Abstract
液体処理装置4は、液体が収容された液体容器2を保持する容器保持部5が設けられ、及び設定温度が異なる複数の恒温領域6a~6cを有する恒温部6と、液体容器2に収容された液体を吸引又は吐出する分注機3と、恒温部6を加熱又は冷却する伝熱部7と、恒温部6の温度に係るデータを出力する温度センサ部11と、伝熱部7に電力を供給する電源部13と、温度センサ部11の出力に基づいて、電源部13が伝熱部7へ供給する電力を制御する制御部12と、恒温領域6a~6cを隔てる第一の熱抵抗部8a及び8bと、を備え、第一の熱抵抗部8a及び8bは、恒温部6よりも大きな熱抵抗値を有し、第一の熱抵抗部8a及び8bによって隔てられる一の恒温領域の温度が当該一の恒温領域の設定温度となるときに第一の熱抵抗部によって隔てられる他の恒温領域の温度が当該他の恒温領域の設定温度となる熱抵抗値を有する。
Description
本開示は、液体処理装置、核酸抽出装置、及び遺伝子検査装置に関する。
遺伝子検査装置では、PCR(polymerase chain reaction)法などによって核酸増幅して、検査を行う前に試料に含まれている核酸の抽出を行う。磁性粒子を担体としたMagtration法による核酸抽出では、試料中のタンパク質を可溶化させる液体処理工程、磁性粒子担体と核酸のリン酸基を結合させる液体処理工程、核酸が結合した磁性粒子から夾雑物を除去する液体処理工程、及び磁性粒子から核酸を溶出させる液体処理工程によって核酸を抽出する。このとき、各液体処理工程、試料の種類や使用する試薬の種類、処理プロトコルの違いによってそれぞれ最適な処理温度が異なる。そのため、信頼性の高い遺伝子検査に向けた核酸抽出を行うためには、液体処理の内容によって液体の温度を迅速に変化させることが可能な液体処理装置が必要となる。
液体処理装置における温度制御に関して、特許文献1には、試料を収容した複数の検査容器に対して個別に設けられた熱源と、当該熱源を制御して試料の温度を個別に調節する制御部を備えた遺伝子検査装置が開示されている。
特許文献2には、液体試料を収容した容器を収納する収納部を備えた温度調節ブロックの異なる箇所を種々の温度にするために、温度調節ブロックに熱伝導的に接触する温度調節装置を少なくとも2個以上備えた実験室用恒温装置が開示されている。
特許文献1に記載されている遺伝子検査装置では、複数の検査容器に入った試料の温度を調節する際に、試料の入った個々の検査容器を保持する伝熱部(熱伝導部)に対して、伝熱部(熱伝導部)を加熱・冷却する熱源と熱源を制御する制御部とがそれぞれ一つずつ必要な構造となっている。そのため複数の温度制御系を搭載することにより、温度制御系に関わる構造の複雑化や、コストの増大が課題となる。また、温度の異なる複数の伝熱部(熱伝導部)の間で伝熱が起きにくくなるように分離した構造となっているため、装置の大型化が課題となる。更に、試料の反応に最適な温度が変化する際に、伝熱部(熱伝導部)を加熱・冷却して温度変化させる構造となっているため、装置の温度昇降に時間が必要となることがスループットを向上させる上で課題となる。
特許文献2に記載されている実験室用恒温装置では、液体試料を収容した容器の収納部を複数備えた温度調節ブロックの異なる箇所を種々の温度にするために、複数の温度調節装置を必要としている。そのため複数の温度制御系を搭載することにより、温度制御系に関わる構造の複雑化や、コストの増大が課題となる。
本発明が解決しようとする課題は、複数の温度で液体処理を行う液体処理装置において、必ずしも複数の温度制御系を必要とせずに、液体処理に最適な温度の複数の恒温部を利用できる液体処理装置を小型の装置で提供することである。
上記課題を解決するために、本開示の液体処理装置は、液体が収容された液体容器を保持する容器保持部が設けられ、設定温度が異なる複数の恒温領域を有する恒温部と、液体容器に収容された液体を吸引又は吐出する分注機と、恒温部を加熱又は冷却する伝熱部と、恒温部の温度に係るデータを出力する温度センサ部と、伝熱部に電力を供給する電源部と、温度センサ部の出力に基づいて、電源部が伝熱部へ供給する電力を制御する制御部と、少なくとも二つの恒温領域を隔てる第一の熱抵抗部と、を備え、第一の熱抵抗部は、恒温部よりも大きな熱抵抗値を有し、第一の熱抵抗部によって隔てられる一の恒温領域の温度が当該一の恒温領域の設定温度となるときに第一の熱抵抗部によって隔てられる他の恒温領域の温度が当該他の恒温領域の設定温度となる熱抵抗値を有する。
また、本開示の核酸抽出装置は、上記した液体処理装置を備える。
また、本開示の遺伝子検査装置は、上記した液体処理装置を備える。
本開示に係る液体処理装置によれば、複数の温度で液体処理を行う液体処理装置において、必ずしも複数の温度制御系を必要とせずに、液体処理に最適な温度の複数の恒温部を利用できる液体処理装置を小型の装置で提供することができる。これにより、液体処理のスループット向上、温度制御系の単純化、装置製造に関わるコスト低減が期待できる。また、液体処理装置を小型化することでマルチウェルプレートを液体容器として使用可能となり、液体容器の交換に関わる工数低減や取り違えリスクの低減が期待できる。

