Abstract

Abstract The invention discloses preparation of a pharmaceutical composition gel preparation against human papillomavirus and gynecological inflammations, comprising the following components by weight: 20-60 parts of Zanthoxylum 5 bungeanum, 20-60 parts of Arternisia argyi, 6-10 parts of Dendrantherna indicurn, 6-10 parts of Aloe vera and 6-10 parts of Campsis grandiflora. The pharmaceutical composition gel preparation of the invention has a good inhibitory effect on all of Staphylococcus aureus, Escherichia coli, mold, Neisseria gonorrhea, and Candida albicans, and is effective in the prevention and treatment of human papillomavirus 10 (HPV) as well as the treatment of gynecological inflammations caused by various reasons.

Description

Preparation of Gel Preparation of Pharmaceutical Composition against Human Papillomavirus and Gynecological Inflammations
BACKGROUND
The present invention relates to preparation of a gel preparation of a pharmaceutical composition.
Studies have shown that human papillomavirus (HPV) plays an important role in the pathogenic process of cervical cancer, and a long-term infection of highrisk HPV is a primary inducing factor of cervical cancer. Therefore, the prevention and treatment of HPV infection is particularly important for the effective inhibition of cervical cancer induced by HPV infection. Gynecological inflammations are common female diseases, and particularly comprise vulvitis, vaginitis, cervicitis and pelvic inflammation. Common symptoms mainly comprise pruritus of vulvae and vagina, some cases have local erosion, with burning pain, urodynia, frequent urination, increased vaginal secretion, redness and swelling, etc. Symptomatic therapies and treatments as early as possible are necessary for protecting the patient's physical and mental health and preventing even infertility and other problems. The invention is specialized and effective in treating HPV infection and gynecological inflammations. No report of a composition composed of Zanthoxylum bungeanum, Artemisia argyi, Dendranthema indicum, Aloe vera, and
Campsis grandiflora of the present invention has been found so far.
(1) Zanthoxylum bungeanum·. a dried mature pericarp of Zanthoxylum schinifolium sied.rt zucc. or Zanthoxylum bungeanum Maxim. Mature fruits are harvested in the fall and dried in the sun, with seeds and impurities removed. [Nature and Flavor]: pungent, and warm. [Channel Tropism]: targeting at spleen,

Useful for cold pain of the stomach duct and abdomen, vomiting and diarrhea and abdominal pain due to parasitic infestation; external use for eczema and pruritus vulvae.
(2) Artemisia argyi·. dried leaves of Artemisia argyi levl.et Vant.d. The leaf is picked in summer before blossom, with impurities removed, and is dried. [Nature and Flavor]: pungent, bitter, warm; and slightly toxic. [Channel Tropism]: targeting at liver, spleen, and kidney channels. [Function and Indications]: warming channels to stop bleeding, dispersing cold and relieving pain; and eliminating dampness and relieving itching in the case of external use. Useful for haematemesis, non-traumatic hemorrhage, metrorrhagia and metrostaxis, menorrhagia, fetal bleeding, cold-pain in the lower abdomen, cold-induced irregular menstruation, uterine cold-induced infertility; and external use for skin itching. Artemisia argyi carbonisatus with vinegar can warm channels to stop bleeding, and is used for haemorrhage due to deficiency-cold.
(3) Dendranthema indicunr. dried capitula of Chrysanthemum indicum L. [Nature and Flavor]: bitter, pungent, and cool. [Channel Tropism]: targeting at lung and liver channels. [Function and Indications]: dispelling wind and clearing heat, detumescence and detoxication. Useful for treating wind-heat common cold, pneumonia, diphtheria, gastroenteritis, hypertension, furuncles, carbuncles, aphthae, phlogosis, eczema, and pemphigus.
(4) Aloe vera: juice concentrates of leaves of Liliaceae, e.g. Aloe barbadmsis Miller, Aloe ferox Miller or other congeners of close related species. [Nature and Flavor]: bitter, and cold. [Channel Tropism]: targeting at liver, stomach,

