Abstract
본 발명은 귀리 유래 β-(1,3; 1,4)-D-글루칸을 유효성분으로 포함하는 면역증강제(adjuvant) 및 이를 포함하는 백신 조성물에 관한 것이다. 본 발명은 귀리를 기질로 한 유산균의 발효 배양액으로부터 분리 정제한 선형의 β-(1,3; 1,4)-D-글루칸을 면역증강제로 사용함으로서 포도상규균 및 백일해를 비롯한 다양한 병원균 유래 항원의 선천성 및 체액성 면역 반응을 현저한 수준으로 지속적으로 증진시킨다. 이에 본 발명은 면역증진 활성, 수급 용이성, 장기 투여시의 안정성이 모두 우수한 효율적인 천연 유래 면역증강제로 유용하게 이용될 수 있다.
Description
본 발명은 귀리를 이용한 유산균의 복합발효배양을 통해 수득한 β-글루칸을 유효성분으로 포함하는 면역증강제 조성물에 관한 것이다.
순수 단백질 정제 항원이나 유전자 재조합 단백질 항원을 이용한 백신은 약독화 또는 생백신에 비하여 안전성이 우수하지만, 체액성 면역과 세포성 면역을 고루 활성화시키지 못해 이를 증진시키기 위한 면역보강제(adjuvants)를 함께 도입하거나 특정 제형(흡입, 경구 또는 패취등)의 적용이 필요하다. 최근에는 유전자 재조합 단백질 항원을 활용하는 다양한 백신이 개발되면서 안전성, 효율성 및 경제성을 갖춘 효율적인 면역보강제 개발이 활발하게 이루어지고 있다.
1세대 백신 면역보강제로는 알루미늄 염 어쥬번트(Alum)와 MPL 계열의 다양한 지질성 면역 보강제가 대표적인데, 알루미늄 염 어쥬번트는 세포성 면역반응인 Th1(helper T cell 1) 보다 체액성 면역반응인 Th2(helper T cell 2)를 주로 활성화시키고 체내 축적 시 신장 기능의 저하와 뇌와 골 조직 결손을 야기하는 부작용을 나타낸다. 한편 지질성 면역보강제는 세포성 및 체액성 면역반응을 모두 활성화시키지만 모두 심한 국소 반응 및 전신 반응과 같은 이상 반응의 문제가 있다.
최근 새로운 면역 보강제 후보물질로 α-글루칸, β-글루칸, β-프락탄, 만난(mannan), 키토산, EPS(exopolysaccharide), 아라비녹실란과 같이 식물과 미생물의 세포벽에 포함된 복합다당류가 주목받고 있다. 이러한 복합다당류는 선천면역 세포 표면에 존재하는 패턴 인식 수용체(pattern recognition receptors, PRR)에 의해 병원체 관련 분자 패턴 (PAMPs_pathogen-associated molecular patterns)으로 인식되어 신호전달경로 활성화, 유전자 발현, 사이토카인 분비, 세포 성숙 및 분화와 같은 일련의 효과를 유발하여 면역 반응을 활성화시키며 독성이 적고 자연 소멸되어 체내 잔류가 없는 특성이 있다.
본 명세서 전체에 걸쳐 다수의 논문 및 특허문헌이 참조되고 그 인용이 표시되어 있다. 인용된 논문 및 특허문헌의 개시 내용은 그 전체로서 본 명세서에 참조로 삽입되어 본 발명이 속하는 기술 분야의 수준 및 본 발명의 내용이 보다 명확하게 설명된다.
본 발명자들은 체액성 면역반응과 세포성 면역반응을 고루 촉진하면서도 안전성이 높고 수급이 용이한 효율적인 천연 유래 면역증강제를 개발하기 위하여 예의 연구 노력하였다. 그 결과, 특정 분자량 범위의 곡물 유래 β-글루칸, 구체적으로는 유산균을 이용한 귀리의 발효 과정을 통해 수득한 선형의 β-(1,3; 1,4)-D-글루칸을 면역증강제로 사용할 경우 장기간 투여에도 독성이 낮고 체내에 축적되지 않으면서도 선천성 면역과 체액성 면역을 현저한 수준으로 지속적으로 증진시킴을 발견함으로써, 본 발명을 완성하게 되었다.
따라서 본 발명의 목적은 β-(1,3; 1,4)-D-글루칸을 유효성분으로 포함하는 면역증강제(adjuvant) 및 이를 포함하는 백신 조성물을 제공하는 데 있다.
본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명, 청구범위 및 도면에 의해 보다 명확하게 된다.
본 발명의 일 양태에 따르면, 본 발명은 β-1,3 글루코사이드 결합 및 β-1,4 글루코사이드 결합을 통해 D-글루코피라노스 단량체가 선형으로 연결된 베타 글루칸 또는 이의 약제학적으로 허용되는 염을 유효성분으로 포함하는 면역증강제(adjuvant) 조성물을 제공한다.
