chain derivative, for a time such that the nucleus is separated from the side chain, this time depending on the severity of the conditions but normally being on the order of a few days. In general, at temperatures between -10 ° C and -I- 50 ,, C, the times employed are 30 days to 12 hours respectively, for example 3 days with 0.1N hydrochloric acid at 20o C.
Then the nucleus can be separated from the reaction products, for example by electrophoresis, chromatography or ion exchange. In one method, the reaction product is treated with a strongly basic anion exchange resin, the resin is eluted with a solvent, and the core-containing fractions are collected. The resin is preferably in the form of acetate and the solvent may be acetic acid.
If these techniques are used, the separated nucleus can generally be identified on a paper chromatogram by the characteristic of lack of antibiotic activity proper at the concentrations used (for example, when the electrophoresis paper is placed in the cell. contact with plates seeded with Staphylococcus aureus (Oxford strain), but active after phenylacetylation by means of phenylacetyl chloride.For this phenylacetylation, the paper can first be subjected to a spray of pyridine M in 50% acetone,
then to a spray of phenylacetyl chloride (1.25% v / v) in acetone. The phenylacetyl derivative can be separated by chromatography and electrophoresis.
To produce the cephalosporin C i nucleus, cephalosporin C i can be prepared. in situ from cephalosporin C. Thus, by treating ce phalosporin C with an acid, it is converted into cephalosporin C. and by continuing the acid treatment, the nucleus of cephalosporin C is obtained. It has been found that hydrochloric acid 0.5 N to N for less than 1 day are the preferred conditions for the production of cephalosporin C. and that the preferred conditions for the production of its nucleus consist of the treatment of cephalosporin C with 0.5 N to N hydrochloric acid for 2 to 4 days.
The cephalosporin C nucleus, obtained according to the invention by acid hydrolysis of cephalosporin C or of a side chain protected derivative of cephalosporin C, can be used for the production of cephalosporin C nuclei, by processing of the cephalosporin C nucleus by a weak tertiary heterocyclic base.
When the invention is carried out starting from a derivative with a protected side chain, dilute hydrochloric acid should be used, especially in a concentration of between 0.1 and 0.3N, in the presence of a water-miscible organic solvent such as acetonitrile. Advantageously, the reaction is carried out in the dark and under an inert atmosphere, for example under nitrogen.
During the treatment of cephalosporin C with an acid, with a concentration within the limits indicated, at least two reactions come into play. These reactions are (1) the hydrolysis of cephalosporin C which cleaves the bond connecting the nucleus to the side chain and (2) the reaction of the acid with the ce phalosporin C molecule producing cephalosporin C '.

The nuclei of cephalosporin C, cephalosporin C. and cephalosporins C.1 also form acid addition salts on the primary amino group, for example of mineral acids and other strong acids, for example of hydrochlorides.
The nucleus of cephalosporin C, which has been called 7-aminocephalosporanic acid, has a steric structure similar to that of the corresponding part of cephalosporin C obtained by fermenting the cephalosporium. It can also be identified by the following properties, after phenylacetylation 1) The fraction of the preparation formed from the nucleus of cephalosporin C migrates slightly faster than cephalosporin C towards the anode, during electrophoresis at pH 7 ( with aqueous collidine acetate as a solvent).
2) The fraction formed from the nucleus of cephalosporin C migrates at a speed equal to about half that of cephalosporin C towards the anode at pH 4.5 (with aqueous pyridine acetate as solvent).
3) The moiety formed from the nucleus of cephalosporin C remains near the point of origin in electrophoresis at pH 4.0 (in aqueous pyridine acetate), showing only relatively little migration to the anode. Under these conditions, cephalosporin C still migrates to the anode almost as quickly as at pH 7.
