Abstract
본 발명은 고분자 물질과 광감응제를 포함하는 광감음성 고분자 필름 제조방법과 상기 광감응성 고분자 필름에 광조사하는 단계를 포함하는 세포의 증식 촉진 방법 및 세포 분리 방법에 관한 것이다. 본 발명에 따른 광감응성 고분자 필름은 하나의 광감응성 고분자 필름에서 세포 배양, 성장 촉진, 세포시트 분리 및 전달의 모든 과정이 광에너지의 밀도와 조사 시간만 조절함에 따라 한 번에 이루어짐으로써 낮은 비용과 간단한 절차, 적은 소요 시간에 따라 이루어 질 수 있다.
Description
본 발명은 고분자 물질과 광감응제를 포함하는 광감음성 고분자 필름 제조방법과 상기 광감응성 고분자 필름에 광조사하는 단계를 포함하는 세포의 증식 촉진 방법 및 세포 분리 방법에 관한 것이다.
광감응제란 광에너지 조사에 의해 활성산소(Reactive oxygen species, ROS)를 발생시키는 물질을 말한다. 활성산소는 세포 분리를 유도하는 분자 기전의 초기 단계에 중요한 역할을 하므로, 최근 광감응제를 이용한 세포 분리의 가능성이 제시되고 있다. 세포 배양의 지지체로 많이 사용되는 고분자로부터의 세포 시트 분리 방법에 있어서 온도감응형 세포 배양 지지체 및 이를 이용한 세포 시트 분리에 관한 기술은 개시되어 있으나, 광감응형 세포 시트 분리 기술 개발은 아직 미비한 실정이다.
활성산소종(Reactive oxygen species, ROS)은 과산화수소(H2O2), 슈퍼옥사이드(O2
-), 하이드록실라디칼(OH · ), 1중항 산소(1O2)와 같은 반응성이 큰 이온 또는 분자를 총칭하는 용어이다. 활성산소종은 산소의 정상적인 대사작용에 의해서 자연스럽게 생기며, 세포신호와 항상성에 중요한 역할을 한다. 활성산소종의 농도는 자외선이나 높은 열에 노출되는 것처럼 환경적인 스트레스로 인하여 매우 빠르게 증가할 수 있다.
즉, 활성산소종은 생물학적 기능에 있어서 필수적인 역할을 하는 물질로서, 상기 활성산소종은 단백질, 전사인자 및 유전자에 직접 작용하여 이들의 구조를 변화시키거나 또는 이들의 기능을 조절함으로써 다수의 신호전달 경로를 조절한다. 또한 일정량의 활성산소종 증가는 세포의 증식을 촉진할 수 있으나, 과량의 활성산소종은 지질, 단백질 및 DNA를 산화적으로 손상시키는 원인이 될 수 있다.
한편, PDT('광역학처리' 또는 '광역학치료')란 광감응제(혹은 '광민감제' 또는 '광감각제'라고도 함)로 알려진 비독성 약물 또는 염료를 암을 포함하는 병변을 가진 환자에게 전신적 또는 국소적으로 투여하는 것으로 정의할 수 있다. 그리하여 빛과 광감응제의 상호작용에 의해 에너지 전달이 일어나면서 국소적인 화학적 효과가 발생된다. 이러한 PDT에 관련된 종래 기술은 활성산소종을 통해 특정 부위의 세포를 사멸시키는 기술이 있었다. 하지만, 이러한 종래 기술은 활성산소종을 통해 세포를 사멸시키는데 연구의 초점이 맞추어져 있었을 뿐, 활성산소종을 통해 세포를 증식시키는 것에 관하여는 현재 연구가 미진한 실정이다.
또한 상기 세포 성장을 촉진시키는 단계와 세포시트 분리 단계는 필요한 광감응제의 양, 광조사 에너지의 수준에 차이가 있어서 각각 별도의 단계로 이루어질 수 있을 뿐, 동일한 광감응제의 양 하에서 광에너지 밀도와 조사 시간의 차이만으로 세포 성장 촉진 및 세포시트 분리와 전달이 모두 가능한 기술 개발은 미비한 실정이다.