본 발명자들은 체액성 면역반응과 세포성 면역반응을 고루 촉진하면서도 안전성이 높고 수급이 용이한 효율적인 천연 유래 면역증강제를 개발하기 위하여 예의 연구 노력하였다. 그 결과, 특정 분자량 범위의 곡물 유래 베타 글루칸, 구체적으로는 유산균을 이용한 귀리의 발효 과정을 통해 수득한 선형의 β-(1,3; 1,4)-D-글루칸을 면역증강제로 사용할 경우 장기간 투여에도 독성이 낮고 체내에 축적되지 않으면서도 선천성 면역과 체액성 면역을 현저한 수준으로 지속적으로 증진시킴을 발견하였다.
본 명세서에서 용어“면역증강제(adjuvant)”는 면역세포의 초기 활성화 과정에서 항원에 대한 면역 반응을 비특이적으로 촉진하는 물질로, 숙주의 면역계에 대해 면역원으로 직접 작용하진 않으나 면역계 세포의 활성을 증진시킴으로써 면역반응을 강화하는 모든 분자를 포괄하는 의미이다. 이에, 본 명세서에서 용어“면역증강제”는“면역보조제”,“면역증진제”또는“어쥬번트”와 동일한 의미로 사용된다.
본 발명에서 사용되는 면역증강제는 항원을 포함하는 백신 조성물과 동시에 투여되거나 또는 시간 간격을 두고 순차적으로 투여될 수 있다. 본 발명의 면역증강제가 백신 조성물과 동시에 투여될 경우 백신 조성물과 혼합된 단일 제형으로 제조되어 하나의 바이얼(vial) 또는 주사기(prefilled syringe) 등에 포장될 수도 있고, 또는 별도의 제형으로 제조되어 개별적으로 동시에 투여될 수도 있다.
본 명세서에서 용어“베타 글루칸(β-glucan)”은 곡물, 박테리아 및 균류 의 세포벽에 자연적으로 존재하는 β-D-글루코스 다당체(글루칸)를 포괄하는 의미이다. 베타 글루칸은 D-글루코피라노스 단량체가 β-글리코사이드 결합(glycosidic bond)을 통해 다당체를 형성한다는 공통점을 제외하고는 유래 물질(생물)에 따라 분자량, 용해도, 점도, 측쇄 여부, 기타 생물학적 및 생리학적 특성이 상이하다.
본 발명에 따르면, 본 발명은 β-1,3 글루코사이드 결합 및 β-1,4 글루코사이드 결합을 통해 D-글루코피라노스 단량체가 선형으로 연결된 특정 평균 분자량 범위의 베타 글루칸을 사용하며, 후술하는 바와 같이 이는 귀리를 기질(matrix)로 한 유산균 발효 배양액으로부터 특이적으로 분리·정제될 수 있다.
본 발명의 구체적인 구현예에 따르면, 상기 베타 글루칸은 380,000 MW (molecular weight) 내지 400,000 MW의 평균 분자량을 가진다. 보다 구체적으로는 385,000 MW 내지 395,000 MW의 평균 분자량을 가지고, 보다 더 구체적으로는 388,000 MW 내지 393,000 MW의 평균 분자량을 가지며, 가장 구체적으로는 약 39100 MW 의 평균 분자량을 가진다.
본 발명의 구체적인 구현예에 따르면, 상기 베타 글루칸은 귀리(Avena sativa), 보리(Hordeum vulgare), 밀(Triticum aestivum) 및 수수(Sorghum bicolor)로 구성된 군으로부터 선택되는 하나 이상의 곡물(cereal)로부터 추출된다. 보다 구체적으로는, 상기 곡물은 귀리(Avena sativa)이다.
본 발명의 구체적인 구현예에 따르면, 본 발명에서 이용되는 베타 글루칸은 귀리를 포함하는 생물학적 시료에 유산균 배양액을 접종하여 발효시킴으로서 수득한다.
본 명세서에서 용어“발효(Fermentation)”는 박테리아나 효모 등의 미생물이 산소 없이 당을 분해하여 생존과 증식에 필요한 에너지 및 화합물을 생산하는 혐기성(anaerobic) 대사과정을 의미한다.
본 명세서에서 용어“유산균(lactic acid bacteria)”은 탄수화물 대사의 주산물로서 유산을 생산하는 박테리아 군을 의미하며, 예를 들어 락토코커스(Lactococcus), 락토바실러스(Lactobacillus), 류코노스톡(Leuconostoc), 프로리오니박테리움(Propionibacterium), 엔테로코커스(Enterococcus), 비피도박테리움(Bifidobacterium), 스트렙토코커스(Streptococcus) 및 페디오코커스 (Pediococcus)를 포함하나, 이에 제한되는 것은 아니다.
보다 구체적으로는, 본 발명에서 이용되는 유산균은 락토바실러스속(Lactobacillus) 균주, 비피도박테리움속(Bifidobacterium) 균주 및 스트렙토코커스속(Streptococci) 균주로 구성된 군으로부터 선택되는 하나 이상의 균주이다.
