Abstract

본 발명은 파수딜과 젤라틴을 이용한 줄기세포 유래 간세포 분화 방법 등에 관한 것으로, 본 발명에 따른 줄기세포 유래 간세포 분화 방법은 줄기세포의 간세포로의 분화 초기 단계에서 파수딜로 줄기세포의 내배엽으로의 분화를 유도하며, 배양용기의 고점도 젤라틴 코팅을 통해 지질방울 생성을 억제하고, 미토콘드리아의 기능을 활성화하며, 내배엽 유전자의 발현을 상향 조절하고, 외배엽 및 중배엽 유전자의 발현을 하향 조절함으로써 줄기세포의 간모세포로의 분화 효율을 향상시켰다. 또한, 분화 성숙(후기) 단계에서 배지에 저점도 젤라틴을 첨가함으로써 지질방울의 축적을 억제하고, 미토콘드리아를 활성화함으로써 간모세포의 간세포로의 분화 효율을 향상시키고, 분화된 간세포의 기능을 향상시킴으로써, 줄기세포의 간세포로의 분화 효율을 향상시켰다. 따라서, 이를 이용하여 수득한 간세포는 간질환의 치료용 세포 치료제, 이식체 등으로 유용하게 이용될 수 있을 것으로 기대된다.

Description

파수딜과 젤라틴을 이용한 줄기세포 유래 간세포 분화 방법
본 발명은 파수딜과 젤라틴을 이용한 줄기세포 유래 간세포 분화 방법 등에 관한 것이다.
본 발명은 2024년 6월 28일에 출원된 대한민국 특허출원 제10-2024-0085745호에 기초한 우선권을 주장하며, 상기 출원들의 명세서 및 도면에 개시된 모든 내용은 원용된다.
일반적으로 분화란 초기 단계의 세포가 각 조직으로서의 특성을 갖게 되는 과정을 말하는데, 그 대표적인 예는 동물의 발생 과정에서 볼 수 있다. 즉, 정자와 난자가 결합하여 만들어진 수정란이라는 하나의 세포가 뼈, 심장, 피부 등의 다양한 조직 세포로 만들어지기 위해서는 '분화'가 일어나야 하는 것이다. 이러한 분화능력을 가지고 있는 줄기세포(stem cell)는 조직을 구성하는 각 세포로 분화되기 전단계의 세포로서, 미분화 상태에서 무한 증식이 가능하며 특정 분화 자극에 의해 다양한 조직의 세포로 분화될 수 있는 잠재적 가능성을 가진 세포를 의미한다.
줄기세포 중 중간엽 줄기세포(mesenchymal stem cell; MSC)는 골수, 탯줄 기질(Umbilical cord matrix), 및 태반과 같은 인간 태아 및 성인 조직, 및 다양한 성인 조직에서 유래한다. 특히, 태아 유래 조직인 탯줄 기질과 태반은 통증 없는 수집 절차, 빠른 자가 재생, 3배엽으로 분화할 수 있는 능력과 같은 탁월한 장점으로 인해 인간 MSC의 훌륭한 공급원이다. 따라서, 최근에는 인간 탯줄 기질 유래 중간엽 줄기세포(human umbilical cord matrix-derived MSCs, hUCM-MSCs)와 이들의 분화세포가 다양한 질병을 치료하기 위한 치료의학에서 조직 재생에 활용되고 있다.
한편, 간은 인간 체중의 3% 정도를 차지하며, 중요한 기능을 수행하는 장기이다. 간을 이루는 핵심세포인 간세포는 글리코겐, 단백질, 지질, 핵산, 비타민류 등의 생합성 및 분해의 기능과 독성이 있는 물질을 해독하는 기능을 나타낸다. 그러나 이런 정상적인 간 기능을 갖지 못하는 간질환 환자의 대부분은 간 조직 손상으로 인해 간세포의 재생 능력이 감소함에 따른다. 현재 국내 간질환 사망자는 10만 명당 1만 천명에 이른다는 보고가 있다. 간질환의 경우, 조직적합성을 갖는 간 조직 이식이 현재까지 가장 효과적인 치료방법으로 알려져 있으나, 공여자를 찾기에 어려움이 따른다는 중대한 단점이 있다. 이에 대한 대안으로 현재 줄기세포에 대한 연구가 활발해지면서 줄기세포를 이용한 세포 치료에 많은 관심이 집중되고 있으나, 줄기세포에서 분화된 세포를 치료에 사용하는 데에는 제약이 있다.
줄기세포에서 분화된 세포를 사용하는 데 가장 큰 장애물 중 하나는 분화 프로토콜의 효율성과 성숙한 세포의 제한된 표현형으로, 예를 들어, 알려진 프로토콜을 이용하여 인간 중간엽 줄기세포로부터 분화시킨 간세포 유사 세포는 전사체 프로파일, 간 기능 및 대사 활동 측면에서 성인 세포보다 태아 간세포와 더 유사한 특성을 나타낸다는 문제점이 있다. 따라서, 이러한 문제점을 해결하기 위해, 유전자 변형, 미세 환경 조정, 사이토카인 및 성장 인자 추가를 이용한 접근법 등을 포함하여 다양한 분화 방법이 개발되고 있다.
한편, 줄기세포의 운명을 결정할 때 전사체 변화는 세포 유형의 분화와 밀접한 관련이 있다. 따라서, 최근에는 RNA 시퀀싱과 같은 시퀀싱 도구를 사용하여 분화된 세포의 전사를 세계적으로 널리 분석하고 있다. 그러나 세포의 운명은 전사 조절만 변경하는 것이 아니고, 세포 분화와 직계 속박(lineage commitment)은 핵과 다양한 생물학적 과정 사이의 통신과 세포질 거대분자 및 세포소기관 상호작용을 포함하는 신호 전달 경로에 의해 영향을 받는다. 특히 줄기세포가 분화할 때 대사의 변화가 동반되며, 이는 줄기세포의 운명 결정에 중요한 역할을 하는 것으로 알려져 있다. 세포 대사에서 미토콘드리아 역할은 세포의 운명과 기능을 결정하는 데 중추적인 역할을 한다. 또한 지질 및 에너지 항상성의 중심에 있는 저장 세포소기관과 관련된 지질방울(lipid droplet; LD)도 줄기세포의 운명 결정과 관련이 있는 것으로 보고되었다. 줄기세포의 세포소기관 상태는 줄기세포의 운명을 결정하는 데 중요하지만, 줄기세포의 세포소기관 상태의 조절이 간 분화에 영향을 미치는지 알려져 있지 않았다.
이러한 배경 하에 본 발명자들은 hUCM-MSC의 간세포로의 분화가 전사체 변화뿐만 아니라 세포소기관의 상태에 의해서도 영향을 받는 것을 확인하였으며, 줄기세포의 간세포로의 분화 초기 단계에서 내배엽으로 분화를 유도하는 파수딜을 고점도의 젤라틴이 코팅된 배양용기에서 사용할 경우, 지질방울의 생성을 억제하고, 미토콘드리아의 기능을 활성화함으로써 줄기세포의 간모세포로의 분화 효율을 향상시킬 수 있고, 분화 성숙(후기) 단계에서 배지에 저점도의 젤라틴을 첨가할 경우, 지질방울의 축적을 줄이고 미토콘드리아를 활성화하여 간모세포의 간세포로의 분화 효율과 분화된 간세포의 기능을 향상시킬 수 있는 것을 확인함으로써 본 발명을 완성하였다.
