Abstract
本発明の課題は、手洗いやうがい以外のヒトコロナウイルス感染予防策を提供することである。本発明に係る発酵乳は、菌体外多糖を産生する乳酸菌により調製されるものであって、ヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させる機能を有する。なお、ここで、乳酸菌は、ラクトバチラシエ(Lactobacillaceae)科乳酸菌であることが好ましく、ラクトバチルス属乳酸菌であることがより好ましく、ラクトバチルス デルブルエッキー 亜種 ブルガリクスであることがさらに好ましく、ラクトバチルス デルブルエッキー 亜種 ブルガリクスOLL1073R-1(FERM BP-10741)であることが特に好ましい。
Description
本発明は、ヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させる発酵乳に関する。また、本発明は、その発酵乳を有効成分とする食品組成物にも関する。
ヒトコロナウイルス(Human Coronavirus:HCoV)には、風邪症候群の原因となるHCoV-229E、HCoV-OC43、HCoV-NL63、HCoV-HKU1、重症肺炎の原因となるMERS-CoV、SARS-CoV、2019年に発生したCOVID-19の原因となるSARS-CoV-2が存在する。風邪症候群の10~15%(流行期においては約35%)はHCoV-229E、HCoV-OC43、HCoV-NL63、HCoV-HKU1を原因とするものであって、主に冬季にその流行のピークが見られる。
ところで、ヒトコロナウイルスの感染予防策として、手洗いやうがいが推奨されているが、その他さらに有効な予防策が渇望されている。本発明の課題は、手洗いやうがい以外の有効なヒトコロナウイルス感染予防策を提供することである。
上記課題を解決するために、本発明者らは、菌体外多糖を産生する乳酸菌により調製される発酵乳に着目し、ヒトの唾液においてヒトコロナウイルスに交叉するIgAの生成量に対するその発酵乳の摂取効果について詳細に検討した。そして、その結果として、ヒトにその発酵乳を摂取させることにより、そのヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させることができることを見出し、本発明を完成させた。
すなわち、本発明は、次の通りとなる。
(1)菌体外多糖を産生する乳酸菌により調製される、ヒトの唾液においてヒトコロナウイルスに交叉するIgA増加用の発酵乳。
(1)菌体外多糖を産生する乳酸菌により調製される、ヒトの唾液においてヒトコロナウイルスに交叉するIgA増加用の発酵乳。
(2)菌体外多糖を産生する乳酸菌により乳原料を発酵させて得られる、(1)に記載の発酵乳。
(3)ヒトコロナウイルスはα型である、(1)または(2)に記載の発酵乳。
(4)乳酸菌は、ラクトバチラシエ(Lactobacillaceae)科乳酸菌である、(1)から(3)のいずれか1つに記載の発酵乳。
(5)ラクトバチラシエ(Lactobacillaceae)科乳酸菌は、ラクトバチルス属乳酸菌である、(4)に記載の発酵乳。
(6)ラクトバチルス属乳酸菌は、以下の配列番号1の塩基配列と95%以上の配列同一性を有する16S rRNA遺伝子を有するラクトバチルス属乳酸菌である、(5)に記載の発酵乳。

(8)ラクトバチルス デルブルエッキー 亜種 ブルガリクスは、ラクトバチルス デルブルエッキー 亜種 ブルガリクスOLL1073R-1(FERM BP-10741)である、(7)に記載の発酵乳。
(9)ストレプトコッカス サーモフィルスを併用して調製される、(1)から(8)のいずれか1つに記載の発酵乳。
(10)ヒトは、高齢のヒトである、(1)から(9)のいずれか1つに記載の発酵乳。なお、ここにいう「高齢のヒト」とは、満年齢65歳以上のヒトである。
(11)無脂乳固形分の濃度は、4.0質量%以上12.0質量%以下の範囲内であり、100g/日以上120g/日以下の範囲内または100mL/日以上120mL/日以下の範囲内の量を1日1回、4週間以上継続して経口摂取させる、(1)から(10)のいずれか1つに記載の発酵乳。

