Das das Ovarialhormon enthaltende Rohfiltrat wird vom Azeton befreit und die hinterbleibende Flüssigkeit erschöpfend mit Chloroform extrahiert. Der mit Wasser aufgenommene und filtrierte Extrakt wird weiter gereinigt, indem man ihn vorsichtig mit geeigneten Metallsalzen, Metalloxyden oder-hydro- xyden oder Metallkolloiden, wie z. B. mit neutraler Bleiazetatlösung oder mit dialysierter Eisenhydroxydlösung versetzt und den Überschuss mit Schwefelwasserstoff wieder entfernt. Hiebei erhält man eine verdünnte Lösung des Ovarialhormons. Will man die für die therapeutische Anwendung wichtigeren konzentrierten Hormonlösungen erhalten, so muss man die Lösung zum Sirup eindampfen und ihn in einem nicht wasserlöslichen Vehikel aufnehmen.
Beispiel 3. Rinderplazenta wird gemahlen und in der Buchnerpresse abgepresst ; der Presssaft wird, gegebenenfalls nach Verdünnen mit Wasser, mit der vierfachen Menge Alkohol versetzt und die auf 400 erwärmte Mischung einige Stunden gerührt. Die Flüssigkeit wird durch Zentrifugieren abgetrennt und der unlösliche Rückstand ein zweites-und drittesmal mit 80% igem Alkohol ei wärmt und ausgerührt.
Die vereinigten alkoholischen Flüssigkeiten enthalten das Ovarialhormon und werden nach den in den Beispielen 1 und 2 angegebenen Verfahren weiter verarbeitet.
Die festen Rückstände enthalten das Hypophysenvorderlappenhormon. Sie werden ebenfalls nach den in den Beispielen 1 und 2 angegebenen Verfahren weiter verarbeitet.
Beispiel 4. Die Vorderlappen frischer Rinderhypophysen werden, zweckmässig bei leicht alkalischer Reaktion, gemahlen und mit unschädlichen Salzlösungen ausgezogen. Die vereinigten Lösungen werden mit 80 Volumprozent Alkohol gefällt und die Fällung, wie in den Beispielen 1-3 angegeben, auf Hypophysenvorderlappenhormon verarbeitet.
Beispiel 5. Frisch abgepresste Milch von Wöchnerinnen in den ersten drei Tagen wird entfettet.
Die entfettete Lösung wird bei 400 stark eingeengt und mit konzentriertem Alkohol gefällt. Nach mehrtägigem Stehen wird die Fällung mit Wasser aufgenommen und das Ungelöste abzentrifugiert. Die wässrige Lösung enthält das Hypophysenvorderlappenhormon.

1. Verfahren zur Darstellung des Hypophysenvorderlappenhormons aus dem Harn schwangerer Frauen oder aus andern das Hormon enthaltenden menschlichen oder tierischen Körperflüssigkeiten oder Organextrakten, darin bestehend, dass man diese Flüssigkeiten, gegebenenfalls nach einer unschädlichen Konzentration und Reinigung, mit wasserlöslichen organischen Flüssigkeiten ausfällt und den erhaltenden Rohniederschlag einer weiteren Verarbeitung zwecks Reinigung und Anreicherung unterwirft.

The crude filtrate containing the ovarian hormone is freed from acetone and the remaining liquid is exhaustively extracted with chloroform. The extracted with water and filtered extract is further purified by carefully treating it with suitable metal salts, metal oxides or hydroxides or metal colloids, such as. B. with neutral lead acetate solution or with dialyzed iron hydroxide solution and the excess is removed with hydrogen sulfide. A dilute solution of the ovarian hormone is obtained. If the concentrated hormone solutions, which are more important for therapeutic use, are to be obtained, the solution must be evaporated into a syrup and absorbed in a water-insoluble vehicle.
Example 3. Bovine placenta is ground and pressed in the Buchner press; four times the amount of alcohol is added to the pressed juice, optionally after dilution with water, and the mixture, heated to 400, is stirred for a few hours. The liquid is separated off by centrifugation and the insoluble residue is warmed up a second and third time with 80% alcohol and stirred.
The combined alcoholic liquids contain the ovarian hormone and are processed further according to the methods given in Examples 1 and 2.
The solid residues contain the anterior pituitary hormone. They are also processed further according to the methods given in Examples 1 and 2.
Example 4. The anterior lobes of fresh bovine pituitary glands are ground, expediently in the event of a slightly alkaline reaction, and extracted with harmless saline solutions. The combined solutions are precipitated with 80 percent by volume of alcohol and the precipitate, as indicated in Examples 1-3, processed on anterior pituitary lobe hormone.
Example 5. Freshly squeezed milk from women who have recently given birth in the first three days is defatted.
The defatted solution is strongly concentrated at 400 and precipitated with concentrated alcohol. After standing for several days, the precipitate is taken up with water and the undissolved material is centrifuged off. The aqueous solution contains the anterior pituitary hormone.
PATENT CLAIMS:
1. A method for the preparation of the anterior pituitary hormone from the urine of pregnant women or from other human or animal body fluids or organ extracts containing the hormone, consisting in precipitating these fluids, optionally after a harmless concentration and purification, with water-soluble organic fluids and the raw precipitate obtained subjected to further processing for cleaning and enrichment.