4) After electrophoresis in 10% acetic acid, elution from the paper and hydrolysis in N hydrochloric acid at 1050 for 16 h, it furnishes a little glycine (50 #tg giving a spot of intensity similar to that glycine stain obtained by hydrolysis of 50 [g of cepha-Iosporin C) but no α-amino-adipic acid. It therefore does not contain the side chain of the cephalosporin C molecule.
5) The fraction formed from the nucleus of cephalosporin C shows a Ru value similar to that of N-phenylacetylcephalosporin C (0.08-0.15) in butanol-ethanol-water (4: 1: 5 by volume) .
6) After chromatography in butanol-ethanol-water, or after electrophoresis, and after having sprayed onto the paper a 0.1% gelatin solution, containing penicillinase (enzyme concentration equal to 10 times that required to inactivate a stain of 50 µg of benzylpenicillin), it gives a stain of apparently undiminished activity when the paper is sprayed with phenylacetyl chloride and pyridine.
7) When a drop is placed on paper, pyridine acetate at pH 7 (2 M concentration of pyridine) is sprayed on the stain, it is suspended in the wet state on the vapor of a solution 2 M pyridine acetate at 37o for 16 h, dried, electrophoresed at pH 7 and phenylacetylated, an active stain (due to the phenylacetyl derivative of the nucleus of cephalosporin <B> C.1 < / B> (pyridine) appears in the neutral position, as well as a spot in a position slightly closer to the anode than that occupied by cephalosporin C.

The nucleus of cephalosporin C., which is the lactone of deacetyl-7-aminocephalosporanic acid, also has a steric structure similar to that of the corresponding part of cephalosporin C obtained by fermentation of cephalosporium. It can be further identified by the following properties, after phenylacetylation.
1) By electrophoresis on paper in a buffer of pyridine acetate (pH 4.5), the nucleus of cephalosporin C i. moves toward the cathode approximately the same distance as the nucleus of cephalosporin C moves toward the anode.
2) By electrophoresis on paper with an acetic acid buffer at 10 <B> () / a, </B> the nucleus of cephalosporin C ,. migrates to the anode two and a half times faster than the nucleus of cephalosporin C.
3) By chromatography in butanol-ethanol-water (4: 1: 5 by volume), the value R ,,. of the nucleus of cephalosporin C ,. is 0.33, while that of cephalosporin C is 0.08-0.15 and that of the nucleus of cephalosporin CA is 0.08.
The nuclei of cephalosporins C. ,, also have a steric structure similar to that of the corresponding part of cephalosporin C obtained by fermentation of the cephalosporium. Cephalosporin C., (pyridine) can be further identified by the following properties: 1) By electrophoresis on paper at pH 4.0 in pyridine acetate buffer, the nucleus of cephalosporin C1 moves towards the cathode on a distance similar to the distance cephalosporin C travels towards the anode.
When electrophoresis is performed in 10 <B>% </B> acetic acid, the nucleus of cephalosporin C_L (pyridine) moves about twice as fast to the cathode as cephalosporin C, t ( pyridine).
2) The nucleus of cephalosporin C @ (pyridine) is not destroyed by the action of penicillinase under conditions in which 50 g of penicillin G is completely destroyed.
3) The nucleus of cephalosporin C., (pyridine) is distinguished from the nucleus of cephalosporin C, on the chromatograms on paper treated with n-butanol-ethanol-water (4: 1: 5 by volume).
The compounds defined above, i.e. the nuclei of cephalosporins C, C, and CA are of particular importance, as they are intermediate products from which other cephalosporin C derivatives endowed with biological activity can be prepared.
The following examples illustrate the invention. Example 1 <I> Preparation of </I> 7-aminocephalosporanic acid <I> (a) Preparation known as derivative of </I> 1-flitoro-2,4-dinitro-benzene (FDNB) <I> and of cephalosporin C. </I>
31.7 g of the sodium salt of ce phalosporin C (purity 85-90 <B>% </B>) was dissolved in 635 ml of water containing 25.5 g of anhydrous sodium bicarbonate. While stirring the solution, 25.5 ml of FDNB dissolved in 635 ml of ethyl alcohol was added. This mixture was stirred in the dark at room temperature for a further 21 / = h. The pH of the solution was then adjusted to 5 with concentrated hydrochloric acid and the alcohol removed by vacuum distillation. Excess FDNB was flushed out at the same time.