(13)菌体外多糖を産生する乳酸菌により調製される発酵乳を、それを必要とするヒトに投与する、ヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させる方法。
(13)菌体外多糖を産生する乳酸菌により調製される発酵乳を、それを必要とするヒトに投与する、ヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させる方法。
(14)ヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させるための、菌体外多糖を産生する乳酸菌により調製される発酵乳の使用。
(15)ヒトの唾液においてヒトコロナウイルスに交叉するIgA増加用の発酵乳の有効成分として使用する、菌体外多糖を産生する乳酸菌により調製される発酵乳。

本発明によれば、菌体外多糖を産生する乳酸菌により調製される発酵乳をヒトに摂取させることにより、そのヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させることができ、延いてはヒトコロナウイルス感染予防を実現することができる。
本発明の実施の形態に係る発酵乳は、菌体外多糖を産生する乳酸菌により調製されるものであって、ヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させる機能を有する。なお、この発酵乳は、ヒトの唾液においてα型ヒトコロナウイルスに交叉するIgAを増加させるのに特に有効である。また、α型ヒトコロナウイルスにはHCoV-229EおよびHCoV-NL63のヒトコロナウイルスが含まれる。また、ここにいうヒトは、免疫機能が低下傾向となる65歳以上のヒトであることが好ましいが、さらに高齢のヒト、例えば、70歳以上のヒトであってもよいし、75歳以上のヒトであってもよい。
なお、乳酸菌は、ラクトバチラシエ(Lactobacillaceae)科乳酸菌であることが好ましい。ラクトバチラシエ科乳酸菌は、特に限定されている場合を除き、下記の参考文献4に記載の区分に従って分類される。

本発明の実施の形態に係るラクトバチルス属乳酸菌は、分類学的にラクトバチルス属の細菌と認定されたものの全ての総称であり、菌種や菌株などで限定されるものではない。なお、ラクトバチルス属乳酸菌はその由来により、植物由来および動物由来のいずれかに分類することもあるが、本発明では植物由来の乳酸菌、動物由来の乳酸菌のいずれも使用することができる。本発明のラクトバチルス属乳酸菌の菌株は、十分な食経験がある発酵乳(例えば、ヨーグルト等)を調製することができるラクトバチルス属乳酸菌に属する1種または2種以上の菌株であることが好ましい。このようなラクトバチルス属乳酸菌としては、以下の配列番号1の塩基配列(つまり、16S rRNA遺伝子の配列の全部又は特徴のある一部(V1領域、V2領域、V3領域、又はV1領域及びV2領域及びV3領域の全部又は一部、又はV1領域及びV2領域及びV3領域を含む一部等))と95%以上の配列同一性を有する16S rRNA遺伝子を有するラクトバチルス属乳酸菌であることが好ましく、同配列番号1の塩基配列と98%以上の配列同一性を有する16S rRNA遺伝子を有するラクトバチルス属乳酸菌であることがより好ましく、同配列番号1の塩基配列と99%以上の配列同一性を有する16S rRNA遺伝子を有するラクトバチルス属乳酸菌であることがさらに好ましい。なお、同配列番号1は、ラクトバチルス デルブルエッキー 亜種 ブルガリクス(Lactobacillus delbrueckii subsp. bulgaricus)OLL1073R-1の16S rRNA遺伝子のV1~V3領域の配列(16S rRNA遺伝子の全長配列において34~535番目までの塩基配列に相当)の塩基配列(「ヌクレオチド配列」と称することもある。)である。

本発明に係る実施の形態において、配列同一性というときは、特に記載した場合を除き、2つの配列を最適に整列させた場合に、配列間で共有する一致した塩基の個数の割合を意味する。塩基配列の同一性に関する解析は、当業者には周知のアルゴリズム又はプログラム(例えば、BLASTN、BLASTP、BLASTX、ClustalW)、により行うことができる。プログラムを用いる場合のパラメーターは、当業者であれば適切に設定することができ、また各プログラムのデフォルトパラメーターを用いてもよい。これらの解析方法の具体的な手法もまた、当業者には周知である。同一性の計算には市販の遺伝子情報処理ソフトウェアを用いてもよい。
また、ラクトバチルス デルブルエッキー 亜種 ブルガリクスOLL1073R-1(以下、「本菌株」と称することがある。)と同一の菌学的性質を有する菌株も利用することができる。その菌学的性質は以下のとおりである。