보다 더 구체적으로는, 상기 락토바실러스속 균주는 락토바실러스 아시도필루스(Lactobacillus acidophilus) 및 락토바실러스 불가리쿠스(Lactobacillus bulgaricus)로 구성된 군으로부터 선택되는 하나 이상이다.
보다 더 구체적으로는, 상기 스트렙토코커스속 균주는 스트렙토코커스 써모필러스(Streptococcus thermophilus)이다.
본 발명에 따르면, 본 발명에서 면역 보강제의 유효성분으로 사용되는 β-글루칸은 귀리를 배양 기질(matrix)로 하여 락토바실러스 아시도필루스, 락토바실러스 불가리쿠스 및 스트렙토코커스 써모필러스 등의 유산균 균주를 복합 공생발효배양(symbiotic fermentation)한 배양액에서 분리 정제할 수 있다.
본 명세서에서 용어“약제학적으로 허용되는 염”은 약학적으로 허용되는 무기산, 유기산, 또는 염기로부터 유도된 염을 포함한다. 적합한 산의 예로는 염산, 브롬산, 황산, 질산, 과염소산, 푸마르산, 말레산, 인산, 글리콜산, 락트산, 살리실산, 숙신산, 톨루엔-p-설폰산, 타르타르산, 아세트산, 트리플루로초산, 시트르산, 메탄설폰산, 포름산, 벤조산, 말론산, 나프탈렌-2-설폰산, 벤젠설폰산 등을 들 수 있다. 적합한 염기로부터 유도된 염은 나트륨 등의 알칼리 금속, 마그네슘 등의 알칼리 토금속, 및 암모늄 등을 포함할 수 있다.
본 발명의 구체적인 구현예에 따르면, 본 발명의 면역증강제(adjuvant) 조성물은 알루미늄 염(aluminum salt)을 추가적으로 포함한다.
본 명세서에서 용어“알루미늄 염”은 3가 양이온 금속인 알루미늄과 음이온 분자가 이온결합을 이루는 금속염 화합물을 의미한다. 보다 구체적으로는, 본 발명에서 추가적인 면역증강제 성분으로 이용되는 알루미늄 염은 알루미늄 하이드록사이드(aluminum hydroxide) 및 알루미늄 포스페이트(aluminum phosphate)로 구성된 군으로부터 선택된다.
본 발명의 또 다른 양태에 따르면, 본 발명은 (a) 하나 이상의 항원 또는 이를 인코딩하는 핵산 분자; 및 (b) 전술한 본 발명의 면역증강제(adjuvant) 조성물을 포함하는 백신 조성물을 제공한다.
본 명세서에서 용어“백신”은 대상체 내에서 종양 또는 바이러스, 진균, 박테리아 등의 감염원 기타 병원체에 대한 보호적 면역 반응을 인위적으로 유도, 촉진 또는 강화하기 위하여 사용하는 약제학적 제제를 통칭하는 의미이다. 백신은 예방용 백신과 치료용 백신으로 구분되며, 대상체에 투여됨으로서 항체 및 면역 림프구(T-세포 및 B-세포)의 생산과 함께 면역 반응을 유도하는 세포 또는 항원을 유효성분으로 함유한다.
본 발명의 백신은 예방 또는 치료하고자 하는 대상 질환의 종류에 따른 적합한 항원을 단백질 형태 뿐 아니라 이를 코딩하는 핵산분자를 유효성분으로 하는 DNA 백신 또는 mRNA 백신의 형태로 이용될 수도 있다.
본 명세서에서, 용어“핵산 분자”및 이를 구성하는“뉴클레오타이드”는 단일가닥 또는 이중가닥 형태로 존재하는 디옥시리보뉴클레오타이드 또는 리보뉴클레오타이드이며, 다르게 특별하게 언급되어 있지 않은 한 자연의 뉴클레오타이드의 유사체(Scheit, Nucleotide Analogs, John Wiley, New York(1980); Uhlman 및 Peyman, Chemical Reviews, 90:543-584(1990)) 및 면역원성을 감소시키기 위한 변이 염기(예를 들어 슈도우리딘(pseudouridine), N1-메틸슈도유리딘(N1-methyl pseudouridine), 5-메틸시토신(5-Methylcytosine) 등)을 포함한다. 본 발명이 DNA 백신 또는 mRNA 백신 형태로 이용될 경우 본 발명의 항원 코딩 유전자를 각각의 유전자 전달체(gene delivery system)에 포함되어 대상체에서 발현시킬 수도 있다.
본 명세서에서 용어“발현시키다”는 대상체가 외래(exogenous) 유전자를 발현하게 하거나 또는 내인성(endogenous) 유전자의 자연적 발현량을 증가시키기 위해 유전자 전달체를 이용하여 인위적으로 이를 도입함으로써 유전자가 대상체 세포 내에서 염색체외 인자로서 또는 염색체 통합 완성에 의해 복제 가능하게 되는 것을 의미한다. 따라서, 용어“발현”은“형질전환(transformation)”, "형질감염(transfection)" 또는 "형질도입(transduction)"과 동일한 의미이다.
