Die Erfindung betrifft ein Verfahren zur Herstellung neuer, 1'-substituierter 17a, 21- (1' -Alkoxy) - - methylendioxysteroide der Pregnanreihe mit der nachstehenden Struktur an den 17, 20-und 21-Stellungen :

Die neuen Verbindungen werden erfindungsgemäss erhalten, indem man das entsprechende, freie 17a, 21-Dihydroxysteroid mit einem Orthoester der Formel : R-C (OR , (H) in der Rund Rl die oben genannte Bedeutung haben, in einem organischen Lösungsmittel bei 60 -1300 C und in Gegenwart eines sauren Katalysators umsetzt und dann für den Fall, dass im Steroidmolekül basisch hydrolysierbare Gruppen aufscheinen, diese gegebenenfalls in alkalischem Medium hydrolysiert.
Zur Durchführung des erfindungsgemässen Verfahrens ist es zweckmässig, in einem Lösungsmittel mit Siedepunkt über 600 C zu arbeiten. Dimethylformamid ist das bevorzugte Lösungsmittel ; man kann aber auch einen Kohlenwasserstoff, wie Benzol und dessen Homologe, Hexan, Isooctan oder auch ein halogenhaltiges Lösungsmittel, wie Chloroform, Tetrachlorkohlenstoff u. ähnl. verwenden.
Der verwendete Katalysator ist von saurer Natur, insbesondere eine aromatische Sulfonsäure, wie

Toluol-, Benzol- oder Naphthalinsulfonsäure oder eine Lewissäure, wie SnCI oderSbCl. Man kann auch Salze von organischen Basen mit starken Säuren, wie Pyridinchlorhydrat oder Pyridintosylat, verwenden.
Der für die Reaktion gewählte Orthoester der Formel 11 ist vorzugsweise der Methyl- oder Äthylorthoester einer Orthocarbonsäure, wie z. B. Orthoessigsäure, Orthopropionsäure, Orthobuttersäure, Ortho-iso-buttersäure, Orthovaleriansäure, Ortho-iso-valeriansäure, Orthocapronsäure, Orthoönanthsäure, Orthocaprilsäure, Orthopelargonsäure, Orthocaprinsäure, Cyclobutanorthocarbonsäure, Cyclo- pentanorthocarbonsäure, Orthohexahydrobenzoesäure, Orthobenzoesäure, Ortho-ss-phenylpropionsäure, Ortho-ss-carbomethoxypropionsäure oder Ortho-ss-carbäthoxypropionsäure. Diese Verbindungen sind bekannt ; sie können nach der Methode von Pinner (Ber.
Bd. 16 ; 1883, S. 1643) aus einem Nitril der Formel R-CN durch Umsetzung mit dem Alkohol R'OH (R und R haben die oben genannte Bedeutung) über das entsprechende Imidoätherchlorhydrat leicht hergestellt werden.
Das erfindungsgemässe Verfahren ist ganz allgemein brauchbar und kann auf irgendein na, 21-Dihydroxy-20-ketopregnan angewendet werden. Im allgemeinen enthalten die Ausgangsverbindungen in 3-Stellung eine oxygenierte Funktion, insbesondere eine Ketogruppe, eine Hydroxygruppe, eine niedrige Acyloxygruppe oder eine niedrige Alkoxygruppe. Andere Substituenten, wie die Hydroxy-, Keto-, Halogen-, Äther- oder Methylgruppen können am Cyclopentanopolyhydrophenanthrengerüst, z. B. an den 1-, 2-, 3-, 4-, 6-, 7-, 9-, 11- und/oder 16-Stellungen haften ; bevorzugte Verbindungen enthalten eine Sauerstoffunktion sowohl in 3- als auch in 11-Stellung und gegebenenfalls ein Halogenatom, vorzugsweise Fluor, in 9-Stellung.
Die Ausgangsverbindungen (17a, 21-Dihydroxy-20-ketopregnane) können gesättigt sein, oder können eine oder mehrere Doppelbindungen, beispielsweise in den 1 : 2-.

Zur Durchführung der Reaktion werden das Ausgangs-17a, 21-diol, der Orthocarbonsäureester und

