Abstract

本発明は、ミトコンドリア活性化作用などの優れた作用を示すアンチエイジング剤に関するものである。
日本における平均寿命は20世紀後半に著しい伸長を遂げ、世界有数の長寿国となった。その一方で、加齢による問題も健在化してきている。例えば、加齢による老化の表現型としては、認知力の低下、筋力の衰え、骨粗鬆症、内臓脂肪の増加、心血管機能の低下、不眠などが挙げられる。
かつては加齢のプロセスは非常に複雑で、介入など不可能であると思われていた。しかし近年、科学の進歩により、加齢は細胞生物学的なプロセスの一つとして介入の可能性があることが明らかにされ、加齢という生物学的プロセスに介入し、加齢に伴う動脈硬化やがんといった加齢関連疾患の発症確率を下げ、生活の質(QOL:Quality Of Life)を維持しつつ健康長寿をめざすアンチエイジング技術が検討されている。
ところで、藻類の中には光合成により多糖類を生合成し、多量に蓄積するものがある。例えば褐藻類が産生するアルギン酸は、カルシウムイオンやマグネシウムイオン等の二価金属イオンによりゲル化するといった性質から、増粘剤、安定剤、ゲル化剤、食物繊維素材などとして、食品分野、医療分野、工業分野で利用されている。
また、例えばアオサ属の大型緑藻類であるミナミアオノリは、全植物の中でもトップクラスの成長速度を有し、バイオマス資源としての利活用が期待されている。ミナミアオノリは、藻体乾燥重量あたり5~15%程度のデンプンやセルロースを含む他、約20~30%のウルバンという多糖類を含む。
例えばウルバンは、ヒアルロン酸と組み合わせて、皮膚加齢の兆候への対処や、皮膚創傷の処置および治療に使用することが検討されている(特許文献1)。
上述したように、藻類から得られるウルバンをヒアルロン酸と組み合わせて皮膚加齢の兆候への対処などに用いることは既に検討されている(特許文献1)。
しかし、特許文献1には、黄藻植物であるAscophyllum nodosum由来のフコイダンリッチ粗抽出物製品であるAscophyscient(登録商標)とヒアルロン酸との組み合わせが、繊維芽細胞の増殖を有意に促進することを示す実験データが記載されているのみである。繊維芽細胞は、結合組織を構成する細胞であり、コラーゲン、エラスチン、ヒアルロン酸といった真皮成分を産生するものではあるが、繊維芽細胞の増殖を促進する成分が実際に肌のハリ等を改善するか否かは不明であるし、また、老化一般を抑制するか否かは勿論不明である。
そこで本発明は、ミトコンドリア活性化作用などの優れた作用を示すアンチエイジング剤を提供することを目的とする。
しかし、特許文献1には、黄藻植物であるAscophyllum nodosum由来のフコイダンリッチ粗抽出物製品であるAscophyscient(登録商標)とヒアルロン酸との組み合わせが、繊維芽細胞の増殖を有意に促進することを示す実験データが記載されているのみである。繊維芽細胞は、結合組織を構成する細胞であり、コラーゲン、エラスチン、ヒアルロン酸といった真皮成分を産生するものではあるが、繊維芽細胞の増殖を促進する成分が実際に肌のハリ等を改善するか否かは不明であるし、また、老化一般を抑制するか否かは勿論不明である。
そこで本発明は、ミトコンドリア活性化作用などの優れた作用を示すアンチエイジング剤を提供することを目的とする。
本発明者らは、上記課題を解決するために鋭意研究を重ねた。その結果、天然ウルバンの特定の誘導体が、客観的に優れたアンチエイジング作用を示すことを見出して、本発明を完成した。

本発明に係るアンチエイジング剤は、老化マーカーの低減、コラーゲンの産生促進、抗酸化酵素の発現の促進、ミトコンドリアの活性化など、客観的で且つ優れたアンチエイジング作用を示す。また、本発明に係るアンチエイジング剤の有効成分であるウルバン誘導体は、グルクロン酸や硫酸化されたラムロース等が重合した多糖類であり、安全性は高いと考えられる。よって本発明は、優れたアンチエイジング剤に関する技術として、産業上非常に優れている。
本発明に係るアンチエイジング剤は、有効成分としてH型ウルバンまたはウルバンのアルカリ金属塩を含有する。
ウルバンは、ミナミアオノリ等のアオノリ属(Enteromorpha sp.)緑藻類や、ミナミアオサ等のアオサ属(Ulva sp.)緑藻類が産生する多糖類であり、グルクロン酸やイズロン酸などのウロン酸と、硫酸基を有するラムノース、ガラクトース、キシロース、グルコース等を含む硫酸化アニオン性多糖類である。ウルバン中、ウロン酸と硫酸化ラムノース等が交互重合している部分が約2/3~3/4あり、その他の部分では硫酸化ラムノース等が主に重合している。以下、天然由来のウルバンを天然ウルバンという場合がある。
天然ウルバンの主な構造単位としては、以下の2つがある:
・1→4型結合によってβ-D-グルクロン酸と3-サルフェートα-L-ラムノースが連結しているウルバノビウロン酸3-サルフェート型A;
・1→4型結合によってα-L-イズロン酸と3-サルフェートα-L-ラムノースが結合しているウルバノビウロン酸3-サルフェート型B。
・1→4型結合によってβ-D-グルクロン酸と3-サルフェートα-L-ラムノースが連結しているウルバノビウロン酸3-サルフェート型A;
・1→4型結合によってα-L-イズロン酸と3-サルフェートα-L-ラムノースが結合しているウルバノビウロン酸3-サルフェート型B。
天然ウルバンは、主にマグネシウムイオン、カルシウムイオン、ナトリウムイオン、及びカリウムイオンを含み、カチオンの内マグネシウムイオンを最も多く含む。但し、アルギン酸などに比べてウルバンはゲル化し難く、マグネシウムイオンを多く含んでいてもゾル状態にあることが多い。
天然ウルバンは、ミナミアオノリ等、ウルバンを含む藻類から常法により抽出することができる。例えば、ウルバンは水溶性であるので、原料藻類を微細化した後、溶媒として水などを加え、80℃以上、120℃以下程度で抽出すればよい。ウルバンは一般的な有機溶媒に対して不溶性を示すため、抽出液にメタノール、エタノール、2-プロパノール等の水混和性有機溶媒を加えれば析出沈殿する。沈殿したウルバンを濾過や遠心分離などで溶媒から分離し、乾燥すればよい。