본 발명자들은 효율적인 줄기세포 유래 간세포 분화 방법을 개발하기 위해, 예의 연구한 결과, 분화 초기 단계에서 내배엽으로 분화를 유도하는 파수딜을 고점도의 젤라틴이 코팅된 배양용기에서 사용할 경우, 지질방울의 생성을 억제하고, 미토콘드리아의 기능을 활성화함으로써 줄기세포의 간모세포로의 분화 효율을 향상시킬 수 있고, 분화 성숙(후기) 단계에서 배지에 저점도의 젤라틴을 첨가할 경우, 지질방울의 축적을 줄이고 미토콘드리아를 활성화하여 간모세포의 간세포로의 분화 효율과 분화된 간세포의 기능을 향상시킬 수 있는 것을 확인함으로써 본 발명을 완성하였다.
따라서, 본 발명의 목적은 줄기세포를 젤라틴이 코팅된 배양용기에서 ROCK 억제제를 포함한 배지에 배양하여 간모세포(hepatoblast)로의 분화를 유도하는 단계를 포함하는, 줄기세포 유래 간세포 분화 방법을 제공하는 것이다.
본 발명의 다른 목적은 (a) ROCK 억제제를 유효성분으로 포함하는 제1조성물; 및 (b) 젤라틴을 유효성분으로 포함하는 제2조성물로 이루어진 군으로부터 선택되는 어느 하나 이상을 포함하는, 간세포 배양용 조성물로서, 상기 제1조성물은 젤라틴이 코팅된 배양용기에 첨가되는 것을 특징으로 하는, 간세포 배양용 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 본 발명에 따른 간세포 배양용 조성물, 및 설명서를 포함하는 간세포 배양용 키트를 제공하는 것이다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 본 발명이 속하는 기술 분야의 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
상기 목적을 달성하기 위하여, 본 발명은 줄기세포를 젤라틴이 코팅된 배양용기에서 ROCK 억제제를 포함한 배지에 배양하여 간모세포(hepatoblast)로의 분화를 유도하는 단계를 포함하는, 줄기세포 유래 간세포 분화 방법을 제공한다.
본 발명의 일 구현예로, 상기 줄기세포는 탯줄 기질, 제대혈, 골수, 지방, 근육, 신경, 피부, 양막, 및 태반으로 이루어진 군으로부터 선택되는 어느 1종 이상의 조직으로부터 유래된 중간엽 줄기세포일 수 있으나, 이에 제한되지 않는다.
본 발명의 다른 구현예로, 상기 ROCK 억제제는 파수딜 (Fasudil), 리파수딜 (Ripasudil), RKI-1447, Y-27632, GSK429286A, 및 Y-30141로 이루어진 군으로부터 선택되는 어느 하나 이상일 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 구현예로, 상기 ROCK 억제제는 1 내지 50 μM의 농도로 배지에 포함될 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 구현예로, 상기 젤라틴은 0.1 내지 10%의 농도로 배양용기에 코팅될 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 구현예로, 상기 간모세포로의 분화를 유도하는 단계는 1 내지 10일 동안 수행할 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 구현예로, 상기 간모세포로의 분화를 유도하는 단계는 하기 단계를 포함할 수 있으나, 이에 제한되지 않는다:
(a-1) 줄기세포를 (고점도 젤라틴이 코팅된 배양용기에서) ROCK 억제제를 포함하는 배지에 배양하여 간 내배엽으로의 분화를 유도하는 단계; 및
(a-2) 상기 간 내배엽을 성장 인자, 골 형성 단백질, 및 wnt 신호 활성자를 포함하는 배지에서 배양하여 간모세포로의 분화를 유도하는 단계,
단, 상기 (a-2) 단계는 젤라틴이 코팅되지 않은 배양용기에서 수행됨.
본 발명의 또 다른 구현예로, 상기 방법은 간모세포를 젤라틴을 포함하는 배지에서 배양하여 간세포로의 분화를 유도하는 단계를 더 포함할 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 구현예로, 상기 젤라틴은 0.1 mg/ml 내지 10 mg/ml의 농도로 배지에 포함될 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 구현예로, 상기 간세포로의 분화를 유도하는 단계는 하기 단계를 포함할 수 있으나, 이에 제한되지 않는다:
(b-1) 상기 간모세포를 온코스타틴 M을 포함하는 배지에서 배양하는 단계; 및
(b-2) 상기 온코스타틴 M을 포함하는 배지에 젤라틴을 첨가하고 추가적으로 배양하여 간세포로의 분화를 유도하는 단계.
또한, 본 발명은 (a) ROCK 억제제를 유효성분으로 포함하는 제1조성물; 및 (b) 젤라틴을 유효성분으로 포함하는 제2조성물로 이루어진 군으로부터 선택되는 어느 하나 이상을 포함하는, 간세포 배양용 조성물로서,
상기 제1조성물은 젤라틴이 코팅된 배양용기에 첨가되는 것을 특징으로 하는, 간세포 배양용 조성물을 제공한다.
본 발명의 일 구현예로, 상기 배양은 줄기세포의 간세포로의 분화를 유도하는 것일 수 있으나, 이에 제한되지 않는다.

본 발명의 일 구현예로, 상기 설명서는 본 발명에 따른 줄기세포 유래 간세포 분화 방법이 기재된 것일 수 있으나, 이에 제한되지 않는다.
본 발명에 따른 줄기세포 유래 간세포 분화 방법은 줄기세포의 간세포로의 분화 초기 단계에서 ROCK 억제제로 줄기세포의 내배엽으로의 분화를 유도하되 이때 발생되는 지질방울의 생성을 고점도 젤라틴이 코팅된 배양용기에서 세포를 배양함으로써 억제하고, 미토콘드리아의 기능을 활성화하며, 내배엽 유전자의 발현을 상향 조절하고, 외배엽 및 중배엽 유전자의 발현을 하향 조절함으로써 줄기세포의 간모세포로의 분화 효율을 향상시켰다. 또한, 분화 성숙(후기) 단계에서 배지에 저점도 젤라틴을 첨가함으로써 지질방울의 축적을 억제하고, 미토콘드리아를 활성화함으로써 간모세포의 간세포로의 분화 효율을 향상시키고, 분화된 간세포의 기능을 향상시킴으로써, 줄기세포의 간세포로의 분화 효율을 향상시켰다. 따라서, 이를 이용하여 수득한 간세포는 간질환의 치료용 세포 치료제, 이식체 등으로 유용하게 이용될 수 있을 것으로 기대된다.

도 4e는 젤라틴 코팅접시에서 파수딜을 처리해 분화시킨 hUCM-MSC에 0.1% 또는 1%의 젤라틴을 첨가하여 분화시킨 후 5일차에 ALB, CYP3A4 단백질의 발현을 확인한 웨스턴 블롯팅을 농도계 분석으로 확인한 결과를 나타낸 것이다.
도 4f는 젤라틴 코팅접시에서 파수딜을 처리해 분화시킨 hUCM-MSC에 0.1% 또는 1%의 젤라틴을 첨가하여 분화시킨 후 5일차에 간세포 특이적 miRNA(miR-122, miR-192)의 발현을 확인한 결과를 나타낸 것이다.
도 5a는 젤라틴 코팅접시에서 파수딜을 처리해 분화시킨 hUCM-MSC에 0.1% 또는 1%의 젤라틴을 첨가하여 분화시킨 후 5일차에 인간 알부민의 분비량을 측정한 결과를 나타낸 것이다 (undiff: 미분화 세포).
도 5b는 젤라틴 코팅접시에서 파수딜을 처리해 분화시킨 hUCM-MSC에 0.1% 또는 1%의 젤라틴을 첨가하여 분화시킨 후 5일차에 CYP3A4 활성을 측정한 결과를 나타낸 것이다.

도 6은 본 발명에 따른 줄기세포 유래 간세포 분화 방법을 개략적으로 나타낸 것이다.