본 발명은 상기와 같은 종래의 기술상의 문제점을 해결하기 위해 안출된 것으로, 고분자 물질과 광감응제를 포함하는 광감음성 고분자 필름 제조방법과 상기 광감응성 고분자 필름에 광조사하는 단계를 포함하는 세포 성장 촉진 및 세포시트 분리와 전달이 모두 가능한 올인원 세포제어 기법에 관한 것이다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당 업계에서 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
본 발명의 일 구체예에서 세포 시트(cell sheet)란, 독립된 단일 세포(single cell)와 구별되는 개념으로서, 단일 세포들이 모여 세포외 기질(Extracellular matrix, ECM)을 공유하며 서로 연결됨으로써 단층 또는 중층의 세포층을 이루고 있는 상태를 의미한다. 체외 배양시 한정된 공간 안에서 세포의 밀집도가 높아져 세포층을 이루는 경우에도, 트립신 등의 화학적 또는 물리학적 기질 분해로 인해 세포층의 상태가 다시 독립된 단일 세포로 전환된다면 세포 시트의 상태를 유지하였다고 정의하기 어렵다. 본 발명에 있어서 세포 시트란, 섬유아세포(fibroblast), 바람직하게는 피부 유래의 섬유아세포(Human neonatal dermal fibroblast)로 구성된 세포층을 의미하나, 여기에 피부각질세포(adult normal human epidermal keratinocyte)로 구성된 단층의 세포층을 중첩하여 다중층 형태의 복합 세포 시트를 제조하는 것도 가능하다.
본 발명에서 상기 세포 시트는 화장품 또는 화장품을 테스트하는 데에 이용할 수 있다.
본 발명의 일 구체예에서 광감응제(Photosensitivie materials)란, 가시광선 등의 광에너지에 의해 분자 상태를 변화시키는 물질을 총칭하는 것으로서, 본 명세서에 있어서 광감응제란 광에너지 조사에 의해 활성산소(Reactive oxygen species, ROS)를 발생시키는 물질을 의미하나, 이에 한정되는 것은 아니다. 본 명세서 사용된 광감응제는 헤마토포피린(Hematoporphyrin, Hp), 아미노 레뷰리닉산(ALA, 5-Aminolevulinic Acid) 및 클로린(Chlorin)이며 바람직하게는 클로린 e6(hlorin e6)일 수 있으나, 이에 한정되는 것은 아니다. 광감응제가 광에너지에 의해 자극되어 발생시키는 활성산소는 세포 분리를 유도하는 분자 기전의 초기 단계에 중요한 역할을 하므로, 이를 이용하여 세포 분리를 유도할 수 있다.
본 발명의 일 구체예에 따르면, (a) 고분자 물질과 광감응제를 용매에 용해시켜 용액을 제조하는 단계; 및 (b) 상기 제조된 용액을 스핀코팅(spin-coating) 방식으로 넓게 펼치는 단계;를 포함하는 광감응성 고분자 필름 제조방법을 제공한다.
상기 스핀코팅(spin-coating) 방식이란 얇고 균일한 층을 제작하기 위해 미세 전자 산업에서 널리 사용되는 기법으로, 유체를 기판 위에 올려놓고 고 속으로 회전시키면서 원심력에 의해 유체를 넓게 펼치는 방식을 말하나 이에 제한되는 것은 아니다.