본 명세서에서 용어 “유전자 전달체”는 원하는 타겟 유전자를 대상 세포에 도입하여 발현시키기 위한 매개체를 의미한다. 이상적인 유전자 전달체는 인체에 무해하고 대량생산이 용이하며 효율적으로 유전자를 전달할 수 있어야 한다.
본 명세서에서, 용어“유전자 전달”은 유전자가 세포 내로 운반되는 것을 의미하며, 유전자의 세포내 침투(transduction)와 동일한 의미를 가진다. 조직 수준에서, 상기 용어 유전자 전달은 유전자의 확산(spread)과 동일한 의미를 가진다. 따라서, 본 발명의 유전자 전달체는 유전자 침투 시스템 및 유전자 확산 시스템으로 기재될 수 있다.
본 발명의 유전자 전달체을 제조하기 위해, 본 발명의 뉴클레오타이드 서열은 적합한 발현 컨스트럭트(expression construct) 내에 존재하고, 프로모터, 시그널 서열, 전사조절인자 결합 위치의 어레이 등의 발현조절 서열에 작동적으로 연결되는 것이 바람직하다. 본 명세서에서, 용어 “작동적으로 연결된”은 핵산 발현조절 서열과 다른 핵산 서열사이의 기능적인 결합을 의미하며, 이에 의해 상기 조절 서열은 상기 다른 핵산 서열의 전사 및/또는 해독을 조절하게 된다.
본 발명의 유전자 전달체는 다양한 형태로 제작할 수 있는 데, 이는 (ⅰ) 내이키드(naked) 재조합 DNA 분자, (ⅱ) 플라스미드, (ⅲ) 바이러스 벡터, (ⅳ) 상기 내이키드 재조합 DNA 분자 또는 플라스미드를 내포하는 리포좀 또는 니오좀의 형태, 또는 (v) mRNA를 내포하는 리포좀 형태로 제작할 수 있다.
본 발명의 구체적인 일 구현예에 따르면, 상기 항원은 포도상구균(Staphylococcus aureus) 유래 항원이다.
보다 구체적으로는, 상기 포도상구균은 메티실린 저항성 포도상구균(methicillin-resistant Staphylococcus aureus, MRSA), 메티실린 민감성 포도상구균(methicillin-sensitive Staphylococcus aureus, MSSA) 또는 병원성 포도상구균일 수 있으며, 보다 더 구체적으로는 메티실린 저항성 포도상구균이다.
본 발명에서 이용되는 포도상구균 유래 항원은 포도상구균 유래 독소(toxin)일 수 있으며, 보다 구체적으로는 Hla(alpha-hemolysin), LukS(Leukocidal toxin S), LukAB(Leukocidal toxin AB) 및 HlgA(gamma-hemolysin)로 구성된 군으로부터 선택되는 하나 이상의 포도상구균 유래 독소일 수 있다.
포도상구균 감염 질환의 예는 연부조직 감염, 화농성 관절염, 화농성 골수염, 중이염, 폐렴, 패혈증, 급성호흡기 감염(acute respiratory tract infection), 카테터의 사용으로 인한 감염, 수술 후 창상 감염, 균혈증, 심내막염 및 식중독을 포함하나, 이에 제한되는 것은 아니다.
본 발명의 구체적인 일 구현예에 따르면, 상기 항원은 백일해균(Bordetella pertussis) 유래 항원일 수 있다. 보다 구체적으로는, 본 발명에서 이용되는 백일해균 유래 항원은 PT(Pertussis toxin), FHA(Filamentous hamagglutinin), PRN (Pertactin) 및 FIM(Fimbriae)로 구성된 군으로부터 선택되는 하나 이상일 수 있으며, 가장 구체적으로는 PT이다.
백일해는 그람 음성의 호기성 간균인 백일해균(Bordetella pertussis) 감염으로 인한 질환으로 주로 영유아에게 발병하는 급성 호흡기 질환이다. 백일해 백신은 파상풍 및 디프테리아 불활화 사백신과 혼합하여 사용되어오고 있으나 전세포 백신(whole-cell vaccine)의 발작, 부종, 발열 등의 부작용이 보고된 이후, PT, FHA 등의 항원 단백질을 이용한 서브유닛 백신이 주로 개발되고 있다.
본 명세서에서 용어“예방”은 질환 또는 질병을 보유하고 있다고 진단된 적은 없으나, 이러한 질환 또는 질병에 걸릴 가능성이 있는 대상체에서 질환 또는 질병의 발생을 억제하는 것을 의미한다.