본 발명자들은 줄기세포의 간세포로의 분화 과정 중 분화 초기 단계에서 내배엽으로 분화를 유도하는 파수딜을 고점도(1%)의 젤라틴이 코팅된 배양용기에서 사용할 경우, 내배엽으로의 분화 과정에서 발생하는 지질방울의 생성이 억제되고, 미토콘드리아의 기능이 활성화됨으로써 줄기세포의 간모세포로의 분화 효율이 향상되었음을 확인하였다. 또한, 분화 성숙(후기) 단계에서 배지에 저점도(0.1%)의 젤라틴을 첨가할 경우, 지질방울의 축적이 감소하고 미토콘드리아가 활성화되어 간모세포의 간세포로의 분화 효율과 분화된 간세포의 기능을 향상시킬 수 있는 것을 확인하였다.
이하, 본 발명을 상세히 설명한다.
본 발명은 줄기세포를 젤라틴이 코팅된 배양 용기에서 ROCK 억제제를 포함한 배지에 배양하여 간모세포(hepatoblast)로의 분화를 유도하는 단계를 포함하는, 줄기세포 유래 간세포 분화 방법을 제공한다.
본 발명에 있어서, 상기 줄기세포는 탯줄 기질, 제대혈, 골수, 지방, 근육, 신경, 피부, 양막, 및 태반으로 이루어진 군으로부터 선택되는 어느 1종 이상의 조직으로부터 유래된 중간엽 줄기세포일 수 있으며, 본 발명의 일 실시예에 따르면, 상기 줄기세포는 인간 탯줄 기질 중간엽 줄기세포일 수 있으나, 이에 제한되지 않는다.
본 발명에 있어서, 상기 ROCK 억제제는 파수딜 (Fasudil), 리파수딜 (Ripasudil), RKI-1447, Y-27632, GSK429286A, 및 Y-30141로 이루어진 군으로부터 선택되는 어느 하나 이상일 수 있으며, 본 발명의 일 실시예에 따르면, 파수딜일 수 있으나, 이에 제한되지 않는다.
상기 파수딜은 줄기세포의 생존력을 증가시킬 수 있을 뿐만 아니라 분화에 사용되는 일반적인 cytokines (단백질 형식)과는 다르게 저분자(Small molecule)로 분화에 효율적인 것을 특징으로 한다.
본 발명에서 사용되는 용어 "젤라틴"이란, 동물의 뼈, 가죽, 힘줄, 연골 등을 구성하는 천연 고분자 단백질인 콜라겐(교원질)을 가수분해하여 얻어지는 유도 단백질이다. 젤라틴의 일반적 구성 성분은 약 단백질(protein): 84 내지 90%, 수분(water): 8 내지 12% 및 무기염류(mineral salts): 2 내지 4%로 구성될 수 있다. 본 발명에서 젤라틴은 다양한 젤 강도(gel strength), 점도(viscosity)를 가질 수 있다. 본 발명에서 젤라틴은 천연물질로부터 분리 또는 합성될 수 있으며, 그 원료의 종류는 한정되지 않으며, 포유동물 또는 어류 등 의 가죽, 뼈 껍질 등으로부터 분리된 콜라겐을 가수분해하여 생산할 수 있다.

본 발명에 있어서, 상기 간모세포로의 분화를 유도하는 단계는 하기 단계를 포함하거나 하기 단계로 이루어진 것일 수 있다:
(a-1) 줄기세포를 (고점도 젤라틴이 코팅된 배양용기에서) ROCK 억제제를 포함하는 배지에 배양하여 간 내배엽으로의 분화를 유도하는 단계; 및
(a-2) 상기 간 내배엽을 성장 인자, 골 형성 단백질, 및 wnt 신호 활성자를 포함하는 배지에서 배양하여 간모세포로의 분화를 유도하는 단계,
단, 상기 (a-2) 단계는 젤라틴이 코팅되지 않은 배양용기에서 수행됨.
본 발명의 일 실시예에 따르면, 상기 (a-1) 단계의 ROCK 억제제를 포함하는 배지는 1 내지 20% 소태아혈청, 0.1 내지 5% 비필수 아미노산, 0.1 내지 5% 항생제-항진균제, 및 0.05 내지 5mM L-아스코르빈산이 보충된 DMEM/F12 배지로, 상기 ROCK 억제제는 1 내지 50 μM, 1 내지 40 μM, 1 내지 30 μM, 1 내지 20 μM, 1 내지 10 μM, 2 내지 40 μM, 2 내지 30 μM, 2 내지 20 μM, 2 내지 10 μM, 5 내지 30 μM, 5 내지 20 μM, 5 내지 10 μM, 10 내지 30 μM, 10 내지 20 μM, 1 μM, 2 μM , 3 μM , 4 μM , 5 μM , 6 μM , 7 μM , 8 μM , 9 μM , 또는 10 μM 의 농도로 배지에 포함될 수 있으나, 이에 제한되지 않는다.
본 발명의 일 실시예에 따르면, 상기 (a-1) 단계는 젤라틴이 코팅된 배양용기에서 1 내지 5일, 1 내지 4일, 1 내지 3일, 1 내지 2일, 2 내지 4일, 2 내지 3일, 3 내지 5일, 3 내지 4일, 1일, 2일, 또는 3일 동안 수행될 수 있으나, 이에 제한되지 않는다.
본 발명의 일 실시예에 따르면, 상기 (a-2) 단계의 성장 인자, 골 형성 단백질, 및 wnt 신호 활성자를 포함하는 배지는 0.01 내지 1% polyvinyl alcohol, 1 내지 50 mM nicotinamide, 1 내지 50 ng/ml hHGF, 0.1 내지 5% ITS, 0.1 내지 5% 페니실린/스트렙토마이신이 보충된 IMDM (Iscove's Modified Dulbecco's Medium, Gibco)일 수 있으나, 이에 제한되지 않는다.
본 발명에서 상기 "성장 인자"는 염기성 섬유아세포 성장 인자(basic fibroblast growth factor; bFGF, FGF2), 상피세포 성장 인자(epidermal growth factor; EGF), 및 인슐린-유사 성장 인자-1(Insulin-like growth factor-1; IGF-1)로 이루어진 군에서 선택된 1종 이상일 수 있으며, 본 발명의 일 실시예에 따르면, 상기 성장 인자는 FGF2(bFGF)일 수 있으나, 이에 제한되지 않는다.
본 발명에서 상기 "골 형성 단백질"은 BMP 패밀리 단백질에 속하는 한 모두 포함될 수 있으며, 예를 들어 BMP1, BMP2, BMP3, BMP4, BMP5, BMP6, BMP7, BMP8, BMP9, 및 BMP10로 이루어진 군에서 선택된 1종 이상일 수 있으며, 본 발명의 일 실시예에 따르면, 상기 골 형성 단백질은 BMP4일 수 있으나, 이에 제한되지 않는다.

본 발명에 있어서, 상기 방법은 간모세포를 젤라틴을 포함하는 배지에서 배양하여 간세포로의 분화를 유도하는 단계를 더 포함할 수 있으며, 상기 간세포로의 분화를 유도하는 단계는 하기 단계를 포함하거나 하기 단계로 이루어진 것일 수 있다:
(b-1) 상기 간모세포를 온코스타틴 M을 포함하는 배지에서 배양하는 단계; 및
(b-2) 상기 온코스타틴 M을 포함하는 배지에 젤라틴을 첨가하고 추가적으로 배양하여 간세포로의 분화를 유도하는 단계.
본 발명에서 용어, "온코스타틴 M(oncostatin M)"은 인터류킨-6(IL-6) 군에 속하는 다발성 사이토카인(pleiotropic cytokine)으로서, 구조와 기능에 있어 백혈병 억제인자(leukemia inhibitory factor; LIF)와 상당히 유사한 것으로 알려져 있다. 간 발달, 혈액생성, 감염, 중추신경계 발달에 있어 중요한 인자로 알려져 있으며, 뼈 형성 및 파괴와도 연관되어 있다는 보고도 있다.