ガラクトースとグルコースを構成糖として含む多糖類を細胞外へ生産するラクトバチラシエ科乳酸菌として、本菌株以外にも、Lactiplantibacillus plantarum C88(参考文献5参照)、Lactobacillus. johnsonii strain 151(参考文献6参照)、L. delbrueckii subsp. bulgaricus NCFB2074(参考7参照文献)、Lacticaseibacillus casei CG11(参考文献8参照)、Lacticaseibacillus rhamnosus E/N(参考文献9参照)、Lactobacillus fermentum TDS030603(参考文献10参照)、Lactobacillus gasseri FR4(参考文献11参照)などが存在する。本発明に係る実施の形態における組成物では、ガラクトースとグルコースを構成糖として含む多糖類を細胞外へ生産するラクトバチラシエ科乳酸菌から産生される菌体外多糖を含むことが好ましく、ガラクトースとグルコースを構成糖とする多糖類を細胞外へ生産するラクトバチラシエ科乳酸菌から産生される菌体外多糖を含むことがより好ましく、ガラクトースとグルコースを構成糖として含み、(好ましくはガラクトースとグルコースを構成糖とし)リンを含有する多糖類を細胞外へ生産するラクトバチラシエ科乳酸菌から産生される菌体外多糖を含むことが特に好ましい。