(5) Campsis grandiflora: dried flowers of Campsis grandiflora (Thunb.) K.Schum. or Campsis radicans (L.) Seem. Flowers are picked when abloom in summer and autumn, and dried. [[Nature and Flavor]: sweet, sour, and cold. [Channel Tropism]: targeting at liver, and pericardium channels. [Function and Indications]: activating blood to promote menstruation, cooling blood and dispelling wind-evil. Useful for irregular menstruation, amenorrhea and abdominal mass, postpartum breast swelling, redness caused by rubella, skin itching, and acnes. SUMMARY
The object of the present invention is to provide a gel preparation against human papillomavirus (HPV) and gynecological inflammations, which has efficacies such as prevention and treatment of human papillomavirus (HPV) and treatment of gynecological diseases. The technical means utilized by the present invention are as follows:
A pharmaceutical composition against human papillomavirus and gynecological inflammations, comprising the following components by weight: 2060 parts of Zanthoxylum bungeanum, 20-60 parts of Artemisia argyi, 6-10 parts of Dendranthema indicum, 6-10 parts of Aloe vera and 6-10 parts of Campsis grandiflora.

Further, the pharmaceutical composition comprises the following components by weight: 30-50 parts of Zanthoxylum bungeanum, 30-50 parts of Artemisia argyi, 8-10 parts of Dendranthema indicum, 8-10 parts of Aloe vera and 8-10 parts of Campsis grandiflora.

A method for preparing a gel preparation against human papillomavirus and gynecological inflammations, comprising the steps of:
a. decocting the pharmaceutical composition of the present invention with water to obtain an aqueous extract;
b. filtering the aqueous extract, adding ethanol to a filtrate, wherein the weight of ethanol added is 1-3 times the weight of the filtrate, subjecting a mixed solution of the filtrate and ethanol to alcohol precipitation at 2-6°C for 12-24 h, then performing rotary evaporation for removing ethanol, and concentrating to a paste;
and
c. adding carbomer, glycerin, chitosan and distilled water to the paste, placing at room temperature for 12-24 h, adjusting pH to 3.9-4.5 with triethanolamine, and stirring uniformly.

Further, in step a, the pharmaceutical composition is immersed in an aqueous solution of acetic acid having a weight of 8-10 times the weight of the pharmaceutical composition for 20-30 minutes, then is decocted with water 2-3 times for 50-60 minutes each, and filtered, and filtrates are combined, wherein the pH value of the aqueous solution of acetic acid is 2.5-3.5.
Further, in step a, the pharmaceutical composition is immersed in an aqueous solution of acetic acid having a weight 10 times the weight of the pharmaceutical composition for 30 minutes, then is decocted with water 3 times for 60 minutes each time, and filtered, and filtrates are combined, wherein the pH value of the aqueous solution of acetic acid is 3.
Further, in step b, ethanol having a weight 2 times the weight of the filtrate is added to the filtrate for alcohol precipitation; and in step c, placing at room temperature for 18 hours, and the pH is adjusted to 4.2 with triethanolamine, followed by stirring uniformly.
Use of the pharmaceutical composition or the gel preparation of the present invention in preparation of a medicament against human papillomavirus, dermatoses or gynecological inflammations.
Compared with the prior art, the pharmaceutical composition and the gel preparation of the present invention are effective in the prevention and treatment of human papillomavirus (HPV)as well as the treatment of gynecological inflammations caused by various reasons, and have an excellent inhibitory effect on all of Staphylococcus aureus, Escherichia coli, mold, Neisseria gonorrhea, and Candida albicans.

Dendranthema indicum, 10 parts of Aloe vera and 10 parts of Campsis grandiflora.