본 발명의 일 실시예에 따르면, 상기 (b-1) 단계의 온코스타틴 M을 포함하는 배지는 0.1 내지 5 μM 덱사메타손, 0.1 내지 5% ITS, 1 내지 50 ng/ml hHGF, 0.1 내지 5% 페니실린/스트렙토마이이신이 보충된 IMDM으로, 상기 온코스타틴 M은 1 내지 50 ng/ml, 1 내지 30 ng/ml, 1 내지 20 ng/ml, 1 내지 10 ng/ml, 5 내지 30 ng/ml, 5 내지 20 ng/ml, 10 내지 20 ng/ml, 20 내지 30 ng/ml, 15 ng/ml, 16 ng/ml, 17 ng/ml, 18 ng/ml, 19 ng/ml, 또는 20 ng/ml의 농도로 배지에 포함될 수 있으나, 이에 제한되지 않는다.

본 발명에서 용어 "배양"은 생체로부터 분리된 세포, 이들의 2차원 또는 3차원적 집합체, 조직 또는 조직의 일부를 체외에서 증식, 성장, 유지 및 분화시키는 것을 의미한다. 이에, 용어 "배양"은 출발 물질(세포, 조직 또는 조직 유사체)을 이용하여 인공적인 환경 하에서 목적 물질을 수득하는 전 과정을 포괄하는 의미이며, 이에 “배양용 조성물"은 "증식용 조성물", "성장용 조성물", "유지용 조성물" 및 "분화 유도용 조성물"을 모두 포함하는 의미이다. 구체적으로, 상기 간세포 배양용 조성물에서 "배양"은 줄기세포의 간세포로의 분화를 유도하여 간세포를 수득하는 것을 의미한다.
본 발명에 있어서, 상기 배양용 조성물은 줄기세포 배양용 기본 배지(basal media) 조성물을 추가적으로 포함할 수 있다. 이러한 기본 배지로는 당업계에서 줄기세포 배양에 이용되는 다양한 배지가 사용될 수 있으며, 예를 들어 IMDM(Iscove's Modified Dulbecco's Medium), α-MEM(Alpha Modification of Eagle's Medium), F12(Nutrient Mixture F-12), 및 DMEM/F12(Dulbecco's Modified Eagle Medium: Nutrient Mixture F12)를 포함하나, 이에 제한되는 것은 아니다. 구체적으로, 본 발명에서 이용되는 줄기세포 배양용 기본 배지는 DMEM/F12 및 IMDM 배지일 수 있으나, 이에 제한되지 않는다.

본 발명에서 상기 제1조성물이 젤라틴이 코팅된 배양용기에 첨가된다는 것은 젤라틴이 코팅된 배양용기에 배지 조성물로 첨가되는 것을 의미하는 것으로, 상기 제1조성물을 배지로 젤라틴이 코팅된 배양용기에서 배양이 이루어지는 것일 수 있다.
본 발명에 있어서, 상기 제1조성물에서 ROCK 억제제는 1 내지 50 μM, 1 내지 40 μM, 1 내지 30 μM, 1 내지 20 μM, 1 내지 10 μM, 2 내지 40 μM, 2 내지 30 μM, 2 내지 20 μM, 2 내지 10 μM, 5 내지 30 μM, 5 내지 20 μM, 5 내지 10 μM, 10 내지 30 μM, 10 내지 20 μM, 1 μM, 2 μM, 3 μM, 4 μM, 5 μM, 6 μM, 7 μM, 8 μM, 9 μM, 또는 10 μM의 농도로 포함될 수 있으나, 이에 제한되지 않는다.

본 발명에 있어서, 상기 제2조성물의 젤라틴은 0.01 내지 1%, 0.01 내지 0.9%, 0.01 내지 0.8%, 0.01 내지 0.7%, 0.01 내지 0.6%, 0.01 내지 0.5%, 0.01 내지 0.4%, 0.01 내지 0.3%, 0.01 내지 0.2%, 0.01 내지 0.1%, 0.01 내지 0.05%, 0.05 내지 0.8%, 0.05 내지 0.6%, 0.05 내지 0.5%, 0.05 내지 0.3%, 0.05 내지 0.1%, 0.1 내지 0.8%, 0.1 내지 0.7%, 0.1 내지 0.6%, 0.1 내지 0.5%, 0.1 내지 0.4%, 0.1 내지 0.3%, 0.1 내지 0.2%, 또는 0.05 내지 0.2%의 농도로 포함될 수 있으며, 바람직하게는 0.1 내지 0.5%의 농도로 포함될 수 있으나, 이에 제한되지 않는다.
본 발명에 있어서, 상기 제1조성물 및 제2조성물은 동시에, 별도로, 또는 순차적으로 간세포의 배양에 사용될 수 있으며, 본 발명의 일 실시예 따르면, 줄기세포로부터 간세포로의 분화 과정 중 줄기세포로부터 간모세포로의 분화를 유도하는 단계, 바람직하게는 줄기세포로부터 간 내배엽으로의 분화를 유도하는 단계에서 제1조성물, 간모세포로부터 간세포로의 분화를 유도하는 단계에서 제2조성물을 사용할 수 있으나, 이에 제한되지 않는다.
또한, 본 발명은 본 발명에 따른 간세포 배양용 조성물, 및 설명서를 포함하는 간세포 배양용 키트를 제공한다.
본 발명에 있어서, 상기 설명서는 본 발명에 따른 줄기세포 유래 간세포 분화 방법이 기재된 것일 수 있으나, 이에 제한되지 않는다.
본 발명에서, "키트"는 본 발명에 따른 방법으로 줄기세포를 간세포로 분화할 수 있도록 하는 도구를 의미한다. 본 발명의 키트에는 상기 물질 외에도 이들의 보관, 처리 방법에 통상적으로 필요한 다른 구성성분, 조성물, 용액, 장치 등이 포함될 수 있다. 구체적인 예로는 이때, 각 구성은 1회 이상 횟수에 제한 없이 적용될 수 있으며, 각 물질을 적용하는 선후에는 제한이 없고, 각 물질의 적용은 동시에 진행될 수도 있고 미시에 진행될 수도 있다.
본 발명에 있어서, 상기 키트는 컨테이너; 지시서; 등을 포함할 수 있다. 상기 컨테이너는 상기 물질을 포장하는 역할을 할 수 있고, 보관 및 고정하는 역할을 할 수도 있다. 상기 컨테이너의 재질은 예컨대, 병, 통 (tub), 작은 봉지 (sachet), 봉투 (envelope), 튜브, 앰플 (ampoule) 등과 같은 형태를 취할 수 있고, 이들은 부분적 또는 전체적으로 플라스틱, 유리, 종이, 호일, 왁스 등으로부터 형성될 수 있다. 상기 용기는 처음에는 용기의 일부이거나 또는 기계적, 접착성, 또는 기타 수단에 의해 용기에 부착될 수 있는, 완전히 또는 부분적으로 분리가 가능한 마개를 장착할 수 있으며, 또한 주사바늘에 의해 내용물에 접근할 수 있는 스토퍼가 장착될 수 있다. 상기 키트는 외부 패키지를 포함할 수 있으며, 외부 패키지는 구성 요소들의 사용에 관한 지시서를 포함할 수 있다.