本発明の実施の形態に係る菌体外多糖は、ラクトバチラシエ科乳酸菌から産生されることが好ましく、ラクトバチルス属乳酸菌から産生されることがより好ましく、ラクトバチルス デルブルエッキーから産生されることがさらに好ましく、ラクトバチルス デルブルエッキー 亜種 ブルガリクスから産生されることがさらに好ましく、ラクトバチルス デルブルエッキー 亜種 ブルガリクスOLL1073R-1(寄託番号:FERM BP-10741)乳酸菌(つまり、OLL1073R-1株)から産生されることが特に好ましい。
また、ここにいう菌体外多糖は、目的の効果を有する限り、特に限定されないが、上述の乳酸菌により原料乳(好ましくは、乳等省令における乳を含む原料)を発酵して生成されるものが好ましい。本発明の実施の形態において用いられる菌体外多糖は、1種でもよく、2種以上の組み合わせであってもよい。また、この菌体外多糖にはリン酸化多糖が含まれることが好ましい。また、この菌体外多糖には非リン酸化多糖が含まれてもよいが、非リン酸化多糖には硫酸化多糖は含まれないことが好ましい。
なお、ここで、「ラクトバチルス デルブルエッキー 亜種 ブルガリクスOLL1073R-1」は、独立行政法人産業技術総合研究所 特許生物寄託センターに寄託番号 FERM BP-10741(寄託日:2006年11月29日)で寄託されている。
本発明の実施の形態に係る発酵乳は、手軽に摂取することができる。また、発酵乳は食経験が豊富で安全性が高く、副作用の心配がないため、摂取者は安心して摂取することができる。
本発明の実施の形態における「発酵乳」とは、上述の通り、乳を発酵させたものをいい、例えば、乳及び乳製品の成分規格等に関する省令(乳等省令)で定義される「発酵乳」、「乳酸菌飲料」、「乳飲料」、「ナチユラルチーズ」等を包含するがこれらに限定されない。例えば、発酵乳は、乳等省令で定義される「発酵乳」、すなわち、生乳、牛乳、特別牛乳、生山羊乳、殺菌山羊乳、生めん羊乳、成分調整牛乳、低脂肪牛乳、無脂肪牛乳および加工乳などの乳またはこれと同等以上の無脂乳固形分を含む乳等を、乳酸菌または酵母で発酵させ、固形状(ハードタイプ)、糊状(ソフトタイプ)または液状(ドリンクタイプ)にしたもの、または、これらを凍結したものをいうが、これに限定されない。本発明の実施の形態に係る発酵乳において、無脂乳固形分の濃度の範囲は、例えば、4.0質量%以上12.0質量%以下の範囲内であることが好ましく、6.0質量%以上10.0質量%以下の範囲内であることがより好ましく、7.0質量%以上9.0質量%以下の範囲内であることがさらに好ましい。また、乳脂肪分の濃度は、例えば、0.2質量%以上4.0%質量以下の範囲内であることが好ましく、0.3質量%以上3.0質量%以下の範囲内であることがより好ましく、0.4質量%以上2.0質量%以下の範囲内であることがさらに好ましい。
発酵乳の典型例としては、ヨーグルトが挙げられる。国際連合食糧農業機関(FAO)/世界保健機構(WHO)で定義されている国際規格にも、「ヨーグルトと称される製品は、Streptococcus thermophilus、Lactobacillus delbrueckii ssp. bulgaricusの両方の菌の乳酸発酵作用により、乳及び脱脂粉乳などの乳製品から作られるもので、最終製品中には前述の2つの菌が多量に生存しているもの」と規定されている。本発明において、「ヨーグルト」とは、前記のFAO/WHOの定義するヨーグルトを包含するものである。このヨーグルトには、例えばプレーンヨーグルト、ハードヨーグルト(セットタイプヨーグルト)、ソフトヨーグルト、ドリンクヨーグルトなどが含まれる。本発明の実施の形態に係るヨーグルトとしては、食べ応えや満足感の観点から、特にハードヨーグルトが好ましい。また、対象が高齢者である場合、誤嚥防止の観点からも、ハードヨーグルトが特に好ましい。
本発明の実施の形態に係る発酵乳の調製方法としては、例えば、「原料乳を殺菌して冷却した後、その原料乳に、上述の乳酸菌を含む乳酸菌スターターを添加して、その乳酸菌スターター入り原料乳を所定の乳酸酸度となるような発酵温度と発酵時間で発酵させる方法」が挙げられる。菌体外多糖は、発酵中にラクトバチルス属乳酸菌により生成される。同方法において、乳酸酸度は、例えば、0.6~1.2である。なお、乳酸酸度は0.6~0.8であることが好ましい。また、発酵温度は、例えば、40~45℃である。また、発酵時間は、例えば2~12時間である。なお、発酵時間は3~8時間であることが好ましい。
乳酸菌スターターとして、ストレプトコッカス属乳酸菌またはストレプトコッカス サーモフィラス(Streptococcus thermophilus)をラクトバチラシエ科乳酸菌、特にラクトバチルス属の乳酸菌と併用することが、生産効率および嗜好性の観点から好ましい。さらに、ラクトバチルス属の乳酸菌がラクトバチルス デルブルエッキー 亜種 ブルガリクス(Lactobacillus delbrueckii subsp. bulgaricus) OLL1073R-1(寄託番号: FERM BP-10741)である場合、サーモフィラス菌は Streptococcus thermophilus 1131や、Streptococcus thermophilus OLS3059であることがより好ましい。Streptococcus thermophilus OLS3059は、独立行政法人産業技術総合研究所 特許生物寄託センターに寄託番号 FERM BP-10740(寄託日:2006年11月29日)で寄託されている。Streptococcus thermophilus 1131は、株式会社明治製の明治ブルガリアヨーグルト「LB81」から入手することができる。