本発明に係るアンチエイジング剤の有効成分であるH型ウルバンは、そのカルボキシ基および硫酸基の少なくとも一部がプロトン化、即ち-CO2Hおよび-OSO3Hの状態にあるウルバンをいう。以下、ウルバンにおけるカルボキシ基および硫酸基を合わせて酸性基という場合がある。具体的には、ウルバンに含まれる総酸性基のモル数に対するプロトン化カルボキシ基(-CO2H)およびプロトン化硫酸基(-OSO3H)の合計モル数の割合が、10mol%以上であるウルバンをいう。当該割合としては、15mol%以上が好ましく、19mol%以上、20mol%または以上30mol%以上がより好ましく、35mol%以上または40mol%以上がより更に好ましい。当該割合の上限は特に制限されず、100mol%であってもよいが、天然ウルバンのカルボキシ基および硫酸基の全てをプロトン化することは難しい場合があるため、当該割合としては95mol%以下または90mol%以下が好ましく、80mol%以下または70mol%以下がより好ましい。なお、例えば、蟻酸の酸解離定数pKaは3.75であるが、硫酸のpKaは-3.0であり、硫酸のKaは蟻酸の106.75倍である等、カルボキシ基に比べて硫酸基の方が酸として強いため、H型ウルバンでは硫酸基よりもカルボキシ基が優先的にプロトン化されていると考えられる。また、プロトン化されている酸性基の割合は、例えば誘導結合プラズマ(ICP)発光分光分析によりウルバン試料に含まれるカチオンを定量し、得られたデータからプロトン量も推定し、カチオンおよびプロトンが全て酸性基に存在していると仮定して求めることができる。
本発明に係るアンチエイジング剤の有効成分であるウルバンアルカリ金属イオン塩は、天然ウルバンのカルボキシ基または硫酸基と塩を形成しているカルシウムイオンやマグネシウムイオン等の二価金属イオンの少なくとも一部が、アルカリ金属イオンに置換されているウルバンをいう。具体的には、ウルバンに含まれる総酸性基のモル数に対する二価イオンのモル数の割合が30mol%以下であり、且つアルカリ金属イオンのモル数の割合が20mol%以上であるウルバンをいう。二価イオンの上記割合としては20mol%以下が好ましく、10mol%以下がより好ましい。アルカリ金属イオンの上記割合としては30mol%以上が好ましく、40mol%以上がより好ましい。当該割合の上限は特に制限されず、100mol%であってもよいが、天然ウルバンの酸性基の全てをアルカリ金属イオン塩にすることは難しい場合があるため、当該割合としては95mol%以下または90mol%以下が好ましく、80mol%以下がより好ましい。本発明に係るH型ウルバンとウルバンのアルカリ金属イオン塩における総酸性基のモル数に対するプロトンおよびアルカリ金属イオンのモル数の割合は、例えば、誘導結合プラズマ(ICP)発光分光分析により求めることができる。ICP発光分光分析は、空気や試料中の水に含まれるC、O、N、Hは測定できないが、以下の手順により対象ウルバン試料の総酸性基のモル数やプロトンのモル数を推定することができ、また、酸性基に対するプロトンやアルカリ金属イオンの割合を算出することが可能になる:(1)遊離カチオンを除去するために、対象ウルバン試料を十分に洗浄し、(2)酸性基が全て金属イオン塩となるように、イオン交換樹脂などを使って洗浄した対象ウルバン試料を処理し、(3)洗浄した対象ウルバン試料と、金属イオン塩化した対象ウルバン試料をICP発光分光分析に付し、(4)両分析データを比較する。
本発明の有効成分は、H型ウルバンであっても、酸性基の一部がアルカリ金属塩および/またはアルカリ土類金属塩であってもよいしウルバンアルカリ金属イオン塩であっても、酸性基の一部がプロトン化されていてもよい。即ち、本発明の有効成分は、酸性基が十分にプロトン化およびアルカリ金属塩化しており、H型ウルバンともアルカリ金属塩ともいえるものであってもよい。
アルカリ金属イオンとしては、例えば、リチウムイオン、ナトリウムイオン、カリウムイオン、ルビジウムイオン、セシウムイオン等が挙げられ、リチウムイオン、ナトリウムイオン、及びカリウムイオンからなる群から選択される1以上のアルカリ金属イオンが好ましく、ナトリウムイオンおよび/またはカリウムイオンがより好ましく、ナトリウムイオンがより更に好ましい。
本発明に係るアンチエイジング剤の有効成分であるH型ウルバンおよびウルバンアルカリ金属イオン塩の分子量は、特に制限されないが、例えば10,000以上とすることができる。当該分子量としては、100,000以上が好ましく、200,000以上がより好ましく、500,000以上がより更に好ましい。当該分子量の上限は特に制限されないが、分子量が過剰に大きいと取り扱いが難しい場合があり得るので、当該分子量としては2,000,000以下が好ましく、1,500,000以下がより好ましく、1,000,000以下がより更に好ましい。
H型ウルバンおよびウルバンアルカリ金属イオン塩は、常法に従って、天然ウルバンに含まれるカチオンをプロトン(H+)またはアルカリ金属イオンに置換することで製造することができる。例えば、天然ウルバンの水溶液にOH型陰イオン交換樹脂を加え、0℃以上、100℃以下で10分間以上、1時間以下処理した後、濾過や遠心分離により陰イオン交換樹脂を除去することにより、天然ウルバンに含まれる硫酸イオンや塩化物イオン等のアニオンを吸着除去する。次に、ウルバン溶液にアルカリイオン型陽イオン交換樹脂を加え、0℃以上、100℃以下で10分間以上、1時間以下処理した後、濾過や遠心分離により陽イオン交換樹脂を除去することにより、ウルバンのアルカリ金属イオン塩を製造することができる。更に、ウルバンアルカリ金属イオン塩の水溶液にH型陽イオン交換樹脂を加え、0℃以上、100℃以下で10分間以上、1時間以下処理した後、濾過や遠心分離により陽イオン交換樹脂を除去することにより、H型ウルバンを製造することができる。H型ウルバンおよびウルバンアルカリ金属イオン塩の水溶液から、凍結乾燥などにより、H型ウルバンおよびウルバンアルカリ金属イオン塩を単離することができる。以下、H型ウルバンおよびウルバンアルカリ金属イオン塩をまとめてウルバン誘導体という場合がある。