상기 용매는 용제라고도 불리는 것으로 어떤 물질을 녹여 용액으로 만들 때 쓰이는 물질을 말한다. 용매는 용액의 매체가 되어 용질을 녹이는 물질로, 주로 액체나 기체상을 띤다. 예를 들어 액체에 물질을 녹여 용액을 만들 때나 액체에 액체가 녹아들어가는 경우에 그 양이 많은 쪽의 액체를 용매라고 하며, 용액 중에서 용매는 용질에 비해 용액을 구성하는 비율이 높다. 본 발명에서 용매는 극성 용매인 것이 바람직하나 이에 제한되는 것은 아니며, 상기 용매는 메탄올, 에탄올, 프로판올, 아세트산, 글리세린, 메틸렌 클로라이드, 아세톤, 에틸 아세테이트, DMSO, DMF, 아세토나이트릴, 피리딘, 플루오르알콜로 이루어진 그룹에서 선택되는 어느 하나 이상일 수 있으며, 바람직하게는 헥사플루오로이소프로판올(hexafluoroisopropanol, HFIP)이나 이에 제한되는 것은 아니다.
또한 상기 고분자물질에 도입되는 광감응제의 농도를 조절하면, 혈관구성세포의 증식을 가능하게 하는 적절한 양의 활성산소종 생성을 유도하여 혈관구성세포의 증식을 촉진시키게 된다. 결국, 이러한 고분자물질에 광감응제가 도입된 상기 광감응성 고분자 필름은 광조사시 혈관구성세포를 증식시키기 위한 적절한 양의 활성산소종 생성을 가능하게 하는 고분자 필름인 것이다. 또한 상기 혈관구성세포를 증식시키기 위한 적절한 양의 활성산소종은 저농도로 생성되는 것이다.
한편, 상기 광감응성 고분자 필름은 적절한 수준의 광조사시 혈관구성세포의 증식 촉진을 가능하게 할 정도의 활성산소종 생성을 가능하게 하는데, 이러한 경우 상기 광감응제는 0.01~10㎍/㎠ 농도인 것이 바람직하다. 상기 광감응제가 상기 농도로 도입되지 않으면 혈관구성세포의 증식을 촉진할 수 있는 정도로 저농도의 활성산소종을 생성할 수 없어 바람직하지 않다.
한편, 상기 혈관구성세포는 인공혈관의 이식에 사용될 수 있는 것이라면 특별한 제한이 있는 것은 아니지만, 바람직하게는 혈관내피세포, 혈관평활근세포, 섬유아세포(fibroblast) 등이 이에 해당할 수 있으며, 특별한 제한이 있는 것은 아니지만 더욱 바람직하게는 혈관내피세포일 수 있다.
한편, 상기 고분자 필름은 광감응제가 도입되어 활성산소종을 생성하는 것을 가능하게 하는 것이라면 특별한 제한 없이 모두 이에 포함될 수 있으나, 본 발명에 적절한 고분자 필름은 폴리우레탄(PU), PLGA(poly(lactic-co-glycolic acid)), PLLA(Poly(L-lactic acid)), PET(polyethylene terephthalate) 및 폴리케톤(PK)으로 이루어지는 군으로부터 선택된 어느 하나 이상인 것이 바람직하다.
또한 상기 광감응제는 특별한 제한이 있는 것은 아니지만 본 발명에 적절한 광감응제는 바람직하게는 헤마토포피린(Hp), 아미노 레뷰리닉산(ALA, 5-Aminolevulinic Acid) 및 클로린(Chlorin)으로 이루어지는 군으로부터 선택되는 어느 하나 이상인 것이 바람직하다.
또한 활성산소종은 적절한 양으로 생성되어 혈관구성세포의 증식을 가능하게 하는 것으로서 특별한 제한이 있는 것은 아니지만 바람직하게는 과산화수소, 슈퍼옥사이드, 하이드록실라디칼, 1중항 산소, 하이드로젠 퍼옥사이드 및 산화질소(NO)로 이루어지는 군으로부터 선택되는 어느 하나 이상일 수 있다.