본 명세서에서 용어“치료”는 (a) 질환, 질병 또는 증상의 발전의 억제; (b) 질환, 질병 또는 증상의 경감; 또는 (c) 질환, 질병 또는 증상을 제거하는 것을 의미한다. 본 발명의 백신 조성물을 대상체에 투여하면 항원에 대한 대상체의 선천성, 세포성 및 체액성 면역반응을 현저한 수준으로 지속적으로 증진시킴으로서 상기 항원을 발현하는 병원체로 인한 증상의 발전을 억제하거나, 이를 제거하거나 또는 경감시키는 역할을 한다. 따라서, 본 발명의 백신 조성물은 그 자체로 이들 질환 치료의 조성물이 될 수도 있고, 혹은 다른 약리성분과 함께 투여되어 상기 질환에 대한 치료 보조제로 적용될 수도 있다. 이에, 본 명세서에서 용어“치료”또는“치료제”는“치료 보조”또는“치료 보조제”의 의미를 포함한다.
본 명세서에서 용어“투여”또는“투여하다”는 본 발명의 조성물의 치료적 유효량을 대상체에 직접적으로 투여함으로써 대상체의 체내에서 동일한 양이 형성되도록 하는 것을 말한다.
본 발명에서 용어“치료적 유효량”은 본 발명의 약제학적 조성물을 투여하고자 하는 개체에게 조성물 내의 약리성분이 치료적 또는 예방적 효과를 제공하기에 충분한 정도로 함유된 조성물의 함량을 의미하며, 이에“예방적 유효량”을 포함하는 의미이다.
본 명세서에서 용어“대상체”는 제한없이 인간, 마우스, 래트, 기니아 피그, 개, 고양이, 말, 소, 돼지, 원숭이, 침팬지, 비비 또는 붉은털 원숭이를 포함한다. 구체적으로는, 본 발명의 대상체는 인간이다.
본 발명의 조성물이 약제학적 조성물로 제조되는 경우, 본 발명의 약제학적 조성물은 약제학적으로 허용되는 담체를 포함한다.
본 발명의 약제학적 조성물에 포함되는 약제학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약제학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
본 발명의 약제학적 조성물은 경구 또는 비경구 투여할 수 있으며, 구체적으로는 비경구로, 보다 구체적으로는 정맥, 피하 또는 복강 투여될 수 있다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다. 본 발명의 약제학적 조성물의 바람직한 투여량은 성인 기준으로 0.001-100 ㎎/kg 범위 내이다.
본 발명의 약제학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액, 시럽제 또는 유화액 형태이거나 엑스제, 산제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
본 발명의 약제학적 조성물이 백신 조성물로 제조될 경우, 본 발명의 적합한 항원과 면역증강제가 선택적으로 조합된 하나의 바이얼(vial) 또는 주사기(prefilled syringe) 등에 포장되거나, 또는 항원과 면역증강제가 별개의 바이얼에 포장되어 사용 직전에 이를 혼합하여(용시조제, bed side mixing) 사용될 수 있다.
본 발명의 또 다른 양태에 따르면, 본 발명은 β-1,3 글루코사이드 결합 및 β-1,4 글루코사이드 결합을 통해 D-글루코피라노스 단량체가 선형으로 연결된 베타 글루칸 또는 이의 약제학적으로 허용되는 염을 대상체에 투여하는 단계를 포함하는 면역 증강 방법을 제공한다.
본 발명의 또 다른 양태에 따르면, 본 발명은 β-1,3 글루코사이드 결합 및 β-1,4 글루코사이드 결합을 통해 D-글루코피라노스 단량체가 선형으로 연결된 베타 글루칸 또는 이의 약제학적으로 허용되는 염의 면역 증강 용도를 제공한다.
본 발명에서 이용되는 베타 글루칸, 이를 면역 증강제로 포함하는 백신 조성물 및 이를 이용하여 예방 또는 치료될 수 있는 질환에 대해서는 이미 상술하였으므로, 과도한 중복을 피하기 위해 그 기재를 생략한다.
본 발명의 특징 및 이점을 요약하면 다음과 같다:
(a) 본 발명은 귀리 유래 β-(1,3; 1,4)-D-글루칸을 유효성분으로 포함하는 면역증강제(adjuvant) 및 이를 포함하는 백신 조성물을 제공한다.
(b) 본 발명은 귀리를 기질로 한 유산균의 발효 배양액으로부터 분리 정제한 선형의 β-(1,3; 1,4)-D-글루칸을 면역증강제로 사용함으로서 포도상규균 및 백일해를 비롯한 다양한 병원균 유래 항원의 선천성 및 체액성 면역 반응을 현저한 수준으로 지속적으로 증진시킨다.
(c) 이에 본 발명은 면역증진 활성, 수급 용이성, 장기 투여시의 안정성이 모두 우수한 효율적인 천연 유래 면역증강제로 유용하게 이용될 수 있다.
도 1은 귀리를 기질(matrix)로 한 유산균 발효 배양액으로부터 β-글루칸을 정제하는 과정을 요약한 모식도이다.