本発明に係るウルバン誘導体は、ミトコンドリアを活性化することが本発明者らの実験により見出されている。ミトコンドリアは、糖質、タンパク質、脂質などの代謝をつかさどり、エネルギー物質であるATPや熱を産生する。よって、本発明に係るウルバン誘導体は、ミトコンドリアの活性化を通じて、恒常的な生命活動を維持するためのエネルギーの供給に寄与したり、また、精神的な安定をもたらす4-アミノ酪酸(GABA)や血糖値を低下させるインスリンの分泌を促進することも考えられる。
また、本発明に係るウルバン誘導体は、本発明者らの実験的知見により、ミトコンドリアの抗酸化酵素の遺伝子発現と生合成を促進することが明らかにされている。生体内の活性酸素は、本来は体内に侵入した細菌やウイルスを攻撃して生体を保護する重要な働きを有するが、過剰な活性酸素は正常な細胞まで攻撃したり、また、活性酸素により生じた過酸化脂質が更に活性酸素を生じるという悪循環に陥ることがある。抗酸化酵素は、過剰な活性酸素を分解して無害化するものであるため、例えば、本発明に係るウルバン誘導体は、ミトコンドリアの抗酸化酵素の活性化を通じてミトコンドリアを活性化する他、健康の維持や肌の状態の維持に寄与することが考えられる。
更に、本発明者らは、本発明に係るウルバン誘導体がコラーゲンの生合成を促進することを実験的に証明している。コラーゲンは、皮膚、腱、軟骨などを構成する繊維状のタンパク質であるが、加齢によりその前駆体からコラーゲンを生成する酵素が減少し、コラーゲンの生合成量が低下して、シワの発生や肌の張りの減少などに繋がることが知られている。よって、本発明に係るウルバン誘導体により、加齢によるシワの発生や肌の張りの減少など、肌の状態が改善されることが考えられる。
本発明に係るアンチエイジング剤は、有効成分としてウルバン誘導体を含有する。有効成分とは、本発明に係るアンチエイジング剤に含まれる成分のうちアンチエイジング効果を発揮する成分をいい、換言すれば、本発明に係るアンチエイジング剤は、アンチエイジング効果が発揮される量のウルバン誘導体を含む。具体的には、特に制限されないが、例えば、本発明に係るアンチエイジング剤におけるウルバン誘導体の割合を10質量%以上、100質量%以下とすることができる。また、本発明に係るアンチエイジング剤が外用剤である場合には、ウルバン誘導体の割合を0.1質量%以上、10質量%以下にすることもできる。
本発明に係る動物の老化の抑制方法は、H型ウルバンまたはウルバンのアルカリ金属イオン塩を有効成分として含む本発明に係るアンチエイジング剤を前記動物に投与する工程を含む。H型ウルバンまたはウルバンのアルカリ金属イオン塩は、H型ウルバンおよびウルバンのアルカリ金属イオン塩の両方を投与してもよいし、H型ウルバンまたはウルバンのアルカリ金属イオン塩の一方を投与してもよい。
本発明に係るアンチエイジング剤の投与頻度や投与量は、投与対象の年齢、性別、状態などに応じて適宜調整すればよく、アンチエイジング効果を発揮できる量を投与対象へ投与する。例えば、1日当たりのアンチエイジング剤の投与量としては1mg/kg体重以上、1g/kg体重以下とすることができる。また、本発明に係るアンチエイジング剤が外用剤である場合には、一日当たりのアンチエイジング剤の塗布量としては0.1mg以上、10mg以下とすることもできる。1日当たりの投与回数や塗布回数は特に限定されず、所望の投与範囲内において、単回または数回に分けて投与または塗布すればよい。
本発明に係るアンチエイジング剤は、ヒトに限らず、ヒト以外の動物にも投与可能である。投与対象動物としては、例えば、ウマ、ウシ、ブタ、ヒツジ、ヤギ、ラクダ、ラマなどの家畜;競走馬などの競技動物;イヌ、ネコなどの愛玩動物;マウス、ラット、モルモット、ウサギなどの実験動物;ニワトリ、アヒル、七面鳥、駝鳥などの家禽などが挙げられる。
本発明に係るアンチエイジング剤の剤形は特に制限されず、例えば、ウルバン誘導体自体であってもよいし、他の成分と組み合わせた組成物であってもよいし、これらの溶液または懸濁液であってもよい。本発明に係るアンチエイジング剤の剤形としては、特に制限されないが、例えば、錠剤、散剤、カプセル剤、糖衣錠、顆粒剤、液剤、外用剤などを挙げることができる。本発明に係るアンチエイジング剤には、剤形に合わせ、薬学上許容される添加剤を用いてもよい。かかる添加剤としては、例えば、賦形剤、崩壊剤、滑沢剤、結合剤、酸化防止剤、着色剤、甘味料、凝集防止剤、防腐剤、有効成分の溶解補助剤、安定化剤などを挙げることができる。
本発明に係るウルバン誘導体は、以下の実施例の通り、ミトコンドリアの活性化、抗酸化作用、コラーゲンの生合成の増強作用、コラーゲンの生合成の増強作用など、客観的で且つ優れたアンチエイジング作用を示す。また、本発明に係るウルバン誘導体は、藻類由来の多糖類をフリー体にしたりアルカリ金属イオン塩にしたものであるので、安全性は高いと考えられる。よって本発明に係るアンチエイジング剤は、アンチエイジング効果に優れた健康食品などとして恒常的に摂取することも可能であり得る。
例えば、本発明に係るウルバン誘導体は、アンチエイジング剤として、一般的な飲食品とすることも可能である。本発明に係るウルバン誘導体を添加する飲食品は特に限定されないが、例えば、乳飲料、清涼飲料、スポーツドリンク、栄養ドリンク、美容ドリンク、液体栄養剤などの飲料;チューインガム、チョコレート、キャンディー、ゼリー、ケーキ、ビスケット、クラッカーなどの菓子類;アイスクリーム、氷菓などの冷菓類;うどん、中華麺、スパゲティー、即席麺などの麺類;蒲鉾、竹輪、半片などの練り製品;ドレッシング、マヨネーズ、ソースなどの調味料;パン、ハム、雑炊、米飯、スープ、各種レトルト食品、各種冷凍食品などが挙げられる。本発明に係るウルバン誘導体を含有する飲食品は、いわゆる健康食品、サプリメント、機能性食品、機能性表示食品、栄養補助食品、特定保健用食品、栄養機能食品、介護食品、スマイルケア食、咀嚼・嚥下補助食品、濃厚流動食品、病者用食品などの用途に用いることができる。
本願は、2022年10月25日に出願された日本国特許出願第2022-170789号に基づく優先権の利益を主張するものである。2022年10月25日に出願された日本国特許出願第2022-170789号の明細書の全内容が、本願に参考のため援用される。