der Katalysator in dem gewählten Lösungsmittel in einem mit absteigendem Kühler versehenen Gefäss gelöst. Im allgemeinen ist die Umsetzung nach l-bis 4stündigem Erhitzen vollständig. Die Lösung wird durch Zugabe von geringen Mengen eines basisch wirkenden Mittels, beispielsweise Pyridin, abgestumpft und das Endprodukt wird aus der Mischung üblicherweise, z. B. durch Eindampfen im Vakuum und Umkristallisieren aus einem geeigneten Lösungsmittel oder durch Chromatographie an einem geeigneten Absorptionsmittel, abgetrennt.
Unter diesen Arbeitsbedingungen verläuft die Umsetzung praktisch quantitativ zur Bildung der l'-substituierten 17cl, 21- (1'-Alkoxy)-methylendioxy-verbindungen. Insbesondere wird die Enolverätherung einer gegebenenfalls vorhandenen #4-3-Ketogruppierung vermiden und Enoläther bilden sich nicht einmal in papierchromatographisch nachweisbaren Spuren. Auch erfolgt, wenn eine 110-Hydroxygruppe vorhanden ist, keine Dehydratisierung in 9 (11)-Stellung.
Die so erhaltenen l'-substituierten 17a, 21- (1'-Alkoxy)-methylendioxysteroide sind in alkalischem Medium beständig. Liegt im Steroidmolekül eine in alkalischem Medium hydrolysierbare Gruppe, wie eine Ester-Cyanhydringruppe vor, so kann man das Endprodukt unter alkalischen Bedingungen hydrolysieren, ohne die substituierte Methylendioxybindung zu zerstören. So, wenn man beispielsweise einen Orthoester der Formel I, worin R eine ss-Carbalkoxy-äthylgruppe bedeutet, am Ende des Verfahrens erhält, so kann man die Verbindung in Gegenwart eines alkalischen Mittels hydrolysieren, um die ss-Carbalkoxyäthylgruppe in die entsprechende ss-Carboxyäthylgruppe zu überführen.
Obwohl die neuen l'-substituierten 17a, 21- (1'-Alkoxy)-äthylendioxy-steroide durch die blockierte Dihydroxyacetonseitenkette sich für zahlreiche Umwandlungen, z. B. für die Herstellung von 2-Methyl- oder 21-Methylderivaten eignen, sind die erfindungsgemäss erhältlichen Verbindungen überhaupt wertvoll, weil überraschenderweise gefunden wurde, dass sie in die bisher nicht zugänglichen 17-Monoester der entsprechenden 17a, 21-Dihydroxysteroide übergeführt werden können.
Ferner sind einige der erfindungsgemäss herstellbaren Orthoester hormonal wirksame Verbindungen, die eine besonders starke entzündungshemmende Wirksamkeit besitzen. Insbesondere weisen die 17cl, 21- - cyclischen Orthoester des Cortisons, Cortisols, Prednisons und Prednisolons mit von 4 bis 7 Kohlenstoffatome enthaltenden Fettsäuren eine lokale, entzündungshemmende Wirkung auf, die vielfach höher als die der entsprechenden Hormone ist. Zum Beispiel ist das 17a, 21- (1'-Methoxy)-n-pentylidendioxy- - A'*-pregnadien-llss-ol-3, 20-dion mindestens 70mal wirksamer als das Prednisolon.
Die folgenden Beispiele erläutern die Erfindung.
Beispiel 1: Eine Mischuhung von 1,5 g Prednison, 4 mg p-Toluosulfonsäure und 600 cm3 Benzol wird einige Minuten destilliert, um gegebenenfalls vorhandene Feuchtigkeitsspuren zu entfernen, dann wird sie mit 2 cams Orthovaleriansäuremethylester, der nach L. G. S. Brooker (Journal of the American Chemical Society, Bd. 57 [1935], S. 2480) hergestellt wurde, behandelt und die Destillation fortgesetzt, bis das Volumen der Lösung etwa 150 cms beträgt. Man fügt einige Tropfen Pyridin hinzu und dampft im Vakuum ein. Der aus Methanol umkristallisierte Rückstand liefert das 17ce, 21- (1'-Methoxy) - -n-pentylidendioxy-#1,4-pregnadien-3,11, 20-trion, F. 200-2030 C ; M = + 1140 (Dioxan).
Beispiel 2 : Von einer Mischung von 2 g Prednisolon, 10 mg Pyridinchlorhydrat, 2 cm3 Ortho- valeriansäuremethylester und 600 cms Isooctan werden in einen mit absteigendem Kühler versehenen Kolben 450 cm des Lösungsmittels abdestilliert, dann stumpft man mit einigen Tropfen Pyridin ab und dampft anschliessend im Vakuum ein. Der in Methanol aufgenommene Rückstand liefert das 17cl, 21- - (l'-Methoxy)-n-pentylidendioxy-A*-pregnadien-llss-ol-3, 20-dion, F. 157-159 C ; M =+64 (Dioxan).
Beispiel 3 : 2, 5 g Prednisolon und 10 mg Pyridintosylat werden in 2,5 cms Dimethylformamid gelöst. Der so erhaltenen Lösung fügt man 2 cms ss-Carbomethoxy-orthopropionsäuremethylester hinzu, der nach L. G. S. Brooker hergestellt wurde. Man erhitzt die Mischung ungefähr 3 h bei 1150 C, dann

gekocht. Man entfernt im Vakuum einen Teil des Lösungsmittels, dann nimmt man die Mischung mit Wasser auf und neutralisiert das überschüssige Carbonat mit 6 cms lobbiger Essigsäure. Die Lösung wird in Eiswasser gegossen, mit 10 cms piger Salzsäure angesäuert und mit Methylenchlorid ausgezogen.
Nach Eindampfen des Lösungsmittels entsteht ein öliger Rückstand, der, nach Aufnehmen in Methanol, schnell kristallisiert. So erhält man 1, 3 g 17 < x, 21- (l'-Methoxy-3'-carboxy)-propylidendioxy-A -pre-

