Abstract
De uitvinding heeft betrekking op een oppervlakteactieve samenstelling bevattende als oppervlakteactief bestanddeel tenminste één rhamnolipide, dat bij voorkeur bevat een rhamnolipide met de algemene formule:(zie fig) waarin R1 = H, alfa - L - rhamnopyranosyl; R2 = H, - CH - CH2 - COOH; R4 ; R3 = (C5-C20)-verzadigd, -mono- of poly-onverzadigd alkyl; R4 = (C5-C20)-verzadigd, - mono- of poly-overzadigd alkyl.
Description
Oppervlakteactieve samenstelling op basis van rhamnolipide De onderhavige uitvinding heeft betrekking op een oppervlakteactieve samenstelling bevattende als oppervlakteactieve stof tenminste één rhamnolipide. In het bijzonder heeft de onderhavige uitvinding betrekking op een dergelijke op een rhamnolipide gebaseerde samenstelling voor het gebruik bij het winnen en behandelen van metalen; papierproduktie; bereiding van verf en coating; olieproduktie en -raffinage; het reinigen, verven, bedrukken en eindbewerken van textiel; de fabricage van bouw- en constructiemateriaal; plastics; levensmiddelen en dranken; en in leerbehandeling. Door de verschillende combinaties van koolhydraten en lipiden, tezamen met structureel verschillende bindingen en verschillende ionische toestanden, bestaan er een groot aantal glycolipiden met een onderling sterk verschillende hydrofiele-lipofiele balans. Het is bekend dat verschillende stammen van Pseudomonas, zoals Pseudomonas aeruginosa, Pseudomonas fluorescens, Pseudomonas putida, Pseudomonas oleovorans, in staat zijn om rhamnolipiden extracellulair uit te scheiden, wanneer zij worden gekweekt op oplosbare, alsook op onoplosbare koolstofoppervlakken. De onderhavige uitvinding heeft betrekking op oppervlakteactieve samenstellingen die deze rhamnolipiden bevatten. Op basis van een uitgebreid onderzoek is gevonden dat deze op rhamnolipiden gebaseerde samenstellingen een uitstekende oppervlakteactieve werking vertonen in polaire en apolaire media. Deze uitstekende oppervlakte-werking is in hoge mate temperatuur onafhankelijk, dat wil zeggen dat deze samenstellingen volgens de onderhavige uitvinding in wezen warmtestabiel zijn. Bovendien zijn de gebruikte rhamnolipiden natuurlijke verbindingen en biologisch afbreekbaar. Het gebruik ervan zal niet een onomkeerbare milieuvervuiling tot gevolg hebben. Bij voorkeur is het rhamnolipide een rhamnolipide met de algemene formule <EMI ID=1.1> Zeer werkzame rhamnolipiden worden verkregen indien het rhamnolipide een di-rhamnolipide is. Indien de substituent R2 waterstof is, bevat het rhamnolipide slechts één lipidegroep. Indien de substituent R2 wordt gevormd door CH - CH2 - COOH, <EMI ID=2.1> bevat het rhamnolipide-molecuul twee lipide-eenheden die onderling verbonden zijn door een esterbinding. De subsituenten R3 en R4 kunnen worden gekozen uit <EMI ID=3.1> onverzadigde alkylgroepen. Voorkeur gaat uit naar onvertakte, verzadigde alkylgroepen met de algemene formule (CH2)X-CH3, waarin x = 4-20. Meer voorkeur gaat uit naar alkylgroepen waarin x = 4 of 6. Een zeer werkzaam farmaceutisch preparaat wordt verkregen indien dit bevat het rhamnolipide (alfa-Lrhamnopyranosyl-(1,2) alfa-L-rhamnopyranosyl)-3hydroxydecoanoyl-3-hydroxydecaanzuur. Aan de hand van een niet-limitatief voorbeeld zullen hierna van een rhamnolipide de isolatie en het gebruik in oppervlakteactieve samenstellingen volgens de uitvinding worden beschreven. Hierbij is van de volgende methoden gebruik gemaakt. 1. Isolatie en karakterisering Uit boorvloeistof van een olieput werden bacteriestammen geïsoleerd die rhamnolipiden kunnen synthetiseren op zowel een vloeibare koolstofbron (glucose) als op een onoplosbare koolstofbron (glycerol, gasolie). Deze geïsoleerde bacteriestammen zijn gekarakteriseerd als Pseudomonasstammen (BBL Minitek, Numerical Determination and Identification System, Becton, Dichinson and Company.) 