本発明により、ビフィドバクテリウム・ブレーベの有用なプレバイオティクス製品を提供することができる。また、ラクト-N-ネオテトラオース骨格を有するオリゴ糖とビフィドバクテリウム・ブレーベを組み合わせた有用なシンバイオティクス製品を提供することができる。
本発明において、プロバイオティクスとプレバイオティクスは、これらを組み合わせて「シンバイオティクス」として使用される。シンバイオティクスは、両者を混合して調製される製剤(腸内細菌製剤)として用いるのが好ましい。
本発明の一態様は、単糖および二糖から選ばれる1種またはそれ以上の糖を含有する、ビフィドバクテリウム・ブレーベのラクト-N-ネオテトラオース骨格を有するオリゴ糖の資化促進用組成物(以下、「本発明の組成物」ということがある)に関する。本発明の組成物は、食品組成物、医薬組成物、または培地組成物等の態様にすることができる。
本発明の組成物は、単糖および二糖から選ばれる1種またはそれ以上の糖を含有する。
本発明の組成物に用いることができる単糖および二糖としては、医療、食品等の分野において用いることが可能な単糖および二糖を、1種または2種以上、自由に選択して用いることができる。具体的には、例えば、グルコース、フルクトース、ガラクトース、ラムノースおよびフコース等の単糖;ラクトース(乳糖)、スクロース、マルトースおよびトレハロース等の二糖等を挙げることができる。好ましくはグルコース、ガラクトース、およびラクトースであり、より好ましくはグルコース、およびラクトースを挙げることができる。単糖および二糖としては、市販品を用いてもよく、適宜製造して取得したものを用いてもよい。
また組成物全体に対する単糖および二糖の含有量は、0.01質量%~100質量%であってよく、0.1質量%~90質量%であってよく、0.2質量%~80質量%であってよく、1質量%~80質量%であってよく、2質量%~70質量%であってよく、5質量%~60質量%であってよい。
本発明の組成物が対象とするビフィドバクテリウム・ブレーベとしては、本発明の効果を損なわない限り、公知または未知のビフィドバクテリウム・ブレーベに属する細菌を1種または2種以上、自由に選択して用いることができる。すなわち、単糖および二糖から選ばれる1種またはそれ以上の糖により、そのラクト-N-ネオテトラオース骨格を有するオリゴ糖の資化を促進することができるものであれば対象とすることができる。本発明の組成物が対象とするビフィドバクテリウム・ブレーベとしては、ラクト-N-ネオテトラオース骨格を有するオリゴ糖の資化能がない、または資化能が低いもの、または通常の資化を有するもののいずれも対象とできるが、好ましくはラクト-N-ネオテトラオース骨格を有するオリゴ糖の資化能がないもの、または資化能が低いものである。
本発明において、「ラクト-N-ネオテトラオース骨格を有するオリゴ糖の資化促進」とは、任意の菌株を同一条件で培養した場合において、糖源がラクト-N-ネオテトラオース骨格を有するオリゴ糖のみである場合と比較して、糖源が単糖または二糖およびラクト-N-ネオテトラオース骨格を有するオリゴ糖である場合に、ラクト-N-ネオテトラオース骨格を有するオリゴ糖を資化する能力が高まることを意味する。
上記寄託株と実質的に同等な株は、例えば、当該寄託株を親株とする派生株であってよい。派生株としては、寄託株から育種された株や寄託株から自然に生じた株が挙げられる。
(1)RAPD(Randomly Amplified Polymorphic DNA)法、PFGE(Pulsed-field gel electrophoresis)法により同一の菌株と判定される菌株(Probiotics in food/Health and nutritional properties and guidelines for evaluation 85 Page43に記載)