また、乳酸菌スターターとしてまたは機能性の付与などを目的として、ラクトバチルス属乳酸菌およびストレプトコッカス属乳酸菌以外の乳酸菌や、ラクトコッカス菌、ビフィズス菌、酵母などの発酵微生物を添加してもよい。
本発明の実施の形態に係る発酵乳を1回の摂取に適切な量で1個包装の形態とすることにより、本発明の実施の形態に係る発酵乳を適切かつ手軽に摂取することができることになり、使用性の面から好ましい。1回の摂取に適切な量は個人差やばらつきがあるものの、無脂乳固形分が8.0質量%である発酵乳である場合には、1回あたりで10mL以上1000mL以下の範囲内であることが好ましく、30mL以上500mL以下の範囲内であることがより好ましく、50mL以上200mL以下の範囲内であることがさらに好ましく、80mL以上120mL以下であることが好ましい。あるいは、1回あたりで10g以上1000g以下の範囲内であることが好ましく、30g以上500g以下の範囲内であることが好ましく、50g以上200g以下の範囲内であることが好ましく、80g以上150g以下の範囲内であることがより好ましく、100g以上120g以下の範囲内であることがさらに好ましい。
本発明の実施の形態では、「1個包装の形態」はあらゆる形態、例えば、蓋付きの容器、キャップ付きのボトル、個袋、パウチ、チューブなどの一般的な包装形態が包含される。本発明の実施の形態では、各個包装、または複数の個包装を含む包装に、本発明の実施の形態に係る発酵乳の用途、効能、摂取方法などの説明を記載すること、および/または、その説明を記載した物等を封入等すること、および/または、別途パンフレットなど、その説明を記載した物等を掲示することなどにより、その用途を明確にすることができる。
本発明の実施の形態に係る発酵乳は、ヒトコロナウイルスの感染予防効果を高める観点から、3日間以上継続して摂取することが好ましく、5日間以上継続して摂取することが好ましく、1週間以上継続して摂取することが好ましく、2週間以上継続して摂取することが好ましく、4週間以上継続して摂取することが好ましく、6週間以上継続して摂取することがより好ましく、8週間以上継続して摂取することがさらに好ましく、10週間以上継続して摂取することがさらに好ましく、12週間以上継続して摂取することがさらに好ましく、24週間以上継続して摂取することがさらに好ましく、36週間以上継続して摂取することが特に好ましい。なお、本発明の実施の形態に係る発酵乳は、十分な食経験があり安全に摂取することができるため、摂取期間の上限は特に制限されず、永久的に継続することが可能であるが、強いて上限を設けるならば、例えば60週間以下である。なお、この上限は、例えば、120週間以下としてもよいし、100週間以下としてもよいし、80週間以下としてもよい。なお、摂取の態様としては、経口摂取であることが好ましい。
ところで、本発明の実施の形態に係る発酵乳に、その用途、効能、機能、有効成分の種類、使用方法などの説明を表示することが好ましい。ここにいう「表示」には、需要者に対して上記効果の説明を知らしめるための全ての表示が含まれる。この表示は、上述の表示内容を想起・類推させ得るような表示であればよく、表示の目的、表示の内容、表示する対象物・媒体などの如何に拘わらない全てのあらゆる表示を含み得る。例えば、製品の包装・容器に上記説明を表示すること、製品に関する広告・価格表もしくは取引書類に上記説明を表示して展示もしくは頒布すること、またはこれらを内容とする情報を電磁気的(インターネットなど)方法により提供することが挙げられる。
本発明の実施の形態に係る発酵乳を包装してなる製品には、例えば、「唾液においてヒトコロナウイルスに交叉するIgAを増加させる」等との表示が付されることが好ましい。
なお、以上のような表示を行うために使用する文言は、上述の例に限定されず、そのような意味と同義である文言であってもかまわない。そのような文言としては、例えば、需要者に対して、「唾液中のIgAを増加させる」、「唾液中のコロナウイルスに対するIgAを増加させる」等の種々の文言が許容され得る。
なお、上述の発酵乳は、特別用途食品、総合栄養食品、栄養補助食品、特定保健用食品、機能性表示食品、加工食品等の形態にすることもできる。また、同発酵乳は、ドリンクヨーグルト以外の飲料や流動食等の組成物に配合することもできる。また、同発酵乳は、乾燥処理、濃縮処理を行うなどしてから用いることもできる。
以下、実施例および比較例に基づいて、本発明をより具体的に説明する。なお、この実施例は、本発明を限定するものではない。
(試験例)
-試験サンプル-
株式会社明治製の明治プロビオヨーグルトR-1ハードタイプを用いて以下の試験を実施した。なお、このハードヨーグルトは、ラクトバチルス デルブルエッキー 亜種 ブルガリクス OLL1073R-1(FERM BP-10741)及びストレプトコッカス サーモフィルスで乳を発酵して得られたものである。ここで、ラクトバチルス デルブルエッキー 亜種 ブルガリクス OLL1073R-1(FERM BP-10741)は、菌体外多糖を産生する乳酸菌である。そして、このハードヨーグルトには、OLL1073R-1(FERM BP-10741)により産生された菌体外多糖が含まれている。また、本ハードヨーグルトは2009年の発売開始以降十分な販売実績を有しており、過去において本製品が原因と考えられる重篤な副作用が報告されたことはない。

