Abstract
La invención proporciona composiciones y métodos novedosos para mejorar la transformación de explantes de embrión extirpados de la semilla monocotiledónea, que puede incluir una o más etapas de preparación del explante, rehidratación del explante, inoculación de y cultivo en conjunto con la bacteria Rhizobiales, la inducción de brotes, la inducción de brotes prolongada o la regeneración de plantas o partes de la planta genéticamente modificadas. Los métodos proporcionados en el presente documento pueden incluir la transformación de al menos una célula vegetal del explante de embrión con un polinucleótido heterólogo mediante la inoculación del explante de embrión con una bacteria Rhizobiales que comprende el polinucleótido heterólogo. Los métodos proporcionados en el presente documento incluyen métodos para regenerar una planta o parte de la planta genéticamente modificada a partir de una célula vegetal o explante transformado o editado. Reivindicación 1: Un método para producir una planta monocotiledónea o parte de la planta genéticamente modificada que comprende: introducir una molécula polinucleotídica heteróloga en al menos una célula de un explante de embrión de semilla monocotiledónea que comprende tejido meristemático mediante la inoculación del explante de embrión con un medio de inoculación que comprende una bacteria Rhizobiales competente para transformar dicha al menos una célula con la molécula polinucleotídica heteróloga, cultivar en conjunto el explante de embrión con la bacteria Rhizobiales en contacto con un medio de cultivo en conjunto, cultivar el explante de embrión en contacto con un primer medio de inducción de brotes que comprende una primera auxina y una primera citoquinina, cultivar el explante de embrión en contacto con un segundo medio de inducción de brotes que comprende la primera auxina o una segunda auxina y la primera citoquinina o una segunda citoquinina, y regenerar la planta monocotiledónea o parte de la planta genéticamente modificada a partir de un explante de embrión.
