Description

Descriptions
A Pharmaceutical Composition for Treating Diarrhea of Piglet and Its Preparation Method
Technical Field
[0001] The present invention relates to a pharmaceutical composition for treating animal diseases, particularly to a pharmaceutical composition for treating diarrhea of piglet and its preparation method.
Background Technology
[0002] Diarrhea of piglet is a type of disease with a relative high incidence rate and mortality rate clinically, which would seriously harm the development of the pig industry. In China, it is a common disease. In addition, it is reported that the annual average incidence rate of piglets under kg reaches up to 46.5%, and the mortality rate reaches up to 15%, and thus causes more than one hundred million in direct economic loss to the pig industry each year. For the Occident and other countries, where the pig industry is more developed, the incidence rate and the mortality rate derived from such disease are also relatively high. No matter in domestic or at abroad, this disease has troubled breeders seriously, especially in intensive pig farms. Once the diarrhea of piglet occurs, it would result in the decrease in surviving rate, the slow growth of piglets, even makes the piglets stop growing, in addition to increasing the expenses for medicine, and thus it affects the feed conversion efficiency, impairs the production performance, as well as causes substantial economic loss to pig farms. There are many types of pathogens which may cause the disease, and the nature of which is complex, including viruses (transmissible gastroenteritis, epidemic diarrhea, and rotavirus, etc.), bacteria (Escherichia coli, Clostridium welchii, Salmonella, and vibrio, etc.), and parasites (Coccidia, and Balantidium coli malmsten, etc.). According to the mode of onset, different symptoms would appear. Most of them have symptoms of acute attack, increased temperature, sensation of chill, and being inclined to crawl into the mow or huddle together, in addition, they would manifest depression, laziness, inactivity, loss of appetite or noneating, as well as have symptoms of difficulty in breathing and watery diarrhea, wherein, the feces are yellowish-white or greyish-white with stench, and there are some undigested feed or curds are mingled with feces. To the later stage, the piglets would have symptoms of dehydration, and their clothing hair becomes crude and yellow and withered, as well as with enophthalmos; meanwhile, for chronic disease, the body temperature would not rise generally or even lower than normal temperature, and the symptoms of low spirits, loss of appetite, moving slowly or standing transfixed to the ground, as well as recurrent diarrhea, serious dehydration, allotriophagy, and being inclined to chew bedding and drink dirty water would be occurred, furthermore, they would suffer from severe anemia, and the visible mucosal becoming pale, and result in death due to systemic failure finally.
[0003] Nowadays, the methods of administration by feeding or injection of chemically synthesized antibacterial or antiviral medicine are frequently adopted to prevent and cure the diarrhea of piglet.
Although these methods can temporarily slow down the progress of disease, they just can cure the symptoms instead of the disease. In addition, such methods require a large amount of capital to be invested, so that they make the efficiency of pig raising decrease. Moreover, some antiviral chemical medicine (such as Moroxydine and Ribavirin, etc.) have been prohibited from treating animal diseases in China. In addition, antibacterial medicines are apt to cause drug residues, and have disadvantages of causing secondary diseases or infection of lentivirus due to incomplete treatment. However, the combined application of traditional Chinese and Western medicine has unique advantages in aspect of antibacterial and antiviral applications, which is at low cost, and has slight toxic and side effects, as well as with extensive sources. Such method not only can save costs, but also can control the disease in a short time, and thus it reduces the death rate of piglets, and improves the efficiency of breeding. Therefore, it is urgent to develop a pharmaceutical composition of traditional Chinese and western medicine for treating diarrhea of piglet, which would be prepared for clinical application.
Summary of the Invention
[0004] The technical problem needs to be addressed by the present invention is to overcome the existing shortcomings of the treatment of diarrhea of piglet, and provide a pharmaceutical composition for treating the diarrhea of piglet which is easy to obtain materials, convenient to prepare, and may reduce drug residues and toxic and side effects, as well as is at low cost and with high treatment effect. This prescription has the advantages of having high overall efficacy, nearly no drug resistance, low residues, and slight toxic and side effects, which would have significant inhibiting effect on the growth and reproduction of many pathogenic microorganisms, and further can achieve the pharmacological effect of killing pathogens, and thus it is of great economic and social significance to the healthy development of the pig industry.