After bringing this mixture to pH 7 with sodium bicarbonate, it was extracted with 3 X 1 / z vol. ether to remove excess FDNB and leave a clear aqueous solution. The pH was then brought to 2.5 with concentrated hydrochloric acid. A sticky precipitate formed, which dissolved on extraction with 3 X 1 /: a vol. ethyl acetate. The ethyl acetate extracts were combined, washed with water, dried over anhydrous sodium sulfate, and evaporated to dryness, leaving 40.3 g of a yellow solid.
A biological assay by comparison with a pure sample of the dinitrophenyl derivative of ce phalosporin C showed that this product is of a purity of about 88%.
<I> (b) Acid hydrolysis of the derivative </I> 2,4-dinitrophenyl <I> (the </I> <I> cephalosporin C. </I>
The crude 2,4-dinitrophenyl derivative of cephalosporin C from the above preparation contains a small proportion of a colored basic impurity, which was removed by dissolving the solid product in acetate. ethyl and washing with 0.2N HCl. Ethyl acetate was then dried and removed in vacuo. 10.1 g of solid was reduced to 9.7 g by this operation.
9.7 g of the derivative of DNP and cephalosporin C were dissolved in a mixture of <B> 100 </B> ml of acetonitrile, 65 ml of water and 34 ml of 1 N hydrochloric acid. This solution was then left to stand in the dark, at room temperature and under nitrogen for 64 h. Then 200 ml of water was added to the solution and the whole was extracted with 5 X 100 ml of ethyl acetate. The aqueous residue (270 ml) of 7-aminocephalosporanic acid was brought to pH 7.2 with saturated sodium bicarbonate solution and cooled to 0 ° C.
<I> (c) Isolation (the </I> 7-arriiriocephalosporanic acid <I> 11I </I> (7-ACA).
After the hydrolysis and extraction with ethyl acetate in step (b) above, an acidic solution remains, mainly containing 7-ACA and 7-ACA lactone. This solution was adjusted to pH 6.7 and passed through a column of Dowex-1 <B> (CI-). </B> The 7-ACA lactone passes through the column and appears in the filtrate. coming out of the resin. The fractions containing 7-ACA give a crystalline precipitate which appears to be the zwitterion form of this material, while the major part of the 7-ACA remains in solution as the hydrochloride. The infrared spectrum of the solid is shown in fig. 1 of the drawing. Solutions of the hydrochloride can be phenylacetylated to provide the benzylcephalosporin.
Example 2 <I> Preparation of the nucleus of cephalosporin C </I> n <I> starting (the </I> cephalosporin <I> C </I> 2 g of sodium salt of cephalosporin C were dissolved in 30 ml of water, the pH was brought to 2.5 by addition of Dowex 50 X 8 (H--), the resin was filtered off, washed with 10 ml of water and 10.2 ml of 1N HCl were added to the combined filtrate and washings The solution was maintained at 20 ° C. for 3 days and passed through a column of Dowex-1 (acetate form). 2.1 cm in diameter X 7 cm.
The percolate was collected in fractions. 5 mL (1-12) and the column eluted with water until a total of 34 fractions were collected. Elution was then started with 0.5N acetic acid and a further 66 fractions were collected. Optical density at 260 mI was measured for each fraction.
Fractions 2-16 were combined and concentrated in vacuo and 312 mg of cephalosporin C, separated in crystalline form. Fractions 36-45 contained most of the nucleus of cephalosporin C, which provided an active phenylacetyl derivative. These fractions were combined and dried under freezing (40 mg). On phenylacetylation, this product gave 250 units of activity on Staph. aureus, per mg of the original product.