以下、実施例を挙げて本発明をより具体的に説明するが、本発明はもとより下記実施例によって制限を受けるものではなく、前・後記の趣旨に適合し得る範囲で適当に変更を加えて実施することも勿論可能であり、それらはいずれも本発明の技術的範囲に包含される。
実施例1: ウルバンAMI塩の製造
(1)天然抽出ウルバンの製造
ミナミアオノリをよく洗い凍結乾燥させた後、ブレンダーで粉砕した。得られた乾燥粉末(1g)に蒸留水(20mL)を加え、90℃で熱水抽出した。上澄み液を回収し、残渣に蒸留水(20mL)を加えて再度熱水抽出する操作を2回繰り返した。回収した水溶液(約60mL)を濃縮後、エタノールを加えてエタノール濃度を75%に調整し、ウルバンを沈殿させた。生じた沈殿を遠心分離で回収し、凍結乾燥することにより、天然抽出ウルバンを得た。
(1)天然抽出ウルバンの製造
ミナミアオノリをよく洗い凍結乾燥させた後、ブレンダーで粉砕した。得られた乾燥粉末(1g)に蒸留水(20mL)を加え、90℃で熱水抽出した。上澄み液を回収し、残渣に蒸留水(20mL)を加えて再度熱水抽出する操作を2回繰り返した。回収した水溶液(約60mL)を濃縮後、エタノールを加えてエタノール濃度を75%に調整し、ウルバンを沈殿させた。生じた沈殿を遠心分離で回収し、凍結乾燥することにより、天然抽出ウルバンを得た。
(2)ウルバンAMI塩の製造
NaOH水溶液を用いて、Cl型陰イオン交換樹脂(「AmberliteTM IRA410Cl」オルガノ社製)をOH型陰イオン交換樹脂にした。天然抽出ウルバンの0.5%水溶液(100g)に、得られたOH型陰イオン交換樹脂(0.5g)を加え、室温で30分間攪拌した。上澄み液を回収し、再度、新たなOH型陰イオン交換樹脂を加えて同様に処理した。OH型陰イオン交換樹脂による処理と上澄み液の回収を計5回繰り返し、ウルバン水溶液に混入する硫酸塩(SO4 2-)や塩化物イオン(Cl-)を除去した。
得られたウルバン水溶液(約100g)に対して、Na型陽イオン交換樹脂(「AmberliteTM IR120BNa」オルガノ社製)(0.5g)を用いて、ウルバンのカチオンサイトの陽イオンを部分的にNaイオンと交換した後、凍結乾燥した。
得られたウルバンAMI塩の分子量を、ゲル浸透クロマトグラフィ(GPC)で測定した。また、得られたウルバンAMI塩に含まれるカチオン濃度を、誘導結合プラズマ(ICP)発光分光分析により測定した。結果を表1に示す。なお、表1中、プロトンの濃度はICP発光分光分析の結果に基づく推定値であり、「アルカリ金属イオン(AMI)+H+の割合」は、ICP発光分光分析の結果から下記式により酸性基濃度を算出し、算出された酸性基濃度に対するアルカリ金属イオンとH+の割合として算出した。なお、得られたウルバンNa塩にプロトンはほとんど含まれていないため、酸性基に対するアルカリ金属イオンの割合は41mol%であるといえる。
酸性基濃度=(Na+濃度+K+濃度+H+濃度)+2×(Ca2+濃度+Mg2+濃度)
NaOH水溶液を用いて、Cl型陰イオン交換樹脂(「AmberliteTM IRA410Cl」オルガノ社製)をOH型陰イオン交換樹脂にした。天然抽出ウルバンの0.5%水溶液(100g)に、得られたOH型陰イオン交換樹脂(0.5g)を加え、室温で30分間攪拌した。上澄み液を回収し、再度、新たなOH型陰イオン交換樹脂を加えて同様に処理した。OH型陰イオン交換樹脂による処理と上澄み液の回収を計5回繰り返し、ウルバン水溶液に混入する硫酸塩(SO4 2-)や塩化物イオン(Cl-)を除去した。
得られたウルバン水溶液(約100g)に対して、Na型陽イオン交換樹脂(「AmberliteTM IR120BNa」オルガノ社製)(0.5g)を用いて、ウルバンのカチオンサイトの陽イオンを部分的にNaイオンと交換した後、凍結乾燥した。
得られたウルバンAMI塩の分子量を、ゲル浸透クロマトグラフィ(GPC)で測定した。また、得られたウルバンAMI塩に含まれるカチオン濃度を、誘導結合プラズマ(ICP)発光分光分析により測定した。結果を表1に示す。なお、表1中、プロトンの濃度はICP発光分光分析の結果に基づく推定値であり、「アルカリ金属イオン(AMI)+H+の割合」は、ICP発光分光分析の結果から下記式により酸性基濃度を算出し、算出された酸性基濃度に対するアルカリ金属イオンとH+の割合として算出した。なお、得られたウルバンNa塩にプロトンはほとんど含まれていないため、酸性基に対するアルカリ金属イオンの割合は41mol%であるといえる。

実施例2: H型ウルバンの製造
天然抽出ウルバンの0.5%水溶液(100g)に、前記OH型陰イオン交換樹脂(0.5g)を加え、室温で30分間攪拌した。上澄み液を回収し、再度、新たなOH型陰イオン交換樹脂を加えて同様に処理した。OH型陰イオン交換樹脂による処理と上澄み液の回収を計5回繰り返し、ウルバン水溶液に混入する硫酸塩(SO4 2-)や塩化物イオン(Cl-)を除去した。
得られたウルバン水溶液(約100g)に対して、Na型陽イオン交換樹脂(「AmberliteTM IR120BNa」オルガノ社製)(0.5g)を加え、室温で30分間攪拌した。更に、上澄み液(90mL)にH型陽イオン交換樹脂(「AmberliteTM IR120BH」オルガノ社製)(0.45g)を加え、室温で30分間攪拌し、ウルバンのカチオンサイトの陽イオンを部分的にHイオンと交換した後、凍結乾燥した。
得られたH型ウルバンを、実施例1(2)と同様に分析した。結果を表1に示す。なお、得られたH型ウルバンにおいて酸性基に対するプロトンの割合は38~42mol%、アルカリ金属イオンの割合は10mol%であった。
天然抽出ウルバンの0.5%水溶液(100g)に、前記OH型陰イオン交換樹脂(0.5g)を加え、室温で30分間攪拌した。上澄み液を回収し、再度、新たなOH型陰イオン交換樹脂を加えて同様に処理した。OH型陰イオン交換樹脂による処理と上澄み液の回収を計5回繰り返し、ウルバン水溶液に混入する硫酸塩(SO4 2-)や塩化物イオン(Cl-)を除去した。
得られたウルバン水溶液(約100g)に対して、Na型陽イオン交換樹脂(「AmberliteTM IR120BNa」オルガノ社製)(0.5g)を加え、室温で30分間攪拌した。更に、上澄み液(90mL)にH型陽イオン交換樹脂(「AmberliteTM IR120BH」オルガノ社製)(0.45g)を加え、室温で30分間攪拌し、ウルバンのカチオンサイトの陽イオンを部分的にHイオンと交換した後、凍結乾燥した。