(2)当該寄託菌株由来の遺伝子のみ保有し、外来由来の遺伝子を持たず、DNAの同一性が95%以上である菌株
(3)当該菌株から育種された株(遺伝子工学的改変、突然変異、自然突然変異を含み、同一の形質を有する株
本発明の組成物におけるビフィドバクテリウム・ブレーベの含有量は、103CFU/gまたはmL~1012CFU/gまたはmLであってよく、104CFU/gまたはmL~1011CFU/gまたはmLであってよく、105CFU/gまたはmL~1010CFU/gまたはmLであってよい。なお、CFUはコロニー形成単位:colony forming unitを意味する。
ビフィドバクテリウム・ブレーベを併用して含有する組成物が成人のヒト(すなわち、16歳を超えるヒト)に投与される場合、ビフィドバクテリウム・ブレーベは、例えば106CFU/g以上、または108CFU/g以上で含有され得る。ビフィドバクテリウム・ブレーベを併用して含有する資化促進用組成物が小児(すなわち、16歳未満のヒト)に投与される場合、ビフィドバクテリウム・ブレーベは、好ましくは約106~約108CFU/gの範囲で含有され得る。
本発明において、ラクト-N-ネオテトラオース骨格を有するオリゴ糖としては、本発明の効果を損なわない限り、公知または未知のラクト-N-ネオテトラオース骨格を有するオリゴ糖を1種または2種以上であり得る。すなわち、単糖および二糖から選ばれる1種またはそれ以上の糖を併用することで、ビフィドバクテリウム・ブレーベの資化能が促進可能なオリゴ糖であり得る。
具体的には、例えば、Lacto-N-neotetraose、Sialyllacto-N-neotetraosec等が挙げられる。
本発明の組成物全体に対するラクト-N-ネオテトラオース骨格を有するオリゴ糖の含有量は、0.001質量%~50質量%であってよく、0.01質量%~40質量%であってよく、0.1質量%~30質量%であってよく、1質量%~20質量%であってよい。
本発明の食品組成物、医薬組成物、培地組成物の単糖および二糖から選ばれる1種またはそれ以上の糖の含有量、ビフィドバクテリウム・ブレーベの含有量、LNnTの含有量は本発明の組成物に記載の含有量を適宜参照することが可能である。
また、本発明の組成物は、有効成分である単糖および二糖から選ばれる1種またはそれ以上の糖とともに、プレバイオティクスであるラクト-N-ネオテトラオース骨格を有するオリゴ糖を併用してもよい。
さらに、ラクト-N-ネオテトラオース骨格を有するオリゴ糖以外の公知または未知のHMOsを1種または2種以上も、自由に選択して用いることができる。なかでも、2’-フコシルラクトース、ラクト-N-フコペンタオースI、ラクト-N-ジフコヘキサオースI、およびラクト-N-テトラオース等が好ましく利用できる
さらに本発明においては、HMOs以外のプレバイオティクスも併せて用いてもよい。
プレバイオティクスは、本発明の組成物とともに摂取させたときに本発明の効果をより発揮し得るものであれば特に制限されないが、例えば、ラクチュロース、ラフィノース、ガラクトオリゴ糖、フラクトオリゴ糖、大豆オリゴ糖、乳果オリゴ糖、キシロオリゴ糖、イソマルオリゴ糖、コーヒー豆マンノオリゴ糖、グルコン酸、ポリデキストロース、イヌリン等が好ましく、その中でも、ラクチュロース、ラフィノース、ガラクトオリゴ糖等がより好ましく、ラクチュロース、ラフィノースおよびガラクトオリゴ糖がさらに好ましい。
本発明の組成物が対象に投与して利用される食品組成物、医薬組成物である場合、本発明の組成物における単糖および二糖の含有量は、本発明の方法における単糖および二糖の投与量についての記載を準用できる。