본 발명의 일 구체예에서 지지체란, 세포가 부착되어 배양될 수 있는 기반이 되는 재료를 의미하며, 바람직하게는 피브린 겔 지지체 또는 하이드로 젤 지지체일 수 있으나 이에 제한되는 것은 아니다. 본 발명에서 광감응성 고분자 지지체란 광감응제가 도입된 고분자 화합물로서의 지지체를 의미하며 본 발명의 목적상 광기능성 고분자 지지체는 세포 배양을 위한 지지체로서 사용될 수 있다.
본 발명에서, 접시는 직접적인 세포의 배양이 이루어질 수 있는 것이면 제한되지 않으나, 바람직하게는 O×P개의 웰을 가지는 플레이트, 디시(Dish) 또는 플라스크(Flask)인 것을 특징으로 할 수 있다. 더욱 바람직하게는 페트리 디쉬(Petri dish)일 수 있다. 이때, O 및 P은 각각 2 이상의 자연수로, 바람직하게는 2 내지 64의 자연수, 더욱 바람직하게는 2 내지 32의 자연수이다. 예시적으로 6/12/24/48/96/1024 웰 플레이트가 사용되거나, 35/60/100 Ψ 디시와 같은 원형 플레이트나 T25/75 플라스크와 같은 배양플라스크가 사용될 수 있다.
본 발명의 일 실시예에서는, 광감응제와 고분자 물질이 함유된 광감응성 고분자 필름을 포함하는, 세포의 생육용 또는 분리용 플레이트를 제공한다.
본 발명의 다른 실시예에서는, 상기 고분자 물질은 폴리우레탄(PU), PLGA(poly(lactic-co-glycolic acid)), PLLA(Poly(L-lactic acid)), PET(polyethylene terephthalate) 및 폴리케톤(PK)으로 이루어지는 군으로부터 선택되는 어느 하나 이상인, 세포의 생육용 또는 분리용 플레이트를 제공한다.

본 발명의 다른 실시예에서는, 상기 (a) 단계에서의 광조사는 광에너지를 0.01㎽/㎠ 내지 0.2㎽/㎠로 5분 내지 1시간 조사하는 것인, 세포의 생육 및 분리 방법을 제공한다.
본 발명의 다른 실시예에서는, 상기 (f) 단계에서의 광조사는 광에너지를 1㎽/㎠ 내지 50㎽/㎠로 조사하는 것인, 세포의 생육 및 분리 방법을 제공한다.
이상에서 본 발명에 대하여 상세하게 설명하였지만 본 발명의 권리범위는 이에 한정되는 것은 아니고, 청구범위에 기재된 본 발명의 기술적 사상을 벗어나지 않는 범위 내에서 다양한 수정 및 변형이 가능하다는 것은 당 기술분야의 통상의 지식을 가진 자에게는 자명할 것이다.
일정량의 활성산소종 증가는 세포의 증식을 촉진할 수 있으나, 과량의 활성산소종은 지질, 단백질 및 DNA를 산화적으로 손상시키는 원인이 될 수 있으며 활성산소종은 세포 분리를 유도하는 분자 기전의 초기 단계에 중요한 역할을 하는 것으로 알려져 있다. 그러나 상기 세포의 증식 단계와 세포 분리 단계에서 필요한 활성산소종의 농도에는 차이가 존재한다. 즉, 세포 성장을 촉진시키는 단계와 세포시트 분리 단계는 필요한 광감응제의 양, 광조사 에너지의 수준에 차이가 있어서 각각 별도의 단계로 이루어질 수 밖에 없는데, 본 발명에 따른 광감응성 고분자 필름과 올인원 세포제어 방법에 따르면 동일한 광감응제를 포함한 광감응성 고분자 필름 하에서 광에너지 밀도와 조사 시간의 차이만으로 세포 배양, 성장 촉진, 세포시트 분리 및 전달의 전 과정이 모두 하나의 광감응성 고분자 필름에서 가능한 효과를 가지고 있다.