得られたH型ウルバンを、実施例1(2)と同様に分析した。結果を表1に示す。なお、得られたH型ウルバンにおいて酸性基に対するプロトンの割合は38~42mol%、アルカリ金属イオンの割合は10mol%であった。
実施例3: ウルバンAMI塩の製造
天然抽出ウルバンの0.5%水溶液(100g)に、前記OH型陰イオン交換樹脂(0.5g)を加え、室温で30分攪拌した。上澄み液を回収し、再度、新たなOH型陰イオン交換樹脂を加えて同様に処理した。OH型陰イオン交換樹脂による処理と上澄み液の回収を計5回繰り返し、ウルバン水溶液に混入する遊離の硫酸イオン(SO4 2-)や塩化物イオン(Cl-)を除去した。
次に、得られたウルバン水溶液(約100g)に対して、H型陽イオン交換樹脂(「AmberliteTM IR120BH」オルガノ社製)(0.5g)を加え、室温で30分攪拌し、樹脂を取り除き、再び30分攪拌した。更に、上澄み液(94.6mL)にNa型陽イオン交換樹脂(「AmberliteTM IR120BNa」オルガノ社製)(47.3g)を用いて、ウルバンのカチオンサイトの陽イオンのほとんどをNaイオンと交換した後、凍結乾燥した。
得られたウルバンAMI塩を、実施例1(2)と同様に分析した。結果を表1に示す。なお、得られたウルバンNa塩にプロトンはほとんど含まれていないため、酸性基に対するアルカリ金属イオン(AMI)の割合は93mol%であるといえる。
天然抽出ウルバンの0.5%水溶液(100g)に、前記OH型陰イオン交換樹脂(0.5g)を加え、室温で30分攪拌した。上澄み液を回収し、再度、新たなOH型陰イオン交換樹脂を加えて同様に処理した。OH型陰イオン交換樹脂による処理と上澄み液の回収を計5回繰り返し、ウルバン水溶液に混入する遊離の硫酸イオン(SO4 2-)や塩化物イオン(Cl-)を除去した。
次に、得られたウルバン水溶液(約100g)に対して、H型陽イオン交換樹脂(「AmberliteTM IR120BH」オルガノ社製)(0.5g)を加え、室温で30分攪拌し、樹脂を取り除き、再び30分攪拌した。更に、上澄み液(94.6mL)にNa型陽イオン交換樹脂(「AmberliteTM IR120BNa」オルガノ社製)(47.3g)を用いて、ウルバンのカチオンサイトの陽イオンのほとんどをNaイオンと交換した後、凍結乾燥した。
得られたウルバンAMI塩を、実施例1(2)と同様に分析した。結果を表1に示す。なお、得られたウルバンNa塩にプロトンはほとんど含まれていないため、酸性基に対するアルカリ金属イオン(AMI)の割合は93mol%であるといえる。
実施例4: 老化マーカーである老化βガラクトシダーゼ活性の測定
MEMα培地に、10%FBS、100μg/mLペニシリン、及び100μg/mLストレプトマイシンを配合した培地を用い、ヒト皮膚線維芽細胞(NB1RGB細胞)を80~89日間培養することにより、老化細胞とした。
天然ウルバン、実施例1のウルバンAMI塩、実施例2のH型ウルバン、又は実施例3のウルバンAMI塩を前記培地に500μg/mLの割合で添加し、前記ヒト皮膚線維芽老化細胞を更に37℃で24時間培養した。比較のために、別途、ウルバン類を添加しない以外は同様にして培養した。
Senescence Detection Kit(abcam社製)を使って、製造元のプロトコルに従って培養後の細胞を固定液で固定して染色することにより、老化マーカーであるsenescence beta-galactosidaseの活性を測定した。senescence beta-galactosidase活性がある細胞(老化細胞)は、青色に染色される。また培養した50個の細胞のうち、青色で染色された細胞の数を計測した。染色細胞の拡大写真を図1に、染色細胞数を図2に示す。なお、図2中、「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で対照群(control)と比べて有意差があることを示す。
MEMα培地に、10%FBS、100μg/mLペニシリン、及び100μg/mLストレプトマイシンを配合した培地を用い、ヒト皮膚線維芽細胞(NB1RGB細胞)を80~89日間培養することにより、老化細胞とした。
天然ウルバン、実施例1のウルバンAMI塩、実施例2のH型ウルバン、又は実施例3のウルバンAMI塩を前記培地に500μg/mLの割合で添加し、前記ヒト皮膚線維芽老化細胞を更に37℃で24時間培養した。比較のために、別途、ウルバン類を添加しない以外は同様にして培養した。
Senescence Detection Kit(abcam社製)を使って、製造元のプロトコルに従って培養後の細胞を固定液で固定して染色することにより、老化マーカーであるsenescence beta-galactosidaseの活性を測定した。senescence beta-galactosidase活性がある細胞(老化細胞)は、青色に染色される。また培養した50個の細胞のうち、青色で染色された細胞の数を計測した。染色細胞の拡大写真を図1に、染色細胞数を図2に示す。なお、図2中、「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で対照群(control)と比べて有意差があることを示す。
図1および図2に示される結果の通り、天然ウルバンを用いても、老化細胞の老化の指標となる老化βガラクトシダーゼの活性を有意に低下させることはできない。
それに対して、カルボキシ基と結合するカウンターカチオンがプロトンおよび/またはナトリウムイオンに置換されているウルバンAMI塩およびH型ウルバンを用いれば、老化βガラクトシダーゼの活性を有意に低下できることが明らかとなった。

実施例5: コラーゲン産生能試験
