Abstract
본 발명은 단백질의 전달을 위한 지질 나노입자에 관한 것으로, 구체적으로는, 치료용 단백질(therapeutic protein) 또는 효소 대체 요법(Enzyme Replacement Therapy, ERT) 효소와 같은 다양한 의약 단백질을 세포 내 또는 체내 전달하기 위한 지질 나노입자에 관한 것이다. 본 발명은 지질 나노입자를 통해 치료용 단백질 또는 ERT 효소 등 다양한 단백질들을 안정적이고 효과적으로 세포 내 전달함으로써 리소좀 축적 질환 등 각종 질환의 치료, 예방에 유용하게 사용될 수 있다.
Description
본 발명은 단백질의 전달을 위한 지질 나노입자에 관한 것으로, 구체적으로는, 치료용 단백질(therapeutic protein) 또는 효소 대체 요법(Enzyme Replacement Therapy, ERT) 효소와 같은 다양한 의약 단백질을 세포 내 또는 체내 전달하기 위한 지질 나노입자에 관한 것이다.
지질나노입자(Lipid Nanoparticle, LNP)는 최근 체내에 mRNA와 같은 생체분자를 효과적으로 전달하기 위한 시스템으로 많이 주목받고 있으며, 이러한 지질나노입자가 물질을 안정적으로 체내 전달하기 위하여, 인지질, 콜레스테롤, 다양한 이온성 지질, PEG 함유 인지질 등을 포함하여 구성된다.
한편, 리소좀 축적 질환(Lysosomal storage disorders, LSDs)은 리소좀 기능의 결함으로 인한 상대적으로 희귀한 유전 대사 질환이다. LSDs는 전형적으로 리소좀에서 대사 산물의 분해에 참여하는 단일 효소의 결핍에 의해 야기된다. 효소 활동 부족으로 인한 산물 축적은 다양한 장기 시스템에 영향을 미치며, 심각한 증상과 조기 사망을 초래할 수 있다. LSDs의 대다수는 또한 진행성 신경퇴행, 중증 인지 손상에서부터 간질성, 거동 장애 및 정신 장애에 이르는 중요한 신경학적 구성 장애를 갖는다.
이러한 리소좀 축적 질환으로 파브리병, 고셔병, 뮤코다당증 등이 대표적인 질환으로, 효소 대체 치료법(ERT)이 표준 치료법으로 자리잡고 있으며, 이러한 효소 대체 치료법은 체내 부족한 효소 단백질을 정기적으로 정맥투여함으로써 세포 내 축적된 유해한 수준의 물질의 농도를 낮춰주는 역할을 한다(Concolino et al., Italian Journal of Pediatrics, 2018).
다만, 이러한 효소 대체 치료법의 치료 효율 향상을 위해서, 원치않는 면역반응 등의 부작용을 저감시키고, 세포 내 효과적이고 안정적인 효소 단백질 전달을 위한 전달 시스템 개발이 요구되는 실정이다. 이를 통하여, 투여되는 효소의 농도를 낮추거나, 항-약물 항체 (Anti-drug antibody) 반응을 최소화할 수 있기 위함이다.
본 발명의 일 예시적 목적은 단백질의 세포 내 또는 체내 전달을 위한 지질 나노입자를 제공하는 것이다.
본 발명의 다른 예시적 목적은 단백질이 봉입된 지질 나노입자를 제공하는 것이다.
본 발명의 또 다른 예시적 목적은 단백질이 봉입된 지질 나노입자를 함유하는 단백질의 세포 내 또는 체내 전달용 조성물을 제공하는 것이다.
본 발명의 또 다른 예시적 목적은 단백질이 봉입된 지질 나노입자를 함유하는 리소좀 축적 질환 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 또 다른 예시적 목적은 단백질이 봉입된 지질 나노입자의 제조방법을 제공하는 것이다.
본 발명의 또 다른 예시적 목적은 ERT 효소 또는 치료용 단백질을 지질 나노입자를 이용하여 세포 내 전달하는 방법을 제공하는 것이다.
본 발명의 또 다른 예시적 목적은 ERT 효소 또는 치료용 단백질이 봉입된 지질 나노입자를 이용한 질환 치료 방법을 제공하는 것이다.
본 명세서에 개시된 발명의 기술적 사상에 따라 이루고자 하는 기술적 과제는 이상에서 언급한 문제점을 해결하기 위한 과제로 제한되지 않으며, 언급되지 않은 또 다른 과제는 아래의 기재로부터 통상의 기술자에게 명확하게 이해될 수 있을 것이다.
이를 구체적으로 설명하면 다음과 같다. 한편, 본 출원에서 개시된 각각의 설명 및 실시형태는 각각의 다른 설명 및 실시 형태에도 적용될 수 있다. 즉, 본 출원에서 개시된 다양한 요소들의 모든 조합이 본 출원의 범주에 속한다. 또한, 하기 기술된 구체적인 서술에 의하여 본 출원의 범주가 제한된다고 볼 수 없다.
본 발명의 목적을 달성하기 위한 일 양태로서, 본 발명은 단백질의 체내 전달을 위한 지질 나노입자를 제공한다. 구체적으로, 본 발명은 수소화된 대두 포스파티딜콜린(HSPC); 콜레스테롤; DOTAP; 및 DSPE-mPEG를 포함하는 지질 혼합물을 포함하는, 단백질의 세포 내 또는 체내 전달을 위한 지질 나노입자를 제공한다.
본 발명의 용어 "지질 나노입자(Lipid Nanoparticle, LNP)"는 인접한 지질 이중층을 갖는 소포체, 예컨대, 구형 소포체를 지칭하며, 표적 위치에 전달되도록 사용될 수 있다. 이러한 지질 나노입자로는, 리포솜, 고체 지질 나노입자(SLN), 나노구조화된 지질 운반체(NLC), 및 단층 막 구조체(미셀) 등을 포함할 수 있다.
본 발명의 지질 나노입자는 하나 이상의 유형의 지질을 포함할 수 있다. 일 예시로, 이온성 지질, PEG-지질, 인지질, 또는 콜레스테롤 등을 포함할 수 있다. 여기서, 이온성 지질은 양이온성 지질일 수 있다. 양이온성 지질로는, 일 예시로, N-(2,3-디올레오일옥시)프로필)-N,N,N-트리메틸암모늄 클로라이드 (DOTAP); N-(2,3-디올레일옥시)프로필)-N,N,N-트리메틸암모늄 클로라이드 (DOTMA); N,N-디스테아릴-N,N-디메틸암모늄 브로마이드 (DDAB); N,N-디올레일-N,N-디메틸암모늄 클로라이드 (DODAC); N-(1,2-디미리스틸옥시프로프-3-일)-N,N-디메틸-N-히드록시에틸 암모늄 브로마이드 (DMRIE); N1,N3,N5-트리스(3-(디도데실아미노)프로필)벤젠-1,3,5-트리카르복스아미드 (TT3), 리포펙타민; 1,2-디리놀레일옥시-N,N-디메틸아미노프로판 (DLinDMA), 1,2-디리놀레닐옥시-N,N-디메틸아미노프로판 (DLenDMA), 디옥타데실디메틸암모늄 (DODMA), 디스테아릴디메틸암모늄 (DSDMA), 3-(N-(N',N'-디메틸아미노에탄)-카르바모일)콜레스테롤 (DC-Chol) 및 1,2-디올레올릴-3-디메틸암모늄 프로판 (DODAP) 등이 있다.