実施例1の液体処理装置の一形態を説明する。図1は、実施例1の液体処理装置の断面構造を示す概要図である。本実施例では、一つの温度制御系によって、三つの恒温領域をそれぞれ異なる設定温度とする構造について説明する。
液体1は、液体容器2に収容され、分注機3によって吸引及び吐出されて分注される。液体処理装置4は、分注機3、液体容器2を保持する容器保持部5を備えた恒温部6、伝熱部7、第一の熱抵抗部8a及び8b、放熱部9、断熱部10、温度センサ部11、制御部12、並びに電源部13を備える。
液体容器2には、液体1を収容する領域が複数設けられている。液体容器2には、液体1を収容する領域が一方向に並んだものを使用してもよいし、二方向に並んだマルチウェルプレートを使用してもよい。本実施例では、液体容器2としてマルチウェルプレートを使用しているとする。
恒温部6には、設定温度が設けられる。設定温度は、環境温度に対して高くてもよく、低くてもよい。恒温部6は、設定温度が異なる複数の領域に分かれている。本実施例では、恒温部6は、設定温度が異なる三つの領域に分かれており、各領域を恒温領域6a、恒温領域6b、及び恒温領域6cとする。例えば、恒温領域6a、恒温領域6b、恒温領域6cの設定温度は、それぞれ80℃、70℃、60℃である。各恒温領域6a~6cが保持する容器保持部5の領域は一つでもよいし、複数であってもよい。各恒温領域6a~6cは、熱伝導率が高い材料で構成され、温度均一性に優れていることが望ましい。これによれば、恒温領域6a~6cが容器保持部5の複数の領域を保持するものであっても、複数の領域内の液体を同じ温度で処理することができる。恒温部6は、例えば銅、アルミニウム、それらの合金で構成されることが望ましい。また、恒温領域6a~6cの全てを同一材料で構成してもよいし、恒温領域6a~6cの少なくとも二つを異なる材料で構成してもよい。
伝熱部7は、恒温部6を加熱又は冷却する。恒温領域6a~6cの設定温度が環境温度よりも高ければ、伝熱部7にはヒータなどの発熱部品を使用できる。恒温領域6a~6cの設定温度が環境温度よりも低ければ、伝熱部7にはペルチェモジュールを使用するなどして冷却すればよい。本実施例では、設定温度の異なる恒温領域6a~6cの個数が三つであるのに対して、伝熱部7の個数は一つである。そして、伝熱部7は、設定温度が環境温度から最も離れている恒温領域6aに接触している。伝熱部7は、恒温領域6aに接触していなくてもよく、恒温領域6aを加熱又は冷却できればよい。
第一の熱抵抗部8aは、設定温度の異なる恒温領域6aと恒温領域6bとの間を隔てて、第一の熱抵抗部8bは、設定温度の異なる恒温領域6bと恒温領域6cとの間を隔てる。第一の熱抵抗部8a及び8bは、隔てている恒温領域のうち、設定温度の高い方から、設定温度の低い方への伝熱量を調整して、両方の恒温領域の温度が設定温度となるのに必要な熱抵抗値を予め備える。本実施例では、第一の熱抵抗部8aは、伝熱部7の加熱によって恒温領域6aが設定温度80℃になっているとき、恒温領域6bが設定温度70℃になる熱抵抗値を持つ。また、第一の熱抵抗部8bは、第一の熱抵抗部8aを介した伝熱で恒温領域6bが設定温度70℃になっているとき、恒温領域6cが設定温度60℃になる熱抵抗値を持つ。
放熱部9は、設定温度が環境温度から最も離れている恒温領域6a以外の恒温領域6b又は6cに備えることができる。本実施例では、恒温領域6a~6cのうち設定温度が環境温度に最も近い恒温領域6cに放熱部9が設置されている。放熱部9には、例えば冷却フィンを用いる。このとき放熱部9は、第一の熱抵抗部8a、第一の熱抵抗部8bの熱抵抗値と共に、恒温領域6a、恒温領域6b、恒温領域6cがそれぞれ設定温度となる放熱性能を持つ。
恒温領域6a、恒温領域6b、恒温領域6cの間に大きな温度差を設けるためには、第一の熱抵抗部8a、及び第一の熱抵抗部8bの熱抵抗値を大きくする必要がある。熱抵抗値を大きくするためには、第一の熱抵抗部8a、及び第一の熱抵抗部8bの見掛けの熱伝導率を小さくするか、厚さを大きくする必要がある。本実施例のように、液体容器2としてマルチウェルプレートが使用され、容器保持部5間の距離がマルチウェルプレートの寸法によって決定される場合では、第一の熱抵抗部8a、及び第一の熱抵抗部8bの厚さを自由に変えることは難しく、恒温領域6a、恒温領域6b、恒温領域6cの間に設定した温度差を設けられない場合がある。そこで、恒温領域6cに放熱部9を設けて環境との熱交換を促進することで、恒温領域6bや恒温領域6cの温度を環境温度に近づけることができる。そして、恒温領域6aが設定温度になっているときに、恒温領域6a、恒温領域6b、恒温領域6cの間に設ける温度差を大きくすることが容易になる。なお、図1の例では、恒温領域6cに放熱部9を設けたが、恒温領域6b及び恒温領域6cに放熱部9を設けてもよい。恒温領域6bに設けられる放熱部9の放熱性能と、恒温領域6cに設けられる放熱部9の放熱性能とを同じにしてもよいし、異ならせてもよい。