天然ウルバン、実施例1のウルバンAMI塩、実施例2のH型ウルバン、又は実施例3のウルバンAMI塩を500μg/mLの割合で添加した培地を用い、ヒト皮膚線維芽老化細胞を更に37℃で72時間培養し、試料を得た。比較のために、別途、ウルバン類を添加しない以外は同様に培養して試料を得た。得られた試料から細胞溶解液を調製し、含まれるタンパク質濃度を揃えた細胞溶解液25μLあたりの細胞層中のコラーゲンの量を、コラーゲン定量キット(コスモバイオ社製)を使用して、製造元のプロトコルに従って評価した。蛍光は、380nm/485nmのフィルターペアを備えた蛍光マイクロプレートリーダー(「Infinite M200」TECAN社製)で測定した。結果を図3に示す。図3中、「*」は、二元配置分散分析に続くTukey’s testによりp<0.05で対照群(control)と比べて有意差があることを示し、「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で有意差があることを示す。
天然ウルバン、実施例1のウルバンAMI塩、実施例2のH型ウルバン、又は実施例3のウルバンAMI塩を500μg/mLの割合で添加した培地を用い、ヒト皮膚線維芽老化細胞を更に37℃で72時間培養し、試料を得た。比較のために、別途、ウルバン類を添加しない以外は同様に培養して試料を得た。得られた試料から細胞溶解液を調製し、含まれるタンパク質濃度を揃えた細胞溶解液25μLあたりの細胞層中のコラーゲンの量を、コラーゲン定量キット(コスモバイオ社製)を使用して、製造元のプロトコルに従って評価した。蛍光は、380nm/485nmのフィルターペアを備えた蛍光マイクロプレートリーダー(「Infinite M200」TECAN社製)で測定した。結果を図3に示す。図3中、「*」は、二元配置分散分析に続くTukey’s testによりp<0.05で対照群(control)と比べて有意差があることを示し、「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で有意差があることを示す。
図3に示される結果の通り、天然ウルバンを用いても、皮膚の張り等に関係するコラーゲンの皮膚細胞による産生量を有意に増加させることはできない。
それに対して、酸性基と結合するカウンターカチオンがアルカリ金属イオン(AMI)に置換されているウルバンAMI塩、及びH型ウルバンを用いれば、皮膚細胞によるコラーゲンの産生量を有意に増加できることが明らかとなった。

実施例6: SOD2生合成量の解析
抗SOD2抗体(Cell Signaling Technology社製)と抗β-actin(sigma社製)を用い、SOD2生合成量を解析した。
具体的には、実施例1のウルバンAMI塩、実施例2のH型ウルバン、又は実施例3のウルバンAMI塩を500μg/mLの割合で添加した培地を用い、ヒト皮膚線維芽老化細胞を更に37℃で72時間培養し、試料を得た。試料からタンパク質溶液を調製し、ポリアクリルアミド電気泳動に付した後、メンブレンに転写し、抗SOD2抗体製品を1000倍に、抗β-アクチン抗体を10000倍に希釈し、メンブレンを浸漬し、4℃で18時間インキュベートした後、洗浄した。次いで、HRP標識抗ウサギおよび抗マウス二次抗体製品(Promega社製)を5000倍に希釈し、メンブレンを浸漬し、常温で1時間インキュベートした後、洗浄した。次に、メンブランをルミノール反応溶液に浸漬し、発光するまで常温で5~10分間インキュベートした後、X線フィルムに感光させて現像した。
SOD2のバンドの蛍光強度を、各試料におけるβ-アクチンのバンドの蛍光強度で補正した。結果を図4に示す。図4中の「Intensity」は、上記補正値を示す。

具体的には、実施例1のウルバンAMI塩、実施例2のH型ウルバン、又は実施例3のウルバンAMI塩を500μg/mLの割合で添加した培地を用い、ヒト皮膚線維芽老化細胞を更に37℃で72時間培養し、試料を得た。試料からタンパク質溶液を調製し、ポリアクリルアミド電気泳動に付した後、メンブレンに転写し、抗SOD2抗体製品を1000倍に、抗β-アクチン抗体を10000倍に希釈し、メンブレンを浸漬し、4℃で18時間インキュベートした後、洗浄した。次いで、HRP標識抗ウサギおよび抗マウス二次抗体製品(Promega社製)を5000倍に希釈し、メンブレンを浸漬し、常温で1時間インキュベートした後、洗浄した。次に、メンブランをルミノール反応溶液に浸漬し、発光するまで常温で5~10分間インキュベートした後、X線フィルムに感光させて現像した。
SOD2のバンドの蛍光強度を、各試料におけるβ-アクチンのバンドの蛍光強度で補正した。結果を図4に示す。図4中の「Intensity」は、上記補正値を示す。
図4に示される結果の通り、ウルバンAMI塩およびH型ウルバンによれば、酸化ストレスにより老化の原因となる活性酸素種を酸素と過酸化水素に不均化するSOD(Superoxide dismutase)のうち、ミトコンドリア内で産生されるSOD2の生合成量を増加できることが証明された。
実施例7: 活性化AKT(リン酸化)の解析
抗リン酸化AKT抗体(Cell Signaling Technology社製)と抗AKT抗体(Cell Signaling Technology社製)を用い、セリン/スレオニンキナーゼ(AKT)の活性化を解析した。
具体的には、天然ウルバンまたはH型ウルバンを100μg/mLの割合で添加した培地を用い、ヒト皮膚線維芽老化細胞を更に37℃で表記の時間培養し、試料を得た。試料からタンパク質溶液を調製し、ポリアクリルアミド電気泳動に付した後、メンブレンに転写し、抗リン酸化AKT抗体製品を1000倍に、抗AKT抗体を1000倍に希釈し、メンブレンを浸漬し、4℃で18時間インキュベートした後、洗浄した。次いで、HRP標識抗ウサギ二次抗体製品(Promega社製)を5000倍に希釈し、メンブレンを浸漬し、常温で1時間インキュベートした後、洗浄した。次に、メンブランをルミノール反応溶液に浸漬し、発光するまで常温で5~10分間インキュベートした後、X線フィルムに感光させて現像した。結果を図5に示す。
抗リン酸化AKT抗体(Cell Signaling Technology社製)と抗AKT抗体(Cell Signaling Technology社製)を用い、セリン/スレオニンキナーゼ(AKT)の活性化を解析した。