도 1은 광감응성 고분자 필름과 적외선 영역 파장대의 광에너지 조사 장비를 사용하는 올인원 세포제어 기법의 전 과정을 나타내는 모식도이다. 본 모식도에서 나타낸 바와 같이, 본 발명에 따른 올인원 세포제어 기법은 광에너지의 조사량과 조사 시간만 조절함으로써, 세포 증식과 세포 시트 분리가 하나의 광감응성 고분자 필름에서 이루어질 수 있다.
도 2는 광감응성 고분자 필름에서 광에너지 밀도 및 조사 시간의 변화에 따른 활성산소 발생 농도를 확인한 결과이다. 즉, 동일한 타입의 광감응성 고분자 필름에서 광에너지 밀도 및 조사 시간의 증가에 따라 활성산소의 발생량이 증가하는 것을 확인한 그래프이다. 특히 도2의 high power density의 DPBF degradation은 20mW/cm2로 조사한 결과를 나타내는 것이다.
도 3은 광에너지 조사 전/후 세포 증식율을 비교한 결과이다. 즉, 광감응성 고분자 필름 위에서 피부유래 섬유아세포(human dermal fibroblasts)를 배양하며 ㎽/㎠의 광에너지를 30분간 조사한 후, 조사 전과 조사 후 5일간 세포 증식율을 확인한 결과 광에너지 조사 후 세포 증식이 촉진되고 유도됨을 확인하였다.
도 4는 광에너지 조사 전/후 광감응성 고분자 필름에서 세포 분리를 확인한 결과이다. 즉, 광감응성 고분자 필름에서 피부유래 섬유아세포(human dermal fibroblasts)를 배양하여 세포시트 형성 유도 후 10-20㎽/㎠의 광에너지 조사 시 필름으로부터 세포가 분리되어 필름 위 부착되어 있는 세포의 비율이 낮아짐을 확인하였다.
도 5는 세포시트 분리 기법을 이용한 하이드로 젤 지지체로의 세포 전달 효과 및 세포 생존 확인한 결과이다. 즉, 세포시트 분리 조건을 이용하여 세포시트가 배양된 광감응성 고분자 필름을 하이드로 젤 지지체로 옮겨 광에너지 조사 시 세포시트가 필름으로부터 분리되어 하이드로 젤로 전달이 가능함을 확인하였다. 도 5에서 녹색 형광은 생존 세포를 표지하는 것으로 세포 손상 없이 전달된 것을 의미한다.
도 6은 폴리우레탄(polyurethane) 고분자로 제작한 광감응성 고분자 필름은 신축성 있고 다공성 구조를 가지고 있어 고농도의 헤마토포르피린(hematoporphyrin) 광감응제를 도입했을 때, 필름으로부터 광감응제가 10㎍/㎠의 경우 약 7.5%, 20㎍/㎠의 경우 약 54.4%가 방출되는 것을 확인한 결과이다.
도 7은 본 발명에서 사용한 폴리케톤(polyurethane) 고분자로 제작한 광감응성 고분자 필름은 고농도의 헤마토포르피린(hematoporphyrin) 광감응제를 도입했을 때, 필름으로부터 광감응제가 10㎍/㎠의 경우 약 2.87%, 50㎍/㎠의 경우 약 5.46%가 방출되는 것을 확인한 결과이다.
도 8은 기존에 세포시트 분리를 위해 사용된 광감응성 고분자 필름에 광에너지 조사 장비로 균일하게 조사할 수 있는 최소 광량을 조사했을 때, 과도한 활성산소종의 발생으로 인한 세포 파괴가 발생하지 않고 세포 증식 촉진에만 영향을 줄 수 있는 낮은 수준의 활성산소 발생량을 얻을 수 있는 최소 광량을 유지하기 어렵다는 것을 보여주는 결과이다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을보다 구체적으로 설명하기 위한 것으로서, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
제조예 1. 광감응성 고분자 필름의 제조
0.1-2㎍/㎠ 농도의 클로린 e6(chlorin e6) 광감응제를 이용하여 광감응성 고분자 필름을 제조하고자 하였다. 구체적으로, 펠렛 형태의 폴리케톤(polyketone, PK)고분자와, 특히 1㎍/㎠ 농도의 클로린 e6(chlorin e6) 광감응제를 HFIP(hexafluoroisopropanol(1,1,1,3,3,3,-Hexafluoro-2-propanol)) 용제에 용해시켜 얻은 용액을 리에틸렌 테레프탈레이트(Polyethyleneterephthalate, PET) 고분자 필름 위에 일정량 분주하여 3000rpm의 회전속도로 약 40초에서 60초 동안 스핀코팅(spin coating)하는 방식으로 광감응성 고분자 필름을 제작하였다.