断熱部10は、恒温部6、伝熱部7、第一の熱抵抗部8a及び8bを覆うように構成される。これにより、恒温部6は、温度均一性を保つことができる。また、環境への放熱を抑えることで伝熱部7の消費エネルギを低く抑えることができる。
温度センサ部11は、恒温部6の温度を測定し、恒温部6の温度に係るデータを制御部12に出力する。制御部12は、温度センサ部11が設置された恒温領域6bの温度が設定温度となるように、温度センサ部11の出力に基づいて、電源部13が伝熱部7に供給する電力を制御する。本実施例では、恒温領域6a~6cのうち、設定温度が中間の恒温領域6bに温度センサ部11が設置されており、恒温領域6bが設定温度70℃となるように伝熱部7に供給する電力が制御される。なお、温度センサ部11が設置される恒温領域は、恒温領域6aであってもよいし、恒温領域6cであってもよい。
恒温領域6a、恒温領域6b、恒温領域6c、及び環境(Ambient)の温度の模式図を図2に示す。本実施例では、環境温度20℃において恒温領域6a、恒温領域6b、恒温領域6cがそれぞれ設定温度80℃、70℃、60℃になる構造となっている。ここで、環境温度が20℃から±10℃ばらつく場合を考える。恒温領域6bは、温度センサ部11の働きによって、環境温度がばらついても設定温度70℃に制御される。一方、恒温領域6aと恒温領域6cとは、環境温度のばらつきに伴って温度が設定温度からばらつく。恒温領域6aと恒温領域6cとの温度のばらつき具合をそれぞれE_6a、E_6cとすると、図2に示したようにE_6aとE_6cは同程度となる。
温度センサ部11の配置の別の例として、設定温度が環境温度から最も離れていて、伝熱部7が加熱している恒温領域6aに温度センサ部11が設置されており、恒温領域6aが設定温度80℃となるように伝熱部7に供給する電力が制御される場合を考える。このときの恒温領域6a、恒温領域6b、恒温領域6c、及び(Ambient)の温度の模式図を図3に示す。環境温度が20℃から±10℃ばらつく場合を考える。恒温領域6aは、温度センサ部11の働きによって、環境温度がばらついても設定温度に制御される。恒温領域6bと恒温領域6cとは、環境温度のばらつきに伴って温度が設定温度からばらつく。恒温領域6bと恒温領域6cとの温度のばらつき具合をそれぞれE_6b、E_6cとすると、図3に示したようにE_6bに対してE_6cが大きくなることが分かる。
環境温度が変化した時に、三つの領域に分かれている恒温領域6a~6cのうちどれかの温度が設定温度から大きく外れるのを防ぐためには、設定温度が中間の恒温領域6bに温度センサ部11を設置することが有効である。一方、三つの領域に分かれている恒温領域6a~6cのうち、どれかを優先的に高精度に温度制御する必要がある場合には、その恒温領域に温度センサ部11を設置することが有効である。以上の工夫は、設定温度の異なる恒温領域6a~6cの数に対して温度制御系の数が少ない構成において、環境温度のばらつきが液体処理のための温度制御性を低下させるのを防ぐ効果が得られる。
環境温度の変化などの要因で複数の恒温領域6a~6cのうちどれかの温度が設定温度から外れている場合に、制御部12は、液体処理が行われる恒温領域の温度が設定温度となるように、電源部13が伝熱部7に供給する電力を修正してもよい。例えば、設定温度が60℃の恒温領域6cで液体処理を行う際には、制御部12は、恒温領域6cが設定温度60℃になるのに必要な電力を電源部13が伝熱部7に供給するように制御する。その後、設定温度が80℃の恒温領域6aで液体処理を行う際には、制御部12は、恒温領域6aが設定温度80℃になるのに必要な電力を電源部13が伝熱部7に供給するように修正してもよい。このとき、それぞれの恒温領域が設定温度になるのに必要な電力供給量は、事前に測定して記録しておくことができる。例えば、図示していない温度センサによって環境温度を測定して、ある恒温部6が設定温度になるのに必要な電力供給量を環境温度ごとに記録しておいてもよい。本実施例では、設定温度の異なる恒温領域6a~6cの温度はそれぞれの設定温度付近にあるので、短時間の温度変更時間で恒温部6の温度をより設定温度に近づけられることが期待できる。
第一の熱抵抗部8a及び8bは、隔てている恒温領域のうち、設定温度の高い方から、設定温度の低い方への伝熱量を調整して、両方の恒温領域の温度が設定温度となるのに必要な熱抵抗値を予め備える。第一の熱抵抗部8a及び8bの構造の例を図4に示す。第一の熱抵抗部8a又は8bの構造の例として、単一の熱抵抗部材801によって所望の熱抵抗値を持たせてもよい。熱抵抗部材801として例えば、恒温部6より熱伝導率の低い一様の固体材料を選択して配置することができる。隔てている恒温領域に温度差を設けるという観点では、恒温部6より熱伝導率の低い固体材料が望ましい。一例として樹脂系材料、ゴム系材料、断熱材が用いられる。熱抵抗部材801として、固体材料の他に気体層としてよい。第一の熱抵抗部8a及び8bの両方が、単一の熱抵抗部材801であってもよいし、第一の熱抵抗部8a又は8bの何れか一方が、単一の熱抵抗部材801であってもよい。