도 3은 정제된 β-글루칸의 동결건조 시료의 외형을 보여주는 그림으로, 본 발명의 방법으로 정제된 시료(PK1)는 동결건조 후 백색의 균질한 분말 형태를 띄는데 반하여, PK2는 동결건조 후 갈색의 비균질 분말이 형성되었다.
도 4는 본 발명의 방법으로 2차 정제된 β-글루칸의 당 정량 결과를 보여주는 그림으로, 정제된 분획별 흡광도 및 총 당량(도 4a), 정제된 β-글루칸에 대한 HPLC 피크(도 4b) 및 시간별 β-글루칸 분획의 종 당량(도 4c)을 각각 나타낸다.

도 7은 메티실린 저항성 포도상구균(methicillin-resistant Staphylococcus aureus, MRSA)의 항원인 LukS, iLukAB, iHla 및 HlgA와 EPS(exopolysaccharide), Alum 및 본 발명의 β-글루칸을 각각 혼합하여 접종한 후 IgG 역가를 측정한 결과를 보여주는 그림이다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실시예 1: β-글루칸의 정제 및 구조분석
β-글루칸의 정제
귀리(Avena sativa)를 기질(matrix)로 하여 발효 배양한 유산균의 배양액으로부터 베타 글루칸을 정제하였다(도 1). 구체적으로, 크리스챤 한센사(Chr. Hansen, Denmark)로부터 구입한 3종의 유산균(Lactobacillus delbrueckii subsp. Lactobacillus bulgaricus, Streptococcus thermophilus)이 포함된 YoFlex® Mild 1.0(Item No.; 703027)과 동사의 4종의 유산균(Bifidobacterium species, Streptococcus thermophilus, Lactobacillus acidophilus, Lactobacillus delbrueckii subsp)이 포함된 FD-DVS ABY-3 Probio-Tec®(Item No.; 669852)를 각각 500U/2,500L 용량 기준으로 투여하여 발효를 진행하였다.
TCA(Trichloroacetic acid) 및 EtOH를 처리하고 20 mM 아세트산 암모늄 완충액에 용해된 상층액을 동결건조하여 β-글루칸 건조 시료를 아세트산 암모늄 완충액에 용해시켰다. 불용성 분획을 원심분리(11000 x g rpm, 5분, 4℃)로 제거하고 EtOH로 침전시킨 수용성 귀리 β-글루칸 시료 약 50 mg을 S-300 컬럼에 로딩하여 20mM 아세트산암모늄 완충액 하에서 정제하였다. 유속은 0.3 ml/min이고 10 분/튜브로 수집하였으며 UV 파장은 220nm이다.
다당류 시료의 가수분해
앰플(5 ml) 당 1.5 mg의 다당류 시료를 1 ml의 3M TFA(Trifluoroacetic acid)에 용해시켰다. 앰플을 질소 대기 하에 밀봉하고 오븐에서 100℃로 유지하여 8시간 동안 다당류를 단당류로 가수분해하였다. 상온으로 냉각 후, 반응 혼합물을 4000 rpm에서 10분 간 원심분리하였다. 이후 상등액을 수집하여 감압 하에서 건조시킨 뒤 가수분해 및 건조된 시료를 50μl 증류수에 용해시키고 다음 실험에 이용하였다.
유도체화 과정
가수분해 및 건조를 통해 수득된 시료 15μl에 35μl의 DDW, 0.3M 액상 NaOH(50μl) 및 PMP의 0.5M 메탄올 용액(50μl)을 첨가하였다. 각 혼합물은 70℃에서 60분 간 반응시켰고, 상온으로 냉각시킨 뒤 50μl의 0.3 M HCl로 중화시켰다. 생성된 용액을 1 ml 클로로포름으로 추출하고 이상의 과정을 3회 반복하였다. 이후 액상층을 0.45μm 막으로 여과한 뒤 수득된 시료 200μl를 진공 농축기로 농축하였다.
HPLC 분석
사용된 분석 컬럼은 C18 컬럼(hydrosphere C18, 4.6 mm i.d. × 250 mm, S-5 im, 12nm)이며, UV 검출을 위한 파장은 245 nm이다. 용출은 1.0 ml/min의 유속 하에 35℃ 수조에서 수행되었다. 이동상 A는 아세토니트릴로, 이동상 B는 0.045% KH2PO4-0.05% 트리에틸아민 완충액(pH 7.0)으로 각각 구성되며, 주입 용적 30μl로 농도구배 용출을 수행하였다.
그 결과, 1차 HPLC 정제에서 크게 두 개의 피크를 얻었으며, 각각 PK1, PK2라고 명명하였다. 총 당(total sugar) 정량 결과 분획 6번 - 9번까지 혼합한 용액에서는 22.59 mg을 얻었으며, 11번 - 18번 피크를 혼합한 용액에서는 28 mg을 결과물로 수득하였다. PK1과 PK2를 각각 동결건조하여 수득한 산물에서 PK1은 백색의 솜과 같은 생성물을 얻었고, PK2는 비결정질의 갈색을 띄는 생성물을 얻었다(도 3). 일반적으로 당의 동결건조 시 PK1과 같이 백색의 결정을 형성하므로, PK1이 β-글루칸인 것으로 판단되었다. PK1의 분획을 다시 HPLC로 분석하였을 때 분획 5번 내지 12번까지의 피크를 회수하고, 이를 총당정량한 결과 26.78mg을 얻었다.