본 발명에 있어서, "포함하는"이라는 용어가 사용될 때, 이는 특별히 반대되는 기재가 없는 한 다른 구성 요소를 제외하는 것이 아니라 다른 구성 요소를 더 포함할 수 있는 것을 의미한다. 본 발명 전체에서 사용되는 정도의 용어 "~(하는) 단계" 또는 "~의 단계"는 "~를 위한 단계"를 의미하지 않는다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실험방법]
1. 세포 배양
아산줄기세포센터(서울 아산생명과학연구원)에서 hUCM-MSC(human umbilical cord matrix-mesenchymal stem cells)를 획득하였다. 줄기세포는 0.1% 젤라틴 코팅 세포 배양 접시에서 DMEM/F12 배지에 10% 소태아혈청 (Fetal Bovine Serum, FBS; GenDEPOT, 미국), 1% 비필수 아미노산(Non-Essential amino acids, NEAA), 1% 항생제-항진균제(Gibco, 미국), 및 0.2mM L-아스코르빈산(L-ascorbic acid)을 보충한 배양액에서 배양하였다. 3-4일마다 1:3~1:5로 희석하여 수동으로 계대배양하였다.
2. 정량적 RT-PCR(Quantitative RT-PCR)
제조업체의 지침에 따라 RNeasy 미니 키트(Qiagen, 미국)를 사용하여 총 RNA를 추출하였다. 상보적 DNA(complementary DNA, cDNA)는 Ultrascript 2.0 cDNA 합성 키트(PCR Biosystems, 영국)를 사용하여 합성하고, qRT-PCR은 CFX Connect Real-Time PCR Detection System(Bio-Rad Laboratories, CA, USA)에서 HOT FIREPol EvaGreen qPCR Supermix (SOLIS BIODYNE, Tartu, Estonia)를 사용하여 수행하였다. 분석을 위해 mRNA 수준을 GAPDH로 정규화하였다. 프라이머 서열은 표 1에 개시하였다.
미분화된 세포 및 분화된 세포에서 간 성숙 miRNA(miR-122 및 miR-192)의 발현을 평가하기 위해 RT-qPCR을 수행하였다. 구체적으로, 제조사의 지침에 따라 miRCURY LNA RT 키트(Qiagen, 독일)를 사용하여 총 RNA로부터 cDNA를 합성하였다. RT-qPCR 분석은 Qiagen에서 구입한 miRNA 특이적 프라이머와 함께 miRCURY LNA SYBR Green PCR 키트(Qiagen, 독일)를 사용하여 수행하였다. 사이클 조건은 다음과 같다: 95°C에서 2분 동안 배양한 다음 95°C에서 10초 동안 변성을 40회 반복, 56°C에서 1분간 어닐링(annealing) 및 연장. 사이클 임계값은 Bio-Rad CFX Maestro software (CFX Maestro, version 1.1; Bio-Rad Laboratories)를 사용하여 결정하였다. 모든 실험은 세 번 반복하였으며, 내부 대조군으로 RNU6B를 사용하였다.
3. 미토콘드리아 DNA 복제수(copy number) 예측
mtDNA(Mitochondrial DNA) 복제수는 Absolute Human Mitochondrial DNA Copy Number Quantification qPCR assay kit(ScienCell, CA, USA)를 사용하여 측정하였다. mtDNA 복제수를 평가하기 전에 QIAamp DNA 117 Mini Kit (Qiagen)를 사용하여 총 DNA를 분리하였다. 간략하게, 핵 특이적 및 미토콘드리아 특이적 프로브를 사용하여 각 샘플에 대한 주기 임계값을 삼중으로 측정하였다. 분석은 제조업체의 지침에 따라 수행하였다.
4. 분비된 인간 알부민(albumin) 검출
분화된 세포에서 분비된 인간 알부민을 제조업체의 지침에 따라 Human Albumin ELISA kit (Bethyl Laboratories, TX, USA)를 사용하여 검출하였다. 알부민 분비는 배양일과 총 세포 수로 정규화하였다.
5. 시험관 내 CYP3A4 활성 측정
효소 활성은 제조업체의 지침에 따라 P450-Glo CYP3A4 키트(Promega, 미국)를 사용하여 측정하였다. 발광은 GloMax 96 Microplate Luminometer (Promega)로 측정하였다. CYP3A4 활성은 각 샘플의 배양일과 이중 가닥 DNA 함량으로 정규화하였다.
6. 시험관 내 간 분화
간 분화는 이전에 보고된 방법(Cells. 2020;9(6), Stem Cell Research & Therapy. 2021;12(1):569) 일부를 수정하여 수행하였다. 간략하게, 줄기세포를 세포 배양 배지에 7000 cell/cm2로 0.1% 또는 1% 젤라틴 코팅 접시에 시딩(seeding)하였다. 코팅 접시에 줄기세포가 부착되도록 1일 동안 배양한 후, 세포를 줄기세포 배양액(DMEM/F12 배지에 10% 소태아혈청 (Fetal Bovine Serum, FBS; GenDEPOT, 미국), 1% 비필수 아미노산(Non-Essential amino acids, NEAA), 1% 항생제-항진균제(Gibco, 미국), 및 0.2mM L-아스코르빈산(L-ascorbic acid)을 보충한 배양액)과 10 μM 파수딜(fasudil, AdooQ Bioscience, CA, USA) 로 3일 동안 전처리하였다. 다음으로, 세포를 1단계 기본 배지, 10 ng/ml FGF2, 20 ng/ml BMP4, 3 μM CHIR99021로 구성된 간모세포(hepatoblast) 유도 배지에서 4일간 배양하였다. 마지막으로, 분화된 세포를 2단계 기본 배지와 20 ng/ml OSM(oncostatin M)으로 구성된 간 성숙 배지에서 8일 동안 배양하였다. 8일 후, 배지를 2단계 기본 배지, 20 ng/ml OSM, 0.1% 젤라틴(1 mg/ml) 또는 1% 젤라틴(10 mg/ml)으로 구성된 간 성숙 배지로 교체하여 5일 동안 배양하였다. 분화 배지는 2일마다 교체하였다.
1단계 기본 배지의 구성은 다음과 같다: 0.1% polyvinyl alcohol(PVA; Sigma Aldrich), 10 mM nicotinamide(Sigma Aldrich), 20 ng/ml hHGF(human hepatocyte growth factor; Peprotech, 미국), 1% ITS(insulin-transferrin-selenium; Gibco), 1% 페니실린/스트렙토마이신(GeneDirx, 대만)이 보충된 IMDM (Iscove's Modified Dulbecco's Medium, Gibco)을 사용하였다.
2단계 기본 배지의 구성은 다음과 같다: 1 μM 덱사메타손(dexamethasone), 1% ITS, 20 ng/ml hHGF, 1% 페니실린/스트렙토마이이신이 보충된 IMDM.
모든 성장 인자는 Peprotech에서 구입하였다.
7. 단백질 추출 및 웨스턴 블롯팅
웨스턴 블롯팅을 위해 세포를 트립신(Trypsine) 처리하고 얼음처럼 차가운 PBS로 세척한 다음 protease and phosphatase inhibitor cocktail (Sigma Aldrich) 이 포함된 RIPA 용해 완충액(50 mM HEPES, pH 7.4, 150 mM NaCl, 1 mM EDTA, 2.5 mM EGTA, 1 mM DTT, 1% Triton X-100)에 용해시켰다. 용해 후, 13,000rpm에서 20분간 원심분리하여 세포 파편을 제거하였다. 그런 다음, Bradford assay를 사용하여 단백질 농도를 측정하였다. 총 세포 단백질(15 μg)을 8-15% SDS-PAGE(sodium dodecyl sulfate-polyacrylamide gel electrophoresis)로 분리한 후 Immobilon PVDF membrane(Immobilon polyvinylidene fluoride membranes; Millipore, MA, USA)으로 옮겼다. 멤브레인을 TBST(Tris-buffered saline with Tween 20; 20 mM Tris-HCl pH 7.4, 150 mM NaCl, 0.1% Tween 20) 중 8% BSA(bovine serum albumin; GenDEPOT)로 차단하고, 항-알부민(Abcam, 영국) 및 항-CYP3A4(Santa Cruz, USA) 1차 항체로 탐침(probe)하였다. TBST로 세척한 후, 1차 항체는 horseradish peroxidase conjugated anti-mouse 2차 항체와 enhanced chemiluminescence detection system(Amersham, UK)을 사용하여 검출하였다.