第一の熱抵抗部8a及び8bの他の構造の例として、恒温領域の表面に設けた凸部802と凹部803によって所望の熱抵抗値を持たせてもよい。この構造の場合、一方の恒温領域の表面に設けた凸部802と他方の恒温領域の表面の間の接触面積を変化させ、凸部802による固体伝導伝熱を調整して所望の熱抵抗値を持たせることができる。凹部803は、熱抵抗部材801と同様に、適切な熱伝導率を持った固体材料や、気体層などを用いてもよい。また恒温部6を一体の部品として凹部803に対応する箇所に穴加工をすることで凸部802と凹部803に相当する構造としてもよい。第一の熱抵抗部8a及び8bの両方が、凸部802及び凹部803を有する構造であってもよいし、第一の熱抵抗部8a又は8bの何れか一方が、凸部802及び凹部803を有する構造であってもよい。
第一の熱抵抗部8a及び8bの構造の他の例を図5に示す。第一の熱抵抗部8a及び8bの他の構造の例として、スペーサ804と締結部材805によって所望の熱抵抗値を持たせてもよい。スペーサ804と締結部材805は、隔てている恒温領域を相対的に所定の位置関係に固定する他、固体伝導伝熱を調整する役割を持たせることができる。スペーサ804によって固体伝導伝熱を調整する方法として、スペーサ804の大きさを変えて伝熱面積を変化させてもよいし、スペーサ804を配置する個数を変えて伝熱面積を変化させてもよいし、スペーサ804を熱伝導率の異なる材料を使用してもよい。例えば、スペーサ804は、締結部材805によって締結する際に熱伝導率の低い樹脂製のワッシャを用いることができる。締結部材805によって固体伝導伝熱を調整する方法として、締結部材805を熱伝導率の異なる材料を使用してもよい。例えば、締結部材805は、樹脂ねじを用いることができる。第一の熱抵抗部8a及び8bの両方が、スペーサ804及び締結部材805を有する構造であってもよいし、第一の熱抵抗部8a又は8bの何れか一方が、スペーサ804及び締結部材805を有する構造であってもよい。また、第一の熱抵抗部8aとして使用したスペーサ804と、第一の熱抵抗部8bとして使用したスペーサ804とを一つの締結部材805で固定してもよい。
第一の熱抵抗部8a及び8bの構造の他の例を図6に示す。第一の熱抵抗部8a及び8bの他の構造の例として、恒温領域の対向する表面に設けた熱輻射部806と熱輻射部807とによって所望の熱抵抗値を持たせてもよい。この構造では、恒温領域の表面間の輻射伝熱を調整することで所望の熱抵抗値を持たせる。熱輻射部806又は熱輻射部807に、放射率を変化させる表面処理を施してもよい。例えば、熱輻射部806又は熱輻射部807は、放射性塗料を用いてもよいし、アルマイト加工などの酸化被膜を形成してもよい。また、表面処理を施す面積によって輻射伝熱を調整して所望の熱抵抗値を持たせてもよい。図6の例では、恒温領域の対向する表面の両方に熱輻射部806及び熱輻射部807を形成したが、一方の面のみに熱輻射部を設けてもよい。また、第一の熱抵抗部8a及び8bの両方が、熱輻射部806又は807を有する構造であってもよいし、第一の熱抵抗部8a又は8bの何れか一方が、熱輻射部806又は807を有する構造であってもよい。
第一の熱抵抗部8a及び8bの他の構造の例として、密閉部808を形成し、密閉部808の内部空間を減圧することで所望の熱抵抗値を持たせてもよい。密閉部808は、減圧しなければ空気による対流と伝導伝熱、恒温領域の表面間輻射伝熱によって熱移動する。密閉部808を減圧していくことで、空気による対流と伝導伝熱の影響を減じて所望の熱抵抗値を持たせてもよい。また、真空断熱パネルのように予め内部を減圧して封止した断熱材を用いてもよい。第一の熱抵抗部8a及び8bの両方が、密閉部808を有する構造であってもよいし、第一の熱抵抗部8a又は8bの何れか一方が、密閉部808を有する構造であってもよい。
第一の熱抵抗部8a及び8bは、上記の構造を用いることで容易に熱抵抗値を変更することができる。恒温領域6a~6cの設定温度を変更する必要が生じた場合でも、上記の構造によって熱抵抗値を変更すれば、全体の構造を大きく変更することなく、短い検討期間で異なる設定温度の恒温領域6a~6cを実現することが可能である。第一の熱抵抗部8a及び8bの各々は、上記の構造からひとつ選択して構成されていてもよいし、複数の構造を組み合わせてもよい。