2. Concentratie en zuivering De door extracellulaire secretie in het medium aanwezige rhamnolipiden werden op verschillende wijzen geconcentreerd en gezuiverd. a) zuur precipitatie De pH van de supernatant werd met behulp van zoutzuur gebracht op een pH 1,5-2,0, waarna werd ingedampt tot 1/10 van het oorspronkelijke volume en het concentraat liet men over de nacht bij 4[deg.]C staan. De <EMI ID=4.1> gecentrifugeerd bij 17.300 x g gedurende 30 minuten bij 4[deg.]C en vervolgens geëxtraheerd met CH2C12, gefiltreerd, drooggedampt, opnieuw gesuspendeerd in water en vervolgens opnieuw geprecipiteerd. b) schuimfractionering Samengeperste lucht werd gebracht tot in de supernatant en gevormd schuim werd opgenomen in een afzonderlijke houder, of gebracht in aangezuurd water (pH 1,5-2,0). c) chromatografie Supernatant werd gebracht op een preparatieve kolom (Amberlit XAD-8 of XAD-2 hars, Rohm & Haas). De kolom was geëquilibreerd met water en werd na absorptie doorspoeld met water. De actieve componenten werden geëlueerd met een lagere alkylalcohol, zoals ethanol of methanol. Het gebruikte oplosmiddel werd afgedampt onder vacuüm en het concentraat werd aangezuurd en geprecipiteerd zoals hiervoor is beschreven. 3. Chemische detectie en karakterisering. Dunne-laag-chromatografie werd analytisch uitgevoerd met silica gel 60F 254 platen (Merck) en preparatief uitgevoerd met Kemika-platen. Voor een optimale scheiding kunnen de navolgende oplosmiddelmengsels worden gebruikt: CHC13-MeOH-azijnzuur-water (25:15:4:2; 12:15:4:2; 25:25:4:2) CHC13-MeOH-azijnzuur (80:15:5) Hexaan-isopropylether-azijnzuur (15:10:1) CHC13-MeOH-NH40H (25%)-water (65:25:4:2) propanol-NH40H (25%) (4:1). VOORBEELD Pseudomonas aeruginosa werd aeroob gekweekt bij een temperatuur van 32[deg.]C. De volgende kweekmedia bleken zeer geschikt voor de rhamnolipide-produktie. a) 5 g glucose, 5 g pepton, 2 g gistextract, 5 g <EMI ID=5.1> oplossing en 1 liter water. b) 20 g glucose, 10 g gistextract, 20 g CaC03 en 1 liter water. Glucose kan worden vervangen door glycerol, waardoor een twee-fasensysteem ontstaat. Glycerol als koolstofbron levert een betere opbrengst in vergelijking tot glucose, maar een meer gecompliceerd proces is vereist. De verandering van de oppervlaktespanning van het kweekmedium naar 28-31 mN/m werd gemeten met behulp van een White's ring tensiometer. Deze verlaging van de oppervlaktespanning is een goede maat voor de opbrengst aan het einde van de fermentatie. De rhamnolipideconcentratie werd spectrofotometrisch gemeten met antron-reagens. Nadat de biomassa was afgescheiden door centrifugeren, werd de supernatant gechromatografeerd op silica gel 60 F 254 platen en op Kemika platen. Glycolipiden konden worden aangetoond door gebruik te maken van de navolgede reagentia, voor lipiden alphanaphtol, voor glycolipiden diphenylamine, en voor koolhydraten en oligosacchariden beta-naphtol-timol en anthron. Gebruikmaking van deze reagentia leverde op, dat de verbindingen van het glycolipide-type zijn. Met alphacyclodextrine, reagens en kleurstof voor vetzuren, kon worden aangetoond dat het lipofiele/hydrofobe deel van het molecuul bestaat uit vetzuurketens met een even aantal koolstofatomen. Negatieve resultaten werden verkregen met reagentia voor onverzadigde lipiden. Met behulp van alkali KMn04 en ammoniacaal AgN03 kon de aanwezigheid van glycosidische bindingen worden aangetoond. Door alkalische hydrolyse werden esterbindingen tussen aanwezige vetzuren gesplitst, en door zure hydrolyse werd de O-glycosidische binding tussen de suikergroep en de OH-groep van het lipide-deel van het molecuul gesplitst. 