The new compound was further purified by electrophoresis of the appropriate fraction from the column of Dowex-1 (acetate). Electrophoresis was performed in a Beckman / spinco Model CP continuous flow paper electrophoresis cell. The buffer used was prepared by adding pyridine to 0.05N acetic acid until the pH rose to 4.0. The cell was operated at constant current (40 mA), the voltage being 880 V.
The sample, in 20 ml buffer, was supplied to the curtain for 24 h and 32 fractions were collected, the fractions being numbered from cathode (1) to anode (32). The volume of each fraction was approximately 12 ml. At the end of the experiment, ninhidrin was sprayed on the paper curtain. This revealed a strong band of very low mobility material which was dragged out of the curtain in fractions 13-15, a band of strongly acidic material which had leaked into the anode wick, and a weak band of a product. (corresponding to α-aminodipic acid) which had migrated to the anode and was entrained out of the curtain in fraction 24.
To determine the position of the new compound, 10 dabs of 10 µl of each fraction were applied to paper and the paper was sprayed with 1 M pyridine in. 50% acetone followed by 2% phenylacetyl chloride in acetone. After contact of the paper with plates inoculated with Staph. aureus and plate incubation,
a large zone of inhibition was found in a position corresponding to fraction 22 and a smaller one in a position corresponding to fraction 23.
By freeze-drying fraction 22, a residue was obtained which was too small to be weighed accurately. In aqueous solution, the ultraviolet absorption spectrum of the product showed a plateau at 260 m # t, and its total weight (43 l, g) was estimated from the extinction at this wavelength, in assuming that the molecular weight of the substance is equal to that of 7-aminocephalosporanic acid, and that its molecular extinction is equal to that of cephalosporin C. The product has been phenyl-acetylated in aqueous acetone against nant a phosphate buffer of pH 6.5.
The activity of the phenylacetyl derivative against Staph. aureus corresponded to 1450 units per mg of original product. Cephalosporin C shows an activity of 8-10 units per mg against the same organism.
The amino group of 6-aminopenicillanic acid (penicillin nucleus) has relatively low basicity, the published isoelectric pH for this compound being 4.3 (Batchelor, Doyle, Naylor and Rolinson, 1959).
This is easy to understand, since the amino group is 3 to both a CON group and a CH - S - group. For analogous reasons, the amino group of 7-aminocephalosporanic acid should be weakly basic and the substance should migrate to the anode at pH 7.
The properties of the new derivative of cephalosporin C (described on pages 5 and 6) indicate that it is in fact 7-aminocephalosporanic acid. The resulting solution, containing 7-aminocephalosporanic acid and other weakly acidic degradation products of cephalosporin C, was neutralized and buffered to pH 7.0 with sodium bicarbonate solution under atmosphere. of C02. The pH was then lowered to 2.0 and 7-phenylacetamidocephalosporanic acid was extracted with butyl acetate, together with N-phenacetyl-cephalosporin C. and N-phenylacetyl-cephalosporin C. butyl acetate separated and extracted with water brought to pH 5.0 to equilibrium by addition of alkali. By paper chromatography of 150 µg of the product in n-butanol-ethanol-water (4: 1: 5 by vol.) It was found to contain three components active against Staph. aureus (see fig. 1).
The main active ingredient is 7-phenylacetamidocephalosporanic acid. The other active ingredients are N-phenylacetyl-cephalosporin C ,, and N-phenyl-acetyl-cephalosporin C, both of which were only partially extracted by butyl acetate.
7-Phenylacetamidocephalosporanic acid was separated from the other active components of the crude mixture (17 units / mg against Staph. Aureus) by chromatography on a Grycksbo paper roll column (IKB-Produkter, Sweden) and distribution to against the current.
