Verfahren zur Herstellung von N- (2, 3-Dimethylphenyl)-anthranilsäure und deren Salzen
Die Erfindung betrifft ein Verfahren zur Herstellung einer N-substituierten Anthranilsäure und ihren Salzen. Im besondern betrifft die Erfindung ein Verfahren zur Herstellung von N- (2, 3-Dimethylphenyl)- - anthranilsäure und ihren Salzen durch selektive Decarboxylierung von neuen Polycarbonsäuren der Formel

worin a und b 0 oder 1 bedeuten, wobei mindestens eines der beiden Symbole a und b 1 ist, oder eines Monocarboxylat-Salzes davon.
Erfindungsgemäss wird die selektive Decarboxylierung durch Erhitzen der Polycarbonsäure oder eines Monocarboxylat-Salzes davon durchgeführt. Die Reaktion kann bei einer Temperatur zwischen etwa 180 und 2500C durchgeführt werden. Temperaturen über 2500C können angewendet werden, sind aber im allgemeinen nicht notwendig oder erwünscht. Die bevorzugte Temperatur für die Reaktion liegt zwischen etwa 200 und 2400C. Zweckmässig wird die Reaktion in einem inerten organischen Lösungsmittel mit hohem Siedepunkt (2000C oder darüber) durchgeführt.
Dies ist insbesondere in solchen Fällen empfehlenswert, in denen ein Monocarboxylat-Salz der Polycarbonsäure als Ausgangsmaterial verwendet wird, weil solche Salze oberhalb der Reaktionstemperatur zum Schmelzen neigen und daher beim Fehlen eines das Salz wenigstens teilweise lösenden Lösungsmittels, nicht leicht zerfallen. Einige Beispiele für geeignete hochsiedende inerte Lösungsmittel sind tertiäre organische Amine, wie Chinolin, Chinaldin, N, N - Dimethyl- anilin und N, N-Diäthylanilin, Kohlenwasserstoffe, wie Diphenyl und Erdöl, und Äther, wie Diphenyl- äther und Triäthylenglykoldimethyläther. Die bevorzugten Lösungsmittel für die Reaktion sind die hochsiedenden tertiären Amine.
Die Reaktion kann in Gegenwart oder in Abwesenheit eines Kupfer enthaltenden Katalysators durchgeführt werden, wobei die Verwendung eines Katalysators bevorzugt wird. Einige Beispiele von geeigneten Kupfer enthaltenden Katalysatoren sind Kupferstaub, Kupferoxyd und Kupfersalze, wie Kupfersulfat und Kupferacetat. Die Reaktionsdauer variiert mit der Temperatur und den übrigen Bedingungen, aber im allgemeinen ist die Reaktion innerhalb einer Dauer von 10 min"2h beendet.
Die bevorzugten Ausgangsmaterialien für das Verfahren sind N- (2', 3'-Dimethylphenyl)-2-aminoisophthalsäure und ihre Monoalkalicarboxylat-Salze.
Die N- (2. 3-Dimethylphenyl) -anthranilsäure kann auf verschiedenen Wegen aus der Reaktionsmischung isoliert werden, u. zw. entweder als freie Säure oder als Salz. Eine der bequemsten Methoden bei Verwendung eines tertiären Amins als Lösungsmittel besteht darin, dass die Reaktions-

N-(2,3-Dimethylphenyl)-anthranilsäure und ihre pharmazeutisch anwendbaren Salze sind als schmerzstillende Mittel und zur Linderung von rheumatischen, arthritischen und andern Entzündungszuständen von Wert. Diese Stoffe sind bei oraler Verabreichung wirksam.
Die Erfindung wird durch die folgenden Beispiele erläutert.
Beispiel 1 : EineMischung aus 1 gN- (2', 3'-Dimethylphenyl)-2-aminoisophthalsäure, 5 ml Chi- nolin und einigen mg Cupriacetat wird mittels eines Ölbades 10 min auf eine Temperatur zwischen 215 und 224 C erhitzt, während welcher Zeit Kohlendioxyd entwickelt wird. Die Mischung wird anschliessend gekühlt und mit 100 ml n-Salzsäure verdünnt. Die saure wässerige Lösung wird von dem gummiartigen Niederschlag abgetrennt, und der Niederschlag wird in n-Natriumhydroxydlösung gelöst. Die alkalische Lösung wird mit Äther extrahiert, der Ätherextrakt wird verworfen und die wässerige Lösung angesäuert.
'Die ausgefällte N- (2, 3-Dimethylphenyl) -anthranilsäure wird gesammelt und aus Methanol umkristalli- siert Fp 229 - 2300C (df.).
Beispiel 2 : 2 g des Mononatriumsalzes von N-(2',3'-Dimethylphenyl)-2-aminoisophthalsäure, 10 ml Chinolin und einige mg Cupriacetat werden mittels eines Ölbades etwa 2 h auf eine Temperatur zwischen 170 und 2400C erhitzt. Die Reaktionsmischung wird gekühlt und mit 100 ml n-Salzsäure verdünnt. Dann wird die saure wässerige Lösung von dem gummiartigen Niederschlag getrennt. Der Niederschlag wird in n-Natriumhydroxydlösung gelöst, die Lösung mit Äther extrahiert und der Ätherextrakt verworfen. Die alkalische Lösung wird mit verdünnter Salzsäure angesäuert und die ausgefällte N- (2, 3-Di- methylphenyl)-anthranilsäure gesammelt und aus Methanol umkristallisiert. Fp 229-2300C (eff. ).
Das Natriumsalz der N- (2, 3-Dimethylphenyl) -anthranilsäure kann dadurch bereitet werden, dass man die freie Säure in Äthanol löst, ein Äquivalent Natriumhydroxyd dazugibt und die Mischung im Vakuum zur Trockne verdampft.
Die im erfindungsgemässen Verfahren als Ausgangsmaterialien verwendeten neuen Polycarbonsäuren können durch Kondensieren eines Salzes der entsprechenden m-carboxylierten o-Halogenbenzoesäure mit 2, 3-Dimethylanilin und Ansäuern des Reaktionsproduktes erhalten werden. Die Monocarboxylat-Salze der Polycarbonsäuren können durch Umsetzung der Polycarbonsäure mit einem Äquivalent einer Base bereitet werden. Diese Arbeitsweisen sind in der folgenden Beschreibung für die Herstellung der in den Beispielen l und 2 verwendeten Ausgangsmaterialien erläutert.

According to the invention, the selective decarboxylation is carried out by heating the polycarboxylic acid or a monocarboxylate salt thereof. The reaction can be carried out in the presence or in the absence of a copper-containing catalyst, the use of a catalyst being preferred. Some examples of suitable copper-containing catalysts are copper dust, copper oxide and copper salts such as copper sulfate and copper acetate. The new polycarboxylic acids used as starting materials in the process according to the invention can be obtained by condensing a salt of the corresponding m-carboxylated o-halobenzoic acid with 2,3-dimethylaniline and acidifying the reaction product. The monocarboxylate salts of the polycarboxylic acids can be prepared by reacting the polycarboxylic acid with one equivalent of a base. These procedures are illustrated in the following description for the preparation of the starting materials used in Examples 1 and 2.