실시예 1. 광감응성 고분자 필름에서의 활성산소 발생 여부 확인
상기 제조예 1의 방법으로 제조된 광감응성 고분자 필름에 광에너지를 조사하였을 때 발생하는 활성산소의 양을 DPBF(1,3-diphenylisobenzofuran)를 이용하여 측정하였다. DPBF(1,3-diphenylisobenzofuran)는 활성산소에 의해 분해되므로, DPBF의 분해 정도를 411nm 파장에 대한 DPBF의 흡광도로 측정함으로써 광에너지 조사에 의해 발생된 활성산소의 양을 확인할 수 있다. DPBF 용액(1.85 X 10-5 M)에 필름이 충분히 잠기도록 넣은 후, 자체 제작한 녹색광 LED(Green LED) 장비로 50 ㎽/㎠의 광에너지를 조사하여 5분 간격으로 위 용액을 회수하여 DPBF의 흡광도를 UV-Vis 분광광도계로 측정하였다. 실험결과 동일한 타입의 광감응성 고분자 필름에서 광에너지 밀도 및 조사 시간의 증가에 따라 활성산소의 발생량이 증가하는 것을 확인할 수 있었으며 이는 도 2에 나타내었다.
실시예 2. 활성산소 발생에 따른 세포 성장 촉진 확인
PDT에 의한 세포증식 유도를 MTT 분석에 의해 평가하였다. 광역학요법은 광감응제를 표적세포에 침투시켜 특정 파장의 광자를 흡수하고 세포내에 존재하는 산소분자에 여기에너지를 전달하도록 함으로써 활성산소종의 농도를 높이는 것이다. 활성산소종 농도가 임계값 이상으로 올라가면 비가역적인 세포사멸이 유도된다. 즉, 상기 제조예 1에서 제조한 광감응성 고분자 필름 위에서 피부유래 섬유아세포(human dermal fibroblasts)를 배양하며 0.05㎽/㎠의 광에너지를 30분간 조사한 후, 조사 전과 조사 후 5일간 세포 증식율을 확인한 결과 0.05㎽/㎠의 광에너지를 30분간 조사시 비가역적인 세포사멸이 유도되는 것이 아니라 세포 증식이 촉진됨을 확인하였다. 이는 도 3에 나타내었다.
실시예 3. 활성산소 발생에 따른 세포 분리 여부 확인
상기 제조예 1의 방법으로 제조된 광감응성 고분자 필름 1.8 ㎠ 면적 당 5 X 104 개의 피부유래 섬유아세포를 접종하고, 24시간 동안 배양하였다. 배양 후에는, 광에너지 발현 장치로 녹색광을 조사하였고, 이 때의 조사광량과 조사 시간에 따른 세포의 변화를 MTT 시험법으로 분석하였다. 예비실험을 통하여 조사광량이 50 ㎽/㎠을 초과할 경우 세포들이 회복 불가능한 손상을 입는다는 것과, 20 ㎽/㎠의 광량을 10분 이상 조사할 경우 광에너지 발현 장치로부터 발생한 고열이 세포에 영향을 준다는 것을 확인하였으므로, 본 실시예에서의 광에너지 조사는 10~20 ㎽/㎠의 광량을 10분 이내로 조사하는 것으로 하였다. 구체적으로 1회 실험에서는 10㎽/㎠의 광량을 5분간 조사하였고, 2회 실험에서는 20㎽/㎠의 광량을 또한 5분간 조사하여 세포시트가 분리되는지 확인하였다. 실험결과 2회 실험에서 모두 세포 분리가 일어남으로 인하여, MTT 시험법을 통한 측정값이 감소하였는 바 광에너지 조사시 세포가 분리되어 필름 위 부착되어 있는 세포의 비율이 낮아지는 것을 확인하였다. 상기 결과를 도 4에 나타내었다.