本発明の組成物を対象に投与することにより、ビフィドバクテリウム・ブレーベのラクト-N-ネオテトラオース骨格を有するオリゴ糖の資化促進効果が期待できる。すなわちプロバイオティクスが腸内で増殖することにより、腸内細菌叢が改善される。腸内細菌叢が改善されることにより、腸内細菌叢に起因する疾患が改善または予防され、治療が可能になり得る。
本実施形態の用途は、治療目的使用であっても、非治療目的使用であってもよい。
「非治療目的」とは、医療行為、すなわち、治療による人体への処置行為を含まない概念である。例えば、健康増進、美容行為等が挙げられる。
「改善」とは、疾患、症状または状態の好転;疾患、症状または状態の悪化防止、遅延;疾患または症状の進行の逆転、防止または遅延をいう。
「予防」とは、適用対象における疾患若しくは症状の発症の防止や遅延、または適用対象の疾患若しくは症状の危険性の低下をいう。
本発明の組成物が非治療目的で使用される場合は、健常者における腸内細菌に起因する疾患の予防が可能になる。腸内細菌叢に起因する疾患としては、炎症性大腸炎、潰瘍性大腸炎、過敏性大腸炎、大腸がん、糖尿病、動脈硬化等が挙げられる。
また、本発明の組成物が非治療目的で使用される場合は、健常者における通常の免疫機能がさらに増進されることにより、健常者における花粉症、喘息、アトピー性皮膚炎等のアレルギー症状の予防が可能になる。
本発明の組成物が治療目的で使用される場合は、非健常者の腸内細菌叢に起因する疾患、又は免疫機能の異常の改善、予防され、治療が可能になり得る。
本発明の組成物が治療目的で使用される場合は、非健常者の保有する腸内細菌叢を改善するために使用され、特に、非健常者の腸内でビフィドバクテリウム・ブレーベを増殖させるために使用され得る。
本発明の一態様は、食品組成物である、本発明の組成物(以下、「本発明の食品組成物」ということがある)に関する。
本発明に係る食品組成物は、そのままでまたは製剤化成分と組み合わせて添加物として公知の飲食品に添加することもできるし、サプリメント等として用いることもできるし、飲食品の原料中に混合して飲食品として用いることもできる。製剤化成分としては、後記<医薬組成物>に記載の製剤担体を参照できる。本発明の食品組成物の製造方法は、特に制限されない。本発明の食品組成物は、単糖および二糖から選ばれる1種またはそれ以上の糖を添加すること以外は、例えば、通常の飲食品と同様の原料を用いて通常の飲食品と同様の方法により製造することができる。
前記即席食品類としては、例えば、即席めん、カップめん、レトルト・調理食品、調理缶詰め、電子レンジ食品、即席スープ・シチュー、即席みそ汁・吸い物、スープ缶詰め、フリーズ・ドライ食品、その他の即席食品等が挙げられる。
前記農産加工品としては、例えば、農産缶詰め、果実缶詰め、ジャム・マーマレード類、漬物、煮豆類、農産乾物類、シリアル(穀物加工品)等が挙げられる。
前記水産加工品としては、例えば、水産缶詰め、魚肉ハム・ソーセージ、水産練り製品、水産珍味類、つくだ煮類等が挙げられる。
前記畜産加工品としては、例えば、畜産缶詰め・ペースト類、畜肉ハム・ソーセージ等が挙げられる。
前記乳・乳製品としては、例えば、発酵乳、加工乳、乳飲料、ヨーグルト類、乳酸菌飲料類、チーズ、アイスクリーム類、調製粉乳類、クリーム、その他の乳製品等が挙げられる。
前記油脂類としては、例えば、バター、マーガリン類、植物油等が挙げられる。
前記基礎調味料としては、例えば、しょうゆ、みそ、ソース類、トマト加工調味料、みりん類、食酢類等が挙げられ、前記複合調味料・食品類として、調理ミックス、カレーの素類、たれ類、ドレッシング類、めんつゆ類、スパイス類、その他の複合調味料等が挙げられる。
