Definitions
- the present invention relates to the medical field, and in particular to a protein-nucleic acid aggregate for delivering biological agents and a preparation method thereof.
- Biomacromolecules such as proteins and nucleic acids form membrane-less organelles or aggregates through weak multivalent interactions and liquid-liquid phase separation (LLPS) mediated by conformational entropy, which participate in the regulation of many key cellular life activities and are closely related to a variety of human diseases. Aggregates formed by LLPS allow the enrichment of biomolecules with different physical and chemical properties, while isolating other biomolecules outside the aggregates. Aggregates, as separate compartments within cells, create an isolated environment for the enriched molecules, thereby separating and regulating biochemical reactions within cells.
- the LLPS phenomenon is not limited to playing a role in biology or pathology, but can also provide inspiring clues for biomimetic applications.
- the aggregates can absorb and recruit biomacromolecules (peptides, proteins, etc.) into the compartments, and have the advantages of efficiently enriching guest molecules and effectively retaining their biological activity, showing the potential as a delivery carrier for biological agents.
- biomacromolecules peptides, proteins, etc.
- the aggregates reversibly and dynamically formed by LLPS are not restricted by the membrane structure, are easy to aggregate, and are affected by environmental factors such as temperature, pH, ionic strength, etc., which limits their application in the field of drug delivery.
- the present invention designs and generates a new delivery carrier, which is composed of protein and nucleic acid agglomerates, wherein the agglomerates are hollow.

- the globular part uses the electrostatic interaction between Arg and the phosphodiester backbone to wrap the DNA molecule around the histone core to form a nucleosome.
- the tail contains a large amount of Arg and Lys, which is the site of post-translational modification.
- H1 is diverse and has genus and tissue specificity.

Description

溶剂为1XPBS(pH 7.4)。