제조예 2. 하이드로젤 지지체 제조
고분자 필름으로부터 피부 유래 단일세포층을 전달할 수 있는 광감응성 세포전달시스템을 개발하기 위해, 온도에 따른 물성의 변화가 없는 피브린 젤(Fibrin gel)을 이용하여 하이드로젤 지지체(Hydrogel)를 제작하였다. 이를 위해, 먼저 10 ㎎/㎖의 피브리노겐(Fibrinogen)과 50 units/㎖의 트롬빈(Thrombon)을 혼합하였고, 혼합액을 37℃에서 1시간 동안 응고시켜는 방법으로 피브린 젤을 제조하였다.
실시예 4. 광감응성 고분자 필름에서 젤 지지체로 세포 전달 여부 확인
상기 제조예 1의 방법으로 제조된 광감응성 고분자 필름 1.1 ㎠ 면적 당 5 X 104 개의 피부유래 섬유아세포를 접종하고, 24시간 동안 배양한 후, 필름에 부착되어 있는 세포층을 피브린 젤과 접촉하도록 위치시키고 광에너지 발현장치로 광에너지 조사는 10~20 ㎽/㎠의 광량을 1분내지 5분간 조사하였다. 광량 조사 1시간 후에 광감응성 고분자 필름 과 피브린 젤에서의 세포의 부착 정도와 활성산소 발생에 의해 분리된 세포의 생존 상태를 세포사 분석법(live/dead cell assay)으로 확인하였다. 세포사 분석법은 두 가지 형광 프로브를 사용하여 세포의 생존여부를 확인하는 방법으로, 살아있는 세포는 칼세인 AM(calcein AM)에 의해 녹색 형광으로 염색되고, 죽은 세포는 EthD-1(ethidium homodimer-1)에 의해 적색 형광으로 염색되는 방법이다. 실험 결과, 광에너지를 조사하지 않은 경우와 비교하여 10~20 ㎽/㎠의 광량을 조사한 경우에서 더 많은 세포가 광기능성 고분자 필름으로부터 피브린 젤 지지체로 손상없이 전달된 것을 관찰하였다. 상기 결과를 도 5에 나타내었다.
비교예 1. 광감응성 고분자 필름의 광감응제 용출 시험
상기 제조예 1의 방법으로 제조된 광감응성 고분자 필름으로부터 도입된 Hp의 용출 정도를 측정하기 위하여 용출시험을 실시하였다. 0.5 ㎖의 DPBS (Dulbecco`s Phosphate Buffered Saline) 용액에 필름이 충분히 잠기도록 넣은 후, 37℃ 항온수조에서 50 rpm의 진동속도로 흔들어 주면서 각 시간별로 필름으로부터 Hp의 용출을 유도하였다. 그 다음 용출용액에서 필름을 제거한 후 DPBS로 용출된 Hp를 1 ㎖의 순수 에탄올(Absolute ethanol)을 첨가하여 재용해 시켜 Hp의 형광스펙트럼을 UV-Vis 분광광도계(UV-Vis spectrophotometer)로 측정하였다.