前記冷凍食品としては、例えば、素材冷凍食品、半調理冷凍食品、調理済冷凍食品等が挙げられる。
前記菓子類としては、例えば、キャラメル、キャンディー、チューインガム、チョコレート、クッキー、ビスケット、ケーキ、パイ、スナック、クラッカー、和菓子、米菓子、豆菓子、デザート菓子、その他の菓子等が挙げられる。
前記飲料としては、例えば、炭酸飲料、天然果汁、果汁飲料、果汁入り清涼飲料、果肉飲料、果粒入り果実飲料、野菜系飲料、豆乳、豆乳飲料、コーヒー飲料、お茶飲料、粉末飲料、濃縮飲料、スポーツ飲料、栄養飲料、アルコール飲料、その他の嗜好飲料等が挙げられる。
上記以外の市販食品としては、例えば、ベビーフード、ふりかけ、お茶潰けのり等が挙げられる。
「表示」行為には、需要者に対して前記用途を知らしめるための全ての行為が含まれ、前記用途を想起・類推させ得るような表現であれば、表示の目的、表示の内容、表示する対象物・媒体等の如何に拘わらず、全て本発明の「表示」行為に該当する。
本発明に係る食品組成物は、飲食品の中でも特に、調製乳に好適に用いることができる。調製乳としては、調製粉乳や調製液状乳が挙げられる。
調製乳としては、乳児用調製乳、フォローアップ用調製乳、低出生体重児用調製乳、小児用調製乳、成人用調製乳、高齢者用調製乳、アレルギー用調製乳、乳糖不耐症用調製乳、先天代謝異常用調製乳等が挙げられる。特に乳児用調製乳、フォローアップ用調製乳、低出生体重児用調製乳、小児用調製乳に用いられることが好ましい。乳幼児用調製乳とは、0~12か月の乳児を対象とする乳児用調製乳、6~9か月以降の乳児および年少幼児(3歳まで)を対象とするフォローアップミルク、出生時の体重が2500g未満の新生児(低出生体重児)を対象とする低出生体重児用調製乳、牛乳アレルギーや乳糖不耐症等の病的状態を有する児の治療に用いられる各種治療用ミルク等を指す。
具体的には、下記工程(A)を含む、調製粉乳の製造方法を提供する:
(A)単糖および二糖から選ばれる1種またはそれ以上の糖(必要に応じて、さらに、ビフィドバクテリウム・ブレーベを含む菌体粉末およびラクト-N-ネオテトラオース骨格を有するオリゴ糖)および乳成分を混合し、粉乳を得る工程。
具体的には、下記工程(A)および(B)を含む、乳幼児用サプリメントの製造方法を提供する:
(A)単糖および二糖から選ばれる1種またはそれ以上の糖(必要に応じて、さらに、ビフィドバクテリウム・ブレーベを含む菌体粉末およびラクト-N-ネオテトラオース骨格を有するオリゴ糖)および賦形剤を混合し、混合物を得る工程;
(B)前記混合物を打錠する工程。
本発明の調製乳が健常者に投与される場合は、乳児用調製乳、フォローアップ用調製乳、低出生体重児用調製乳、小児用調製乳、成人用調製乳、高齢者用調製乳に用いられ、上記の非治療目的として使用され得る。
本発明の一態様は、培地組成物である、本発明の組成物(以下、「本発明の培地組成物」ということがある)に関する。
本発明に係る組成物は、そのままでまたは製剤化成分と組み合わせて添加物として公知の培地に添加することもできるし、培地の原料中に混合して培地組成物として用いることもできる。培地組成物は、ビフィドバクテリウム・ブレーベ用の培地組成物であってよい。製剤化成分としては、後記<医薬組成物>に記載の製剤担体を参照できる。本発明の培地組成物の製造方法は、特に制限されない。本発明の培地組成物は、単糖および二糖から選ばれる1種またはそれ以上の糖を添加すること以外は、例えば、通常の培地と同様の原料を用いて通常の培地と同様の方法により製造することができる。