HPLC로 정제한 PK1의 구조를 확인하기 위하여 다당류를 단당류로 절단할 수 있는 TFA를 처리하여 HPLC로 분석하였고, 글루코스 표준품과 비교하였을 때 동일한 시간대에 용출되는 것을 확인하였다. 이에 PK1은 글루코스 단량체로 구성된 다당류로 예측되었으며, 귀리로부터 수용성으로 용출될 수 있는 다당류로서 글리코사이드 결합으로 이뤄진 당류는 β-글루칸이므로, PK1은 β-글루칸으로 구성되었음을 알 수 있었다.
이후, 본 발명의 β-글루칸(PK1)의 분자량을 확인하기 위하여 추가적인 HPLC 분석을 수행하였다. 대조군으로 다양한 크기의 β-글루칸 MW standard(MW = 391,000, Cat No. P-MWBGS, Megazyme, Ireland)와 글루코스(Cat No. MB-G4398 , Mbcell, 한국)의 RT(Retention Time)를 사용하였다. 대조군 시료는 증류수를 이용하여 1g/L의 농도로 제조한 후, 0.2μm, MCE Syringe Filter로 정제하여 준비하였다. β-글루칸(PK1) 역시 0.2μm, MCE Syringe Filter로 정제하여 시료를 준비하였다. PK1의 분자량을 확인하기 위하여 HPLC 분석을 진행하였다. HPLC 분석은 Vanquish Flex UHPLC Systems를 기본으로 당분석을 위한 RefractoMax 521 굴절률 검출기 (ERC, 미국)와 분자량 확인을 위한 크기배제 컬럼인 TSKgel UP-SW2000(Cat No. 083HA00117H, TOSOH, 일본) 으로 구성하여 진행하였다. 굴절률 검출기를 35℃ 설정하고 이동상 용매는 20 mM 아세트산 암모늄 완충액을 0.2 ml/min 의 속도로 흘려주고 1시간 이상 안정화를 시킨 후 결과 수집 시간은 30분으로 하여 분석을 진행하였다. 음성 대조군으로 이동상 용매를 사용하였으며, 대조군으로 β-글루칸 MW standard와 글루코스를 사용하였다. β-글루칸(PK1)의 결과(HPLC 크로마토그램)을 음성 대조군과 대조군의 결과와 중첩시켜 각 시료의 피크 RT를 비교함으로써 PK1의 분자량을 유추하였다. 그 결과, RT(Retention Time) 10분대 피크가 β-글루칸(PK1)으로 예상되었으며(도 2d), β-글루칸 MW standard의 peak 패턴을 기반으로 RT 10 분대의 피크가 β-글루칸(MW = 391,000)임을 확인하였다(도 2f).
실시예 2: MRSA에 대한 β-글루칸의 면역증진효과
β-글루칸의 항체 생성능 확인
메티실린 저항성 포도상구균(MRSA)의 항원인 LukS, iLukAB, iHla 및 HlgA와 세포외다당류(EPS), 알럼(Alum), β-글루칸을 각각 혼합한 백신 조성물의 면역원성을 확인하였다. 이들 백신 후보물질을 2주 간격으로 토끼에 3회 접종한 후 채혈하여 항체가를 ELISA로 측정하였다.
그 결과, 세 그룹 모두에서 항체의 생성이 확인되었으며, 이중 β-글루칸에서 IgG 역가가 가장 높음을 확인하였다(도 7). 아울러, MRSA의 상기 각 항원과 β-글루칸 또는 Alum 및 β-글루칸을 각각 혼합하여 래빗에 근육 또는 피하 접종 후 MRSA 균주인 USA 300을 감염시켜 각 개체별 체중의 변화를 측정한 결과, 각 그룹별 체중 변화의 차이는 크지 않았다(도 8).
2차 인 비보 실험
래빗(1.5-2kg 수컷)을 이용한 동물실험 절차는 도 9에 나타내었다. 시험물질을 2주 간격으로 3회 근육주사(intramuscular injection) 하였으며 각 투여 7일째에 혈액을 채취하였다. 3회째 접종 후 7일이 경과한 시점에 USA 300을 감염시키고 항체 역가를 측정하였다.

실험동물은 PBS 100μl를 투여한 대조군(n=5, 수컷), 상기 표 1의 4가지 항원과 β-글루칸 및 Alum을 투여한 군(n=5, 수컷); 4가지 항원과 MF59을 투여한 군(n=5, 수컷); 및 4가지 항원과 알럼을 투여한 군(n=4, 수컷)으로 나누었다.
