Abstract

Novi biopostupak za izradu 7-aminodesacetoksi cefalosporanske kiseline (7-ADCK) koji se sastoji od stupnjeva 1) održavanja u podlozi za gajenje koja je sposobna da podržava njegov rast, soj Penicillium chrysogenum-a koji proizvodi izopenicilin N i dodavanja u podlogu za gajenje adipatne sirovine koja se sastoji od adipinske kiseline, ili jedne ili više od njenih soli i estara koje su podesne da ih soj Penicillium chrysogenum asimiluje i koristi da proizvodi adipoil-6-amino penicilansku kiselinu (adipoil-6-APK), pri čemu se proizvodi adipoil-6-APK; u kome je soj Penicillium-chrysogenum-a bio transformisan pomoću DNK koja kodira aktivnost encima ekspandaze koji je sposoban da primi adipoil-6-APK kao supstrat, posle čega se kao rezultat njegove ekspresije, adipoil-6-APK proizveden pomoću soja isto tako posle toga in situ širi u prstenu da se obrazuje adipoil-7-ADCK; i 2) dovođenja u dodir adipoil-7-ADCK sa adipoil acilazom pri čemu se adipoil bočni lanac odstranjuje i stvara se 7-ADCK proizvod i potom se proizvod izoluje. - Prijava sadrži još 3 zavisna zahteva.A new bio-process for the manufacture of 7-aminodesacetoxy cephalosporanic acid (7-ADCK) consisting of stage 1) maintenance in a growth medium capable of supporting its growth, a strain of Penicillium chrysogenum producing isopenicillin N and adding to the adipate culture medium a raw material consisting of adipic acid, or one or more of its salts and esters, suitable for the assimilation and use of Penicillium chrysogenum to produce adipoyl-6-amino penicillanic acid (adipoyl-6-APK), thereby producing adipoyl -6-APK; in which a strain of Penicillium-chrysogenum was transformed by DNA encoding the activity of an expandase enzyme capable of receiving adipoyl-6-APK as a substrate, after which, as a result of its expression, adipoyl-6-APK was produced by the strain of this in situ propagates in the ring to form adipoyl-7-ADCK; and 2) contacting adipoyl-7-ADCK with adipoyl acylase whereby the adipoyl side chain is removed and a 7-ADCK product is formed and the product is then isolated. - The application contains 3 more dependent requests.