以上の形態によって本実施例では、恒温部6が異なる設定温度の複数の領域に分かれていても、伝熱部7、温度センサ部11、制御部12、電源部13からなる一つの温度制御系によって、複数の恒温領域6a~6cの温度を異なる設定温度にすることができる。これにより、本実施例の液体処理装置4では、異なる設定温度が設定された複数の恒温領域6a~6cが用意されているため、恒温部の温度昇降を待つ必要がなく、液体処理によって異なる最適な温度の恒温領域6a~6cを直ちに利用でき、液体処理のスループット向上が期待できる。また、制御系を単純化し、装置製造に関わるコストの低減が期待できる。
また、設定温度の異なる恒温領域間を分離して、実質的に断熱して熱的に独立させる構造と比べ、第一の熱抵抗部8a及び8bを介した恒温領域6a~6c間の伝熱を調整する構造によって、液体処理装置全体を小型に構築することができる。これにより、液体容器としてマルチウェルプレートを使用することが可能となり、液体容器の交換に関わる工数低減、取り違えリスクの低減が期待できる。
実施例2の液体処理装置の一形態を説明する。図7は、実施例2の液体処理装置の断面構造を示す概要図である。本実施例では、複数の温度制御系によって、三つの恒温領域をそれぞれ異なる設定温度とする構造について説明する。実施例1と重複する要素については適宜説明を省略する。
伝熱部7は、恒温部6を加熱又は冷却する。恒温領域6a~6cの設定温度が環境温度よりも高ければ、伝熱部7にはヒータなどの発熱部品を使用できる。恒温領域6a~6cの設定温度が環境温度よりも低ければ、伝熱部7にはペルチェモジュールを使用するなどして冷却すればよい。本実施例では、設定温度の異なる恒温領域6a、恒温領域6b、恒温領域6cをそれぞれ伝熱部7a、伝熱部7b、伝熱部7cで加熱又は冷却する。
本実施例では、恒温領域6a、恒温領域6b、恒温領域6cに対してそれぞれ温度センサ部11a、温度センサ部11b、温度センサ部11cが設置されている。各温度センサ部11a、温度センサ部11b、温度センサ部11cが、それぞれ恒温領域6a、恒温領域6b、恒温領域6cの温度を測定する。温度センサ部11a、温度センサ部11b、温度センサ部11cによる測定結果は、それぞれ制御部12a、制御部12b、制御部12cに入力される。制御部12a、制御部12b、制御部12cのそれぞれは、恒温領域6a、恒温領域6b、恒温領域6cの温度がそれぞれの設定温度となるように、電源部13a、電源部13b、電源部13cが伝熱部7a、伝熱部7b、伝熱部7cに供給する電力を制御する。このように、本実施例では、恒温領域6aに対して、伝熱部7a、温度センサ部11a、制御部12a、電源部13aからなる温度制御系、恒温領域6bに対して、伝熱部7b、温度センサ部11b、制御部12b、電源部13bからなる温度制御系、恒温領域6cに対して、伝熱部7c、温度センサ部11c、制御部12c、電源部13cからなる温度制御系、が設けられる。なお、プロセッサやメモリを有する一つのコントローラが制御部12a、制御部12b、及び制御部12cの機能を実行してもよい。
本実施例のように、設定温度の異なる複数の恒温領域6a~6cのそれぞれに対して、伝熱部、温度センサ部、制御部、電源部からなる温度制御系を複数備えた構造としてもよい。本実施例では、設定温度の異なる三つの恒温領域6a~6cに対して同数の温度制御系を備えた構造を示している。この構成に限らず、温度制御系の数は、一つ以上で、かつ設定温度の異なる恒温領域6a~6cの数よりも少なくしてもよい。温度制御系の数を一つとした構造は実施例1に相当する。
以上の形態によって本実施例では、恒温部6が異なる設定温度の複数の領域に分かれていても、複数の温度制御系によって、複数の恒温領域6a~6cの温度を異なる設定温度にすることができる。これにより、本実施例の液体処理装置4では、異なる設定温度の複数の恒温領域6a~6cが用意されているため、恒温部の温度昇降を待つ必要がなく、液体処理によって異なる最適な温度の恒温領域6a~6cを直ちに利用でき、液体処理のスループット向上が期待できる。また、温度制御系の数を、異なる設定温度の恒温領域6a~6cの数よりも少なくすれば、制御系を単純化し、装置製造に関わるコストの低減が期待できる。

実施例3の液体処理装置の一形態を説明する。図8は、実施例3の液体処理装置の断面構造を示す概要図である。本実施例では、一つの温度制御系によって、三つの恒温領域6a~6cをそれぞれ異なる設定温度とする構造であって、一つの伝熱部が複数の恒温領域6a~6cを加熱又は冷却する構造について説明する。実施例1と重複する要素については適宜説明を省略する。図8の例では、温度センサ部11は、恒温領域6aに設置されているが、温度センサ部11は、恒温領域6bに設置されてもよいし、恒温領域6cに設置されてもよい。