또한, 다른 지질 성분과 함께 구성될 수 있다. 일 예시로, 포스파티딜콜린 (PC) 부류 (예컨대, 1,2-디스테아로일-sn-글리세로-3-포스포콜린 (DSPC), 및 1,2-디올레오일-sn-글리세로-3-포스포에탄올아민 (DOPE), 스테롤 (예컨대, 콜레스테롤) 및 폴리에틸렌 글리콜 (PEG)-지질 접합체 (예컨대, 1,2-디스테아로일-sn-글리세로-3-포스포에탄올아민-N-[폴레이트(폴리에틸렌 글리콜)-2000 (DSPE-PEG2000), 1,2-디스테아로일-sn-글리세로-3-포스포에탄올아민-N-메톡시(폴리에틸렌 글리콜) (DSPE-mPEG), L-알파-포스파티딜콜린 (HSPC), 디팔미토일포스파티딜콜린 (DPPC) 및 1,2-디미리스토일-sn-글리세로-3-포스포에탄올아민-N-[메톡시(폴리에틸렌 글리콜)-2000 (C14-PEG2000))에 속하는 다른 지질 분자를 포함할 수 있다.
구체적으로, 본 발명의 지질 나노입자는 HSPC; 콜레스테롤; DOTAP; 및 DSPE-mPEG를 포함하는 지질 혼합물을 포함하는 것일 수 있다.
지질 나노입자 내에 이러한 각 지질들의 비율은 특별히 제한되지 않으나, 예컨대, HSPC는 지질 나노입자 내 지질 혼합물의 8 내지 15 몰%, 콜레스테롤은 지질 나노입자 내 지질 혼합물의 30 내지 50 몰%, DOTAP는 지질 나노입자 내 지질 혼합물의 40 내지 60 몰%, DSPE-mPEG는 지질 나노입자 내 지질 혼합물의 0.5 내지 5 몰%로 포함하는 것일 수 있다. 보다 바람직하게는, HSPC는 지질 나노입자 내 지질 혼합물의 9 몰%, 콜레스테롤은 지질 나노입자 내 지질 혼합물의 40 몰%, DOTAP는 지질 나노입자 내 지질 혼합물의 50 몰%, DSPE-mPEG는 지질 나노입자 내 지질 혼합물의 1 몰%로 포함하는 것일 수 있다.
또한, 상기 지질 혼합물은 HSPC:콜레스테롤:DOTAP:DSPE-mPEG의 몰비가 8-15:30-50:40-60:0.5-5로 구성되는 것일 수 있고, 바람직하게는 지질 혼합물은 HSPC:콜레스테롤:DOTAP:DSPE-mPEG의 몰비가 8-10:35-45:45-55:0.5-1.5로 구성되는 것일 수 있으며, 보다 바람직하게는 지질 혼합물은 HSPC:콜레스테롤:DOTAP:DSPE-mPEG의 몰비가 9:40:50:1로 구성되는 것으나, 이에 제한되지 않는다.
상기 범위 내에서 지질 나노입자에 봉입되는 단백질의 종류에 따라 최적의 몰비 또는 몰%를 적절히 선택할 수 있으며, 일 예시로, 인간 이두로네이트-2-설파타제(Human Iduronate-2-sulfatase, IDS)를 지질 나노입자에 봉입하는 경우 지질 혼합물은 바람직하게는 HSPC:콜레스테롤:DOTAP:DSPE-mPEG의 몰비가 9:40:50:1로 구성되는 것일 수 있다.
본 발명에 있어서, 지질 나노입자의 입자 크기는 약물 방출 속도, 생체 분포, 점막 접착력, 세포의 물 흡수 및 나노입자 내부로의 완충제 교환 및 단백질 확산에 영향을 줄 수 있다. 일 예시로, LNP 직경은 10 내지 500 nm 범위 일 수 있으며, 바람직하게는 70 내지 120 nm 범위일 수 있고, 보다 바람직하게는 75 내지 95 nm 범위일 수 있다.
본 발명에 있어서, 지질 나노입자의 다분자 지수(Poly Dispersity Index, PDI)는 지질 혼합물의 분자 또는 입자 크기의 이질성을 측정한 것으로, 일 예시로, 지질 나노입자의 PDI는 0.1 내지 0.4 범위일 수 있으며, 바람직하게는 0.1 내지 0.2 범위일 수 있고, 더 바람직하게는 0.15 내지 0.18 범위일 수 있다.
본 발명에 있어서, 상기 단백질은 특별히 제한없이 적용 가능하나, 일 예로, 효소 또는 치료용 단백질(therapeutic protein)을 포함할 수 있다. 상기 치료용 단백질은 질병 또는 장애를 치료 또는 예방하기 위한 단백질을 의미하며, 재조합 단백질, 항체, 바이오시밀러, 바이오베터 등일 수 있다. 본 발명에 있어서, 상기 효소는 특별히 제한없이 적용 가능하나, 체내 결핍되거나 부재한 효소일 수 있고, 특히, 상기 효소는 효소 대체 요법(enzyme replacement therapy, ERT) 효소일 수 있다.

상기 "지질 나노입자" 및 "단백질"은 전술한 바와 같다.
본 발명의 용어 "효소 대체 요법(Enzyme Replacement Thearpy)"은 체내 결핍된 효소를 환자의 정맥에 직접 투여해주는 치료법이다. 리소좀 축적 장애(LSD)로 불리우는 파브리병, 고셔병, 폼페병, 헌터증후군 등에 대표적인 치료법으로 알려져 있으며, 아데노신 디아미나아제 결핍증 (adenosine deaminase deficiency)으로부터 초래되는 중증 복합 면역결핍(ADA-SCID)과 같은 희귀 선천성 증후군의 치료에도 사용되고 있다. 또한, 효소 대체 요법은 기질 감소 치료법, 유전자 치료, 골수 유래 줄기세포 이식 등에도 효소 또는 단백질 결핍시 활용될 수 있다.