また、培地としては、例えば、ビフィドバクテリウム・ブレーベ等のビフィドバクテリウム属細菌の培養に通常用いられる培地に単糖および二糖から選ばれる1種またはそれ以上の糖を添加して用いることができる。ビフィドバクテリウム属細菌の培養に通常用いられる培地としては、強化クロストリジア培地(Reinforced Clostridial medium)、MRS培地(de Man, Rogosa, and Sharpe medium)、mMRS培地(modified MRS medium)、TOSP培地(TOS propionate medium)、TOSP Mup培地(TOS propionate mupirocin medium)が挙げられる。
本発明に係る組成物は、そのままで医薬品または医薬部外品(以下、「医薬組成物」ともいう)とすることもできるし、医薬品または医薬部外品の原料中に混合して医薬組成物として用いることもできる。医薬組成物は、単糖および二糖から選ばれる1種またはそれ以上の糖を添加すること以外は、例えば、通常の医薬組成物と同様の原料を用いて通常の医薬組成物と同様の方法により製造することができる。
なお、製剤化は剤形に応じて、適宜、公知の方法により実施できる。
なお、経口投与用の液剤の場合に使用する担体としては、水等の溶剤、矯味矯臭剤等が挙げられる。
本発明の一態様は、ビフィドバクテリウム・ブレーベのラクト-N-ネオテトラオース骨格を有するオリゴ糖の資化を促進させる方法であって、ラクト-N-ネオテトラオース骨格を有するオリゴ糖と、単糖および二糖から選ばれる1種またはそれ以上の糖とを共存させることを含む、方法(以下、「本発明の方法」ということがある)に関する。
すなわち、本発明の方法は、ラクト-N-ネオテトラオース骨格を有するオリゴ糖と、単糖および二糖から選ばれる1種またはそれ以上の糖とを共存させる工程を含む方法である。
本試験例は、単糖または二糖によるビフィドバクテリウム・ブレーベのラクト-N-ネオテトラオースの資化促進効果を評価するために行った。
(培地調製)
糖なしMRS培地(表1)を80mL調製し、pH6.5に調整した後に121度15分で滅菌した。LNnT液、Glucose液、Lactose液、およびGalactose液(表2)をそれぞれ調製し、0.22μmフィルター(Merck Millipore社)を用いてフィルター滅菌した。得られた各調製液を、終濃度がそれぞれLNnT1%(w/v)、Glucose、Lactose、またはGalactoseについては0.2%(w/v)となるよう滅菌後の糖なしMRS培地に添加し、合計7種類の培地を調製した(表3)。
マルチウェルプレート(Falcon社)を用いて、フィドバクテリウム・ブレーベFERM BP-11175、ビフィドバクテリウム・ブレーベNITE BP-02622をMRS培地(Difco社)にシステイン(関東化学株式会社)を0.05%添加した培地で前培養した。その後、前培養を行った各菌株を、3連で上記調製した7種類の各培地に対してそれぞれ1×109 cfu/ml程度を1%接種し、37℃、12時間培養し、波長可変型吸光マイクロプレートリーダー(TECAN社・サンライズレインボーサーモRC-R)を用いて30分毎に濁度(OD600nm)を測定した。菌体未接種の培養液をベースラインとした。3連の培養をそれぞれ3回測定し、平均値を算出した。
図1に、各菌株の各培地における増殖曲線を示す。培養開始時と比較して濁度の値が高くなったものは、ビフィズス菌の菌数が増加したものと考えられるため、糖を資化していると判断した。