8. 분화된 세포의 세포소기관 분석
굴절률 단층 촬영(refractive index(RI) tomography)을 이용한 무표지(Label-free) 광학 회절 단층 촬영(optical diffraction tomography, ODT)은 ODT 현미경(HT-X1; Tomocube Inc, 한국)을 사용하여 hUCM-MSC에서 수행하였다. ODT는 3차원 RI 단층 촬영을 사용하여 37℃ 온도에서 5% CO2의 제어된 대기에서 450nm LED(light-emitting diode)로 조명된 다양한 각도에서 캡처한 48개의 중첩된 2차원 홀로그램에서 단일 hUCM-MSC를 재구성하였다. 세포의 3차원 RI 단층 촬영 재구성을 위해 Mach-Zehnder interferometer가 통합된 HT-X1 현미경을 사용하였다. LD 정량화 및 체적 분석은 TomoCube의 TomoAnalysis 소프트웨어를 사용하여 수행하였으며 정밀도를 보장하기 위해 형광 염색을 사용하였다. 미토콘드리아 라벨링용 250 nM MitoTracker 염료(Invitrogen, CA, 미국)와 Biotium LipidSpot 488 지질방울 염색(1:1000 희석)을 각각 미토콘드리아와 LD를 염색하는데 사용하였다. 생세포 염색은 제조업체의 지침에 따라 수행하였다.
또한, 공초점 레이저 스캐닝 현미경인 Zeiss LSM 880(Carl Zeiss, 독일)을 사용하여 세포를 관찰하였다. 구체적으로, 세포를 4% 포름알데히드로 하룻밤 동안 고정한 뒤, PBST(Tween 20이 포함된 PBS)로 세척하고, 0.5% 트리톤 X-100으로 투과시킨 다음 1% BSA가 포함된 PBST로 차단하였다. 그런 다음 샘플을 Biotium LipidSpot 488 지질방울 염색으로 염색하였다. 핵은 10분 동안 NucBlue 고정 세포 레디프로브 시약(DAPI; Invitrogen)으로 상대 염색하고, 형광 신호는 Zeiss LSM 880 공초점 레이저 주사 현미경을 사용하여 검출하였다.
9. 해마 분석(Seahorse assay): ATP 생성률을 정량화하는 실험
분화된 세포에서 OCR(oxygen consumption rate)을 측정하기 위해, 줄기세포를 0.1% 또는 1% 젤라틴 코팅된 XFe24 세포 배양 플레이트(Agilent Technologies, Santa Clara, CA, USA)에 7000 cell/cm2로 시딩한 다음 분화를 유도하였다. 미토콘드리아 OCR은 XF24 extracellular flux analyzer (Agilent Technologies)에서 XF Cell MitoStress test kit를 사용하여 측정하였다. OCR 값은 세포 DNA의 양에 따라 정규화하였다.
10. 통계 분석
통계 분석은 GraphPad Prism 버전 6.0(GraphPad Software, 미국)을 사용하여 수행하였다. 3개 이상의 데이터 세트에 대한 비교는 일원 분산 분석 또는 이원 분산 분석(ANOVA)과 Bonferroni의 다중 비교 테스트(Bonferroni's multiple comparison test)를 통해 수행하였다. 두 그룹 간 비교는 two-tailed Student's t-test를 사용하여 수행하였다. P값이 0.05 미만이면 통계적으로 유의미한 것으로 간주하였다.
[실시예]
실시예 1. 간 분화의 초기 단계에 영향을 미치는 분화된 세포의 세포소기관 상태
이전 연구에서 ROCK 억제제인 파수딜(fasudil)이 세포 분화의 초기 단계에서 중요한 과정인 hUCM-MSC의 간 내배엽으로의 분화에 어떤 영향을 미치는지 확인한 결과, ROCK 억제제가 줄기세포 생존력을 향상시킬 뿐만 아니라 인간 만능 줄기세포(human pluripotent stem cell; hPSC)의 내배엽으로의 유도를 촉진하는 것으로 확인되었다. 또한, 분화에 사용되는 일반적인 단백질(ex. cytokine)만 사용한 경우보다 저분자(파수딜)을 사용할 경우 분화 효율이 증가하는 것으로 나타났다(Cell Stem Cell. 2009;4(4):348-58). 따라서, 파수딜이 hUCM-MSC의 간 분화에 미치는 영향을 확인하기 위해 먼저 파수딜의 적정 농도와 처리 시간을 확인하였다. 그 결과, 도 1a 및 도 1b에 나타낸 바와 같이, 기존에 보고된 hPSC에 사용된 파수딜의 농도(Molecular metabolism. 2017;6(7):640-50)와는 달리 10 μM의 고농도에서 내배엽 마커(GATA4, SOX17, FOXA2)가 유의하게 증가하였으며, 72시간 처리 시 효과적인 것으로 나타났다 (P < 0.05). 또한, 줄기세포 운명의 대사와 조절에 중요한 것으로 알려진 분화세포 내 LD(lipid droplet), 미토콘드리아와 같은 세포소기관의 상태를 확인하기 위해 단층촬영 분석을 실시하였다. 그 결과, 도 1c에 나타낸 바와 같이, 줄기세포가 조금 더 난형 모양으로 변화하고 미토콘드리아 형태가 달라졌다. 또한, 도 1c 및 도 1d에 나타낸 바와 같이, 세포 내부에서 LD가 유도되었고, LD의 수가 증가하였다(P <0.001).
다음으로, 도 1e에 나타낸 바와 같이, 간 내배엽의 다음 단계에 대한 평가로 파수딜의 사용이 간모세포(hepatoblast)의 분화 효율을 증가시키는지 여부를 확인하였다. 그 결과, 도 1f에 나타낸 바와 같이, 분화된 세포의 표현형이 대조군과 파수딜 처리군 모두에서 유사한 것으로 나타났다. 그러나, 도 1g에 나타낸 바와 같이, 간세포 관련 유전자(AFP 및 HNF4A)의 발현이 파수딜 처리군에서는 억제된 것으로 나타났다(P <0.001). 내배엽 유전자 상향조절에도 불구하고 간세포 분화 효율이 감소한 이유를 확인하기 위해 미토콘드리아 기능 검사를 수행하였다. ATP (adenosine triphosphate) 생성과 같은 미토콘드리아 기능은 분화 효율과 관련이 있으며, 과도한 LD가 유도되면 감소한다. 따라서, 파수딜에 의해 유도된 LD가 미토콘드리아 기능에 영향을 미칠 것으로 가정하고 이를 확인하기 위해 파수딜을 처리한 세포에서 미토콘드리아의 기능을 확인하였다. 확인 결과, 예상대로, 도 1h에 나타낸 바와 같이, 분화 3일차에 파수딜을 처리한 세포에서 미토콘드리아 기능이 감소하였다(P <0.001).
상기 결과로부터, 파수딜이 hUCM-MSC의 간 분화 초기 단계에 전사적으로 영향을 미치지만 세포소기관 상태로 인해 내배엽의 다음 단계에는 영향을 미치지 않는다는 것을 알 수 있었다.