본 발명에 있어서, 일 예시로써, 상기 효소는 리소좀 축적 장애의 치료에 사용될 수 있는 리소좀 효소로써, 예를 들어, 이두설파제(idursulfase), 라로니다제(laronidase), 엘로설파제 알파(elosulfase alpha), 갈설파제(galsulfase), 베스트로니다제 알파(vestronidase alpha), 아갈시다제 베타(agalsidase beta), 아갈시다제 알파(agalsidase alpha), 페구니갈시다제 알파(pegunigalsidase alpha), 이미글루세라제(imiglucerase), 탈리글루세라제 알파(taliglucerase alpha), 알글루세라제(alglucerase), 벨라글루세라제(velaglucerase), 알글루코시다제 알파(alglucosidase alpha), 아발글루코시다제 알파(avalglucosidase alpha), 세벨리파제 알파(sebelipase alpha), 셀리포나제 알파(cerliponase alpha), 또는 아데노신 디아미나제(adenosine deaminase)일 수 있으나, 이에 제한되지 않는다.
구체적으로, 상기 효소는 효소 유전자를 벡터에 클로닝하여 발현 플라스마드를 제작한 후, 숙주세포로 발현 플라스미드를 형질도입하여 배양시킨 후 생산성이 높은 효소 발현 단일 클론 세포주를 선발 및 배양한 이후 단백질을 분리 및 정제함으로써 수득할 수 있다.
본 발명에 있어서, 상기 효소는 리소좀 축적 질환을 치료 또는 예방하기 위한 것일 수 있다. 리소좀은 체내에서 생성되는 물질 중 필요한 성분은 분배하고 필요 없는 성분은 배출시키는 역할을 하는 세포 내 소기관이다. 그러나 체내 효소가 부족하게 되면, 리소좀은 이 역할을 제대로 수행하지 못하며, 이 경우 몸에 불순물이 축적되어 신체 조직과 장기에 손상을 일으키게 된다.
이러한 리소좀 축적 질환은 50여가지 이상의 질환이 알려져 있다. 유전적 결함으로 인하여 리소좀 내에 특정 효소의 생산이 저해됨으로써, 체내 산물이 축적되어 결국 골격, 뇌, 피부, 심장, 중추신경계 등에 심각한 영향을 미치는 질환이다.
구체적으로 상기 리소좀 축적 질환은 MPS I, MPS II, MPS IVA, MPS VI, MPS VII, 파브리병, 고셔병, 고셔병 타입 I, 폼페병, 리소좀산 지질분해효소 결핍증(wolman disease, CESD), 신경 세로이드 리포푸신증(CLN2 질환), 또는 중증 복합 면역결핍일 수 있으나, 이에 제한되지 않는다.
일 예시로, 파브리병(Fabry disease)은 리소좀 효소인 α-갈락토시다제 A (α-Gal A)의 α-Gal A 유전자 (GLA) 내 변이로 인한 결핍에 의해 야기되는, 글리코스핑고리피드 대사의 진행성, X-연관된 선천적 오류이다. X-연관된 질병임에도 불구하고, 여성은 다양한 정도의 임상 증상을 나타낼 수 있다. 파브리병은 발병율이 남성에서는 40,000명 중 1명, 전체 인구에서는 117,000명 중 1명으로 추산되는 희귀 질병이다. 파브리병 환자에 있어서 치료받지 않는 경우의 예상 수명은 단축되며, 신장, 심장 및/또는 중추 신경계에 영향을 미치는 혈관 질환으로 인해 대체로 40대 또는 50대에 사망에 이른다. 효소 결핍은 전신에 걸쳐 혈관 내피 및 내장 조직에 기질인 글로보트리아오실세라미드 (GL-3)의 세포내 축적을 일으킨다.
파브리병으로 인한 심장 질환은 대부분의 남성에서 또한 다수의 여성에서 발생한다. 초기 심장 질환 소견은 좌심실 확대, 판막 침범 및 전도 이상을 포함한다. 승모판폐쇄부전증은 전형적으로 아동기 또는 청소년기에 나타나는 가장 빈도가 높은 판막 병변이다. 뇌혈관 증상은 주로 다병소성 소혈관 침범에 기인하며, 혈전증, 일과성 뇌허혈성 발작, 뇌기저동맥 허혈증 및 동맥류, 발작, 편측마비, 편측 감각소실, 실어증, 미로장애 또는 뇌출혈을 포함할 수 있다. 뇌혈관 증상의 평균 발병 연령은 33.8세이다. 성격 변화 및 정신병적 행동은 나이가 들면서 나타날 수 있다.
파브리병에 대해 현재 FDA-승인된 치료법은 효소 대체 요법 (ERT)으로, 아갈시다제 알파 (레플라갈 (Replagal)®; 셔 휴먼 지네틱 써라피즈 (Shire Human Genetic Therapies)), 아갈시다제 베타 (파브라자임 (Fabrazyme)®; 젠자임 코포레이션 (Genzyme Corporation)) 및 페구니갈시다제 알파 (엘파브리오 (Elfabrio)®; 프로탈릭스 바이오테라뷰틱스 (Protalix BioTherapeutics, Inc) 등이 있다. 이들 유형의 ERT는 환자의 불충분한 α-Gal A 활성을 효소의 재조합 형태를 정맥내 투여함으로써 보충하도록 하는 것이다. ERT가 많은 상황에서 유효하지만, 치료에는 또한 한계가 있다. ERT는 뇌졸중의 위험을 감소시키는 것으로 입증된 바가 없으며, 심장 근육은 느리게 반응하고, 신장의 세포 유형 일부에서 GL-3 제거는 제한적이다. 일부 환자는 또한 ERT에 대한 면역 반응을 나타낸다.
다른 예시로, 고셔병(Gaucher Disease) 또한 대표적인 리소좀 축적 질환으로 용해효소제의 가수분해 효소인 베타-글루코세레브로시다아제(β-glucocerebrosidase; β-GC)의 결핍으로 인해 간, 비장, 중추신경계, 골격 및 폐 등 주로 세망내피계(reticuloendothelial system)에 스핑고지질(sphingolipid)의 일종인 당지질 글루코세레브로사이드 (glucocerebroside)가 축적되어 발생하는 질환으로 지질 축적 질환 중 가장 흔하다.
고셔병은 GBA gene의 돌연변이에 의한 질병으로 상염색체 열성으로 유전되며, GBA 효소 활성도의 결핍 정도에 따라 다양한 중증도의 임상 양상을 보인다. 고셔병은 Knudsen과 Kaplan의 분류에 따라 중추신경계 침범 유무, 질병의 진행 속도 및 발병 연령에 따라 세 가지 임상 유형으로 나뉘었는데, 최근에는 이들을 신경계 비침투형(non-neuronopathic) 급성(acute) 혹은 만성(chronic) 신경계 침투형(neuronopathic)의 세 군으로 분류하고 있다. 이때, 신경계 비침투형은 1형으로 불리고, 급성 신경계 침투형은 2형, 만성 신경계 침투형은 3형으로 불린다.