A preparation method for the above-mentioned gel preparation is as follows:

c. adding Carbomer 940, glycerin, chitosan and distilled water to the paste, placing at room temperature for 20 h for a sufficient swelling, adjusting pH to 4.3 with triethanolamine, and stirring uniformly.
Example 5
A pharmaceutical composition against human papillomavirus and gynecological inflammations, comprising the following components by weight: 60 parts of Zanthoxylum bungeanum, 60 parts of Artemisia argyi, 9 parts of Dendranthema indicum, 9 parts of Aloe vera and 9 parts of Campsis grandiflora.
A gel preparation against human papillomavirus and gynecological inflammations, prepared from the pharmaceutical composition of this example.
A preparation method for the above-mentioned gel preparation is as follows:
a. immersing the pharmaceutical composition of this example in an aqueous solution of acetic acid having a pH of 3.0 and having a weight 10 times the weight of the composition for 30 minutes, then decocting with water 3 times for 60 minutes each time, filtering, combining the filtrates, so as to obtain an aqueous extract;

b. filtering the aqueous extract, adding ethanol to a filtrate, wherein the weight of ethanol added is 1 time the weight of the filtrate, subjecting a mixed solution of the filtrate and ethanol to alcohol precipitation at 4°C for 24 h, then performing rotary evaporation for removing ethanol, and concentrating to a paste;
and
c. adding Carbomer 940, glycerin, chitosan and distilled water to the paste, placing at room temperature for 20 h for a sufficient swelling, adjusting pH to 4.5 with triethanolamine, and stirring uniformly.
The above-mentioned examples were subjected to microbial inhibition experiments, and the experimental results are shown in Table 1.

It can be seen from Table 1 that the gel preparations of the examples of the present invention are effective in microbial inhibition and the treatment of gynecological inflammations.
Clinical Trials

The gel preparation of the present invention for preventing and treating human papillomavirus (HPV) and treating gynecological diseases has been used for treating and observing 93 patients or volunteers. Five above-mentioned example samples prepared were applied to four different types of patients or volunteers respectively, as follows:
A: 11 human papillomavirus (HPV) positive cases (with main symptoms: vaginal irregular bleeding and abnormal leucorrhea; vascular tumor embolus, deep muscle infiltration in the cervix, and lymph node metastasis therebetween; and obvious lumps in the cervix) in total with an average age of 52.3.
B: 31 vulvitis (with main symptoms: vulvar redness, swelling and congestion, skin itching, pain, burning sensation, and aggravated pain during urination or sexual intercourse) cases in total with an average age of 33.7.
C: 32 vaginitis (with main symptoms: frequent urination, urodynia, increased vaginal secretion in a soybean curb residue-like or milk curd-like form with an off15 odour; and itching and burning pain in vulva) cases in total with an average age of 29.6.
D: 19 eczema (with main symptoms: thick and millet-sized papules, papulovesicles or blisters on the skin surface, basal flushing, and strong itching) cases in total with an average age of 27.6.
Each of example samples was applied to not less than 10 cases of the above four types of cases. Results: Group A: 8 cases were cured, 2 cases were improved, 1 case was ineffective, and the total effective rate was 90.9%; Group B: 27 cases were cured, 3 cases were improved, 1 case was ineffective, and the total effective rate was 96.8%; Group C: 28 cases were cured, 4 cases were improved,
2019100620 10 Jun 2019 and the total effective rate was 100%; and Group D: 16 cases were cured, 2 cases were improved, 1 case was ineffective, and the total effective rate was 94.7%.
The results show that the application of the gel dosage form prepared by the traditional Chinese medicine composition of the present invention is safe and effective in use, has good efficacy on a human papillomavirus (HPV) positive condition, vulvitis, vaginitis and eczema, and can eliminate the patient’s pain and effectively improve the quality of life. A few typical cases are listed as follows: Case 1: DING, female, 51 years old, diagnosed as human papillomavirus (HPV) positive by the hospital. 2 ml of the gel (Example 5) prepared from the traditional Chinese medicine composition of the present invention, containing Zanthoxylum bungeanum, Artemisia argyi and the like, was placed intravaginally, 3 times per day for 60 days, the result of human papillomavirus (HPV) turned negative, and the patient was cured.
Case 2: SUN, female, 34 years old, diagnosed as vulvitis by the hospital. 2 ml of the gel (Example 4) prepared from the traditional Chinese medicine composition of the present invention, containing Zanthoxylum bungeanum, Artemisia argyi and the like, was placed intravaginally, 2 times per day for 15 days, various symptoms of vulvitis were eliminated, and the patient was cured.
Case 3: ZHANG, female, 32 years old, diagnosed as vaginitis by the hospital. 2 ml of the gel (Example 3) prepared from the traditional Chinese medicine composition of the present invention, containing Zanthoxylum bungeanum, Artemisia argyi and the like, was placed intravaginally, 2 times per day for 10 days, various symptoms of vaginitis were eliminated, and the patient was cured.