실시예 2. 젤라틴 점도가 hUCM-MSC의 간모세포 유도에 미치는 영향
파수딜 처리 결과를 바탕으로 미토콘드리아 기능이 분화 효율과 상관관계가 있는 것으로 가정하였다. 따라서, 다음 단계로 ECM(extracellular matrix) 성분을 통해 미토콘드리아 기능을 강화하고자 하였다. 또한, 본 실험 결과에서 확인된 바와 같이, 0.1% 또는 1% 젤라틴 코팅 접시에서 hUCM-MSC를 배양할 경우, 도 2a에 나타낸 바와 같이, 기저(basal) 산화 인산화, 최대(maximal) 산화 인산화, ATP 생성, 및 양성자 누출(proton leak)과 같은 미토콘드리아 활성화 수준이 증가하였다(P <0.001). 이전에 진행된 연구 결과에 따르면, 분화된 세포에서 산화적 인산화 수준과 ATP 생산의 증가는 특정 계통 분화에 필요한 것으로 확인되었다(Dynamic changes in mitochondrial biogenesis and antioxidant enzymes during the spontaneous differentiation of human embryonic stem cells. 2006;348(4):1472-8, Glycolytic network restructuring integral to the energetics of embryonic stem cell cardiac differentiation. 2010;48(4):725-34). 따라서, ECM으로 사용되는 젤라틴이 파수딜과 함께 분화 효율을 시너지적으로 향상시킬 수 있을 것으로 예상되었다.
젤라틴이 간 분화에 미치는 영향을 평가하기 위해 도 2b에 나타낸 바와 같이, 0.1% 또는 1% 젤라틴 코팅 접시에서 hUCM-MSC를 배양하고 이러한 조건에서 분화를 진행시켰다. 도 2c에 나타낸 바와 같이, 분화된 세포의 표현형은 7일차에 모든 그룹에서 유사하게 나타났다. 다음으로 분화 0일, 3일, 7일차에 간 내배엽 및 간모세포와 관련된 유전자 발현의 역학을 분석하였다. 그 결과, 도 2d 및 도 2e에 나타낸 바와 같이, 1% 젤라틴과 파수딜 처리군에서 GATA4를 제외한 내배엽-관련 유전자(SOX17, FOXA2)와 간모세포-관련 유전자(AFP, HNF4A)가 다른 군에 비해 유의하게 상향 조절되는 것으로 나타났다. 그러나, 도 2e에 나타낸 바와 같이, 흥미롭게도 간모세포 분화가 진행됨에 따라 1% 젤라틴 및 파수딜 그룹과 달리 0.1% 젤라틴 및 파수딜 그룹에서는 간모세포 마커(AFP 및 HNF4A)가 감소하였다(#a 및 #b, P <0.01).
저점도 젤라틴 그룹과 고점도 젤라틴 그룹에서 관찰된 분화 효율의 차이를 확인하기 위해 줄기세포의 초기 분화 운명을 확인하였다. 특히, 파수딜은 뉴런과 심근세포를 포함하는 외배엽과 중배엽에서 유래하는 세포의 분화를 위한 프로모터(promoter)로 알려져 있다. 따라서 분화 3일차에 분화된 세포에서 외배엽(PAX6, SOX1, OTX2) 및 중배엽(MIXL2, CDX2) 마커의 유전자 발현을 확인하였다. 그 결과, 도 2f에 나타낸 바와 같이, 1% 젤라틴과 파수딜 그룹에서 외배엽 유전자(PAX6 및 OTX2)의 발현이 억제된 것으로 나타났다(P <0.05). 또한, 도 2g에 나타낸 바와 같이, 1% 젤라틴과 파수딜 그룹에서 중배엽 유전자 MIXL2는 하향 조절된 것으로 나타났다(P <0.01).
상기 결과로부터 고점도 젤라틴이 외배엽 및 중배엽 관련 유전자의 발현을 억제하여 내배엽의 운명을 유도하는 것을 알 수 있었다.
젤라틴 점도에 따른 줄기세포의 운명 결정이 세포소기관 상태에 영향을 받을 수 있다는 점을 고려하여, 다음으로 고점도 젤라틴이 분화된 세포의 세포소기관에 미치는 영향을 확인하였다.
실시예 3. 고점도 젤라틴은 LD의 유도를 억제하고 미토콘드리아 기능을 향상시켰다.
고점도 젤라틴이 세포소기관 조절을 통해 줄기세포의 운명에 미치는 영향을 분석하기 위해 단층 촬영 및 미토콘드리아 기능 분석을 수행하였다. 단층 촬영 분석은 공초점 레이저 주사 현미경(confocal laser scanning microscopy)보다 세포소기관을 더 정확하게 관찰할 수 있는 것으로 알려져 있다. 따라서 분화 3일차에 분화된 세포의 미토콘드리아의 LD와 형태를 단층 촬영으로 분석하였다. 그 결과, 도 3a 및 도 3b에 나타낸 바와 같이, 1% 젤라틴과 파수딜을 처리한 그룹은 0.1% 젤라틴과 파수딜을 처리한 그룹보다 LD 유도가 낮은 것으로 나타났으며(P <0.001), 이는 고점도 젤라틴이 파수딜에 의해 유도된 LD 생성을 억제한다는 것을 시사한다. 또한, 도 3a에 나타낸 바와 같이, 0.1% 젤라틴과 파수딜 그룹에서는 미토콘드리아 형태가 과융합 유도되는 것으로 나타났다.
다음으로, 과도한 LD 유도 및 과융합 형태의 미토콘드리아가 세포 미토콘드리아 기능에 어떤 영향을 미치는지 확인하기 위해, 해마 분석을 수행하였다. 그 결과, 도 3c 및 도 3d에 나타낸 바와 같이, 1% 젤라틴과 파수딜 그룹에서 전체 OCR 수치가 가장 높게 나타났다(P <0.05). 또한, 모든 그룹에서 mtDNA 복제수를 측정하였다. 이전 연구에서 분화를 지원하기 위해 mtDNA 수준이 점차 증가하는 것으로 확인되었다. 따라서, 분화가 효율적일수록 mtDNA 복제수가 더 높아질 것으로 예상되었다. 도 3e에 나타낸 바와 같이, 본 실험에서는 1% 젤라틴과 파수딜 그룹에서 mtDNA 복제수가 더 높은 것으로 나타났다(P <0.001).
요약하면, 1% 젤라틴과 파수딜 그룹에서 향상된 분화 효율은 외배엽 및 중배엽 유전자 발현의 하향 조절뿐만 아니라 LD 생성 감소 및 활성화된 미토콘드리아 기능을 포함한 세포소기관 상태의 조절에 기인한 것으로 볼 수 있다.
실시예 4. HLC(hepatocyte-like cell)의 효율성과 기능을 시너지적으로 향상시키는 저점도 젤라틴
마지막으로, 젤라틴이 간세포 성숙에 미치는 영향을 확인하기 위해, 1% 젤라틴 코팅과 파수딜을 사용하여 유도한 간모세포 도 4a에 나타낸 바와 같이, HLC로 분화시켰다. 이 단계에서는 접시를 코팅하지 않고 2단계 분화 배지에 젤라틴을 추가하여 사용하였다. 구체적으로, 8일 동안 OSM을 사용하여 간모세포로부터 HLC를 유도하였고, 5일 동안 0%(젤라틴을 첨가하지 않음), 0.1%, 또는 1%의 다양한 점도로 젤라틴을 첨가하여 성숙을 진행하였다. 도 4b에 나타낸 바와 같이, 분화 20일차에 HLC 표현형은 모든 그룹에서 유사했다. 그러나 전사 분석 결과, 도 4c에 나타낸 바와 같이, 저점도(0.1%) 젤라틴 그룹에서 다른 그룹에 비해 성숙 간세포 관련 유전자(ALB, CYP3A4, CYP1A2, HNF1A 및 HNF4A)의 발현이 유의하게 증가한 것을 나타났다(P <0.01). 또한, 도 4d 및 도 4e에 나타낸 바와 같이, 웨스턴 블롯팅을 통해 이들 단백질(ALB(알부민) 및 CYP3A4)의 발현을 확인한 결과, 0.1% 젤라틴 그룹의 ALB 수치는 0% 젤라틴 그룹과 비슷했으나, CYP3A4 발현은 현저히 높은 것으로 나타났다(P <0.01). 또한, 성숙 효율은 miR-122 및 miR-192와 같은 간세포 특이적 miRNA의 발현을 통해 평가하였다. 그 결과, 도 4f에 나타낸 바와 같이, 0.1% 젤라틴이 존재할 때 간세포 특이적 miRNA가 유의하게 상향 조절되는 것으로 나타났다.
