Abstract
Reivindicación 1: Un método para la cuantificación de un inhibidor polianiónico de la PCR en una muestra que comprende: a) Preparar una serie de diluciones de una muestra que comprende al menos cuatro miembros; b) Realizar adiciones a cada miembro de la serie de diluciones de una cantidad constante de un ADN molde distinguible del ADN de la célula hospedadora; c) Realizar un ensayo de PCR en cada miembro de la serie de diluciones y en la cantidad constante del ADN molde en ausencia de cualquier muestra; d) Generar una curva patrón del inhibidor polianiónico; e) Comparar los resultados del ensayo de PCR de la serie de diluciones con los resultados del ensayo de PCR de la cantidad constante del ADN molde en ausencia de cualquier muestra; y f) Identificar la concentración de inhibidor polianiónico de la PCR en la muestra. Reivindicación 9: Un método para retirar un inhibidor polianiónico de la PCR de una solución de tampón que comprende: a) preparar una solución de tampón de una especie tamponante ácida, una especie tamponante básica o una combinación de las mismas; b) poner en contacto la solución de tampón con una resina de modo mixto; y c) separar la solución de tampón de la impureza polianiónica, retirando de este modo la impureza polianiónica de la solución de tampón. Reivindicación 28: Un método para retirar una impureza polianiónica de tampón de una solución de proteínas que comprende: a) ajustar el pH de una solución de proteínas que contiene una impureza aniónica de tampón a un pH inferior al punto isoeléctrico de la proteína en no más de 4 unidades de pH; b) poner en contacto la solución de proteínas con un medio de intercambio aniónico; y c) separar la proteína de la impureza aniónica de tampón. Reivindicación 29: Un método para retirar una impureza polianiónica de tampón de una solución de proteínas que comprende: a) ajustar el pH de una solución de proteínas que contiene una impureza aniónica de tampón a un pH inferior al punto isoeléctrico de la proteína en no más de 4 unidades de pH; b) poner en contacto la solución de proteínas con una resina de modo mixto; y c) separar la proteína de la impureza aniónica de tampón. Reivindicación 33: Un método para la cuantificación de un inhibidor polianiónico de la PCR en una muestra que comprende: (a) poner en contacto una muestra que comprende un inhibidor polianiónico de la PCR con al menos una alícuota de un compuesto policatiónico; (b) añadir un colorante indicador polianiónico en una cantidad suficiente para detectar la forma libre del colorante; y (c) cuantificar el inhibidor polianiónico de la PCR basándose en la cantidad de compuesto policatiónico necesaria para detectar la forma libre del colorante indicador polianiónico. Reivindicación 34: Un método de retirada de una impureza polianiónica en una muestra que comprende: (a) poner en contacto un fluido que comprende una impureza polianiónica con un contraión policatiónico; y (b) separar el fluido de la impureza polianiónica que forma complejo con el contraión policatiónico, retirando de este modo la impureza polianiónica del fluido.
