Abstract

Se describen compuestos que tienen una actividad inhibidora de la proteasa del VHC. En otra realización describe las composiciones farmacéuticas que comprenden a dichos compuestos así como también a los métodos para utilizarlos para tratar trastornos asociados con la proteasa del VHC. Reivindicación 1: Un compuesto, o enantiómero, estereoisómero, rotámero, tautómero, racemato o prodroga de dicho compuesto, o sal o solvato aceptable para uso farmacéutico de dicha prodroga, dicho compuesto tiene la fórmula general (1) donde: Y se selecciona del grupo formado por las siguientes porciones: alquilo, alquilarilo, heteroalquilo, aril-heteroarilo, alquil-heteroarilo, cicloalquilo, alquiloxi, alquil-ariloxi, ariloxi, heteroariloxi, heterocicloalquiloxi, cicloalquiloxi, alquilamino, arilamino, alquil-arilamino, arilamino, heteroarilamino, cicloalquilamino y heterocicloalquilamino, con la condición de que Y puede estar opcionalmente sustituida con X11 o X12; X11 es alquilo, alquenilo, alquinilo, cicloalquilo, cicloalquil-alquilo, heterociclilo, heterociclilalquilo, arilo, alquilarilo, arilalquilo, heteroarilo, alquilheteroarilo o heteroarilalquilo, con la condición de que X11 puede, adicionalmente, estar sustituido por X12. X12 es hidroxi, alcoxi, tio, alquiltio, ariltio, amino, alquilamino, arilamino, alquilsulfonilo, arilsulfonilo, alquilsulfonamido, arilsulfonamido, carboxilo, carbalcoxi (el término "carbalcoxi" significa éster), carboxamido (el término "carboxamido" significa amida), alcoxicarbonilamino, alcoxicarboniloxi, alquilureido, arilureido, halógeno, ciano, o nitro, con la condición de que dicho alquilo, alcoxi y arilo pueden, además, estar opcionalmente sustituidos con porciones seleccionadas de manera independiente, de X12; R1 se selecciona de las estructura del grupo de fórmulas (2) donde k es un número entre 0 y 5, que puede ser igual o diferente, R11 denota sustituyentes opcionales en el anillo, cada uno de dichos sustituyentes se selecciona, de manera independiente, del grupo formado por alquilo, alquenilo, alquinilo, arilo, cicloalquilo, alquil-arilo, heteroalquilo, heteroarilo, aril-heteroarilo, alquil-heteroarilo, alquiloxi, alquil-ariloxi, ariloxi, heteroariloxi, heterocicloalquiloxi, cicloalquiloxi, alquilamino, arilamino, alquil-arilamino, arilamino, heteroarilamino, cicloalquilamino, heterocicloalquilamino, hidroxi, tio, alquiltio, ariltio, amino, alquilsulfonilo, arilsulfonilo, alquilsulfonamido, arilsulfonamido, carboxilo, carbalcoxi, carboxamido, alcoxicarbonilamino, alcoxicarboniloxi, alquilureido, arilureido, halógeno, ciano, y nitro, con la condición de que R11 cuando R11 es distinto de H puede estar, además, opcionalmente sustituido con X11 o X12; Z se selecciona entre O, N, CH o CR; W puede estar presente o ausente y, si W está presente, W se selecciona entre C=O, C=S, C(=N-CN), o SO2; Q puede estar presente o ausente, y cuando W está presente, Q es CH, N, P, (CH2)p, (CHR)p, (CRR')p, O, NR, S, o SO2; y cuando Q está ausente, M puede estar presente o ausente; cuando Q y M están ausentes, A está directamente unido a L; A es O, CH2, (CHR)p, (CHR-CHR')p, (CRR')p, NR, S, SO2 o un enlace; E es CH, N; CR o un enlace doble hacia A, L o G; G puede estar presente o ausente, y cuando G está presente, G es (CH2)p, (CHR)p, o (CRR')p, y cuando G está ausente, J está presente y E está directamente conectado con el átomo de C en la fórmula (1) tal como está unido G; J puede estar presente o ausente, y cuando J está presente, J es (CH2)p, (CHR)p, o (CRR')p, SO2, NH, NR u O; y cuando J está ausente, G está presente y E está directamente enlazado a N, como se ilustra en la fórmula (1) tal como está unido J; L puede estar presente o ausente, y cuando L está presente, L es CH, CR, O, S o NR; y cuando L está ausente, entonces M puede estar presente o ausente; y si M está presente y L ausente, entonces, M está enlazado de manera directa e independiente a E; y J está enlazado de manera directa e independiente a E; M puede estar presente o ausente, y cuando M está presente, M es O, NR, S, SO2, (CH2)p, (CHR)p, (CHR-CHR')p, o (CRR')p, p es un número entre 0 y 6; y R, R', R2, R3, y R4 se seleccionan, de manera independiente, del grupo formado por H, alquilo C1-10, alquenilo C2-10, cicloalquilo C3-8, heterocicloalquilo C3-8, alcoxi, ariloxi, alquiltio, ariltio, amino, amido, éster, ácido carboxílico, carbamato, urea, cetona, aldehído, ciano, nitro, halógeno; (cicloalquil)alquilo, y (heterocicloalquil) alquilo, donde dicho cicloalquilo está formado por tres a ocho átomos de C, y entre 0 y 6 átomos de O, N, S o P, y dicho alquilo tiene de 1 a 6 átomos de C; arilo, heteroarilo, alquil-arilo y alquil-heteroarilo; donde cualquiera de las porciones alquilo, heteroalquilo, alquenilo, heteroalquenilo, arilo, heteroarilo, cicloalquilo y heterocicloalquilo anteriormente mencionadas puede estar sustituida, de manera independiente y opcional; dicho término "sustituida" se refiere a una sustitución opcional con una o más porciones seleccionadas del grupo formado por alquilo, alquenilo, alquinilo, arilo, aralquilo, cicloalquilo, heterociclico, halógeno, hidroxi, tio, alcoxi, ariloxi, alquiltio, ariltio, amino, amido, éster, ácido carboxílico, carbamato, urea, cetona, aldehído, ciano, nitro, sulfonamido, sulfóxido, sulfona, sulfonil urea, hidracida e hidroxamato; además, donde dicha unidad N-C-G-L-J-N representa una estructura de anillo cíclico de 5 o 6 miembros con la condición de que cuando dicha unida N-C-G-E-L-J-N representa una estructura de anillo cíclico de 5 miembros, o cuando la estructura de anillo bicíclico en la fórmula (1) que comprende N, C, G, E, L, J, N, A, Q y M representan una estructura de anilo cíclico de 5 miembros, entonces, dicha estructura de anillo cíclico de 5 miembros carece de un grupo carbonilo formando parte del anillo cíclico.