효소대체요법(Enzyme replacement therapy)이 고셔병 치료에 효과가 입증된 전형적인 치료법으로, 특히 신경계 비침투형 및 만성 신경계 침투형 고셔병에서 간비장비대, 혈액학적 소견에 크게 도움이 되는 것으로 알려져 있다. 고셔병은 재조합 효소 치료의 효시가 되는 질환으로, acid β-glucosidase enzyme, imiglucerase (Cerezyme®, Genzyme Co.)가 DNA 재조합기술(recombinant DNA technology)을 통해 1994년부터 상용화 되어 최근까지 신경계 비침투형인 type I 고셔병의 표준적인 치료법(standard treatment)으로 사용되고 있다.
국내에서도 이미글루세라제(imiglucerase)의 바이오시밀러(biosimilar)인 Abcertin®(Isu Abxis, 성남, 한국)이 개발되어 상용화되어 사용하고 있으며, 이외 VPRIV® (Takeda Pharmaceutical Company, Tokyo, Japan)도 재조합 효소제로 사용되고 있다. 이들 재조합 효소는 세망내피계의 증상을 호전시켜, 간/비장 비대의 호전을 가져온다. 그러나 이들 효소제는 뇌혈관장벽을 통과하지 못하기 때문에 신경계 침투형 고셔병 환자의 신경계 증상 진행을 막을 수는 없다. 또한, 복부나 종격동 등의 부위에 림프절의 비대가 심한 환자에서도 치료효과가 제한적인 것으로 알려져 있다.
또 다른 예시로, 뮤코다당증 (MPS) I, II, IVA, VI, VII 등이 있다.
뮤코다당증 I형(MPS I)은 100,000명 출생 중 1명의 추정된 발생률을 갖는 드문 열성 유전병이다(Moore D et al., 2008, Orphanet Journal of Rare Diseases 3). MPS I은 아주 흔한 복합체 다당류 헤파란 설페이트 및 데르마탄 설페이트의 리포솜 이화작용을 위해 필요한 효소인 α-l-이두로니다아제(IDUA)의 결핍에 의해 유발된다. 글리코사미노글리칸(glycosaminoglycan, GAG)으로 지칭되는 이들 다당류는 MPS I 환자의 조직에서 축적되어, 특유의 저장 병변 및 다양한 질환 후유증을 유발한다. 환자는 저신장, 뼈 및 관절 기형, 거친 얼굴 특징, 간비종대, 분문판 질환, 폐쇄 수면 무호흡, 재발성 상기도 감염, 청각 장애, 손목 터널 증후군, 및 각막 혼탁으로 인한 시각 장애를 나타낼 수 있다(Beck M, et al., 2014, The natural history of MPS I: global perspectives from the MPS I Registry. Genetics in medicine: official journal of the American College of Medical Genetics 16(10):759-765). 또한, 많은 환자는 중추 신경계에서 GAG 저장과 관련된 증상이 발생하며, 이는 수두증, 척수 압박 및, 일부 환자에서, 인지 장애를 포함할 수 있다.
뮤코다당증 II(헌터 증후군/MPS II)형은 100,000명의 남아 신생아 당 0.5 내지 1.3명으로 발생하는 희귀한 X-연관 열성 유전 질환이다. 이 진행성 및 파괴성(devastating) 질환은 IDS 유전자의 유전자 돌연변이에 의해 유발되며, 이는 헤파란 설페이트(heparan sulfate) 및 더마탄 설페이트(dermatan sulfate)의 리소좀 이화작용에 필요한 효소인 리소좀 저장 효소 이두로네이트-2-설파타제(Iduronate-2-sulfatase)의 결핍으로 이어진다.
GAG(글리코사미노글리칸)로 불리는 이러한 유비쿼터스(ubiquitous) 다당류는 MPS II 환자의 조직과 기관에 축적되어, 특유의 저장 병변 및 다양한 질환 후유증을 유발한다. 이 환자 집단에서 이환율 및 사망률이 높다. 사망은 (신경인지 악화를 특징으로 하는) 중증 표현형을 갖는 환자에서는 11.7세의 평균 연령, 및 경증 또는 약화된 표현형을 갖는 환자에서는 21.7세의 평균 연령에서 발생하는 것으로 보고되어 있다.
뮤코다당증 IVA형(MPS IVA; 모르키오 A 증후군)은 N-아세틸갈락토사민-6-설페이트 설파타제(GALNS)의 결핍으로 인한 상염색체 열성 라이소좀 저장 장애이다(Khan, 등, Mol Genet Metab., 2017; 120(1-2): 78-95). 상기 효소의 결핍은 글리코사미노글리칸(GAG), 콘드로이틴 6-설페이트(C6S) 및 케라탄 설페이트(KS)의 점진적인 축적을 초래하여 불완전한 골화와 연속적인 성장 불균형에 의한 전신의 독특한 골격 이형성증을 야기하고, 결과적으로 짧은 목과 몸통, 경부 척수 압박, 기관 폐쇄, 돌출흉, 관절 이완, 척추후측만증, 외반고 및 외반슬을 초래한다. 상기 질병의 다른 임상 소견으로는 청력 상실, 심장 판막 병발 및 각막 혼탁을 포함할 수 있다. 200개 이상의 상이한 돌연변이가 환자에서 확인되었으며 미국에서의 유병률은 약 250,000명 중 1명이다.
본 발명에 따른 지질 나노입자를 효소 대체 요법에 사용함으로써, 상기 리소좀 축적 질환의 치료가 가능할 수 있다. 따라서, 이러한 관점에서, 본 발명은 ERT 효소를 이용한 질환의 치료 방법을 제공한다. 또한, 상기 리소좀 축적 질환 외에도, 체내에 결핍되거나 수준이 낮은 단백질을 효과적으로 전달하여줌으로써 각종 질환의 치료와 예방이 가능하다. 따라서, 치료용 단백질을 탑재한 본 발명의 지질 나노입자를 이용하여 다양한 질환의 치료방법을 제공할 수 있다. 이러한 치료방법은 지질 나노입자 내에 탑재한 ERT 효소, 또는 치료용 단백질을 세포 내 전달함으로써 달성될 수 있다.
본 발명의 목적을 달성하기 위한 다른 양태로서, 본 발명은 상기 단백질 또는 효소가 봉입된 지질 나노입자를 함유하는 단백질의 세포 내 또는 체내 전달용 조성물을 제공한다.
상기 "단백질" 및 "지질 나노입자"는 전술한 바와 같다.
본 발명에 있어서, 상기 조성물은 단백질 또는 효소가 봉입된 지질 나노입자를 세포 내 또는 체내 전달 효율을 높이기 위한 물질을 추가로 포함하거나 추가적인 개량을 통해 조성물의 형태를 변형할 수 있다.