伝熱部7は、恒温部6を加熱又は冷却する。恒温領域6a~6cの設定温度が環境温度よりも高ければ、伝熱部7にはヒータなどの発熱部品を使用できる。恒温領域6a~6cの設定温度が環境温度よりも低ければ、伝熱部7にはペルチェモジュールを使用するなどして冷却すればよい。本実施例では、設定温度の異なる恒温領域6a~6cの個数が三つであるのに対して、伝熱部7の個数は一つでよい。また伝熱部7には、伝熱部7の温度を均一化するために熱伝導部材を含んでいてもよい。本実施例の伝熱部7は、ヒータ701と熱伝導部材702とを有する。熱伝導部材702は、熱伝導率の高い材料で構成されることが望ましい。例えば銅、アルミニウム、それらの合金で構成されることが望ましい。
第二の熱抵抗部14a~14cは、伝熱部7と恒温部6との間に位置し、伝熱部7から恒温部6への伝熱量を調整して、恒温領域6a~6cの温度が設定温度となるのに必要な熱抵抗値を予め備える。本実施例では、第二の熱抵抗部14aは、伝熱部7から恒温領域6aへの伝熱量を調整し、恒温領域6aが設定温度80℃になる熱抵抗値を持つ。第二の熱抵抗部14bは、伝熱部7から恒温領域6bへの伝熱量を調整し、恒温領域6bが設定温度70℃になる熱抵抗値を持つ。第二の熱抵抗部14cは、伝熱部7から恒温領域6cへの伝熱量を調整し、恒温領域6cが設定温度60℃になる熱抵抗値を持つ。第二の熱抵抗部14a~14cには、図4、図5、図6で示した第一の熱抵抗部8a及び8bの構造と同様の構造を用いて熱抵抗値を定めることができる。
第二の熱抵抗部14a~14cの構造の例として、単一の熱抵抗部材によって所望の熱抵抗値を持たせてもよい。熱抵抗部材として例えば、恒温部6より熱伝導率の低い一様の固体材料を選択して配置することができる。恒温部6と伝熱部7とに温度差を設ける点では、恒温部6より熱伝導率の低い固体材料が望ましい。一例として樹脂系材料、ゴム系材料、断熱材が用いられる。熱抵抗部材として、固体材料の他に気体層としてもよい。
第二の熱抵抗部14a~14cの他の構造の例として、恒温部6の表面又は伝熱部7(熱伝導部材702)の表面に設けた凸部と凹部によって所望の熱抵抗値を持たせてもよい。恒温部6の表面に凸部と凹部を設ける場合、恒温部6の表面に設けた凸部と伝熱部7(熱伝導部材702)の表面との間の接触面積を変化させ、凸部による固体伝導伝熱を調整して所望の熱抵抗値を持たせることができる。凹部は、熱抵抗部材と同様に、適切な熱伝導率を持った固体材料や、気体層などを用いてもよい。また、恒温部6と第二の熱抵抗部14a~14cとを一体の部品とし、穴加工をすることで凸部と凹部に相当する構造としてもよい。この場合、第二の熱抵抗部14a~14cと一体となった恒温部6と、伝熱部7との間の接触面を実質的に平面で構成できるため組立が容易となる。
第二の熱抵抗部14a~14cの他の構造の例として、スペーサと締結部材によって所望の熱抵抗値を持たせてもよい。スペーサと締結部材は、恒温部6と伝熱部7を相対的に所定の位置関係に固定する他、固体伝導伝熱を調整する役割を持たせることができる。スペーサによって固体伝導伝熱を調整する方法として、スペーサの大きさを変えて伝熱面積を変化させてもよいし、スペーサを配置する個数を変えて伝熱面積を変化させてもよいし、スペーサを熱伝導率の異なる材料を使用してもよい。例えば、スペーサは、締結部材によって締結する際に熱伝導率の低い樹脂製のワッシャを用いることができる。締結部材によって固体伝導伝熱を調整する方法として、締結部材を熱伝導率の異なる材料を使用してもよい。例えば、締結部材は、樹脂ねじを用いることができる。
第二の熱抵抗部14a~14cの他の構造の例として、恒温部6又は伝熱部7の表面に設けた熱輻射部によって所望の熱抵抗値を持たせてもよい。この構造では、恒温部6と伝熱部7の表面間の輻射伝熱を調整することで所望の熱抵抗値を持たせる。熱輻射部として、放射率を変化させる表面処理を施してもよい。例えば放射性塗料を用いてもよいし、アルマイト加工などの酸化被膜を形成してもよい。また、表面処理を施す面積によって輻射伝熱を調整して所望の熱抵抗値を持たせてもよい。
第二の熱抵抗部14a~14cの他の構造の例として、密閉部を形成し内部を減圧することで所望の熱抵抗値を持たせてもよい。密閉部は、減圧しなければ空気による対流と伝導伝熱、恒温部6と伝熱部7の表面間輻射伝熱によって熱移動する。密閉部を減圧していくことで、空気による対流と伝導伝熱の影響を減じて所望の熱抵抗値を持たせてもよい。また、真空断熱パネルのように予め内部を減圧して封止した断熱材を用いてもよい。