gnadien-11ss-ol-3, 20-dion, F. 160 - 1620C ; [aHV = +600 (Dioxan).
Beispiel 4: 3 g Hydrocortison, 10mg p-Toluolsulfonsäure, 3 cms Dimethylformamid und 3 cm Orthovaleriansäurepropylester, der durch Umsetzung von Valeronitril mit n-Propanol und Behandlung des entstehenden Propylimidoäthers mit n-Propanol hergestellt wurde, werden in einem mit absteigendem Kühler versehenen Kolben auf 1100 C erhitzt. Nach 4 h wird die Lösung durch Zugabe einiger Tropfen Pyridin abgestumpft und dann im Vakuum eingedampft. Der in Methanol aufgenommene Rückstand liefert 17cl, 21-(1'-Propoxy)-n-pentylidendioxy-#4-pregnen-11ss-ol-3, 20-dion, F. 200-205 C ; M = = +102 (Dioxan).
Beispiel 5: In einem mit absteigendem Kühler versehenen Kolben setzt man eine Mischung von 1, 5 g Prednison, 3 mg p-Toluolsulfonsäure und 600 cm* wasserfreiem Benzol ein, und man fügt tropfenweise eine Lösung von 1, 2 g ss-Carboxy-orthopropionsäuremethylester in 50 cm3 Benzol hinzu. Man destilliert etwa 400 cms Lösungsmittel ab, dann stumpft man mit einigen Tropfen Pyridin ab und dampft im Vakuum ein. Der in wenig Methanol aufgenommene Rückstand liefert 17ct, 21- (l'-Methoxy-3'-carb- oxy)-propylidendioxy-#1,4-pregnadien-3, 11, 20-trion; F. 132-135 C; [α]D20=+95 (Dioxan). In gleicher Weise erhält man durch Umsetzung von Prednison mit Orthobenzoesäureäthylester, der nach L. G. S.
Brooker hergestellt wurde, in Benzol 17α,21-(1'-Äthoxy)-benzylidendioxy-#1,4 -pregnadien- - 3, 11, 20-trion.
Beispiel 6 : Eine Mischung von 2 g Cortison, 10 mg Sulfosalicylsäure, 20 cm* Orthoessigsäuremethylester und 600 cm* Isooctan werden in einen mit absteigendem Kühler versehenen Kolben gebracht. Man destilliert etwa 450 cm* des Lösungsmittels ab, stumpft dann mit einigen Tropfen Trimethylamin ab ; anschliessend dampft man das Lösungsmittel im Vakuum ab. Der in Methanol aufgenommene Rückstand liefert 17a, 21-(1'-Methoxy)-äthylidendioxy-#4-pregnen-3,11,20-trion, F. 210 - 2140 Ci Mp = + 1500 (Dioxan).
Auf gleiche Weise erhält man durch Umsetzung von Cortison mit Orthophenylpropionsäuremethylester, der aus Phenylpropionitril durch Umsetzung mit Methanol und nachfolgendes Behandeln des so erhaltenen Methylimidoäthers mit Methanol hergestellt wurde, in Isooctan 17ce, 21- - (l'-Methoxy-3'-phenyl)-propylidendioxy-A -pregnen-3, 11, 20-trion.
Beispiel 7 : Eine Mischung von 1, 5 g A*-Pregnen-17a, 21-diol-3, 20-dion, 4 mg p-Toluolsulfonsäure und 600 cms Toluol wird einige Minuten destilliert, um gegebenenfalls vorhandene Feuchtigkeitsspuren zu entfernen, dann wird sie mit 2 cm3 Orthocapronsäuremethylester, der nach L. G. S. Brooker hergestellt wurde, behandelt und die Destillation wird fortgesetzt, bis das Volumen der Lösung etwa 150 cm* beträgt. Man fügt einige Tropfen Pyridin hinzu und dampft im Vakuum ein. Auf gleiche Weise erhält man durch Umsetzung von .'" -Pregnen-17a, 21-diol-3,20-dion mit Orthohexahydrobenzoesäuremethylester, der aus Hexahydrobenzonitril durch Umsetzung mit Methanol und nachfolgendes Behandeln des so erhaltenen Methylimidoäthers mit Methanol hergestellt wurde, in Toluol 17a, 21- (1'-Methoxy-1'-cyclohexyl)-me- thylendioxy-#4-pregnen-3, 20-dion.
Beispiel 8 : Eine Mischung von 0,5 g Triamcinolon, 2 mg p-Toluolsulfonsäure, 3 cm3 Dimethylformamid und 2 cm3 Orthovaleriansäuremethylester wird 4-5 h bei 110 - 1300 C erhitzt. Nach Neutralisierung mit Pyridin und völliger Abdampfung des Lösungsmittels im Vakuum wird der ölige Rückstand an 15 g synthetischem Magnesiumsilikat (bekannt unter dem Handelsnamen"Florisil") chro-