본 발명의 목적을 달성하기 위한 다른 양태로서, 본 발명은 상기 단백질이 봉입된 지질 나노입자를 함유하는 리소좀 축적 질환 예방 또는 치료용 조성물을 제공한다.

본 발명의 용어 "예방"은 본 발명의 약학적 조성물의 투여를 통해 리소좀 축적 질환의 발병을 억제 또는 지연시키는 모든 행위를 의미하며, 본 발명의 용어 "치료"는 본 발명의 약학 조성물을 투여함으로써 질병이 호전 또는 완화되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명의 용어 "약학적 조성물"은 질병의 예방 또는 치료를 목적으로 제조된 것을 의미하며, 각각 통상의 방법에 따라 다양한 형태로 제형화하여 사용될 수 있다. 예컨대, 투여 경로에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽 등의 경구형 제형으로 제형화할 수 있고, 외용제, 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 구체적으로, 상기 투여 경로는 국소 경로, 경구 경로, 정맥 내 경로, 근육 내 경로, 및 점막 조직을 통한 직접 흡수를 포함하는 임의의 적절한 경로일 수 있으며, 두 가지 이상의 경로를 조합하여 사용할 수도 있다. 두 가지 이상 경로의 조합의 예는 투여 경로에 따른 두 가지 이상의 제형의 약물이 조합된 경우로서 예컨대 1차로 어느 한 약물은 정맥 내 경로로 투여하고 2차로 다른 약물은 국소 경로로 투여하는 경우이다.
본 발명의 약학적 조성물은, 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 또는 희석제를 추가로 포함하는 암의 치료 또는 예방용 약학 조성물의 형태로 제조될 수 있는데, 상기 담체는 비자연적 담체(non-naturally occuring carrier)를 포함할 수 있다. 구체적으로, 상기 약학 조성물은, 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 약학적 조성물의 구체적인 제제화와 관련하여서는 당업계에 공지되어 있으며, 예컨대 문헌[Remington's Pharmaceutical Sciences(19th ed., 1995)] 등을 참조할 수 있다. 상기 문헌은 본 명세서의 일부로서 간주된다.
본 발명의 약학적 조성물은 쥐, 개, 고양이, 소, 말, 돼지, 인간 등의 동물에 다양한 경로를 통해 투여될 수 있으며, 인간인 경우가 바람직할 수 있다. 투여의 모든 방식은 예상될 수 있으며, 예를 들어 경구, 정맥, 동맥, 근육 또는 피하 주사에 의해 투여될 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 치료 방법은 종래 알려져있는 치료요법과의 조합 또는 종래 알려져 있는 다른 치료제와의 병용 투여 또한 포함한다.
본 발명의 목적을 달성하기 위한 다른 양태로서, 본 발명은 단백질이 봉입된 지질 나노입자의 제조방법 및 이러한 제조방법으로 얻어진 나노입자를 제공한다. 구체적으로, 상기 단백질이 봉입된 지질 나노입자의 제조방법은, (a) HSPC; 콜레스테롤; DOTAP; 및 DSPE-mPEG를 포함하는 지질 혼합물을 제조하는 단계; (b) 상기 지질 혼합물을 알코올류 용매에 용해시킨 제1용액을 미세유체(microfluidics) 시스템의 주입구에 주입하는 단계; (c) 단백질이 함유된 제2용액을 미세유체 시스템의 다른 주입구에 주입하는 단계; 및 (d) 단백질이 봉입된 지질 나노입자를 정제하는 단계를 포함한다.
상기 "단백질" 및 "지질 나노입자"는 전술한 바와 같다.
본 발명에 있어서, 상기 알코올류 용매는 메탄올, 에탄올, 프로필알콜 또는 변성알콜을 포함할 수 있으나 이에 제한되지 않으며, 바람직하게는 에탄올일 수 있다.
본 발명에 있어서, 상기 미세유체 시스템의 알코올상(제1용액) 및 수성상(제2용액)의 유속 비율은 1:3 내지 1:5의 비율로 진행되는 것일 수 있으나 이에 제한되지 않으며, 바람직하게는 1:3의 비율로 진행되는 것일 수 있다.
본 발명에 있어서, 상기 미세유체 시스템의 유속은 9 내지 18 ml/분의 속도로 진행되는 것일 수 있으나 이에 제한되지 않으며, 바람직하게는 15 ml/분의 속도로 진행되는 것일 수 있다.
본 발명에 있어서, 상기 (d) 단계의 지질 나노입자를 정제하는 단계는 상기 (b) 단계의 알코올류 용매 및 봉입되지 않은 단백질을 제거시키는 단계를 포함하는 것일 수 있다. 상기 지질 나노입자의 정제는 컬럼이 장착된 접선 흐름 여과 시스템을 사용하여 정제하는 것일 수 있으며, 구체적으로, 지질 나노입자를 컬럼을 통해 순환시키고, 컬럼에서 빠져나가는 투과액과 동일한 속도로 신선한 버퍼(20 mM 소듐 포스페이트, 137 mM 소듐 클로라이드, pH 6±0.2)를 첨가하면서 정제하는 것일 수 있다.

이러한 상기 치료방법은 지질 나노입자에 봉입된 ERT 효소, 또는 치료용 단백질을 세포 내로 전달함으로써 달성될 수 있다.
본 발명은 지질 나노입자를 통해 치료용 단백질 또는 ERT 효소 등 다양한 단백질들을 안정적이고 효과적으로 세포 내 전달함으로써 리소좀 축적 질환 등 각종 질환의 치료, 예방에 유용하게 사용될 수 있다.

이하, 본 발명을 하기 실시예를 통하여 보다 상세하게 설명한다. 그러나 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예만으로 한정되는 것은 아니다.
실시예 1. 단백질이 봉입(encapsulation)된 지질 나노입자 제조
1.1. 효소 단백질 생산
1.1.1. IDS 생산 세포주 개발
인간 이두로네이트-2-설파타제(Human Iduronate-2-sulfatase, 이하 'IDS') 유전자를 합성한 후 팬젠 고유 발현벡터 pPGXII(대한민국등록특허 10-1385223호 도 2 참고)에 클로닝하여 발현 플라스미드를 제작하였다. CHO DG44 숙주세포주로 발현 플라스미드의 형질도입은 24-웰 규모로 수행하였으며, 전기 천공법(Electroporation)을 이용하여 형질도입시켰다. 37℃, 5% CO2 배양기에서 배양한 후 세포들이 충분히 자랐을 때 형질 전환된 세포들만 생장할 수 있도록 선택배지에서 배양하였다. 약 2주 후 세포가 충분히 성장하였을 때 배양액의 일부를 취하여 ELISA 분석법으로 IDS 발현 세포군을 선별하였다.