다음으로, 단백질 합성과 해독에서 간세포의 주요 역할을 고려하여 알부민 분비 및 CYP3A4 활성을 측정하였다. 이전 연구 결과와 일치하게, 도 5a 및 도 5b에 나타낸 바와 같이, 0.1% 젤라틴을 첨가한 HLC에서 간 기능이 유의적으로 향상된 것으로 나타났다(P <0.001). 전반적으로, 간세포의 사양(specification) 및 성숙은 간모세포 분화와 대조적으로 저점도 젤라틴에서 더 효율적인 것으로 나타났다. 따라서, 이를 이해하기 위해 20일차 HLC의 LD와 mtDNA 복제수에 초점을 맞춰 분화된 세포소기관의 상태를 분석하였다. 공초점 영상 분석 결과, 도 5c 및 도 5d에 나타낸 바와 같이, 0.1% 젤라틴을 간세포 성숙에 사용한 경우 LD가 더 적게 유도되는 것을 확인했으며(P <0.05), 도 5e에 나타낸 바와 같이, mtDNA 복제수의 유의미한 증가가 동일한 그룹에서 확인되었다 (P <0.001).
상기 결과는 저점도 젤라틴이 초기 분화 단계에서의 역할과 달리 HLC의 성숙에 유리하다는 것을 의미하며, 세포소기관 상태가 간세포의 효율성과 기능을 결정하는 데 중추적인 역할을 한다는 것을 강조한다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
본 발명에 따른 줄기세포 유래 간세포 분화 방법은 줄기세포의 간세포로의 분화 초기 단계에서 ROCK 억제제로 줄기세포의 내배엽으로의 분화를 유도하되 이때 발생되는 지질방울의 생성을 고점도 젤라틴이 코팅된 배양용기에서 세포를 배양함으로써 억제하고, 미토콘드리아의 기능을 활성화하며, 내배엽 유전자의 발현을 상향 조절하고, 외배엽 및 중배엽 유전자의 발현을 하향 조절함으로써 줄기세포의 간모세포로의 분화 효율을 향상시켰다. 또한, 분화 성숙(후기) 단계에서 배지에 저점도 젤라틴을 첨가함으로써 지질방울의 축적을 억제하고, 미토콘드리아를 활성화함으로써 간모세포의 간세포로의 분화 효율을 향상시키고, 분화된 간세포의 기능을 향상시킴으로써, 줄기세포의 간세포로의 분화 효율을 향상시켰다. 따라서, 이를 이용하여 수득한 간세포는 간질환의 치료용 세포 치료제, 이식체 등으로 유용하게 이용될 수 있을 것으로 기대되는 바, 산업상 이용가능성이 있다.

Claims (21)

  1. 줄기세포를 젤라틴이 코팅된 배양용기에서 ROCK 억제제를 포함한 배지에 배양하여 간모세포(hepatoblast)로의 분화를 유도하는 단계를 포함하는, 줄기세포 유래 간세포 분화 방법.
  2. 제1항에 있어서,
    상기 줄기세포는 탯줄 기질, 제대혈, 골수, 지방, 근육, 신경, 피부, 양막, 및 태반으로 이루어진 군으로부터 선택되는 어느 1종 이상의 조직으로부터 유래된 중간엽 줄기세포인 것을 특징으로 하는, 줄기세포 유래 간세포 분화 방법.
  3. 제1항에 있어서,
    상기 ROCK 억제제는 파수딜 (Fasudil), 리파수딜 (Ripasudil), RKI-1447, Y-27632, GSK429286A, 및 Y-30141로 이루어진 군으로부터 선택되는 어느 하나 이상인 것을 특징으로 하는, 줄기세포 유래 간세포 분화 방법.
  4. 제1항에 있어서,
    상기 ROCK 억제제는 1 내지 50 μM의 농도로 배지에 포함되는 것을 특징으로 하는, 줄기세포 유래 간세포 분화 방법.
  5. 제1항에 있어서,
    상기 젤라틴은 0.1 내지 10%의 농도로 배양용기에 코팅된 것을 특징으로 하는, 줄기세포 유래 간세포 분화 방법.
  6. 제1항에 있어서,
    상기 간모세포로의 분화를 유도하는 단계는 1 내지 10일 동안 수행하는 것을 특징으로 하는, 줄기세포 유래 간세포 분화 방법.
  7. 제1항에 있어서,
    상기 간모세포로의 분화를 유도하는 단계는 하기 단계를 포함하는 것을 특징으로 하는, 줄기세포 유래 간세포 분화 방법:
    (a-1) 줄기세포를 ROCK 억제제를 포함하는 배지에서 배양하여 간 내배엽으로의 분화를 유도하는 단계; 및
    (a-2) 상기 간 내배엽을 성장 인자, 골 형성 단백질, 및 wnt 신호 활성자를 포함하는 배지에서 배양하여 간모세포로의 분화를 유도하는 단계,
    단, 상기 (a-2) 단계는 젤라틴이 코팅되지 않은 배양용기에서 수행됨.
  8. 제1항에 있어서,
    상기 방법은 간모세포를 젤라틴을 포함하는 배지에서 배양하여 간세포로의 분화를 유도하는 단계를 더 포함하는 것을 특징으로 하는, 줄기세포 유래 간세포 분화 방법.
  9. 제8항에 있어서,
    상기 젤라틴은 0.1 mg/ml 내지 10 mg/ml의 농도로 배지에 포함되는 것을 특징으로 하는, 줄기세포 유래 간세포 분화 방법.
  10. 제8항에 있어서,
    상기 간세포로의 분화를 유도하는 단계는 하기 단계를 포함하는 것을 특징으로 하는, 줄기세포 유래 간세포 분화 방법:
    (b-1) 상기 간모세포를 온코스타틴 M을 포함하는 배지에서 배양하는 단계; 및
    (b-2) 상기 온코스타틴 M을 포함하는 배지에 젤라틴을 첨가하고 추가적으로 배양하여 간세포로의 분화를 유도하는 단계.
  11. (a) ROCK 억제제를 유효성분으로 포함하는 제1조성물; 및 (b) 젤라틴을 유효성분으로 포함하는 제2조성물로 이루어진 군으로부터 선택되는 어느 하나 이상을 포함하는, 간세포 배양용 조성물로서,
    상기 제1조성물은 젤라틴이 코팅된 배양용기에 첨가되는 것을 특징으로 하는, 간세포 배양용 조성물.
  12. 제11항에 있어서,
    상기 배양은 줄기세포의 간세포로의 분화를 유도하는 것을 특징으로 하는, 간세포 배양용 조성물.
  13. 제12항에 있어서,
    상기 줄기세포는 탯줄 기질, 제대혈, 골수, 지방, 근육, 신경, 피부, 양막, 및 태반으로 이루어진 군으로부터 선택되는 어느 1종 이상의 조직으로부터 유래된 중간엽 줄기세포인 것을 특징으로 하는, 간세포 배양용 조성물.
  14. 제11항에 있어서,
    상기 ROCK 억제제는 파수딜 (Fasudil), 리파수딜 (Ripasudil), RKI-1447, Y-27632, GSK429286A, 및 Y-30141로 이루어진 군으로부터 선택되는 어느 하나 이상인 것을 특징으로 하는, 간세포 배양용 조성물.