特別養護老人ホームAには上述の明治プロビオヨーグルトR-1ハードタイプを数年にわたり提供し続けており、入居者は毎日1個(112g)を摂取し続けていた。特別養護老人ホームAの入居者41名(76歳~102歳(平均年齢86.7歳))と、対照として、上述の明治プロビオヨーグルトR-1ハードタイプを提供しなかった介護老人保健施設Bの入居者49名(76歳~99歳(平均年齢90.4歳))を、解析対象として選定した。そして、特別養護老人ホームAに上述の明治プロビオヨーグルトR-1ハードタイプを提供開始してから4年8か月後に各老人ホームの入居者から唾液を採取すると共に、唾液分泌速度を測定した。なお、この際、唾液採取直前の2ケ月に風邪を罹患した入居者については解析対象から除外した。これは、風邪罹患により唾液中IgA分泌速度が亢進した可能性を排除するためである。この結果、特別養護老人ホームAの解析対象数は変わらず41名、介護老人保健施設Bの解析対象数は43名となった。

次に、採取した各唾液中の総IgA濃度を測定すると共に、α型ヒトコロナウイルスであるHCoV-229EおよびHCoV-NL63に交叉する唾液中のIgA濃度をELISA法に従って測定し、上述の各IgA濃度の値に唾液分泌速度を乗じて各IgA分泌速度を算出した。なお、α型ヒトコロナウイルスに交叉するIgA濃度は、任意に選抜した唾液検体の混合物の値を1arbitrary units(AU)/mLとし、その相対値(AU/mL)で表した。
なお、ELISA法に従ったα型ヒトコロナウイルスに交叉する唾液中のIgA濃度の測定方法は以下の通りであった。

(2)前処理
(2-1)唾液処理
各唾液を解凍した後、その唾液を4℃の温度下、3,000gの遠心力で5分間、遠心処理し、上清を得た。以下、この上清を試験に供した。

(2-2)標準試料の調製
任意に選抜した各被験者の唾液を等量ずつ混合して標準試料を調製した。上述の通り、α型ヒトコロナウイルスに交叉するIgA濃度については、この標準試料のそれが1arbitrary units(AU)/mLとなる。次に、この標準試料を適量ずつ分注し、使用するまで凍結保存した。
任意に選抜した各被験者の唾液を等量ずつ混合して標準試料を調製した。上述の通り、α型ヒトコロナウイルスに交叉するIgA濃度については、この標準試料のそれが1arbitrary units(AU)/mLとなる。次に、この標準試料を適量ずつ分注し、使用するまで凍結保存した。
(3)ELISA手順
(3-1)抗原固相化
上述の各抗原をCoating Bufferで1μg/mLに調整した。次に、その溶液を96ウェルプレートの各ウェルに80μLずつ注ぎ入れた。次いで、その96ウェルプレートをシールして4℃で一晩静置した。なお、非特異的反応の確認のため、抗原なしのウェルも作製した。