ELISA 분석 결과를 바탕으로 세포군을 선별하여 발현 효율이 높고 안정적인 세포주(stable cell line) 확보를 위해 단일 클론 선별(single clonal selection)을 수행하였다. 배양배지에 0.5 세포/웰(cell/well)이 되도록 96-웰 플레이트에 분주하였고, 약 4주 후 형성된 콜로니(colony)를 분석하여 발현 효율이 높은 세포주를 선별하여 배양하였다. 충분한 수의 세포가 확보되었을 때 ELISA 분석법으로 단일 클론 세포주의 발현 효율을 비교 분석하였고, 생산성이 높은 단일 클론 세포주를 후보 세포주로 선발하였다. 단일 클론 세포주는 90일의 장기 계대배양 동안 안정적으로 단백질을 발현함을 확인하였고, 이 중 생산성이 가장 높은 세포주를 최종 세포주로 선발하였다. 선발된 최종세포주는 충분한 세포수가 확보되었을 때 바이알에 분주하여 연구용세포은행을 제조한 후 액체질소 저장소에 보관하였다.
1.1.2. IDS 생산을 위한 세포 배양
IDS 생산 세포주를 해동한 후 계대배양 배지(EX-CELL (+ 8 mM L-glutamine) + 1% 2X Feed A+)를 이용하여 2 내지 3일 간격으로 계대배양을 수행하였다. 계대배양 배지는 8 mM L-글루타민(L-glutamine)이 첨가된 EX-CELL 배지에 1% 2X Feed A+ 첨가물을 첨가하여 사용하였다.
총 15일간의 종배양을 통해 충분한 수의 세포를 확보한 후 7.5 L 바이오리액터(New Brunswick scientific, Bioflo320)에서 5 L 배양액 규모로 IDS를 생산하였다. 생산 시 1 × 106 cells/mL 농도로 세포를 접종하였고, 생산배지는 8 mM L-글루타민이 첨가된 EX-CELL 배지에 12.5% 2X Feed A+ 첨가물을 첨가하여 사용하였다. 배양조건은 배양온도 37℃, DO(용존산소, Dissolved oxygen) 30%, pH 7.0 내지 7.2, 그리고 교반 속도 150 rpm으로 설정하였고, 총 11일 동안 배양을 진행하였다. 배양 기간 중 세포수 및 생존율은 Vi-CELLTM counter를 이용하여 분석하였고, pH 및 포도당(glucose)과 젖산(lactate)의 함량을 매일 측정하여 모니터링한 후 포도당 함량은 20 mM 이상으로 유지되도록 하였다. 배양 종료 후 바이오리액터로부터 배양액을 수확한 후 C0HC depth filter를 이용하여 세포를 제거하였고, 0.22 μm PES 필터로 제균하였다.
1.1.3. IDS 단백질 정제
세포 배양액을 이용하여 IDS 단백질을 분리 및 정제하였다. Capto MMC 레진이 충전된 컬럼을 이용하여 크로마토그래피를 수행하였다. AKTA Pure (GE healthcare)에 컬럼을 장착하고 20 mM 소듐 아세테이트(Sodium acetate)/150 mM 소듐 클로라이드(Sodium chloride) (pH 4.3) 평형 버퍼로 컬럼을 평형화한 후 동일한 pH로 조절한 배양액을 로딩하였다. 이후 컬럼을 세척하고 20 mM 소듐 아세테이트/150 mM 소듐 클로라이드 (pH 5.1) 용출 버퍼로 IDS 단백질을 용출하였다. 용출액으로 Blue 세파로스 6 FF을 이용한 크로마토그래피를 수행하였다. AKTA Pure (GE healthcare)에 컬럼을 설치하고 20 mM 소듐 아세테이트/50 mM 소듐 클로라이드 (pH 4.0) 평형 버퍼로 컬럼을 평형화한 후 희석 버퍼(20 mM 소듐 아세테이트 (pH 4.0))로 1/3 희석한 시료를 로딩하였다.
이후 컬럼을 세척하고 20 mM 소듐 포스페이트(Sodium phosphate) (pH 6.6) 용출버퍼로 IDS 단백질을 용출하였다. 용출된 IDS 단백질로 Q 세파로스 FF를 이용한 크로마토그래피를 수행하였다. AKTA Pure (GE healthcare)에 컬럼을 설치하고 20 mM 소듐 포스페이트 (pH 6.6) 평형 버퍼로 컬럼을 평형화한 후 시료를 로딩하였다. 세척 버퍼(50 mM 아세트산 (pH 3.0))로 컬럼을 세척한 후 50 mM 아세트산/150 mM 소듐 클로라이드 (pH 3.0) 용출 버퍼를 흘려주어 IDS 단백질을 확보하였다. 다음 단계의 정제를 수행하기 직전 10% 아세트산을 이용하여 Q 세파로스 FF 용출액의 pH를 3.5로 낮춘 후 상온에서 180분 동안 반응시켜 바이러스 불활화 단계를 수행하였다. 바이러스 불활화가 종료된 직후 시료를 희석 버퍼(50 mM 소듐 아세테이트/4,430 mM 소듐 클로라이드 (pH 4.0))로 1/3 희석한 후 Phenyl 세파로스 크로마토그래피를 수행하였다. AKTA Pure (GE healthcare)에 컬럼을 설치하고 50 mM 소듐 아세테이트/3,000 mM 소듐 클로라이드 (pH 4.0) 평형 버퍼로 컬럼을 평형화한 후 시료를 로딩하였다. 이후 컬럼을 세척하고 20 mM 소듐 포스페이트 (pH 6.0) 용출 버퍼로 IDS 단백질을 용출하였다. 용출된 IDS 단백질은 UF/DF 과정을 통해 약 2 mg/mL의 농도, 20 mM 소듐 포스페이트/137 mM 소듐 클로라이드 (pH 6.0) 버퍼 상태로 제조하였고, 시료를 회수한 후 Polysorbate 20을 0.022%가 되게 첨가하였다.
1.2. 지질 나노입자 제조
양이온성 지질 혼합물(cationic lipid mixture)을 특정 농도, 주로 총 지질 2.5 내지 7.5 mg/mL 범위에서 에탄올을 용해시켜 미세유체 시스템을 이용하여 지질나노입자(Lipid Nanoparticle, LNP)를 제조하였다.
구체적으로, 상기 선택된 지질 혼합물을 무진메디 사에서 자체 제작한 미세유체 시스템(herringbone -shaped channel을 이용한 micromixer system) 두 입구 중 하나에 주입하고, HSPC(hydrogenated soybean phosphatidiycholine) 15% 이하, 콜레스테롤(cholesterol) 50% 이하, DOTAP(Dioleoyl-3-trimethylammonium propane) 60% 이하, 및 DSPE-mPEG 5% 이하의 조성을 갖도록 양이온성 지질 나노입자를 제조하였다.