実施例4の液体処理装置の第三の熱抵抗部15a及び15bの一形態を説明する。図9は、実施例4の第三の熱抵抗部15a及び15bの断面構造を示す概要図である。設定温度の異なる恒温領域6a、恒温領域6b、恒温領域6cは、第一の熱抵抗部8a及び8bにより隔てられている。恒温領域6a、恒温領域6b、恒温領域6cの間の設定温度の差に対して、第一の熱抵抗部8a及び8bに十分な熱抵抗値が与えられない場合、恒温領域6a、恒温領域6b、及び恒温領域6cの温度は設定温度から外れる。このような場合であっても、液体1の温度を液体処理に最適な温度にするために、液体容器2と、恒温部6の容器保持部5との間に第三の熱抵抗部15a及び15bを設けることができる。第三の熱抵抗部15aは、液体容器2と恒温領域6bとの間に設けられ、第三の熱抵抗部15bは、液体容器2と恒温領域6cとの間に設けられる。なお、図9の例では、第三の熱抵抗部15a及び15bの両方を設けたが、第三の熱抵抗部15a又は15bの何れか一方のみを設けてもよい。また、第三の熱抵抗部を、液体容器2と恒温領域6aとの間に設けてもよい。第三の熱抵抗部15a及び15bは、恒温部6の温度が液体1の液体処理に最適な温度よりも環境温度から遠いときに、恒温部6と液体容器2の間の伝熱量を調整して、液体1の温度が液体処理に最適な所定温度となるのに必要な熱抵抗値を予め備える。第三の熱抵抗部15a及び15bには、図4、図5、図6で示した第一の熱抵抗部8a及び8bの構造と同様の構造を用いて熱抵抗値を定めることができる。
第三の熱抵抗部15a及び15bの構造の例として、単一の熱抵抗部材によって所望の熱抵抗値を持たせてもよい。熱抵抗部材として例えば、恒温部6より熱伝導率の低い一様の固体材料を選択して配置することができる。恒温部6と液体1に温度差を設ける点では、恒温部6より熱伝導率の低い固体材料が望ましい。一例として樹脂系材料、ゴム系材料、断熱材が用いられる。熱抵抗部材として、固体材料の他に気体層としてよい。
第三の熱抵抗部15a及び15bの他の構造の例として、恒温部6の表面に設けた凸部と凹部によって所望の熱抵抗値を持たせてもよい。恒温部6の表面に凸部と凹部を設ける場合、恒温部6の表面に設けた凸部と液体容器2の表面の間の接触面積を変化させ、凸部による固体伝導伝熱を調整して所望の熱抵抗値を持たせることができる。凹部は、熱抵抗部材と同様に、適切な熱伝導率を持った固体材料や、気体層などを用いてもよい。また恒温部6と第三の熱抵抗部15a及び15bとを一体の部品として穴加工をすることで凸部と凹部に相当する構造としてもよい。
第三の熱抵抗部15a及び15bの他の構造の例として、スペーサによって所望の熱抵抗値を持たせてもよい。スペーサは、恒温部6と液体容器2を相対的に所定の位置関係に固定する他、固体伝導伝熱を調整する役割を持たせることができる。スペーサによって固体伝導伝熱を調整する方法として、スペーサの大きさを変えて伝熱面積を変化させてもよいし、スペーサを配置する個数を変えて伝熱面積を変化させてもよいし、スペーサを熱伝導率の異なる材料を使用してもよい。
第三の熱抵抗部15a及び15bの他の構造の例として、恒温部6の表面に設けた熱輻射部によって所望の熱抵抗値を持たせてもよい。この構造では、恒温部6の表面の輻射伝熱を調整することで所望の熱抵抗値を持たせる。熱輻射部として、放射率を変化させる表面処理を施してもよい。例えば放射性塗料を用いてもよいし、アルマイト加工などの酸化被膜を形成してもよい。また、表面処理を施す面積によって輻射伝熱を調整して所望の熱抵抗値を持たせてもよい。
第三の熱抵抗部15a及び15bの他の構造の例として、密閉部を形成し内部を減圧することで所望の熱抵抗値を持たせてもよい。密閉部は、減圧しなければ空気による対流と伝導伝熱、恒温部6と液体容器2の表面間輻射伝熱によって熱移動する。密閉部を減圧していくことで、空気による対流と伝導伝熱の影響を減じて所望の熱抵抗値を持たせてもよい。また、真空断熱パネルのように予め内部を減圧して封止した断熱材を用いてもよい。
以上の形態によって本実施例では、恒温部6が異なる設定温度の複数の領域に分かれていても、一つの温度制御系によって、複数の恒温領域6a~6cの温度を異なる設定温度にすることができる。これにより、本実施例の液体処理装置4では、異なる設定温度の複数の恒温領域6a~6cが用意されているため、恒温部の温度昇降を待つ必要がなく、液体処理によって異なる最適な温度の恒温領域6a~6cを直ちに利用でき、液体処理のスループット向上が期待できる。また、制御系を単純化し、装置製造に関わるコストの低減が期待できる。
また、設定温度の異なる恒温部間を分離して、実質的に断熱して熱的に独立させる構造と比べ、第一の熱抵抗部8a及び8bを介した恒温領域6a~6c間の伝熱を調整することによって、液体処理装置全体を小型に構築することができる。これにより、液体容器としてマルチウェルプレートを使用することが可能となり、液体容器の交換に関わる工数低減、取り違えリスクの低減が期待できる。
なお、本開示は上記した実施形態に限定されるものではなく、様々な変形例が含まれる。例えば、上記した実施形態は本開示を分かりやすく説明するために詳細に説明したものであり、必ずしも説明した全ての構成を備えるものに限定されるものではない。また、各実施形態の構成の一部について、他の構成の追加・削除・置換をすることが可能である。
例えば、実施例1では伝熱部は一つであり、実施例2では伝熱部は3つであったが、本開示の伝熱部の個数は限定されず、例えば、3つの恒温領域に対して2つの伝熱部を設けてもよい。この場合、2つの伝熱部の少なくとも一つは、設定温度が最も環境温度から離れている恒温領域を加熱又は冷却する。
