'La présente invention a pour objet la prépara- tion d'une tétracycline neutre à tolérance thérapeutique améliorée,.et plus particulièrement, elle a pour objet un procédé perfectionné pour purifier la tétracycline neutre de manière à obtenir un produit présentant une apparence visuelle améliorée, une faible coloration, et pratique-, ment dépourvu de goût.
Une tétracycline"cristalline insipide est très demandée pour servir dans diverses préparations pharma- ceutiques telles que sirops, suspensions pour administra- tion buccale, gouttes pédiatriques, etc... La tétracy- cline neutre habituellement préparée, que ce soit par fermentation d'un microorganisme approprié ou par déchlo- ruration réductive de la chrorotétracycline, contient diverses impuretés fortement basiques et des matières colorées qui lui sont associées. Bien que cette matière convienne très bien à de nombreux usages, par exemple en tablettes, capsules, onguents, etc., on ne peut pas l'uti- liser facilement par voie buccale à cause du goût amer qui lui est associé, et qui rend le produit indésirable à employer dans les sirops, suspensions pour usage buccal et gouttes liquides pédiatriques.
Suivant l'invention,le procédé de préparation d'une tétracycline.neutre douée d'une tolérance thérapeu- tique améliorée consiste à ajuster le pH d'une solution aqueuse ou bouillie de matière chargée de tétracycline, à un niveau maximum de 2,0 environ, à.ajouter un ou plu- sieurs sulfites alcalins et/ou du sulfite d'ammonium, à rajuster le pH entre 2,0 et 3,0 environ, et à récupérer la tétracycline neutre ainsi précipitée. citron ou paille, très réfringents, qui sont relativement insipides, et tel quel le produit amélioré peut servir largement dans des préparations pharmaceutiques à usage bmccal, telles que sirops de tétracycline, suspensions à usage buccal, et gouttes liquides pédiatriques.
On réalise de préférence la présente invention en ajustant d'abord le pH d'une solution aqueuse ou d'une bouillie de tétracycline à 0,5 à 2,0 environ, et de pré- férence à 1,0 environ. On peut procéder à cet ajustement au moyen de tous les acides usuels tels que l'acide chlor- hydrique et l'acide sulfurique. Le traitement à l'amide solubilise la tétracycline et la fait passer en solution.
On peut ajouter un adjuvant de filtration et filtrer la solution. On ajoute à la solution un sulfite alcalin ou d'ammonium, ou un mélange de ceux-ci, et on rajuste le pH à environ 2,0 - 3,0 au moyen d'un caustique faible, et la tétracycline neutre purifiée précipite. On filtre ensuite le produit purifié, en le débarrassant des matiè- res colorées qui l'accompagnent, et on obtient un pro- duit stable de couleur claire, pratiquement exempe de l'amertume qui est associée à la tétracycline telle qu'on la produit habituellement.
Le sulfite alcalin ou d'ammonium peut être pré- sent en quantités variant de 0,9 à 15 % environ en poids par rapport à la tétracycline présente. Mais on a trouvé que pour les usages commerciaux, une gamme de 5 à 10 % environ est préférable pour des raisons économiques.
La présente invention n'est pas limitée en ce qui concerne la source de tétracycline brute. Autrement dit, la tétracycline préparée par la réduction catalyti- que de la chlorotétracycline au moyen d'un catalyseur au palladium en présence du 2-éthoxyéthanol et de la triéthylamine, par exemple de la façon expliquée dans le

Journal of the American Chemical Society, volume 75 page 4621, peut avantageusement être utilisée. On peut aussi utiliser avec la même facilité la tétracycline obte- nue par fermentation d'un microorganisme approprié,'ainsi qu'il est expliqué dans le brevet américain n02.734.018 du 7 février 1956.
Un procédé d'hydrogénolyse de la chlo- rotétracycline qui donne naissance à un produit particuliè. rement propre à servir suivant la présente invention, est décrit dans le brevet américain n02.731.497 du 17 janvier 1956.
On décrira l'invention plus en détail à propos des exemples précis suivants.
EXEMPLE 1
On délaie dans 500 cm3 d'eau 50 g de chlorhy- drate de tétracycline préparé par déchlorur,ation réducti- ve de la chlortétracycline. On ajuste le pH de la bouillie à 1,0 avec de l'acide chlorhydrique concentré.
On ajoute, avec agitation, 2,5 g de silicate de magné- sium. On ajoute 5,0 g de terre d'infusoires et on filtre la solution. On rince le gâteau filtre à l'eau et on ajoute l'eau de lavage au filtrat. On ajoute au filtrat 2,6 g de sulfite de sodium dans 10 cm3 d'eau. On ajuste le filtrat à pH 2,8 avec 75 cm3 de soude à 3,5 %. On agite la solution, on la laisse reposer 3 heures, on fil- tre et on sèche. On délaie à nouveau le produit dans 35 cm3 d'eau, on filtre et on sèche sous vide à 50 C pendant 23 heures. On obtient un rendement de 89 % en té- tracycline neutre pratiquement insipide.
EXEMPLE 2
On délaie, dans 710 litres d'eau désionisée, 80 kg de tétracycline préparée par déchloruration réduc- tive de la chlorotétracycline. On ajuste la bouillie à pH 1,0 - 1,3 avec de l'acide chlorhydrique concentré. On

ajoute 4 kg de silicate de magnésium et l'on agite le mélange. On ajoute 4 kg de terre d'infusoires avec agi- tation, et ensuite on filtre le mélange. On lave le gâteau filtre avec 90 litres d'eau désionisée que l'on rajoute au filtrat. On traite le filtrat par une solu- tion de 40 litres d'eau désionisée contenant 3,8 kg de sulfite de sodium anhydre, qualité réactif. On ajoute rapidement cette solution avec une bonne agitation. Après addition de sulfite, le pH est de 1,5 à 1,6.
On ajuste la solution à pH 2,8- 3,0 avec une solution de soude à 3,5 % et on agite le mélange. On filtre le produit, on le délaie dans 20 litres d'eau désionisée, et on filtre.
On sèche le gâteau filtre à 21 C pendant 24 heures envi- ron. On obtient une tétracycline neutre relativement in- sipide, avec un rendement de 78 %.
REVENDICATIONS 1. Procédé pour la préparation d'une tétracycline neutre à tolérance thérapeutique améliorée, caractérisé en ce que l'on ajuste à un pH non supérieur à 2,0 envi- ron, une solution ou bouillie aqueuse de matière conte- nant de la tétracycline, que l'on y ajoute un ou plu- sieurs sulfites alcalins et/ou du sulfite d'ammonium que l'on rajuste le pH entre 2,0 et 3,0 environ et que l'on récupère la tétracycline neutre ainsi précipitée.