Case 4: WANG, male, 29 years old, diagnosed as skin eczema by the hospital. The gel (Example 2) prepared from the traditional Chinese medicine composition of the present invention containing Zanthoxylum bungeanum, Artemisia argyi and the like was administered via external application 3 times per 5 day for 10 days, various symptoms of skin eczema were eliminated, and the patient was cured.
The examples described above are only intended to describe the preferred embodiments of the present invention, without limiting the scope of the present invention. All variations and modifications made to the technical solutions of the 10 present invention by an ordinary person skilled in the art without departing from the design principle shall fall within the scope defined by the claims of the present invention.

  1. Claims
    The claims defining the invention are as follows:
    1. A pharmaceutical composition against human papillomavirus and gynecological inflammations, comprising the following components by weight: 20-60 parts of
    5 Zanthoxylum bungeanum, 20-60 parts of Artemisia argyi, 6-10 parts of Dendranthema indicum, 6-10 parts of Aloe vera and 6-10 parts of Campsis grandiflora.
  2. 2. The pharmaceutical composition against human papillomavirus and gynecological inflammations according to claim 1, comprising the following
    10 components by weight: 30-50 parts of Zanthoxylum bungeanum, 30-50 parts of Artemisia argyi, 8-10 parts of Dendranthema indicum, 8-10 parts of Aloe vera and 8-10 parts of Campsis grandiflora.
  3. 3. The pharmaceutical composition against human papillomavirus and gynecological inflammations according to claim 2, comprising the following
    15 components by weight: 45 parts of Zanthoxylum bungeanum, 45 parts of Artemisia argyi, 9 parts of Dendranthema indicum, and 9 parts of Aloe vera, and 9 parts of Campsis grandiflora.
  4. 4. A gel preparation against human papillomavirus and gynecological inflammations, prepared from the pharmaceutical composition according to any

  5. 5. A method for preparing a gel preparation against human papillomavirus and gynecological inflammations, comprising the steps of:
    a. decocting the pharmaceutical composition according to any one of claims 1 to 3 with water to obtain an aqueous extract;

    b. filtering the aqueous extract, adding ethanol to a filtrate, wherein the weight of ethanol added is 1-3 times the weight of the filtrate, subjecting a mixed solution of the filtrate and ethanol to alcohol precipitation at 2-6°C for 12-24 h, then performing rotary evaporation for removing ethanol, and concentrating to a paste;
    5 and
    c. adding carbomer, glycerin, chitosan and distilled water to the paste, placing at room temperature for 12-24 h, adjusting pH to 3.9-4.5 with triethanolamine, and stirring uniformly.
  6. 6. The method for preparing a gel preparation against human papillomavirus and
    10 gynecological inflammations according to claim 5, wherein in step a, the pharmaceutical composition is immersed in an aqueous solution of acetic acid having a weight 8-10 times the weight of the pharmaceutical composition for 20-30 minutes, then is decocted with water 2-3 times for 50-60 minutes each time, and filtered, and filtrates are combined, wherein the pH value of the aqueous solution
    15 of acetic acid is 2.5-3.5.

The method for preparing a gel preparation against human papillomavirus and gynecological inflammations according to claim 6, wherein in step a, the pharmaceutical composition is immersed in an aqueous solution of acetic acid having a weight 10 times the weight of the pharmaceutical composition for 30
    20 minutes, then is decocted with water 3 times for 60 minutes each time, and filtered, and filtrates are combined, wherein the pH value of the aqueous solution of acetic acid is 3.
  8. 8. The method for preparing a gel preparation against human papillomavirus and gynecological inflammations according to claim 5, characterized in that: in step b,

    5 the gel preparation according to claim 4, in preparation of a medicament against human papillomavirus, dermatoses or gynecological inflammations.