[0008] Another purpose of the present invention is to provide a preparation method for the medicine administrated by drenching 1 of the pharmaceutical composition for treating diarrhea of piglet: Weigh and take raw materials as per the ratios of the pharmaceutical composition for treating diarrhea of piglet, then, pulverize the Herba Potentillae Chinensis, the Polygonum Hydropiper L, the white Hibiscus, the Radix pulsatillae, the Cortex Fraxini, the Purslane, the Radix Curcumae, and the Rhizoma Atractylodis, after that, decoct them with water, and then filter the dregs and take the filtrate, finally, concentrate the filtrate into 0.66-0.70g/ml of crude medicine;
The present invention is a pharmaceutical composition for treating diarrhea of piglet. For this prescription, the Herba Potentillae Chinensis refers to the root or the whole herb with root whole of the Herba Potentillae Chinensis (rosaceous plant), which is bitter in taste and cold in nature, and has the efficacy of clearing heat and removing toxicity, hemostasis and relieving diarrhea, and further can achieve better treatment effect on abdominal pain and diarrhea, and lingering dysentery especially. The white Hibiscus has the efficacy of clearing dampness and heat, cooling the blood, and thus it can treat the dysentery, further would enhance the therapeutic effect of the Herba Potentillae Chinensis if administrated with the Herba Potentillae Chinensis. In addition, the white Hibiscus has the efficacy of clearing heat and drying dampness, cooling blood for hemostasis, which can achieve significant therapeutic effect on damp-heat dysentery or diarrhea. Hibiscus is divided into two types: i.e., the red Hibiscus and the white Hibiscus, wherein, the red Hibiscus has the therapeutic effect on hematochezia with syndrome of intestinal wind, and the white Hibiscus has the therapeutic effect on white diarrhea. Thus, the white Hibiscus can achieve better therapeutic effect on diarrhea. The Polygonum Hydropiper L refers to the dry whole herb of the Polygonum pubescens Blume (polygonaceae plants), which is pungent in taste and warm in nature, and is distributed in the lung, liver, and large intestine meridians. This medicine has the efficacy of detoxication, eliminating dampness, dissipating blood stasis, and hemostasis, which can achieve practical effect for treating dysentery, gastroenteritis, and diarrhea, etc. The Radix pulsatillae has the efficacy of clearing heat and removing toxicity, and cooling blood for checking diarrhea through invading the blood system, which is conducive to eliminate dampness heat in the gastrointestinal tract, and thus it constitutes the priority medicine for treating damp-heat diarrhea; In addition, the Cortex Fraxini is bitter in taste and cold in nature, which is distributed in the liver, gallbladder, and large intestine meridians, and has the efficacy of clearing heat and drying dampness and removing liver-fire for improving eyesight. This medicine can treat damp-heat diarrhea, and is often administrated with Radix pulsatillae, etc. The Purslane has the efficacy of clearing heat and removing toxicity, dispelling blood and detumescence, which can achieve therapeutic effect on heat dysentery, pyaemia, and heat strangury, etc. This medicine is especially suitable for treating diarrhea or dysentery caused by dampness-heat. The Radix Curcumae is pungent and bitter in taste, and cold in nature, which is distributed in the liver, heart, and lung meridians, and has the efficacy of cooling blood and clearing away the heart fire, as well as promoting qi circulation and resolving depression. This medicine can achieve therapeutic effect on chest and abdomen pain caused by damp-heat diseases and stagnation of qi and blood. The Rhizoma Atractylodis pungent and bitter in taste, and warm in nature, which is distributed in the spleen and stomach meridians, and has the efficacy of drying damp and strengthening spleen, and relieving exterior syndrome by diaphoresis. The Montmorillonite, also known as microcrystalline kaolinite, which is an aluminosilicate and is appropriate for treating acute and chronic diarrhea of humans and animals clinically. The montmorillonite powder, which is the mature product of montmorillonite in the field of medicine, has almost become synonymous with montmorillonite. This medicine has the efficacy of checking diarrhea. Furthermore, relevant studies on Montmorillonite show that it has a good adsorption effect on Escherichia coli, Vibrio cholerae, Campylobacter jejuni, Staphylococcus aureus, rotavirus and cholate; which can fix the bacterial toxins; furthermore, the Montmorillonite only adsorbs and fixes the pathogenic charged pathogens with mitochondrial encoding protein (CS31 A) on the surface, instead of the normal flora without CS31 A on the surface. The heterogeneous static dissipation of the Montmorillonite makes it able to adsorb various pathogenic factor existed in the digestive tract. And the electrostatic bonding of the Montmorillonite and the mucus protein of the digestive tract, can increase the amount of mucus and improve the quality of the mucus, as well as improve the cohesion and elasticity of the mucus, and thus it can protect and repair the mucous membrane of the digestive tract. In addition, the Montmorillonite plays better effect on diarrhea caused by various causes, whether it is bacterial, viral, or dyspeptic diarrhea, or is the diarrhea with dysbacteriosis, and enterotoxin syndrome. Furthermore, the Montmorillonite would not enter the blood, which can be excluded from the body completely, and without residues absolutely. Meanwhile, any bacteria or virus cannot produce drug resistance to the Montmorillonite, and thus it can be deemed as a green animal health care product. In addition, the Cerebiogen tablet is a safe and effective active bacterial preparation, which is free from toxic and side effects. This medicine can be mainly used for treating human and animal diarrhea, acute and chronic enteritis, acute and chronic dysentery and gastrointestinal function disorder, etc., especially is conducive to treating the diarrhea of piglet.