Elution from the paper column was performed with n-butanol saturated with water. In a preliminary experiment, the sodium salt of 7-phenylacetamidocephalosporanic acid was obtained from the paper column as a powder having an activity of 167 units / mg. This product is clearly unclean.

After eight transfers in solvent I (10 ml of the upper layer and 20 ml of the lower layer), the concentration of 7-phenylacetamidocephalosporanic acid reached a maximum between tubes 6 and 7, while the other active components were found in tubes 0 and 1. The material collected in tubes 5, 6 and 7 assayed approximately 50 units per mg. Paper chromatography of this material in n-butanol-water-ethanol (4: 5: 1 by vol.) Showed that 7-phenylacetamidocephalosporanic acid was the only active substance present.
After eight transfers in Solvent II (20 ml of each layer), the sodium phenylacetamidocephalosporanate showed a peak between tubes 3 and 4, but most of the remainder of the active material remained in tube 0.
7-Phenylacetamidocephalosporanic acid moved towards the anode at almost the same rate as penicillin G, electrophoresed on paper in collidine acetate buffer at pH 7. Incubation with aqueous pyridine at 37 , 7-phenylacetamidocephalosporanic acid has been partially converted to an active derivative of the type of ce phalosporin CA (pyridine). This derivative behaved as a neutral substance in electrophoresis at pH 7.0.
About 10 µg of 7-phenylacetamidocephalosporanic acid was not appreciably inactivated by a solution of penicillinase containing 10 times the amount of enzyme fully inactivating 50 µg of penicillin G under normal conditions. alike.
Example 4 <I> Preparation of the nucleus of </I> cephalosporin C = 1 (pyridine) <I> from cephalosporin </I> C.1 (pyridine) Cephalosporin C_1 (pyridine) is maintained in 0.5 N or 1 N hydrochloric acid at room temperature for 3 days or more.
When the products of this reaction are analyzed by electrophoresis on paper in a buffer of pyridine acetate (pH 4.0) followed by contacting the paper with agar plates seeded with Staph. In addition, a single active zone due to untransformed cephalosporin C A (pyridine) is revealed, at the concentration used. Cephalosporin CA (pyridine) behaves as if it has no net charge at pH 4.0.
However, duplicate strips of paper sprayed with pyridine M in 50% aqueous acetone and 0.125% phenylacetyl chloride in dry acetone,
reveal an additional active zone on the inoculated plates, due to the products of the phenylacetylation of the nucleus of cephalosporin C, (pyridine). The cephalosporin CA nucleus behaves like a base at pH 4.0. It migrates towards the cathode a little greater than the distance by which cephalosporin C itself migrates towards the anode in the same experiment. It remains in a neutral position in the electrophoresis at pH 7.
This behavior is consistent with the predicted properties of the nucleus of cephalosporin CA (pyridine), (see position 9 of the definition of the nucleus of cephalosporin C). A compound with the same properties was formed by incubating the nucleus of cephalosporin C with 2 M pyridine acetate at pH 7 for 16 h. Upon phenylacetylation, this compound provided an active phenylacetyl derivative. The properties of the nucleus of cephalosporin CA have been studied and they have been shown to coincide with the properties described above for this nucleus.
Example 5 Preparation <I> of </I> drowned <I> of cephalosporin C, </I> <I> from </I> cephalospotin C, 50 mg of cephalosporin C were maintained in 1.33 ml of 0.5 N or 1 N HCl at 20o C for 2 or 3 days. The nucleus of cephalosporin C ,. was also formed by contacting cephalosporin C with an equal weight of Do-wex 50 X 8 (-H +) ion exchange resin for 15-48 h at 20o C.
Duplicate bands from ionograms carried out in a pH 4.5 pyridine acetate buffer, sprayed with phenylacetyl chloride and brought into contact with a test organism, or sprayed with ninhydrin revealed that the position of the cephalosporin C nucleus. corresponded to a yellow area on the ninhydrin-treated bands.