ビフィドバクテリウム・ブレーベFERM BP-11175、ビフィドバクテリウム・ブレーベNITE BP-02622のいずれの菌株においても、LNnTを単独の糖源として用いた場合、濁度の値は経時的にほとんど変化しないか、緩やかに上昇するに留まった。一方、LNnTと単糖または二糖(グルコース、ガラクトース、ラクトースのいずれか)との組み合わせを糖源として用いた場合は、糖源がLNnTのみである場合や、糖源が単糖または二糖のみの場合と比較して、培養時間が経過するに従い、高い濁度の値を示した。グルコース、ガラクトース、ラクトースから選択されるいずれかとLNnTとを組み合わせることによって、ビフィドバクテリウム・ブレーベFERM BP-11175、ビフィドバクテリウム・ブレーベNITE BP-02622の持つLNnTの資化能が高まった、すなわちLnNTの資化が促進されることが確認された。
ビフィドバクテリウム・ブレーベFERM BP-11175、またはビフィドバクテリウム・ブレーベNITE BP-02622を、MRS液体培地3mLに添加し、37℃で16時間嫌気培養後、培養液を濃縮し、凍結乾燥を行い、細菌の凍結乾燥粉末(菌末)を得る。菌末と、単糖または二糖(グルコース、ガラクトース、またはラクトース)と、ラクト-N-ネオテトラオースを均一に混合して組成物を得る。当該組成物20gを200gの水に溶かし、ビフィドバクテリウム・ブレーベ、単糖または二糖、およびLNnTを含む組成物を得る。当該組成物を対象に投与することにより、腸内において、ビフィドバクテリウム・ブレーベによるLNnTの資化促進が期待できる。
ビフィドバクテリウム・ブレーベFERM BP-11175、またはビフィドバクテリウム・ブレーベNITE BP-02622を、MRS液体培地3mLに添加し、37℃で16時間嫌気培養後、培養液を濃縮し、凍結乾燥を行い、細菌の凍結乾燥粉末(菌末)を得る。次に、単糖または二糖(グルコース、ガラクトース、またはラクトース)、ラクト-N-ネオテトラオース、および結晶セルロースを撹拌造粒機に投入し混合する。その後、精製水を加え造粒、造粒物を乾燥し、ビフィドバクテリウム・ブレーベ、単糖または二糖、およびLNnTを含有し、賦形剤を含有してなる造粒物(医薬品組成物)を得る。当該造粒物を対象に投与することにより、腸内において、ビフィドバクテリウム・ブレーベによるLNnTの資化促進が期待できる。

- 単糖および二糖から選ばれる1種またはそれ以上の糖を含有する、ビフィドバクテリウム・ブレーベのラクト-N-ネオテトラオース骨格を有するオリゴ糖の資化促進用組成物。
- 前記単糖が、グルコースおよびガラクトースから選ばれる1種または2種であり、前記二糖が、ラクトースである、請求項1に記載の組成物。

- ビフィドバクテリウム・ブレーベのラクト-N-ネオテトラオース骨格を有するオリゴ糖の資化を促進させる方法であって、
ラクト-N-ネオテトラオース骨格を有するオリゴ糖と、
単糖および二糖から選ばれる1種またはそれ以上の糖とを共存させることを含む、方法。 - 前記単糖が、グルコースおよびガラクトースから選ばれる1種または2種であり、前記二糖が、ラクトースである、請求項8に記載の方法。
- 単糖および二糖から選ばれる1種またはそれ以上の糖と、
ラクト-N-ネオテトラオース骨格を有するオリゴ糖と、
ビフィドバクテリウム・ブレーベと、
を含有する組成物。 - 前記単糖が、グルコースおよびガラクトースから選ばれる1種または2種であり、前記二糖が、ラクトースである、請求項10に記載の組成物。
- 前記ラクト-N-ネオテトラオース骨格を有するオリゴ糖が、ラクト-N-ネオテトラオースである、請求項10または11に記載の組成物。
- 前記ビフィドバクテリウム・ブレーベが、ビフィドバクテリウム・ブレーベNITE BP-02622またはビフィドバクテリウム・ブレーベFERM BP-11175である、請求項10~12のいずれか一項に記載の組成物。