また、実施例3では、恒温領域6a~6cのそれぞれに第二の熱抵抗部14a~14cを設けたが、本開示の第二の熱抵抗部の個数は限定されず、恒温領域6a~6cから選択した一つ以上の恒温領域に第二の熱抵抗部を設けてもよい。
また、図10に示すように、核酸抽出装置101は、上記した実施例1~4の液体処理装置4を備える。そして、遺伝子検査装置110は、核酸抽出装置101を備える。核酸抽出装置101及び遺伝子検査装置110は、上記した実施例1~4の液体処理装置4を備えるため、上記した効果と同様の効果を得ることができる。

Claims (14)
- 液体が収容された液体容器を保持する容器保持部が設けられ、設定温度が異なる複数の恒温領域を有する恒温部と、
前記液体容器に収容された前記液体を吸引又は吐出する分注機と、
前記恒温部を加熱又は冷却する伝熱部と、
前記恒温部の温度に係るデータを出力する温度センサ部と、
前記伝熱部に電力を供給する電源部と、
前記温度センサ部の出力に基づいて、前記電源部が前記伝熱部へ供給する電力を制御する制御部と、
少なくとも二つの前記恒温領域を隔てる第一の熱抵抗部と、を備え、
前記第一の熱抵抗部は、前記恒温部よりも大きな熱抵抗値を有し、前記第一の熱抵抗部によって隔てられる一の恒温領域の温度が当該一の恒温領域の設定温度となるときに前記第一の熱抵抗部によって隔てられる他の恒温領域の温度が当該他の恒温領域の設定温度となる熱抵抗値を有する
ことを特徴とする液体処理装置。 - 前記第一の熱抵抗部は、前記恒温部より熱伝導率の低い一様の固体材料、気体層、スペーサ、締結部材、前記恒温部の表面に施された凹凸構造、前記恒温部の表面に施された輻射性処理、又は密閉されて減圧された空間のいずれか、若しくはこれらの組み合わせで構成される
ことを特徴とする請求項1記載の液体処理装置。 - 前記伝熱部の数が、前記恒温領域の数よりも少ない
ことを特徴とする請求項1に記載の液体処理装置。 - 前記伝熱部は複数であり、前記複数の伝熱部の少なくとも一つは、設定温度が最も環境温度から離れている前記恒温領域を加熱又は冷却する
ことを特徴とする請求項1に記載の液体処理装置。 - 前記伝熱部は一つであり、当該伝熱部は、設定温度が最も環境温度から離れている前記恒温領域を加熱又は冷却する
ことを特徴とする請求項1に記載の液体処理装置。 - 前記複数の恒温領域のうちの少なくとも二つの恒温領域に設けられる第二の熱抵抗部をさらに備え、
前記伝熱部は、前記第二の熱抵抗部が設けられた前記恒温領域を前記第二の熱抵抗部を介して加熱又は冷却し、
前記第二の熱抵抗部の各々は、一の前記第二の熱抵抗部が設けられる前記恒温領域の温度が当該恒温領域の設定温度となるときに、他の前記第二の熱抵抗部が設けられる前記恒温領域の温度が当該恒温領域の設定温度となる熱抵抗値を有する
ことを特徴とする請求項1に記載の液体処理装置。 - 前記第二の熱抵抗部の少なくとも一つは、前記恒温部より熱伝導率の低い一様の固体材料、気体層、スペーサ、締結部材、前記恒温部又は前記伝熱部の表面に施された凹凸構造、前記恒温部又は前記伝熱部の表面に施された輻射性処理、密閉されて減圧された空間のいずれか、若しくはこれらの組み合わせで構成される
ことを特徴とする請求項6に記載の液体処理装置。 - 前記容器保持部に保持される前記液体容器と前記恒温部との間に設けられる第三の熱抵抗部をさらに備え、
前記第三の熱抵抗部は、前記第三の熱抵抗部が設けられない恒温領域の温度が当該恒温領域の設定温度となるときに前記第三の熱抵抗部が設けられる恒温領域で処理される液体の温度が所定温度となる熱抵抗値を有する
ことを特徴とする請求項1に記載の液体処理装置。 - 前記第三の熱抵抗部は、前記恒温部より熱伝導率の低い一様の固体材料、気体層、スペーサ、締結部材、前記恒温部の表面に施された凹凸構造、前記恒温部の表面に施された輻射性処理、密閉されて減圧された空間のいずれか、若しくはこれらの組み合わせで構成される
ことを特徴とする請求項8に記載の液体処理装置。 - 設定温度が環境温度から最も離れている前記恒温領域以外の前記恒温領域の少なくとも一つに設けられた放熱部をさらに備える
ことを特徴とする請求項1に記載の液体処理装置。 - 前記複数の恒温領域は、三つ以上であり、
前記温度センサ部は、設定温度が環境温度から最も離れている前記恒温領域及び設定温度が環境温度に最も近い前記恒温領域以外の前記恒温領域の温度を測定する
ことを特徴とする請求項1に記載の液体処理装置。 - 前記制御部は、設定温度が異なる前記複数の恒温領域で液体処理を行う際に、前記温度センサ部が出力するデータに基づいて前記温度センサ部が設けられる前記恒温領域が設定温度となるように前記伝熱部に供給する電力を制御する
ことを特徴とする請求項11に記載の液体処理装置。 - 請求項1~12の何れか1項に記載の液体処理装置を備える
ことを特徴とする核酸抽出装置。 - 請求項1~12の何れか1項に記載の液体処理装置を備える
ことを特徴とする遺伝子検査装置。