10 volumina van de supernatant die de rhamnolipiden bevat, werden geleid over een amberlit XAD-8 of XAD-2 kolom (Rohm & Haas). De kolom werd gewassen met water. De rhamnolipiden werden geëlueerd met 100% MeOH. De fracties die de rhamnolipiden bevatten werden drooggedampt en daarna opgenomen in dezelfde hoeveelheid zuiver water. 1 N HC1 werd toegevoegd om de rhamnolipiden neer te slaan. De neergeslagen rhamnolipiden werden gecentrifugeerd bij 3.000 toeren per minuut gedurende 10 minuten. De neergeslagen rhamnolipiden werden opnieuw gewassen met zuiver water en vervolgens gecentrifugeerd bij 3.000 toeren per minuut gedurende 10 minuten. Met behulp van 1/10 N NaOH werd de pH van de neergeslagen lipiden ingesteld op 7,2. Na vriesdrogen werd 10 g van het gevriesdroogde preparaat opgelost in 50 ml propanol en gebracht op een silica-kolom (Waters HPLC, volume 500 ml) die was geëquilibreerd met hexaan. Met behulp van 5 liter propanol werden verontreinigingen geëlueerd. De rhamnolipiden werden geëlueerd onder gebruikmaking van het oplosmiddelmengsel propanol-25% NH40H (4:1). De actieve fracties werden drooggedampt en opgelost in water, neergeslagen met 1 N HCL, gecentrifugeerd bij 3.000 toeren per minuut, en de neerslagen werden met behulp van 0,1 N NaOH ingesteld op pH 7,2 en gevriesdroogd. De verkregen, zuivere rhamnolipiden werden gebruikt voor het bereiden van de bio-oppervlakteactieve samenstellingen volgens de uitvinding. Deze samenstellingen kunnen bestaan uit polaire en apolaire oplossingen, dispersies, emulsies, suspensies en mengsels daarvan. Afhankelijk van de uiteindelijke toepassing bevatten de bio-oppervlakteactieve samenstellingen vanaf 0,05 tot 10 gew.% actief rhamnolipide. Bij voorkeur echter is de rhamnolipide-concentratie vanaf 0,1 tot 2,0 gew.%, meer bij voorkeur vanaf 0,1 tot 1,0 gew.%. De ondergrens wordt bepaald door de activiteit van het aanwezige rhamnolipide en de bovengrens voornamelijk door kostenoverwegingen. Formulerinqsvoorbeeld Waterige oplossingen werden bereid met 0,00001 tot 1,0 gew.% rhamnolipide. Het rhamnolipide was (alfa-Lrhamnopyranosyl-(1,2) alfa-L-rhamnopyranosyl)-3hydroxydecoanoyl-3-hydroxydecaanzuur, en was geïdentificeerd met <EMI ID=6.1> ii) massaspectrofotometriegegevens. <EMI ID=7.1> [deg.]: toerekeningen kunnen omgekeerd zijn <EMI ID=8.1> suiker-deel <EMI ID=9.1> lipide-deel <EMI ID=10.1> massaspectrum <EMI ID=11.1> terminaal lipide en rhamnose Bij het winnen van metalen kunnen deze samenstellingen op basis van rhamnolipide worden gebruikt voor het concentreren van erts. In de bewerking en behandeling van metaal kunnen deze samenstellingen worden gebruikt voor het snijden en vormen, gieten, verwijderen van roest en aanslag, en bij het plateren. In de papierproduktie kunnen deze samenstellingen overeenkomstig de uitvinding worden gebruikt voor behandeling van pulp, in papiermachines voor het ontschuimen, bij het instellen van het kleurniveau en de dispersie, en tijdens het kalanderen. In verf- en coatingbereiding kunnen de rhamnolipiden worden gebruikt bij de bereiding van pigmenten en in latex-verven en verder in wassen. Bij de produktie en raffinage van olie kunnen deze oppervlakteactieve stoffen worden gebruikt in boorvloeistoffen, tijdens het overlopen van producerende bronnen, gedurende de olieproduktie door deëmulsificatie van de ruwe olie, bij het herwinnen van secundaire olie en tenslotte in gerede produkten. Met betrekking tot textiel kunnen deze rhamnolipiden worden gebruikt bij de fabricage van vezels, in verf- en drukprocessen en verder bij de eindbewerking van textiel. In de landbouw kunnen deze bio-oppervlakteactieve rhamnolipiden worden gebruikt in kunstmeststoffen, zoals fosfaat-kunstmest, en in sproeitoepassingen voor biociden, zoals pesticiden en herbiciden. In bouw- en constructiematerialen kunnen deze rhamnolipiden worden gebruikt voor