1.3. 단백질이 봉입(encapsulation)된 지질 나노입자 제조
지질 나노입자를 통한 단백질의 봉입은 헤링본 형태의 마이크로믹서 칩의 미세유체(microfluidics) 시스템을 통해 진행되었다. 미세유체(Microfluidics) 시스템 유속에 따른 지질 나노입자 봉입의 전체적인 개략도는 도 1에 나타내었다.
선택된 지질 혼합물을 특정 농도, 주로 총 지질 2.5 내지 7.5 mg/mL 범위에서 에탄올에 용해시켜 미세유체 시스템 두 입구 중 하나에 주입하고, 단백질이 녹아 있는 수상(20 mM 소듐 포스페이트, 137 mM 소듐 클로라이드, pH 6 ± 0.2)은 2차 주입구에 주입하였다. HSPC 15% 이하, 콜레스테롤 50% 이하, DOTAP 60% 이하, 및 DSPE-mPEG 5% 이하의 조성을 갖도록 양이온성 지질 나노입자를 제조하였고, 각 지질 조성에 따라 제조되는 지질나노입자의 크기(size), PDI(polydispersity index, 다분산 지수) 및 봉입률(encapsulation efficiency)은 하기 표 1에 나타난 바와 같다.

유속 비율(알코올상과 수성상 사이의 비율) 및 총 유속(2개의 주입구가 칩을 통해 주입되는 속도)을 포함하여 미세유체 시스템 소프트웨어를 통해 여러 생산 매개변수를 제어하였다. 구체적으로, 알코올상(alcohol phase) : 수성상(aqueous phase) = 1:3 내지 1:5의 유속 비율과 9 내지 18 ml/분의 유속(ml/min)을 테스트하여 최적의 생산 변수를 선택하였다(표 2 및 표 3).

1.4. 지질 나노입자 정제
공극의 크기(Pore size)가 750kD인 mPES (modified polyethersulfone) hollow fiber 컬럼이 장착된 접선 흐름 여과 시스템 (Repligen사 KrosFlo® KR2i TFF System)을 사용하여 제조된 지질 나노입자를 정제하였다. 에탄올 및 지질 나노입자에 봉입되지 않은 단백질의 제거를 위해, 지질 나노입자 샘플을 컬럼을 통해 순환시키고, 컬럼에서 빠져나가는 투과액과 동일한 속도로 신선한 buffer (20 mM 소듐 포스페이트, 137 mM 소듐 클로라이드, pH 6 ± 0.2)를 첨가하면서 샘플을 정제하였다.
실시예 2. IDS-LNP의 세포 내 이입(Intracellular uptake)
뮤코다당증 II(헌터 증후군/MPS II)형 환자에서 결핍되는 효소 단백질인 IDS를 양이온성 지질 나노입자에 봉입한 [이두로네이트-2-설파타제 단백질 - 양이온성 지질 나노입자] (이하, 'IDS/LNP')를 실시예 1의 방법으로 준비하였다.
정상인 사람의 섬유아세포(fibroblast)(CCD-986Sk, Korean Cell line Bank, KCLB21947) 및 헌터 증후군 환자의 섬유아세포(GM00615, Coriell Institute, 이하 '환자 세포')에 IDS 및 IDS/LNP를 1.25 내지 80 nM의 다양한 농도에서 6시간 처리한 후, 세포 내 이입(uptake)된 단백질 양을 ELISA 방법으로 정량하였다.
구체적으로, 실험에 사용한 세포는 5% CO2, 37℃에서 정상인 사람의 섬유아세포는 10% 소 태아 혈청 (FBS) 및 1% 페니실린-스트렙토마이신 항생제가 보충된 IMDM (Iscove's Modified Dulbecco's Medium) 배지에서, 헌터 증후군 환자세포는 10% 소 태아 혈청(FBS)이 보충된 MEM 배지(Minimum Essential Medium with Earle's salts, L-glutamine, sodium bicarbonate)에서 각각 배양하였다. 세포는 IDS/LNP 처리 24시간 전에 60 mm2 세포 배양 플레이트(2.5×105 세포/플레이트)에 접종한 세포를 이용하였고, 약물 처리 당일 5% FBS-IMDM 또는 MEM 배양 배지에 IDS 대조약(ELAPRASE), IDS 및 IDS/LNP를 첨가하여 37℃에서 24시간 배양하였다. 실험에 사용한 환자 세포의 계대수는 10 미만인 것을 사용하였다.
24시간 반응 후에 배양액을 제거한 후 PBS로 1 내지 2회 세척한 다음 0.25% Trypsin-EDTA 처리, 원심분리, PBS 세척 과정을 거친 후 세포만 수집하였다. 수집된 세포를 프로테아제 억제제 및 포스파타제 억제제 (Sigma)가 포함된 RIPA 버퍼에서 용해하였고, 원심 분리하여 상층액만 모아 분석용 샘플로 사용하였다. 분석용 샘플(전체 세포 용해물)은 Human Iduronate 2-Sulfatase/IDS ELISA 키트(R&D systems, Cat. #DY2449-05)로 세포 내 이입된 IDS 및 BCA(Bicinchoninic acid) 단백질 분석 키트(Thermo Scientific)로 단백질 양을 각각 정량하여 단백질 mg당 세포에 이입된 IDS 단백질 ng으로 나타내었다.
그 결과, IDS만을 처리한 경우에 비해 IDS/LNP를 처리한 경우 정상 및 환자 세포 내에 이입된 IDS 단백질 양이 유의적으로 높은 것을 확인하였다(도 2).
실시예 3. IDS/LNP의 세포 활성(Cellular activity) 확인
뮤코다당증 II(헌터증후군/MPS II) 형 환자에서는 IDS의 결핍으로 세포 내 글리코사미노글리칸(glycosaminoglycan, GAG)으로 지칭되는 다당류가 축적되며 일어나는 질환으로, 헤파란 설페이트(heparan sulfate)는 대표적인 GAG의 일종이다.
IDS/LNP의 세포 활성을 측정하기 위하여 환자 세포에 IDS 단백질 및 IDS/LNP를 5 내지 40 nM의 농도로 24시간 처리한 후, 세포 내 축적된 헤파란 설페이트의 감소량을 ELISA 방법으로 정량하였다.
구체적으로, 실험에 사용한 환자 세포는 5% CO2 하에서 37℃에서 10% 소 태아 혈청(FBS)이 보충된 MEM 배지(Minimum Essential Medium with Earle's salts, L-glutamine, sodium bicarbonate)에서 배양하였다. 세포는 IDS/LNP 처리 24시간 전에 60mm2 세포 배양 플레이트(2.5×105 세포/플레이트)에 접종한 세포를 이용하였고, 약물 처리 당일 5% FBS-MEM 배양배지에 IDS 대조약(ELAPRASE), IDS 및 IDS/LNP를 첨가하여 37℃에서 24시간 배양하였다. 실험에 사용한 환자 세포의 계대수는 10 미만인 것을 사용하였다.