具体的には、天然ウルバンまたはH型ウルバンを100μg/mLの割合で添加した培地を用い、ヒト皮膚線維芽老化細胞を更に37℃で表記の時間培養し、試料を得た。試料からタンパク質溶液を調製し、ポリアクリルアミド電気泳動に付した後、メンブレンに転写し、抗リン酸化AKT抗体製品を1000倍に、抗AKT抗体を1000倍に希釈し、メンブレンを浸漬し、4℃で18時間インキュベートした後、洗浄した。次いで、HRP標識抗ウサギ二次抗体製品(Promega社製)を5000倍に希釈し、メンブレンを浸漬し、常温で1時間インキュベートした後、洗浄した。次に、メンブランをルミノール反応溶液に浸漬し、発光するまで常温で5~10分間インキュベートした後、X線フィルムに感光させて現像した。結果を図5に示す。
AKTの活性化により細胞の増殖が促進されるため、老化により低下している細胞の増殖をH型ウルバンが促進している可能性がある。また、天然ウルバンにはAKTを活性化する働きがないため、H型ウルバンにすることにより、この効果は初めて発現する。
実施例8: ミトコンドリア活性試験
天然ウルバン、実施例1のウルバンAMI塩、実施例2のH型ウルバン、又は実施例3のウルバンAMI塩を前記培地に500μg/mLの割合で添加し、前記ヒト皮膚線維芽老化細胞を更に37℃で24時間培養した。比較のために、別途、ウルバン類を添加しない以外は同様にして培養した。
次いで、ミトコンドリアの活性を、ミトコンドリア膜電位検出試薬(「TMRM(tetramethylrhodamine methyl ester)」Thermo社製)を使って、製造元のプロトコルに従って、TMRM染色により測定した。蛍光は、535nm/590nmのフィルターペアを使用して、蛍光マイクロプレートリーダー(「Infinite M200」TECAN社製)で測定した。
ミトコンドリアの活性が高い場合には膜電位差が維持され、低分子蛍光色素であるTMRMが集積して、より強い蛍光が発せられ、活性が低下すると膜電位差も低下し、TMRMが集積せずに弱い蛍光が発せられる。よって、ミトコンドリアの活性は、535nm/590nmで測定された波長の蛍光強度として測定できる。結果を、対照例に対するulvan等の処理例の比として図6に示す。図6中、「*」は、二元配置分散分析に続くTukey’s testによりp<0.05で有意差があることを示す。「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で有意差があることを示す。

図6に示される結果の通り、H型ウルバンおよびウルバンAMI塩は、エネルギー生産、延いては細胞の活性に関係するミトコンドリアの活性を有意に改善できることが明らかとなった。
実施例9: ATP産生量
天然ウルバン、実施例1のウルバンAMI塩、実施例2のH型ウルバン、又は実施例3のウルバンAMI塩を前記培地に500μg/mLの割合で添加し、前記ヒト皮膚線維芽老化細胞を更に37℃で24時間培養し、試料を得た。比較のために、別途、ウルバン類を添加しない以外は同様にして培養した。次いでATPの産生量については、CellTiter-Glo(登録商標)2.0 Assay(promega社製)を使って、製造元のプロトコルに従って測定した。発光はPromega GloMax 20/20 Luminometerにより測定した。結果を、対照例に対するulvan等の処理例の比として図7に示す。「*」は、二元配置分散分析に続くTukey’s testによりp<0.05で有意差があることを示す。「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で有意差があることを示す。

図7に示される結果の通り、天然ウルバンとウルバン41mol%AMI塩は、皮膚細胞における生体内化学反応のエネルギー伝達物質であるATPの産生量を有意に増加せしめることができ、H型ウルバンおよびウルバン93mol%AMI塩は、かかるATP産生量を更に増加させることができることが明らかとなった。
実施例10: 抗酸化活性測定
天然ウルバン、ウルバンHを前記培地に図8に示される割合で添加し、前記ヒト皮膚線維芽老化細胞を更に37℃で48時間培養し、試料を得た。比較のために、別途、ウルバン等を添加しない以外は同様に培養して試料を得た。

図8に示される結果の通り、天然ウルバンでは抗酸化タンパク質量を有意には増加させることができないのに対して、H型ウルバンによれば、抗酸化タンパク質量を有意に増加できることが示された。
実施例11: mRNAの発現解析
ウルバン、H型ウルバン、又はウルバン93mol%AMI塩を500μg/mLの割合で添加した培地を用い、ヒト表皮角化細胞(NHEK細胞(AD:adult))(クラボウ社製)を更に37℃で24時間培養し、培養開始から24時間後に試料を採取し、高効率リアルタイムPCR用マスターミックス(「THUNDERBIRD(登録商標)Next SYBR(登録商標)qPCR Mix」東洋紡社製)を使って、表皮防御因子であるFilaggrinとミトコンドリアの抗酸化酵素であるスーパーオキシドジスムターゼ2(SOD2)の遺伝子発現を定量した。全RNAは、内部標準としてβ-アクチン相補DNAを使用して、各反応でノーマライズした。結果を図9に示す。図9中、「*」は、二元配置分散分析に続くTukey’s testによりp<0.05で有意差があることを示す。「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で有意差があることを示す。

実施例12: H型ウルバンの製造
実施例1(1)の天然ウルバンと実施例2のH型ウルバン(H+割合:38~42mol%)を混合し、酸性基に対するプロトンの割合が19~21mol%、アルカリ金属イオンの割合が15mol%であるH型ウルバンを調製した。

実施例13: ウルバンAMI塩の製造
実施例1(1)の天然ウルバンと実施例1のウルバンAMI塩(AMI割合:41mol%)を混合し、酸性基に対するアルカリ金属イオンの割合が30mol%であるウルバンAMI塩を調製した。
実施例1(1)の天然ウルバンと実施例1のウルバンAMI塩(AMI割合:41mol%)を混合し、酸性基に対するアルカリ金属イオンの割合が30mol%であるウルバンAMI塩を調製した。
実施例14: mRNAの発現解析