plaveisels, zoals bij de verbetering van de bindende kracht van asfalt aan gravel en zand, en in beton. Voor plastics kunnen deze bio-oppervlakteactieve stoffen worden gebruikt in emulsiepolymerisatie, in schuimplastic, in plastic voorwerpen als anti-statische stoffen en in plastic-coating en laminaat. Voor voedingsmiddelen en dranken kunnen deze oppervlakteactieve stoffen worden gebruikt bij de voedselbereiding, voor fruit en groenten, zoals bijvoorbeeld wascoatings, in gebak en ijs, bij de kristallisatie van suiker door het wassen te verbeteren, en bij het koken van vetten en oliën. Voor reiniging kunnen deze rhamnolipiden worden gebruikt in detergentia en sanitaire samenstellingen. Tenslotte kunnen de rhamnolipiden worden gebruikt in de leerbehandeling voor het behandelen van huid, voor het looien, voor de behandeling van vellen en voor het drogen van leer. Proefvoorbeeld 1 De overeenkomstig het voorbeeld bereide rhamnolipiden werden opgelost in een concentratie 500 mg/10 liter water en gebruikt in een wasmachine voor het wassen van kleding met vetvlekken. Gedurende het wassen werd geen excessieve schuimvorming waargenomen, en de vetvlekken werden verwijderd. Proefvoorbeeld 2 Een waterige oplossing van het rhamnolipide volgens het voorbeeld van 1:10.000 werd gebracht (5 ml) op een preparatieve HPLC kolom voor de verwijdering van organische materialen. Na elueren was het door standaardprocedures niet te verwijderen organische materiaal verdwenen. Proefvoorbeeld 3 Een met olie verontreinigd oppervlak werd gesproeid met een 0,5% kleine w/v oplossing van het rhamnolipide volgens de uitvinding. De geëmulsificeerde olie werd van het oppervlak gespoeld met water. Proefvoorbeeld 4 Om te onderzoeken of de bio-oppervlakteactieve stoffen volgens de uitvinding in staat zijn met olie verontreinigd zand, rotssteen en water zoals zeewater te reinigen werd de oppervlaktespanning van 0,01 en 0,05 gewichtsprocentige oppervlakteactieve oplossingen van synthetisch pekel (15 g natriumchloride per m<2> + 200 pbm CH2+) gemeten. Bovendien werd de interfaciale spanning gemeten van deze bio-oppervlakteactieve oplossingen tegen kerosine D2 olie, een commercieel verkrijgbaar koolwaterstof mengsel. Bovendien werden deze metingen ook uitgevoerd na de biooppervlakteactieve oplossingen aan een warmtebehandeling <EMI ID=12.1> Een 0,01 gewichtsprocent oplossing in pekel reduceert de interfaciale spanning tegen kerosine (D2 olie) van 33,5 mN/m tot een waarde van 0,22 mN/m. De warmtebehandeling had geen effect op de verkregen resultaten. Rhamnolipide oppervlakteactieve stoffen zijn in staat de interfaciale spanning van het systeem pekel/olie te reduceren van 33,5 mN/m tot 0,5-0,2 mN/m (8,5 tot reductie). Bij de lage concentratie van 0,01 gewichtsprocent/vol in pekel wordt de oppervlaktespanning teruggebracht van aanvankelijk 71,0 mN/m tot 28-30 mN/m (ongeveer 60%). Proefvoorbeeld 5 De resultaten van de laboratoriumproef voor verbeterde olieherwinning tonen de efficiency aan van de rhamnolipide-oplossing in de secundaire fase van het terugwinnen van olie, bij de zogeheten gemodificeerde waterstroom, die een verplaatsing van de oliebron omvat. Additionele olie die werd teruggewonnen in relatie tot de waterstroom is 10-26% van de oorspronkelijke middelen, of 25-54% van de residu-verzadiging met opname van 2-3 P.V. (porievolume) van de rhamnolipide oplossing (0,01 mg/ml). Aan de verplaatsingscondities is voldaan, zoals getoond in de diagrammen 1 en 2, door - regulering van de mobiliteit van de geïnjecteerde oplossing tot 0,1 m/dag; en - een tijdelijke afsluiting van de produktie zodat het "stromen" van de bron kan worden gerealiseerd. In praktische situaties betekent dit een 6 maal langer durende produktiecyclus in vergelijking met de waterstroom zelf.