70시간 이상의 용출시험 결과, 신축성 있고 다공성 구조를 가지고 있어 고농도의 헤마토포르피린(hematoporphyrin) 광감응제를 도입했을 때, 필름으로부터 광감응제가 10㎍/㎠의 경우 약 7.5%, 20㎍/㎠의 경우 약 54.4%가 방출되는 것을 확인하였으며, 본 발명에서 사용한 폴리케톤(polyurethane) 고분자로 제작한 광감응성 고분자 필름은 고농도의 헤마토포르피린(hematoporphyrin) 광감응제를 도입했을 때, 필름으로부터 광감응제가 10㎍/㎠의 경우 약 2.87%, 50㎍/㎠의 경우 약 5.46%가 방출되는 것을 확인하였다. 이는 도 6 및 7에 나타내었다. 즉, 헤마토포르피린(hematoporphyrin) 광감응제와 초록 빛(green light) 사용을 통해 세포 시트를 분리하기 위해서는 광감응제 농도가 10㎍/㎠ 이상의 필름이 요구되는데, 본 발명인 광감응성 고분자 필름 제조방법 따른 필름과 달리 폴리우레탄 필름은 필름 안에 광감응제가 고정되어 있지 못하고 상당한 양이 용출되기 때문에 이로 인한 광감응제 농도 차이에 따른 세포 사멸 등 부작용을 야기할 수 있으며, 균일한 양의 활성산소종 발생이 어려운 단점이 있다.
비교예 2. 광감응성 고분자 필름의 세포증식 비교 실험
세포 분리를 위한 광기능성 고분자 필름에 세포 증식 촉진을 위한 광에너지 수준을 조사하는 경우에 세포 증식 촉진이 일어나는지 비교 시험을 진행하였다.
즉, 세포 분리를 위한 광기능성 고분자 필름에 광에너지를 조사하였을 때 발생하는 활성산소의 양을 DPBF(1,3-diphenylisobenzofuran)를 이용하여 측정하였다. DPBF(1,3-diphenylisobenzofuran)는 활성산소에 의해 분해되므로, DPBF의 분해 정도를 411nm 파장에 대한 DPBF의 흡광도로 측정함으로써 광에너지 조사에 의해 발생된 활성산소의 양을 확인할 수 있다. DPBF 용액(1.85 X 10-5 M)에 필름이 충분히 잠기도록 넣은 후, 자체 제작한 녹색광 LED(Green LED) 장비로 50 ㎽/㎠의 광에너지를 조사하여 5분 간격으로 위 용액을 회수하여 DPBF의 흡광도를 UV-Vis 분광광도계로 측정하였다.
실험 결과 세포증식 촉진을 유도하기 위해서는 최소한 30분의 광에너지를 조사해야 하는데, 세포시트 분리에 사용한 광기능성 고분자 필름에 광에너지 조사 장비로 균일하게 조사할 수 있는 최소 광량을 조사하였다. 그 결과 도 8에서 보여주는 것처럼 광에너지가 너무 작아 세포증식촉진에 영향을 준 낮은 레벨의 활성산소 발생량을 맞추기 어려워 세포증식 촉진 연구에 적용하기 어렵다는 것을 확인할 수 있었다.
결론
상기 실시예 및 비교예에 따를 때, 세포 증식을 위한 광감응성 고분자 필름과 세포 시트 분리를 위한 광감응성 고분자 필름은 각각 광감응제의 종류와 농도에 차이가 있어 하나의 광감응성 고분자 필름에서 세포 증식과 세포 시트 분리가 이루어지지 못하고, 별도의 광감응성 고분자 필름에서 각 단계가 별도로 이루어짐에 따라 고비용과 복잡한 절차, 긴 소요 시간이 필요한 문제가 있는데 비하여 본원발명에 따를 때, 하나의 광감응성 고분자 필름에서 세포 배양, 성장 촉진, 세포시트 분리 및 전달의 모든 과정이 광에너지의 밀도와 조사 시간만 조절함에 따라 한 번에 이루어짐으로써 낮은 비용과 간단한 절차, 적은 소요 시간에 따라 이루어 질 수 있다는 것을 확인할 수 있었다.