実施例11と同様にして、天然ウルバン、実施例2のH型ウルバン(H+割合:38~42mol%)、実施例12のH型ウルバン(H+割合:19~21mol%)、実施例1のウルバンAMI塩(AMI割合:41mol%)および実施例13のウルバンAMI塩(AMI割合:30mol%)を添加した培地におけるヒト表皮角化細胞由来のスーパーオキシドジスムターゼ2(SOD2)の遺伝子発現を定量した。H型ウルバンの結果を図10(1)に、ウルバンAMI塩の結果を図10(2)に示す。図10中、「*」は、二元配置分散分析に続くTukey’s testによりp<0.05で有意差があることを示す。「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で有意差があることを示す。
図10に示される結果の通り、天然ウルバンではミトコンドリアの抗酸化酵素であるSOD2の発現を有意には促進させることができないのに対して、プロトン割合が比較的低いH型ウルバンおよびAMI割合が比較的低いウルバンAMI塩であっても、SOD2の発現を有意に増加できることが示された。
実施例15: K型ウルバンAMI塩の製造
実施例1(2)と同様にして得られたウルバン水溶液(約100g)に対して、H型陽イオン交換樹脂(「AmberliteTM IR120BH」オルガノ社製,0.5g)を加え、室温で30分攪拌した。樹脂を取り除き、再び30分攪拌した。別途、硝酸カリウムを用い、H型陽イオン交換樹脂(「AmberliteTM IR120BH」オルガノ社製)のH+をK+に交換し、K型陽イオン交換樹脂を調製した。樹脂を取り除いたウルバン水溶液(98mL)にK型陽イオン交換樹脂(1.0g)を加え、室温で30分攪拌してウルバンのカチオンサイトの陽イオンをカリウムイオンに交換した。樹脂を取り除き、再び30分攪拌した後、凍結乾燥して、K型ウルバンAMI塩を得た。
得られたK型ウルバンAMI塩を実施例1(2)と同様に分析したところ、酸性基に対するアルカリ金属イオンの割合は59mol%であった。分析結果を表3に示す。
実施例1(2)と同様にして得られたウルバン水溶液(約100g)に対して、H型陽イオン交換樹脂(「AmberliteTM IR120BH」オルガノ社製,0.5g)を加え、室温で30分攪拌した。樹脂を取り除き、再び30分攪拌した。別途、硝酸カリウムを用い、H型陽イオン交換樹脂(「AmberliteTM IR120BH」オルガノ社製)のH+をK+に交換し、K型陽イオン交換樹脂を調製した。樹脂を取り除いたウルバン水溶液(98mL)にK型陽イオン交換樹脂(1.0g)を加え、室温で30分攪拌してウルバンのカチオンサイトの陽イオンをカリウムイオンに交換した。樹脂を取り除き、再び30分攪拌した後、凍結乾燥して、K型ウルバンAMI塩を得た。
得られたK型ウルバンAMI塩を実施例1(2)と同様に分析したところ、酸性基に対するアルカリ金属イオンの割合は59mol%であった。分析結果を表3に示す。
実施例16: K型ウルバンAMI塩の製造
K型陽イオン交換樹脂の使用量を1.0gから2.5gに変更した以外は実施例12と同様にして、K型ウルバンAMI塩を得た。
得られたK型ウルバンAMI塩を実施例1(2)と同様に分析したところ、酸性基に対するアルカリ金属イオンの割合は69mol%であった。分析結果を表3に示す。
K型陽イオン交換樹脂の使用量を1.0gから2.5gに変更した以外は実施例12と同様にして、K型ウルバンAMI塩を得た。
得られたK型ウルバンAMI塩を実施例1(2)と同様に分析したところ、酸性基に対するアルカリ金属イオンの割合は69mol%であった。分析結果を表3に示す。
実施例17: mRNAの発現解析
実施例11と同様にして、天然ウルバン、及び実施例15,16のK型ウルバンAMI塩を添加した培地におけるヒト表皮角化細胞由来のスーパーオキシドジスムターゼ2(SOD2)の遺伝子発現を定量した。結果を図11に示す。図11中、「*」は、二元配置分散分析に続くTukey’s testによりp<0.05で有意差があることを示す。「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で有意差があることを示す。
実施例11と同様にして、天然ウルバン、及び実施例15,16のK型ウルバンAMI塩を添加した培地におけるヒト表皮角化細胞由来のスーパーオキシドジスムターゼ2(SOD2)の遺伝子発現を定量した。結果を図11に示す。図11中、「*」は、二元配置分散分析に続くTukey’s testによりp<0.05で有意差があることを示す。「**」は、二元配置分散分析に続くTukey’s testによりp<0.01で有意差があることを示す。
図11に示される結果の通り、カリウムイオンが比較的多いウルバンAMI塩もSOD2の発現を有意に増加できることが示され、AMIの割合が大きいほど効果が高い傾向があることが示された。
Claims (23)
- H型ウルバンまたはウルバンのアルカリ金属イオン塩を有効成分として含有することを特徴とするアンチエイジング剤。
- 前記アルカリ金属がNaおよび/またはKである請求項1に記載のアンチエイジング剤。
- 前記H型ウルバンのカルボキシ基および硫酸基の15mol%以上がプロトン化されている請求項1に記載のアンチエイジング剤。
- 前記ウルバンのアルカリ金属イオン塩のカルボキシ基および硫酸基の30mol%以上がアルカリ金属イオンと塩を形成している請求項1に記載のアンチエイジング剤。
- ミトコンドリア活性化作用を示す請求項1に記載のアンチエイジング剤。
- 抗酸化作用を示す請求項1に記載のアンチエイジング剤。
- コラーゲン生合成増強作用を示す請求項1に記載のアンチエイジング剤。
- 老化を抑制するためのH型ウルバンまたはウルバンのアルカリ金属イオン塩の使用。
- 前記アルカリ金属がNaおよび/またはKである請求項8に記載の使用。
- 前記H型ウルバンのカルボキシ基および硫酸基の15mol%以上がプロトン化されている請求項8に記載の使用。
- 前記ウルバンのアルカリ金属イオン塩のカルボキシ基および硫酸基の30mol%以上がアルカリ金属イオンと塩を形成している請求項8に記載の使用。

- 動物の老化を抑制するための方法であって、
