Abstract
Las cepas de Sphingomonas tienen polisacáridos extracelulares (por ejemplo, gellan, diutan) que están unidos firmemente a la superficie de la célula. Esta union puede limitar la produccion de polisacáridos, al impedir la asimilacion de nutrientes en la célula, debido a los sitios limitados para la biosíntesis de polisacáridos en la superficie de la célula. Con mutaciones de supresion, se han inactivado dos genes para la biosíntesis de polisacáridos, denominados gelM y gelN en las cepas productoras de gellan, y dpsM y dpsN en la cepas productoras de diutan, y se ha demostrado que producen polisacáridos que no están unidos firmemente a la superficie de la célula, es decir, toman la forma de limo. Se demostro el carácter de limo a través de la capacidad de las células de centrifugarse y la ausencia de agrupamientos, lo que puede observarse bajo el microscopio o en suspensiones diluidas. Las mutantes productoras de limo de diutan presentaron una productividad algo mayor, y el diutan tuvo una reología significativamente mejorada.