도 10에서 보는 바와 같이, β-글루칸 및 Alum을 면역보강제로 사용한 군에서 모든 PFT 유전자 재조합 항원에 대한 항체가 차수별로 높게 상승되었다.
각 그룹의 체중 변화에는 유의한 차이가 관찰되지 않았다(도 11).

생존률 및 신장 농 형성
각 그룹에 3회째 접종 후 7일이 경과한 시점에 USA 300을 감염시키고 생존률을 측정하였다. 그 결과, PBS 투여 대조군(n=5)의 생존률이 60%인 반면, 4가지 항원과 β-글루칸 및 Alum을 투여한 군(n=5) 및 4가지 항원과 MF59을 투여한 군(n=4)은 100%의 생존률을 보였고 4가지 항원과 Alum을 투여한 군(n=2)은 50%의 생존률을 보였다(도 13).
아울러, USA 300 감염 후 7일째까지 살아있는 토끼의 신장을 채취하여 관찰한 결과를 도 12에 나타내었다. PBS를 접종한 군에서는 살아남은 3마리를 분석하였고, 모두 농양이 관찰되었다. 4가지 항원과 β-글루칸을 혼합한 후 접종한 군에서는 5마리를 분석하였고, 이중 2마리는 1개의 농양이 관찰되었으며, 1마리는 농양이 관찰되지 않았다. 4가지 항원과 MF59를 혼합하여 접종한 군에서는 4마리를 분석하였고, 2마리에서 농양이 관찰되지 않았다. 4가지 항원과 Alum을 혼합하여 접종한 군에서는 1마리만 생존하였고, 농양이 관찰되지는 않았다. PBS군과 비교하였을 때 β-글루칸군에서 농양의 발생이 감소한 것을 확인할 수 있었다.
실시예 3: 백일해에 대한 β-글루칸의 면역증진효과

면역보강제로서 β-글루칸을 투여 후 상기 표 3의 각 그룹별 사이토카인 생성을 비교하였다. 그 결과, 경구 투여군 보다 복강 투여군에서 사이토카인(IFN-γ, TNF-α, IL-6) 분비량이 높다(도 14).
β-글루칸에 의한 체액성 면역반응 증진효과
하기 표 4와 같이 유전자 재조합 백일해(pertussis toxin, PT) 항원인 EPRS-PtxAM, hRID-PtxAm, LysRS-PtxAM과 면역보강제로 Alum 및 β-글루칸(저농도, 고농도)을 마우스에 접종하였다.

융합 펩타이드의 종류에 따른 비교 결과(그룹 2 내지 4) LysRS-PT 유전자 재조합 항원에서 항체 역가가 가장 높았으며(그룹 4), 면역보강제에 따른 비교 결과(그룹 5 내지 7) 고용량 β-글루칸 처리군(그룹 7)이 가장 현저한 체액성 면역반응을 나타냈다(도 15). 특히 β-글루칸 처리군에서는 Alum 어쥬번트를 사용한 경우에는 관찰되지 않는 IgG 2a 서브그룹 반응을 보임으로서 β-글루칸이 DTaP 백신에서 세포매개 면역반응을 효율적으로 향상시킬 수 있음을 확인하였다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.

- β-1,3 글루코사이드 결합 및 β-1,4 글루코사이드 결합을 통해 D-글루코피라노스 단량체가 선형으로 연결된 베타 글루칸 또는 이의 약제학적으로 허용되는 염을 유효성분으로 포함하는 면역증강제(adjuvant) 조성물.
- 제 1 항에 있어서, 상기 베타 글루칸은 380,000 MW(molecular weight) 내지 400,000 MW의 평균 분자량을 가지는 것을 특징으로 하는 조성물.
- 제 1 항에 있어서, 상기 베타 글루칸은 귀리(Avena sativa), 보리(Hordeum vulgare), 밀(Triticum aestivum) 및 수수(Sorghum bicolor)로 구성된 군으로부터 선택되는 하나 이상의 곡물(cereal)로부터 추출되는 것을 특징으로 하는 조성물.
- 제 3 항에 있어서, 상기 곡물은 귀리(avena sativa)인 것을 특징으로 하는 조성물.

- (a) 하나 이상의 항원 또는 이를 인코딩하는 핵산 분자; 및(b) 제 1 항 내지 제 10 항 중 어느 한 항의 면역증강제(adjuvant) 조성물을 포함하는 백신 조성물.
- 제 11 항에 있어서, 상기 항원은 포도상구균(Staphylococcus aureus) 유래 항원인 것을 특징으로 하는 조성물.
- 제 12 항에 있어서, 상기 포도상구균은 메티실린 저항성 포도상구균(methicillin-resistant Staphylococcus aureus, MRSA)인 것을 특징으로 하는 조성물.
- 제 11 항에 있어서, 상기 항원은 백일해균(Bordetella pertussis) 유래 항원인 것을 특징으로 하는 조성물.