24시간 반응 후에 배양액을 제거한 후 PBS로 1 내지 2회 세척한 다음 0.25% Trypsin-EDTA 처리, 원심분리, PBS 세척 과정을 거친 후 세포만 수집하였다. 수집된 세포를 프로테아제 억제제 및 포스파타제 억제제(Sigma)가 포함된 RIPA 버퍼에서 용해하였고, 원심분리하여 상층액만 모아 분석용 샘플로 사용하였다. 분석용 샘플(전체 세포 용해물)은 Human HS (Heparan Sulfate) ELISA 키트(MyBioSource, Cat. # MBS2515971)로 세포 내 헤파란 설페이트 및 BCA 단백질 분석 키트(Thermo Scientific)로 단백질 양을 각각 정량하여 HS 농도 (ng/ml)를 단백질 농도(mg/ml)로 나누어 보정한 HS 함량(ng/mg)으로 나타내었다.
그 결과, IDS/LNP 처리군의 활성이 IDS 단백질 처리군에 비하여 유의적으로 높은 것을 확인하였다(도 3).
이상으로 본 발명의 내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
Claims (23)
- 수소화된 대두 포스파티딜콜린(HSPC); 콜레스테롤; DOTAP; 및 DSPE-mPEG를 포함하는 지질 혼합물을 포함하는, 단백질의 세포 내 또는 체내 전달을 위한 지질 나노입자.
- 제1항에 있어서,상기 단백질은 효소 또는 치료용 단백질인, 지질 나노입자.
- 제1항에 있어서,상기 HSPC를 상기 지질 혼합물의 8 내지 15몰%로 포함하는, 지질 나노입자.
- 제1항에 있어서,상기 콜레스테롤을 상기 지질 혼합물의 30 내지 50몰%로 포함하는, 지질 나노입자.
- 제1항에 있어서,상기 지질 혼합물은 HSPC:콜레스테롤:DOTAP:DSPE-mPEG의 몰비가 8-15:30-50:40-60:0.5-5로 구성되는 것인, 지질 나노입자.
- HSPC; 콜레스테롤; DOTAP; 및 DSPE-mPEG를 포함하는 지질 혼합물을 포함하는, 단백질이 봉입된(encapsulated) 지질 나노입자.
- 제6항에 있어서,상기 단백질은 효소 또는 치료용 단백질인, 지질 나노입자.
- 제7항에 있어서,상기 효소는 효소 대체 요법(enzyme replacement therapy, ERT) 효소인 것인, 지질 나노입자.
- 제8항에 있어서,상기 효소는 리소좀 효소인 것인, 지질 나노입자.
- 제9항에 있어서,상기 효소는 이두설파제(idursulfase), 라로니다제(laronidase), 엘로설파제 알파(elosulfase alpha), 갈설파제(galsulfase), 베스트로니다제 알파(vestronidase alpha), 아갈시다제 베타(agalsidase beta), 아갈시다제 알파(agalsidase alpha), 페구니갈시다제 알파(pegunigalsidase alpha), 이미글루세라제(imiglucerase), 탈리글루세라제 알파(taliglucerase alpha), 알글루세라제(alglucerase), 벨라글루세라제(velaglucerase), 알글루코시다제 알파(alglucosidase alpha), 아발글루코시다제 알파(avalglucosidase alpha), 세벨리파제 알파(sebelipase alpha), 셀리포나제 알파(cerliponase alpha), 또는 아데노신 디아미나제(adenosine deaminase)인, 지질 나노입자.
- 제6항에 있어서,상기 지질 혼합물은 HSPC:콜레스테롤:DOTAP:DSPE-mPEG의 몰비가 8-15:30-50:40-60:0.5-5로 구성되는 것인, 지질 나노입자.
- 제6항에 있어서,상기 지질 나노입자는 리소좀 축적 질환을 치료하기 위한 것인, 지질 나노입자.
- 제12항에 있어서,상기 리소좀 축적 질환은 MPS I, MPS II, MPS IVA, MPS VI, MPS VII, 파브리병, 고셔병, 고셔병 타입 I, 폼페병, 리소좀산 지질분해효소 결핍증(wolman disease, CESD), 신경 세로이드 리포푸신증(CLN2 질환), 또는 중증 복합 면역결핍인 것인, 지질 나노입자.
- 제6항의 지질 나노입자를 함유하는 단백질의 세포 내 또는 체내 전달용 조성물.
- 제6항의 지질 나노입자를 함유하는 리소좀 축적 질환 예방 또는 치료용 약학적 조성물.
- 제15항에 있어서,상기 리소좀 축적 질환은 MPS I, MPS II, MPS IVA, MPS VI, MPS VII, 파브리병, 고셔병, 고셔병 타입 I, 폼페병, 리소좀산 지질분해효소 결핍증(wolman disease, CESD), 신경 세로이드 리포푸신증(CLN2 질환), 또는 중증 복합 면역결핍인 것인, 약학적 조성물.
- (a) HSPC; 콜레스테롤; DOTAP; 및 DSPE-mPEG를 포함하는 지질 혼합물을 제조하는 단계;(b) 상기 지질 혼합물을 알코올류 용매에 용해시킨 제1용액을 미세유체(microfluidics) 시스템의 주입구에 주입하는 단계;(c) 단백질이 함유된 제2용액을 미세유체 시스템의 다른 주입구에 주입하는 단계; 및(d) 단백질이 봉입된 지질 나노입자를 정제하는 단계를 포함하는, 단백질이 봉입된 지질 나노입자의 제조방법.
- 제17항에 있어서,상기 (a) 단계의 상기 지질 혼합물은 HSPC:콜레스테롤:DOTAP:DSPE-mPEG의 몰비가 8-15:30-50:40-60:0.5-5로 포함되는 것인, 제조방법.
- 제17항에 있어서,상기 미세유체 시스템의 알코올상 및 수성상의 유속 비율은 1:3 내지 1:5의 비율로 진행되는 것인, 제조방법.
- 제17항에 있어서,상기 미세유체 시스템의 유속은 9 내지 18 ml/분의 속도로 진행되는 것인, 제조방법.
- 제17항에 있어서,상기 (d) 단계의 지질 나노입자를 정제하는 단계는, 상기 (b) 단계의 알코올류 용매 및 지질 나노입자에 봉입되지 않은 단백질을 제거시키는 단계를 포함하는 것인, 제조방법.
- ERT 효소 또는 치료용 단백질을 지질 나노입자를 이용하여 세포 내 전달하는 방법.
- ERT 효소 또는 치료용 단백질이 봉입된 지질 나노입자를 이용한 질환 치료 방법.