[0009] The combination of all medicines would have the efficacy of clearing heat and removing toxicity, eliminating dampness and checking diarrhea.
[0010] The characteristics of the prescription include that: this prescription has the efficacy of clearing heat and removing toxicity, eliminating dampness and relieving diarrhea, meanwhile, it has the effect of regulating the activity of Qi and removing exterior pathogens, as well as treating both the exterior and the interior.
[0011] The aforesaid medicines are organically combined, dialectically compatible and mutually assisted to enhance the efficacy clearing heat, eliminating dampness and checking diarrhea of the prescription, and further achieve the purpose of curing diarrhea of piglet. Through scientific compatibility, the medicine administrated by drenching 1 shall be administrated in the morning, while the medicine administrated by drenching 2 shall be administrated in the afternoon. In addition, both medicines hall be administrated separately at a certain time interval. Moreover, the present invention realized the organically combination of the traditional Chinese compound decoction, the montmorillonite and the Cerebiogen, etc, and thus it can more effectively prevent and treat various diarrhea of piglet caused by bacteria and viruses. In addition, the treatment effect is reliable, safe and non-toxic, which can enhance the immunity of animals and improve the efficiency of raising. The clinical treatment tests proved that the cure rate of diarrhea of piglet of this prescription reached up to 90.23%, and the effective rate reached up to 98.70%.

Wherein, take 5 days for administration as a course of treatment,, and the specific therapeutic effects are divided into three levels: Cure: All symptoms of diarrhea of piglet disappeared; Improved: Partial symptoms of diarrhea of piglet disappeared; Invalid: There is no obvious improvement to symptoms of diarrhea of piglet.
[0027] For the past few years, we have treated thousands of piglets suffered from diarrhea in several pig farms and veterinary hospitals, etc. After being treated for one course of treatment, the cure rate reached up to 92.17% and the effective rate reached up to 99.40%.
[0028] Clinical efficacy trial 1. Method: selected five pig farms where the diarrhea of piglet occurred, and selected 1266 piglets suffered from diarrhea in total, which are at the age of 7-30 days. Then, divided the diseased piglets into four groups, i.e., the group of the present invention, the group administered the prescription composed of the Radix pulsatillae, the group administered the montmorillonite and the Cerebiogen and the control group.
[0029] The group of the present invention refers to a pharmaceutical composition for treating diarrhea of piglet prepared according to the Embodiments 3 and 4.
[0030] The decoction composed of the Radix pulsatillae administered by the group administered the prescription composed of the Radix pulsatillae is derived from the Treatise on Febrile Diseases, which comprises 60 parts of Radix pulsatillae, 30 parts of Cortex phellodendri, 45 parts of Coptis chinensis, and 60 parts of Cortex fraxini, all of which shall be put in a decoction pot, then, add 1200 parts of water, and decoct them for 30 minutes, after that, remove the residues, and then take the filtrate, the obtained filtrate is 287 parts, and per milliliter of filtrate contains 0.68 g of crude drug.
[0031] The group administered the montmorillonite powder and the Cerebiogen refers to the group which administers the medicine administrated by drenching 2 of the present invention, which comprises 5 parts of montmorillonite powder and 5 parts of Cerebiogen tablets. Then, add water as 5 times the total amount of the medicine, i.e., 50 parts of water, and thus, the medical solution contains about 0.20 g of crude medicine per milliliter.
[0032] The control group did not receive any medication.
[0033] The treatment time is a course of treatment, that is, 5 days. The medicine shall be administered once a day in the morning and afternoon, and the dosage is according to the criterion of 4ml/kg of weight.
