Abstract

Un método de ensayo comercial con biomarcadores para el diagnostico de multiples enfermedades que es simple, rápido, economico y prometedor. Por medio de la presente, pueden detectarse biomarcadores basados en miARN en especímenes de heces humanas. El método de la presente permite amplificar el miARN directamente a partir de los especímenes de heces, sin que sea necesario extraer el miARN con antelacion. La expresion diferencial de un microARN específico en las heces de pacientes que padecen un cáncer colorrectal o un adenoma es indicativa de la posibilidad de usar el microARN fecal como un biomarcador potencial para la deteccion de las neoplasias colorrectales. El método de la presente tiene relevancia para el diagnostico, el pronostico y la terapia de los cánceres gastroenterologicos/colorrectales y de las enfermedades gastrointestinales adquiridas o hereditarias. Reivindicacion 1: Un método para aislar y amplificar uno o más microARN (miARN) a partir de una muestra biologica, que comprende los siguientes pasos: mezclar una muestra fecal con una solucion de una sal, agua libre de ARNasa o ambas para formar una suspension; centrifugar la suspension; separar el sobrenadante de la suspension centrifugada; medir la concentracion de ARN/miARN total en el sobrenadante usando un espectrofotometro; transcribir ADNc a partir del ARN total usando uno o más cebadores específicos para el miARN; y amplificar el ADNc transcripto usando un ensayo de reaccion en cadena de la polimerasa (PCR), con el fin de obtener uno o más miARN amplificados.