上述の各抗原をCoating Bufferで1μg/mLに調整した。次に、その溶液を96ウェルプレートの各ウェルに80μLずつ注ぎ入れた。次いで、その96ウェルプレートをシールして4℃で一晩静置した。なお、非特異的反応の確認のため、抗原なしのウェルも作製した。
(3-2)ブロッキング
抗原を固相化した96ウェルプレートをWash Bufferで3回洗浄した後、その96ウェルプレートの各ウェルにChonBlock blocking bufferを200μLずつ注ぎ入れた。その後、その96ウェルプレートをシールして37℃で1時間静置した。

(3-3)一次抗体反応
ブロッキング処理済みの96ウェルプレートをWash Bufferで3回洗浄した後、その96ウェルプレートの各ウェルに、ChonBlock antibody dilution bufferで希釈(50~200倍程度)した唾液を80μLずつ注ぎ入れた。その後、その96ウェルプレートをシールして37℃で2時間静置した。
ブロッキング処理済みの96ウェルプレートをWash Bufferで3回洗浄した後、その96ウェルプレートの各ウェルに、ChonBlock antibody dilution bufferで希釈(50~200倍程度)した唾液を80μLずつ注ぎ入れた。その後、その96ウェルプレートをシールして37℃で2時間静置した。
(3-4)二次抗体反応
一次抗体反応処理後の96ウェルプレートをWash Bufferで5回洗浄した後、その96ウェルプレートの各ウェルに、ChonBlock antibody dilution bufferで10,000倍希釈した HPR2次抗体を80μLずつ注ぎ入れた。その後、その96ウェルプレートをシールして37℃で1時間静置した。
一次抗体反応処理後の96ウェルプレートをWash Bufferで5回洗浄した後、その96ウェルプレートの各ウェルに、ChonBlock antibody dilution bufferで10,000倍希釈した HPR2次抗体を80μLずつ注ぎ入れた。その後、その96ウェルプレートをシールして37℃で1時間静置した。
(3-5)発光・測定
TMB Substrate SolutionのA液およびB液を等量ずつ分注しておいた。また、二次抗体反応処理後の96ウェルプレートをWash Bufferで5回洗浄した。上述のA液とB液を混合し、その混合液を80μLずつ、洗浄済みの96ウェルプレートの各ウェルに注ぎ入れた。次に、その96ウェルプレートをシールせずに常温で遮光して30分間静置した後に、各ウェルに40μLの2N H2SO4を注ぎ入れて各ウェルにおける反応を停止させた。最後に、各ウェル中の試料に対して450nm~570nmで光学密度(OD)測定を行った。

-各種データの統計学的処理方法-
IgA分泌速度の老人ホーム間の差は、Mann-Whitney U Testで解析した。

図1に示されるように、唾液中の総IgA分泌速度については老人ホーム間で差がなかったが、HCoV-229EおよびHCoV-NL63に交叉する唾液中のIgA分泌速度については介護老人保健施設Bの入居者よりも特別養護老人ホームAの入居者の方が有意に高かった。なお、HCoV-229Eに対するp値は0.05未満であり(p<0.05)、HCoV-NL63に対するp値は0.1未満であった(p<0.1)。したがって、明治プロビオヨーグルトR-1ハードタイプが、ヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させることが確認された。
図1に示されるように、唾液中の総IgA分泌速度については老人ホーム間で差がなかったが、HCoV-229EおよびHCoV-NL63に交叉する唾液中のIgA分泌速度については介護老人保健施設Bの入居者よりも特別養護老人ホームAの入居者の方が有意に高かった。なお、HCoV-229Eに対するp値は0.05未満であり(p<0.05)、HCoV-NL63に対するp値は0.1未満であった(p<0.1)。したがって、明治プロビオヨーグルトR-1ハードタイプが、ヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させることが確認された。
本発明に係る発酵乳は、菌体外多糖を産生する乳酸菌により調製されるものであって、ヒトの唾液においてヒトコロナウイルスに交叉するIgAを増加させる機能を有する。

- 菌体外多糖を産生する乳酸菌により調製される、ヒトの唾液においてヒトコロナウイルスに交叉するIgA増加用の発酵乳。
- 前記菌体外多糖を産生する乳酸菌により乳原料を発酵させて得られる
